Phylogeny and evolutionary history of the silkworm显示文摘The silkworm,Bombyx mori, played an important role in the old Silk Road that connected ancient Asia and Europe. However,to date, there have been few studies of the origins and domestication of this species using molecular methods. In this study,DNA sequences of mitochondrial and nuclear loci were used to infer the phylogeny and evolutionary history of the domesticated silkworm and its relatives. All of the phylogenetic analyses indicated a close relationship between the domesticated silkworm and the Chinese wild silkworm.Domestication was estimated to have occurred about 4100 years ago(ya),and the radiation of the different geographic strains of B.mori about 2000 ya.The Chinese wild silkworm and the Japanese wild silkworm split about 23600 ya.These estimates are in good agreement with the fossil evidence and historical records. In addition,we show that the domesticated silkworm experienced a population expansion around 1000 ya.The divergence times and the population dynamics of silkworms presented in this study will be useful for studies of lepidopteran phylogenetics,in the genetic analysis of domestic animals, and for understanding the spread of human civilizations. | SUN Wei YU HongSong SHEN YiHong BANNO Yutaka XIANG ZhongHuai ZHANG Ze | 2012 | Science China(Life Sciences)2012,55,6: | 6
Spatio-temporal variation in rock exhumation linked to large-scale shear zones in the southeastern Tibetan Plateau显示文摘Crustal-scale shear zones are believed to have played an important role in the tectonic and landscape evolution of orogens. However, the variation of long-term rock exhumation between the interior of shear zones and adjacent regions has not been documented in detail. In this study, we obtained new zircon U-Pb, biotite ^(40)Ar/^(39)Ar, zircon and apatite(U-Th)/He data, and conducted inverse thermal history modeling from two age-elevation profiles(the Pianma and Tu'er profiles) in the southeastern Tibetan Plateau. Our goal is to constrain the exhumation history of the Gaoligong and Chongshan shear zones and adjacent regions, so as to explore the effect of the shear zones on exhumation and their thermal effect on cooling that should not be ignored. Our results suggest that during the interval of 18–11 Ma the exhumation rates of rocks within the Gaoligong shear zone are anomalously high compared with those outside of. The rapid cooling during 18–11 Ma appears to be restricted to the shear zone, likely due to localized thermal effects of shearing and exhumation. After 11 Ma, both the areas within and outside of the shear zones experienced a similar two-stage exhumation history: slower cooling until the early Pliocene, and then a rapid increase in cooling rate since the early Pliocene. Our results indicate a synchronized exhumation but with spatially varied exhumation rates. Our study also highlights the important role of large-scale shear zones in exposing rocks, and thus the importance of the structural context when interpreting thermochronological data in the southeastern margin of the Tibetan Plateau. | Yukui GE Jing LIU-ZENG Jinyu ZHANG Wei WANG Yuntao TIAN Matthew FOX Lingsen ZENG Xiaoming SHEN Heng WANG Ze SU Kejia XIE | 2020 | Science China Earth Sciences2020,63,4: | 5
Atomic-scale mechanism of the θ’’→θ’ phase transformation in Al-Cu alloys显示文摘The phase transformation of θ’’→θ’ in an Al-5.7 Cu alloy was investigated by aberration-corrected scanning transmission electron microscopy, and the tranformation mode of θ’’→θ’ during aging treatment was clarified. In the presence of the θ’ phases, θ’ was found to be formed by in-situ transformation fromθ’’ with the same plate shape, size and broad faces. The transformation starts from multiple sites within the θ’ precipitate and the whole θ’ phase finally forms as the preferential θ’ sections grow and connect with each other. Antiphase domain boundaries are also found in some θ’ precipitates when the disregistry exists between different θ’ sections. | Zhenju Shen Qingqing Ding Chunhui Liu Jiangwei Wang He Tian Jixue Li Ze Zhang | 2017 | Journal of Materials Science & Technology2017,33,10: | 3
