2Super-resolution fluorescence-assisted diffraction computational tomography reveals the threedimensional landscape of the cellular organelle interactome显示文摘The emergence of super-resolution(SR)fluorescence microscopy has rejuvenated the search for new cellular substructures.However,SR fluorescence microscopy achieves high contrast at the expense of a holistic view of the interacting partners and surrounding environment.Thus,we developed SR fluorescence-assisted diffraction computational tomography(SR-FACT),which combines label-free three-dimensional optical diffraction tomography(ODT)with two-dimensional fluorescence Hessian structured illumination microscopy.The ODT module is capable of resolving the mitochondria,lipid droplets,the nuclear membrane,chromosomes,the tubular endoplasmic reticulum,and lysosomes.Using dual-mode correlated live-cell imaging for a prolonged period of time,we observed novel subcellular structures named dark-vacuole bodies,the majority of which originate from densely populated perinuclear regions,and intensively interact with organelles such as the mitochondria and the nuclear membrane before ultimately collapsing into the plasma membrane.This work demonstrates the unique capabilities of SR-FACT,which suggests its wide applicability in cell biology in general.Dashan Dong Xiaoshuai Huang Liuju Li Heng Mao Yanquan Mo Guangyi Zhang Zhe Zhang Jiayu Shen Wei Liu Zeming Wu Guanghui Liu Yanmei Liu Hong Yang Qihuang Gong Kebin Shi Liangyi Chen 2020Light(Science & Applications)2020,9,1:7
3Single small molecule-assembled nanoparticles mediate efficient oral drug delivery显示文摘Oral drug delivery,which requires surviving the harsh environment in the gastrointestinal(Gl)tract and penetrating the intestinal epithelium,has not bee n achieved using simple formulatio n nan oparticles(NPs).Medici nal natural products(MNPs)have bee n widely used in traditi onal medicine for disease management through oral consumption.However,most pharmacologically active compounds within MNPs do not have the properties suitable for oral applicatio ns.We hypothesize that some MNPs contain n atural nano materials that can convert those compounds into oral formulations by forming NPs.After screening 66 MNPs,we identified five classes of small molecules that form NPs,many of which are capable of efficient drug encapsulation and Gl penetration.We show that one of them,dehydrotrametenolic acid(DTA),is capable of mediating oral delivery for effective disease treatment.We determined that DTA NPs assemble through hydrogen bonding and penetra怕the Gl tract via apical sodium-depe ndent bile acid tran sporter.Our study reveals a no vel class of single comp orient,small molecule-assembled NPs for oral drug delivery,and suggests a n ovel approach to modernizi ng MNPs through nano material discovery.Xin Yang Chao Ma Zeming Chen Jun Liu Fuyao Liu Rongbin Xie Haitian Zhao Gang Deng Ann TChen Ningbo Gong Lei Yao Pengjian Zuo Kangkang Zhi Jiacheng Wang Xiaobin Gao Jing Wang Louzhen Fan Jiangbing Zhou 2019Nano Research2019,12,10:4

5Microplastic pollution and enrichment of distinct microbiota in sediment of mangrove in Zhujiang River estuary,China显示文摘The microbial communities colonized on microplastics(MPs)have attracted widespread attention.However,few studies focused on the MPs impacts on mangrove ecosystems,particularly on bacterial communities.We investigated the MPs pollution in mangrove of Zhujiang(Pearl)River estuary(ZRE).To study the potential risk posed by MPs to the mangrove ecosystems,the differences in bacterial communities,functions,and complexity between MPs and sediment samples were reported for the first time.Microplastics(2991±1586 items/kg dry weight(dw))in sediment were mainly fibers and polyethylene,mostly transparent,and in size less than 0.5 mm.Bacterial communities and functions significantly differed from MPs in mangrove sediment.Compared with sediment,MPs significantly enriched members of Proteobacteria,Bacteroidetes,and Actinobacteria,as well as the bacteria associated with plastic-degrading and human diseases on their surface,suggesting that microbial communities on MPs may promote MPs degradation and the spread of diseases,posing potential risk to mangrove ecosystems and human health.Although bacteria on MPs exhibited a lower diversity,the co-occurrence network analysis indicated that network of bacteria colonized on MPs was bigger and more complex than those of mangrove sediment,illustrating that MPs can act as a distinct habitat in this special ecosystem.This study provides a new perspective for increasing our understanding of microplastic pollution in mangrove ecosystems.Muting YAN Xiaofeng CHEN Wei CHU Weixin LI Minqian LI Zeming CAI Han GONG 2023Journal of Oceanology and Limnology2023,41,1:1

