1Get effective polyclonal antisera in one month显示文摘According to the traditional immunization procedure, after the first injection of the sample A (emulsion of aimed antigen and Freund's complete adjuvant) to immunize rabbit, successive injections of the sample B (emulsion of aimed antigen and Freund's incomplete adjuvant) were followed every 2-4 weeks. In general,high titer of the corresponding polyclonal antisera will be observed after 4-5 injections of sample B in 3-4months. This report presents a simply modified procedure that was able to stimulate the antisera formation in one month and achieve enough avidity to satisfy either Western blot or immunohistochemistry analysis.It just applied an additional injection of the sample A to the rabbit at the 3rd day after the primary immunization injection. You could gain the high titer of the antisera right after the first sample B injection in one month. 3Shear wave elastography results correlate with liver fibrosis histology and liver function reserve显示文摘AIM: To evaluate the correlation of shear wave elastography(SWE) results with liver fibrosis histology and quantitative function reserve.METHODS: Weekly subcutaneous injection of 60% carbon tetrachloride(1.5 m L/kg) was given to 12 canines for 24 wk to induce experimental liver fibrosis, with olive oil given to 2 control canines. At 24 wk, liver condition was evaluated using clinical biochemistry assays, SWE imaging, lidocaine metabolite monoethylglycine-xylidide(MEGX) test, and histologic fibrosis grading. Clinical biochemistry assays were performed at the institutional central laboratory for routine liver function evaluation. Liver stiffness was measured in triplicate from three different intercostal spaces and expressed as mean liver stiffness modulus(LSM). Plasma concentrations of lidocaine and its metabolite MEGX were determined using high-performance liquid chromatography repeated in duplicate. Liver biopsy samples were fixed in 10% formaldehyde, and liver fibrosis was graded using the modified histological activity index Knodell score(F0-F4). Correlations among histologic grading, LSM, and MEGX measures were analyzed with the Pearson linear correlation coefficient.RESULTS: At 24 wk liver fibrosis histologic grading was as follows: F0, n = 2(control); F1, n = 0; F2, n = 3; F3, n = 7; and F4, n = 2. SWE LSM was positively correlated with histologic grading(r = 0.835, P < 0.001). Specifically, the F4 group had a significantly higher elastic modulus than the F3, F2, and F0 groups(P = 0.002, P = 0.003, and P = 0.006, respectively), and the F3 group also had a significantly higher modulus than the control F0 group(P = 0.039). LSM was negatively associated with plasma MEGX concentrations at 30 min(r =-0.642; P = 0.013) and 60 min(r =-0.651; P = 0.012), time to ? of the maximum concentration(r =-0.538; P = 0.047), and the area under the curve(r =-0.636; P = 0.014). 4Intercropping of Sonchus asper and Vicia faba affects plant cadmium accumulation and root responses显示文摘The cadmium(Cd)pollution of farmland soil is serious in the world.The present study investigated the effects of intercropping Vicia faba and the hyperaccumulator Sonchus asper on the Cd accumulation and root responses(morphology and secreted organic acids)of plants grown on soil from a mining area in Yunnan Province,China,under different Cd stress levels(0,50,100,and 200 mg kg-1).Intercropping increased the biomass of both S.asper and V.faba,as well as the Cd accumulation and Cd transfer coefficient from roots to shoots of S.asper,but decreased those of V.faba in the 200 mg kg-1Cd treatment.The Cd concentrations in roots,shoots,and grains from intercropped V.faba plants were positively correlated(P<0.05)with the N,N-diethyl phenylacetamide-extractable Cd content in soil.In the 50 mg kg-1Cd treatment,intercropping decreased citric and malic acids in root secretions of S.asper.Intercropped V.faba secreted more citric,oxalic,acetic,and malic acid compared with S.asper.Intercropping also increased root length and root surface area of both S.asper and V.faba.At 50 mg kg-1Cd,root length for S.asper was higher than that at 0 mg kg-1Cd,whereas V.faba roots had significantly decreased length and mean diameter.Intercropping of S.asper and V.faba is suggested as an in situ phytoremediation strategy of Cd-contaminated soils and may improve the food quality of V.faba.Yanqun ZU Li QIN Fangdong ZHAN Jiong WU Yuan LI Jianjun CHEN Jixiu WANG Wenyou HU 2020Pedosphere2020,30,4:6