Acoustic emission characteristics and stress release rate of coal samples in different dynamic destruction time显示文摘Considering the importance of the prediction of rock burst disasters, and in order to grasp the law of acoustic emission(AE) of coal samples in different dynamic destruction time, the SH-II AE monitoring system was adopted to monitor the failure process of coal samples. The study of the change rule of the AE numbers, energy, ‘b' value and spectrum in the micro crack propagation process of the coal samples shows that as dynamic damage time went by, AE presented high-energy counts and the accumulated counts increased during the compression phase. The AE energy and cumulative counts increased during the elastic stage. The AE blank area increased gradually and the blank lines were more and more obvious in the molding stage. The AE counts and energy showed a trend of decrease in the residual damage phase.AE ‘b' values gradually became sparse, and the large scale cracks percentage compared with micro cracks decreased and the degree of damage decreased. The AE frequency spectrum peak went from the residual damage phase to the molding phase, and finally it was nearly stable, besides the bandwidth of the main frequency is gradually narrowed. Also, the frequency peak changed from single peak frequency to bi-peak frequency and to the single peak frequency. Uniaxial compressive strength is more sensitive than the elastic modulus to dynamic damage time. | Xiao Fukun Liu Gang Zhang Ze Shen Zhiliang Zhang Fengrui Wang Yifei | 2016 | International Journal of Mining Science and Technology2016,26,6: | 3
RNA interference targeting ecdysone receptor blocks the larval-pupal transition in Henosepilachna vigintioctopunctata显示文摘Henosepilachna vigintioctopunctata is a serious insect pest which attacks a large number of nightshades and cucurbits in Asian countries,Brazil and Australia.Prolonged application of traditional pesticides has caused environmental pollution and exerted deleterious effects on human health.Finding new approaches with high target specificity and low environmental contamination has become an urgent task.RNA interference(RNAi)induced by double-stranded RNA(dsRNA)is expected to be applicable to managing this pest. Here we evaluated the effects of Escherichia co/Z-expressed dsRNAs targeting ecdvsone receptor(EcR)gene via dietary delivery in laboratory and foliar spraying in a greenhouse. The target transcript was successfully knocked down when the 4th-instar larvae had fed on potatofoliage dipped with dsEcR in a laboratory bioassay.Around 85%of the HvEcR RNAi larvae remained as prepupae or became abnormal pupae, and failed to emerge into adults.Ingestion of ds£c7?-immersed foliage by the 3rd-instar larvae effectuated a comparable RNAi response and brought about more severe defects:all the resultant larvae arrested development, remained as prepupae and finally died. For assay in the greenhouse,a ds£c7?-contained E.coli suspension was directly sprayed to the foliage of greenhouse-growing potato plants and the 3rd-and 4th-instar larvae were transferred to the leaves.High RNAi efficacy was obtained and identical RNAi phenotypes were observed in treated larvae. In addition, spraying dsEcR reduced leaf damage. Our results indicate a possibility of practical application of dsEcR as an environmentally friendly RNA pesticide to control H.vigintioctopunctata larvae. | Jian-Jian Wu Li-Li Mu Wei-Nan Kang Long-Ji Ze Chen-Hui Shen Lin Jin Ahmad Ali Anjum Guo-Qing Li | 2021 | Insect Science2021,28,2: | 3

Diffusion of helium in FCT zircon显示文摘To gain a better quantitative understanding of zircon(U-Th)/He ages and evaluate the applicability of zircon(U-Th)/He dating, the diffusion characteristics, He diffusion kinetics, helium partial retention zone(HePRZ), closure temperature, and(U-Th)/He ages were investigated using high-precision laboratory step heating experiments based on the thermally activated diffusion process. The ln(D/a^2) in Fish Canyon Tuff(FCT) zircons determined from laboratory step heating experiments was negatively correlated with reciprocal temperature, as expected for thermally activated volume diffusion. The zircon activation energies