9Brain-targeting,acid-responsive antioxidant nanoparticles for stroke treatment and drug delivery显示文摘Stroke is the leading cause of death and disability.Currently,there is no effective pharmacological treatment for this disease,which can be partially attributed to the inability to efficiently deliver therapeutics to the brain.Here we report the development of natural compound-derived nanoparticles(NPs),which function both as a potent therapeutic agent for stroke treatment and as an efficient carrier for drug delivery to the ischemic brain.First,we screened a collection of natural nanomaterials and identified betulinic acid(BA)as one of the most potent antioxidants for stroke treatment.Next,we engineered BA NPs for preferential drug release in acidic ischemic tissue through chemically converting BA to betulinic amine(BAM)and for targeted drug delivery through surface conjugation of AMD3100,a CXCR4 antagonist.The resulting AMD3100-conjugated BAM NPs,or A-BAM NPs,were then assessed as a therapeutic agent for stroke treatment and as a carrier for delivery of NA1,a neuroprotective peptide.We show that intravenous administration of A-BAM NPs effectively improved recovery from stroke and its efficacy was further enhanced when NA1 was encapsulated.Due to their multifunctionality and significant efficacy,we anticipate that A-BAM NPs have the potential to be translated both as a therapeutic agent and as a drug carrier to improve the treatment of stroke.Shenqi Zhang Bin Peng Zeming Chen Jiang Yu Gang Deng Youmei Bao Chao Ma Fengyi Du Wendy C.Sheu W.Taylor Kimberly J.Marc Simard Daniel Coman Qianxue Chen Fahmeed Hyder Jiangbing Zhou Kevin N.Sheth 2022Bioactive Materials2022,7,10:1
10Boron Nanosheet-Supported Rh Catalysts for Hydrogen Evolution:A New Territory for the Strong Metal-Support Interaction Effect显示文摘High-efficiency electrochemical hydrogen evolution reaction(HER)offers a promising strategy to address energy and environmental crisis.Platinum is the most effective electrocatalyst for the HER.However,challenging scarcity,valuableness,and poor electrochemical stability still hinder its wide application.Here,we designed an outstanding HER electrocatalyst,highly dispersed rhodium(Rh)nanoparticles with an average diameter of only 3 nm supported on boron(B)nanosheets.The HER catalytic activity is even comparable to that of commercial platinum catalysts,with an overpotential of only 66 mV in 0.5 M H_(2)SO_(4) and 101 mV in 1 M KOH to reach the current density of 10 mA cm−2.Meanwhile,the catalyst exhibited impressive electrochemical durability during long-term electrochemical processes in acidic and alkaline media,even the simu-lated seawater environment.Theoretical calculations unraveled that the structure-activity relationship between B(104)crystal plane and Rh(111)crystal plane is beneficial to the release of hydrogen,and surface O plays a vital role in the catalysis process.Our work may gain insights into the development of supported metal catalysts with robust catalytic performance through precise engineering of the strong metal-supported interaction effect.Keng Chen Zeming Wang Liang Wang Xiuzhen Wu Bingjie Hu Zheng Liu Minghong Wu 2021Nano-Micro Letters2021,13,10:1

12Petrogenesis and Tectonic Implications of the Latest Cretaceous Intrusive Rocks from the Eastern Gangdese Belt,Southeast Tibet显示文摘The latest Cretaceous magmatic activity in the eastern segment of the Lhasa terrane provides important insights for tracking the magma source and geodynamic setting of the eastern Gangdese batholith,eastward of eastern Himalayan Syntaxis.Detailed petrological,geochemical and geochronological studies of the intrusive rocks(monzodiorites and granodiorites)of the eastern Gangdese batholith are presented with monzodiorites and granodiorites giving zircon U-Pb crystallization dates of 70-66 Ma and 71-66 Ma withεHf(t)values of−4.8 to+6.2 and−1.9 to+5.3,respectively.These rocks are metaluminous to weakly peraluminous I-type granites showing geochemically arc-related features of enrichment in LREEs and some LILEs,e.g.,Rb,Th,and U,and depletion in HREEs and some HFSEs,e.g.,Nb,Ta,and Ti.The rocks are interpreted to be derived from partial melting of mantle material and juvenile crust,respectively,which are proposed to be triggered by Neo-Tethyan slab rollback during northward subduction,with both experiencing ancient crustal contamination.The studied intrusive rocks formed in a transitional geodynamic setting caused by Neo-Tethyan oceanic flat subduction to slab rollback beneath the eastern Gangdese belt during the latest Cretaceous.CHEN Yanfei CHEN Xuanhua ZHANG Zeming SHAO Zhaogang TIAN Zuolin DONG Xin QIN Shengkai YUAN Yuelei 2022Acta Geologica Sinica(English Edition)2022,96,3:1