7Factors Affecting Patient Satisfaction with Community Health Service under the Gatekeeper System:A Cross-sectional Study in Nanjing,China显示文摘The gatekeeper policy has been implemented for approximately ten years on a pilot population in China. It is necessary to assess the satisfaction of patients utilizing community health service(CHS) under the gatekeeper system. Our study showed that the cognition of gatekeeper policy was associated with four dimensions including doctor‐patient relationships, information and support, organization of care, and accessibility(P < 0.001). One or more factors such as gender and self‐perceived health scores also affected their satisfaction. General practitioners must be prepared to focus on these aspects of information and support, organization of care, and accessibility as indicators of potential opportunities for improvement. 8Predicting densities and elastic moduli of SiO_(2)-based glasses by machine learning显示文摘Chemical design of SiO_(2)-based glasses with high elastic moduli and low weight is of great interest.However,it is difficult to find a universal expression to predict the elastic moduli according to the glass composition before synthesis since the elastic moduli are a complex function of interatomic bonds and their ordering at different length scales.Here we show that the densities and elastic moduli of SiO_(2)-based glasses can be efficiently predicted by machine learning(ML)techniques across a complex compositional space with multiple(>10)types of additive oxides besides SiO_(2).Our machine learning approach relies on a training set generated by high-throughput molecular dynamic(MD)simulations,a set of elaborately constructed descriptors that bridges the empirical statistical modeling with the fundamental physics of interatomic bonding,and a statistical learning/predicting model developed by implementing least absolute shrinkage and selection operator with a gradient boost machine(GBM-LASSO).Yong-Jie Hu Ge Zhao Mingfei Zhang Bin Bin Tyler Del Rose Qian Zhao Qun Zu Yang Chen Xuekun Sun Maarten de Jong Liang Qi 2020npj Computational Materials2020,,1:2
9Genetically modified CD7-targeting allogeneic CAR-T cell therapy with enhanced efficacy for relapsed/refractory CD7-positive hematological malignancies:a phase I clinical study显示文摘Chimeric antigen receptor(CAR)-T cell therapy against T cell malignancies faces major challenges including fratricide between CAR-T cells and product contamination from the blasts.Allogeneic CAR-T cells,generated from healthy donor T cells,can provide ready-to-use,blast-free therapeutic products,but their application could be complicated by graft-versus-host disease(GvHD)and host rejection.Here we developed healthy donor-derived,CD7-targeting CAR-T cells(RD13-01)with genetic modifications to resist fratricide,GvHD and allogeneic rejection,as well as to potentiate antitumor function.A phase I clinical trial(NCT04538599)was conducted with twelve patients recruited(eleven with T cell leukemia/lymphoma,and one with CD7-expressing acute myeloid leukemia).All patients achieved pre-set end points and eleven proceeded to efficacy evaluation.No dose-limiting toxicity,GvHD,immune effector cell-associated neurotoxicity or severe cytokine release syndrome(grade≥3)were observed.28 days post infusion,81.8%of patients(9/11)showed objective responses and the complete response rate was 63.6%(7/11,including the patient with AML).3 of the responding patients were bridged to allogeneic hematopoietic stem cell transplantation.With a median follow-up of 10.5 months,4 patients remained in complete