ranged from 144 to 184 kJ mol^(-1) with a mean of 169±12 kJ mol^(-1). The closure temperatures ranged from 144 to 216°C(a cooling rate of 10°C Ma^(-1) and an effective grain radius of 38–60 μm) with an average of 176±18°C. The calculated closure temperature increased with increasing cooling rate, yielding an average zircon He closure temperature of ~136°C at a slow cooling rate of 0.1°C Ma^(-1), whereas the closure value was ~199°C at a cooling rate of 100°C Ma^(-1). The closure temperature increased with the equivalent spherical radius assuming a constant cooling rate. The He ages from FCT zircons were negligibly affected by grain size because of the rapid cooling. He preserved in the zircon was sensitive to temperature and holding time, and the temperature range for zircon HePRZ gradually decreased with increasing holding time. The(U-Th)/He ages from 26 FCT zircons yielded an algorithmic mean of 28.3±0.3 Ma(S.E.) and a geometric mean of 28.4±0.3 Ma(S.E.), consistent with the ages of 28.4±1.9 Ma reported by other laboratories. The FCT zircons were characterized by rapid cooling, young(U-Th)/He ages with good reproducibility, and low alpha doses. Weak correlations between the He ages and effective uranium(eU) concentrations from the FCT zircons indicated radiation damage did not significantly affect He diffusivity. | Shun YU Wen CHEN Jingbo SUN Ze SHEN | 2019 | Science China Earth Sciences2019,62,4: | 1
Late Mesozoic Tectono-Thermal History in the South Margin of Great Xing’an Range,NE China:Insights from Zircon and Apatite(U-Th)/He Ages显示文摘The Late Mesozoic tectono-thermal evolution and geodynamic setting of the Great Xing’an Range(GXR),particularly in the south margin, are still ambiguous. In this study,we present original low-temperature thermochronological data of six granitoid samples collected from Maanzi pluton in the south margin of the GXR.The apatite and zircon(U-Th)/He ages vary of 70.8 ± 4.3-119.0 ±7.0 Ma and 120.0 ± 7.1-146.7 ± 8.7 Ma,respectively.Further numerical inverse modeling results reveal that the granitoid plutons experienced a rapid cooling and exhumation stage during Early Cretaceous with a rate of ~5.0-6.0 ℃/Ma and ~0.14-0.17 mm/yr,respectively,including the western minor pluton emplaced during Early Permian and the major pluton crystallized in latest Jurassic, corresponding to the extensional tectonics in NE China. The rapid exhumation processes in the south margin of the GXR during the Early Cretaceous could be related to both the rollback of Paleo-Pacific oceanic plate and the collapse of thickened crust in the Mongol-Okhotsk Orogen. | Wen Zhang Shuangfeng Zhao Jingbo Sun Ze Shen Wen Chen | 2022 | Journal of Earth Science2022,33,1: | 1

Autophagy inhibitors increase the susceptibility of KRAS-mutant human colorectal cancer cells to a combined treatment of 2-deoxy-D-glucose and lovastatin显示文摘RAS-driven colorectal cancer relies on glucose metabolism to support uncontrolled growth. However, monotherapy with glycolysis inhibitors like 2-deoxy-D-glucose causes limited effectiveness.Recent studies suggest that anti-tumor effects of glycolysis inhibition could be improved by combination treatment with inhibitors of oxidative phosphorylation. In this study we investigated the effect of a combination of 2-deoxy-D-glucose with lovastatin(a known inhibitor of mevalonate pathway and oxidative phosphorylation)on growth of KRAS-mutant human colorectal cancer cell lines HCT116 and LoVo.A combination of lovastatin(>3.75μM)and 2-deoxy-D-glucose(>1.25 mM)synergistically reduced cell viability, arrested cells in the G2/M phase, and induced apoptosis. The combined treatment also reduced cellular oxygen consumption and extracellular acidification rate, resulting in decreased production of ATP and lower steady-state ATP levels.Energy depletion markedly activated AMPK,inhibited mTOR and RAS signaling pathways, eventually inducing autophagy, the cellular pro-survival process under metabolic stress, whereas inhibition of autophagy by chloroquine(6.25μM)enhanced the cytotoxic effect of the combination of lovastatin and 2-deoxy-D-glucose.These in vitro experiment results were reproduced in a nude mouse xenograft model of HCT116 cells. Our findings suggest that concurrently targeting glycolysis, oxidative phosphorylation, and autophagy may be a promising regimen for the management of RAS-driven colorectal cancers. | Xiao-ming Huang Jia-jun Huang Jing-jing Du Na Zhang Ze Long You Yang Fang-fang Zhong Bo-wen Zheng Yun-fu Shen Zhe Huang Xiang Qin Jun-he Chen Qian-yu Lin Wan-jun Lin Wen-zhe Ma | 2021 | Acta Pharmacologica Sinica2021,42,11: | 1