13Rare-Earth Metal Alkyl Complexes with 3-Arylamido-Functionalized Indolyl Ligands:Synthesis,Characterization and Reactivity显示文摘A series of dinuclear rare-earth metal alkyl complexes{[μ-η^2:η^1:η^1-3-(LNCH)(CH2SiMe3)Ind]RE-(CH2SiMe3)(THF)}2(L^1=2-tBuC6H4,RE=Y,Gd,Dy,Er,Yb;L^2=2,4,6-Me3C6H2,RE=Dy,Er;Ind=indolyl)and{[μ-η^2:η^1:η^1-3-(LNCH2)Ind]RE(CH2SiMe3)(THF)}2(L^1,RE=Y,Dy,Er,Yb;L^2,RE=Er,Yb)bearing 3-arylamido functionalized indolyl ligands having diverse bonding modes with metal ions were synthesized either by the insertion reaction of the imino group to the RE-C bond or by the alkane elimination reaction.In the preparation of above complexes,rare-earth metal alkyl complexes{[μ-η^5:η^1:η^1-3-(L^2 NCH)(CH2SiMe3)Ind]Gd(CH2SiMe3)(THF)}2 with aμ-η^5:η^1:η^1 coordination mode to the gadolinium ion and{[μ-η^3:η^1:η^1-3-(L^2NCH2)Ind]Dy(CH2SiMe3)(THF)}2 with aμ-η^3:η^1:η^1 coordination mode to the dysprosium ion were unexpectedly isolated.The reactions of 3-(L^2N=CH)lnd with Er(CH2SiMe3)3(THF)2 at room temperature,generated a tetranuclear imino-indolyl erbium intermediate{[μ-η^1:η^1-3-(L^2N=CH)Ind]Er(CH2SiMe3)2(THF)}4,which can transform into the amido functionalized indolyl erbium complex in hot toluene.Moreover,the reactivities of the newly synthesized ytterbium complex with N-heterocyclic compounds were investigated,affording the corresponding products of the mixed pyridyl-indolyl,imidazolyl-indolyl,and ortho-metalated complexes.The yttrium complexes showed a high regioselectivity and steroselectivity for the isoprene polymerization with 1,4-trans selectivity up to 91.7%and 1,4-cis selectivity up to 96.1%in the presence of cocatalysts,respectively.Xiancui Zhu Yuzhe Jiang Jun Chen Shaowu Wang Zeming Huang Shan Zhu Xu Zhao Wenrun Yue Jun Zhang Weikang Wu Xiangyang Zhong 2020Chinese Journal of Chemistry2020,38,5:1
14A Unique Ru-N_(4)-P Coordinated Structure Synergistically Waking Up the Nonmetal P Active Site for Hydrogen Production显示文摘Numerous experiments have demonstrated that the metal atom is the active center of monoatomic catalysts for hydrogen evolution reaction(HER),while the active sites of nonmetal doped atoms are often neglected.By combining theoretical prediction and experimental verification,we designed a unique ternary Ru-N_(4)-P coordination structure constructed by monodispersed Ru atoms supported on N,P dual-doped graphene for highly efficient hydrogen evolution in acid solution.The density functional theory calculations indicate that the charge polarization will lead to the most charge accumulation at P atoms,which results in a distinct nonmetallic P active sites with the moderate H∗adsorption energy.Notably,these P atoms mainly supply highly efficient catalytic sites with ultrasmall absorption energy of 0.007 eV.Correspondingly,the Ru-N_(4)-P demonstrated outstanding HER performance not only in an acidic condition but also in alkaline environment.Notably,the performance of Ru-NPC catalyst at high current is even superior to the commercial Pt/C catalysts,whether in acidic or alkaline medium.Our in situ synchrotron radiation infrared spectra demonstrate that a P-H_(ads) intermediate is continually emerging on the Ru-NPC catalyst,actively proving the nonmetallic P catalytically active site in HER that is very different with previously reported metallic sites.Chuanqiang Wu Shiqing Ding Daobin Liu Dongdong Li Shuangming Chen Huijuan Wang Zeming Qi Binghui Ge Li Song 2020Research2020,,1:1

20Reactivity,Regioselectivity,and Synthetic Application of 2-Pyrenyl Units in Scholl Reactions显示文摘We herein report the reactivity and regioselectivity of 2-pyrenyl as a coupling unit in Scholl reactions.On the basis of the Scholl reactions of hexaarylbenzene substrates,we have found that pyrenyl units are preferably oxidized over naphthyl and phenyl units under appropriate Scholl reaction conditions,allowing divergent synthesis through a highly controllable intramolecular coupling sequence.We find that the C1 and C3 positions of the 2-pyrenyl unit are the favorable sites for intramolecular coupling while C4 is not reactive to allow further coupling.The reactivity and regioselectivity pattern can be explained by the spin density distribution,which shows that carbon-carbon bonds preferably form at sites with higher positive spin density.Guided by these findings,we successfully synthesized a double helicene and a sextuple helicene through the controlled Scholl reactions of 2-pyrenyl units.Sai Ho Pun Ethan Chi Ho Wen Zeming Xia Han Chen Felix RFischer Qian Miao 2023CCS Chemistry2023,5,9:0