remission.Cytomegalovirus(CMV)and/or Epstein-Barr virus(EBV)reactivation was observed in several patients,and one died from EBV-associated diffuse large B-cell lymphoma(DLBCL).Expansion of CD7-negative normal T cells was detected post infusion.In summary,we present the first report of a Phase I clinical trial using healthy donor-derived CD7-targeting allogeneic CAR-T cells to treat CD7^(+)hematological malignancies.Our results demonstrated the encouraging safety and efficacy profiles of the RD13-01 allogeneic CAR-T cells for CD7^(+)tumors.Yongxian Hu Yali Zhou Mingming Zhang Houli Zhao Guoqing Wei Wengang Ge Qu Cui Qitian Mu Gong Chen Lu Han Tingting Guo Jiazhen Cui Xiaoyan Jiang Xiujun Zheng Shuhui Yu Xiaolong Li Xingwang Zhang Mingxi Chen Xiuju Li Ming Gao Kang Wang Cheng Zu Hao Zhang Xiaohong He Yanbin Wang Dongrui Wang Jiangtao Ren He Huang 2022Cell Research2022,32,11:2
10A Thermochemical Study on Complex Nickel(Ⅱ) L-α-Histidine显示文摘The formation enthalpy of complex nickel(Ⅱ) histidine(His) in water was determined by means of microcalorimetry in the temperature range of 298 15-323 15 K. The standard enthalpy of the formation of Ni(His) 2+ 2(aq) was calculated. 11Compositional dependence of hydrogenation performance of Ti-Zr-Hf-Mo-Nb high-entropy alloys for hydrogen/tritium storage显示文摘A series of Ti-Zr-Hf-Mo-Nb high-entropy alloys with different Mo concentrations were developed as candidate materials for hydrogen/tritium storage in solid phase.The crystal structures and hydrogenation properties of the Ti-Zr-Hf-Mo-Nb alloys were investigated by X-ray diffraction and differential scanning calorimetry techniques.All the alloys have a body-centred cubic single phase structure.The results demonstrate that the cell volume of the Ti-Zr-Hf-Mo-Nb hydride decreases with increasing Mo concentration,which reduces their thermal stability.The theoretical calculation proposes that the lower binding energy of the Ti-Zr-Hf-Mo-Nb hydride decreases the thermal stability of Ti-Zr-Hf-Mo-Nb alloys with higher Mo content.The great hydrogenation performance for all the Ti-Zr-Hf-Mo-Nb alloys is owing to their reversible single-phase transformation during the hydrogen absorption-desorption cycle,which would be bene ficial to improving the hydrogen recycling rate and preventing the disproportionation.The compositional dependence of the hydrogenation performance of the Ti-Zr-Hf-Mo-Nb alloys was established and will be useful in designing novel hydrogen/tritium storage materials to satisfy the requirements of diffe rent application fields in hydrogen,solar thermal and nuclear energy.Huahai Shen Jutao Hu Pengcheng Li Gang Huang Jianwei Zhang Jinchao Zhang Yiwu Mao Haiyan Xiao Xiaosong Zhou Xiaotao Zu Xinggui Long Shuming Peng 2020Journal of Materials Science & Technology2020,52,20:2
12Gaseous emissions from compressed natural gas buses in urban road and highway tests in China显示文摘The natural gas vehicle market is rapidly developing throughout the world, and the majority of such vehicles operate on compressed natural gas(CNG). However, most studies on the emission characteristics of CNG vehicles rely on laboratory chassis dynamometer measurements, which do not accurately represent actual road driving conditions. To further investigate the emission characteristics of CNG vehicles, two CNG city buses and two CNG coaches were tested on public urban roads and highway sections. Our results show that when speeds of 0–10 km/hr were increased to 10–20 km/hr, the CO_2, CO, nitrogen oxide(NO_x), and total hydrocarbon(THC) emission factors decreased by(71.6 ± 4.3)%,(65.6 ± 9.5)%,(64.9 ± 9.2)% and(67.8 ± 0.3)%, respectively. In this study, The Beijing city buses with stricter emission standards(Euro Ⅳ) did not have lower emission factors than the Chongqing coaches with Euro Ⅱ emission standards. Both the higher emission factors at 0–10 km/hr speeds and the higher percentage of driving in the low-speed regime during the entire road cycle may have contributed to the higher CO_2 and CO emission factors of these city buses.