Mesh‑free semi‑quantitative variance underestimation elimination method in Monte Caro algorithm显示文摘The inter-cycle correlation of fission source distributions(FSDs)in the Monte Carlo power iteration process results in variance underestimation of tallied physical quantities, especially in large local tallies. This study provides a mesh-free semiquantitative variance underestimation elimination method to obtain a credible confidence interval for the tallied results. This method comprises two procedures:Estimation and Elimination. The FSD inter-cycle correlation length is estimated in the Estimation procedure using the Sliced Wasserstein distance algorithm. The batch method was then used in the elimination procedure. The FSD inter-cycle correlation length was proved to be the optimum batch length to eliminate the variance underestimation problem. We exemplified this method using the OECD sphere array model and 3D PWR BEAVRS model. The results showed that the average variance underestimation ratios of local tallies declined from 37 to 87%to within±5%in these models. | Peng‑Fei Shen Xiao‑Dong Huo Ze‑Guang Li Zeng Shao Hai‑Feng Yang Peng Zhang Kan Wang | 2023 | Nuclear Science and Techniques2023,34,1: | 0
Safety and immunogenicity of a mosaic vaccine booster against Omicron and other SARS-CoV-2 variants:a randomized phase 2 trial显示文摘An ongoing randomized,double-blind,controlled phase 2 trial was conducted to evaluate the safety and immunogenicity of a mosaic-type recombinant vaccine candidate, named NVSI-06-09,as a booster dose in subjects aged 18 years and older from the United Arab Emirates(UAE),who had administered two or three doses of inactivated vaccine BBIBP-CorV at least 6 months prior to enrollment. The participants were randomly assigned with 1:1 to receive a booster dose of NVSI-06-09 or BBIBP-CorV.The primary outcomes were immunogenicity and safety against severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)Omicron variant, and the exploratory outcome was cross-immunogenicity against other circulating strains.Between May 25 and 30,2022,516 adults received booster vaccination with 260 in NVSI-06-09 group and 256 in BBIBP-CorV group.Interim results showed a similar safety profile between two booster groups, with low incidence of adverse reactions of grade 1 or 2.For immunogenicity,by day 14 post-booster,the fold rises in neutralizing antibody geometric mean titers(GMTs)from baseline elicited by NVSI-06-09 were remarkably higher than those by BBIBP-CorV against the prototype strain(19.67 vs 4.47-fold),Omicron BA.1.1(42.35 vs 3.78-fold),BA.2(25.09 vs 2.91-fold),BA.4(22.42 vs 2.69-fold),and BA.5 variants(27.06 vs 4.73-fold).Similarly,the neutralizing GMTs boosted by NVSI-06-09 against Beta and Delta variants were also 6.60-fold and 7.17-fold higher than those by BBIBP-CorV.Our findings indicated that a booster dose of NVSI-06-09 was well-tolerated and elicited broad-spectrum neutralizing responses against divergent SARS-CoV-2 variants, including Omicron and its sub-lineages. | Nawal Al Kaabi Yun Kai Yang Yu Liang Ke Xu Xue Feng Zhang Yun Kang Yu Qin Jin Jun Wei Hou Jing Zhang Tian Yang Salah Hussein Mohamed Saif ElDein Ze Hua Lei Hao Zhang Shuai Shao Zhao Ming Liu Ning Liu Xiang Zheng Ji Guo Su Sen Sen Yang Xiangfeng Cong Yao Tan Wenwen Lei Xue Jun Gao Zhiwei Jiang Hui Wang Meng Li Hanadi Mekki Mekki Walid Zaher Sally Mahmoud Xue Zhang Chang Qu Dan Ying Liu Jing Zhang Mengjie Yang Islam Eltantawy Peng Xiao Fu Jie Shen Jin Juan Wu Zi Bo Han Li Fang Du Fang Tang Shi Chen Zhi Jing Ma Fan Zheng Ya Nan Hou Xin Yu Li Xin Li Zhao Nian Wang Jin Liang Yin Xiao Yan Mao Jin Zhang Liang Qu Yun Tao Zhang Xiao Ming Yang Guizhen Wu Qi Ming Li | 2023 | Signal Transduction and Targeted Therapy2023,8,2: | 0
