| 1 | Jianpijiedu Fang improves survival of hepatocarcinoma mice by affecting phosphatase and tensin homolog, phosphoinositide 3-kinase, and focal adhesion kinase显示文摘OBJECTIVE: To investigate the effect of Jianpijiedu Fang (JPJDF) on phosphatase and tensin homolog (PTEN), phosphoinositide 3-kinase (PI3K), and focal adhesion kinase (FAK), and on the survival of hepatocellular carcinoma (HCC) nude mice. METHODS: Forty male nude mice were randomly divided into 4 groups. Human HCC tissue was implanted in the livers of three groups. After 24 h, the three groups were treated respectively with JPJDF (37.5 g/kg), saline (20 mL/kg) and Tegafur (FT-207, 160 mg/kg) once a day for 10 weeks. The control group without implanting the tissue was concurrently treated with saline (20 mL/kg). The survival data and body weight of all mice were recorded, and expression levels of PTEN, PI3K and FAK in normal tissue and cancer tissue of the livers were eval-uated with immunohistochemical method. RESULTS: The cumulative survival rate of the mice in the JPJDF group was higher than those of the other groups. The rate of weight loss was the lowest in JPJDF group. The survivability and weight loss rate in FT-207 group were the poorest in all groups. The expression intensity of PTEN was higher in normal tissues than in cancer tissues, and lower in the normal tissues of HCC models than in that of mice without HCC. The PTEN expression intensity in normal tissue and cancer tissue from mice treated with FT-207 were lower than that from the mice treated with JPJDF or saline.The expression intensity of PI3K was higher in cancer tissue than in normal tissue. The PI3K expression intensity was the lowest in normal tissue and cancer tissue from mice treated with JPJDF, and the intensity from mice treated with FT-207 was the highest. In mice treated with JPJDF, the expression intensity of FAK was higher in the normal tissue and lower in the cancer tissue than those of the other treatment groups. | 2 | Sleep Deprivation Selectively Down-Regulates Astrocytic 5-HT2B Receptors and Triggers Depressive-Like Behaviors via Stimulating P2X7 Receptors in Mice显示文摘Chronic loss of sleep damages health and disturbs the quality of life.Long-lasting sleep deprivation(SD)as well as sleep abnormalities are substantial risk factors for major depressive disorder,although the underlying mechanisms are not clear.Here,we showed that chronic SD in mice promotes a gradual elevation of extracellular ATP,which activates astroglial P2X7 receptors(P2X7Rs).Activated P2X7Rs,in turn,selectively down-regulated the expression of 5-HT2B receptors(5-HT2BRs)in astrocytes.Stimulation of P2X7Rs induced by SD selectively suppressed the phosphorylation of AKT and FoxO3 a in astrocytes,but not in neurons.The overexpression of FoxO3a in astrocytes inhibited the expression of 5-HT2BRs.Down-regulation of 5-HT2BsRs instigated by SD suppressed the activation of STAT3 and relieved the inhibition of Ca2+-dependent phospholipase A2.This latter cascade promoted the release of arachidonic acid and prostaglandin E2.The depression-like behaviors induced by SD were alleviated in P2X7R-KO mice.Our study reveals the mechanism underlying chronic SD-induced depression-like behaviors and suggests 5-HT2BRs as a key target for exploring therapeutic strategies aimed at the depression evoked by sleep disorders. | Maosheng Xia Zexiong Li Shuai Li Shanshan Liang Xiaowei Li Beina Chen Manman Zhang Chengyi Dong Alexei Verkhratsky Dawei Guan Baoman Li | 2020 | Neuroscience Bulletin2020,36,11: | 10 |

| 4 | Effect of polypeptide 2B1 on condition of dampness pattern in rats in terms of Traditional Chinese Medicine显示文摘OBJECTIVE: This study investigated how polypeptide 2B1 is involved in regulating and governing dampness in rat models with dampness pattern defined in terms ofTraditional Chinese Medicine.METHODS: We randomly divided 48 SPF10-week-old male Sprague-Dawley(SD) rats into a normal group, normal + Aristolochic acid I(AA-I) for5 min group, normal + AA-I for 60 min group,dampness pattern group(DS-Group), dampness pattern + AA-I for 5 min group, and dampness pattern + AA-I for 60 min group. Groups were then treated accordingly. | 5 | Correlation between pathogenesis of dampness syndrome and Interleukin-2,Interleukin-8 in rats显示文摘OBJECTIVE: To explore the pathogenesis of dampness syndrome by detecting the changes of interleukin-2 (IL-2) and interleukin-8 (IL-8) levels. METHODS: Female Sprague Dawley rats were divided randomly into five groups according to the random number table: a normal group (GroupⅠ), an external dampness group (GroupⅡ), an internal dampness group (Group Ⅲ), and an external and internal dampness group (GroupⅣ). Twenty days after the model made, IL-2 and IL-8 levels were detected by radioimmunoassay method. RESULTS: The IL-2 and IL-8 levels among groups were significant (F=3.102, P<0.05; F=2.657, P<0.05, respectively). The level of IL-2 in Group Ⅱ and Group Ⅲ were higher than that in GroupⅠ(P<0.05, P<0.01, respectively), especially higher in the Group Ⅲ compared with GroupⅡ (P<0.05). The lev- el of IL-8 in Group Ⅲ were higher than those inGroupⅠ, GroupⅡ and GroupⅣ (P<0.05, P<0.01, P< 0.05, respectively). In the GroupⅢ, the 24-hour wa ter and body weight were higher than that in the GroupⅣ (all P<0.05), and spontaneous movemen frequency was higher than those in GroupⅡ and GroupⅣ (P<0.05). | 6 | Multi-scale finite element simulation on large deformation behavior of wood under axial and transverse compression conditions显示文摘Multi-scale finite element method is adopted to simulate wood compression behavior under axial and transverse loading. Representative volume elements (RVE) of wood microfibril and cell are proposed to analyze orthotropic mechanical behavior. Lignin, hemicellulose and crystalline-amorphous cellulose core of spruce are concerned in spruce nanoscale model. The equivalent elastic modulus and yield strength of the microfibril are gained by the RVE simulation. The anisotropism of the crystalline-amorphous cellulose core brings the microfibril buckling deformation during compression loading. The failure mechanism of the cell-wall under axial compression is related to the distribution of amorphous cellulose and crystalline cellulose. According to the spruce cell observation by scanning electron microscope, numerical model of spruce cell is established using simplified circular hole and regular hexagon arrangement respectively. Axial and transverse compression loadings are taken into account in the numerical simulations. It indicates that the compression stress-strain curves of the numerical simulation are consistent with the experimental results. The wood microstructure arrangement has an important effect on the stress plateau during compression process. Cell-wall buckling in axial compression induces the stress value drops rapidly. The wide stress plateau duration means wood is with large energy dissipation under a low stress level. The numerical results show that loading velocity affects greatly wood microstructure failure modes in axial loading. For low velocity axial compression, shear sliding is the main failure mode. For high velocity axial compression, wood occur fold and collapse. | 8 | Chromosome-level assembly of triploid genome of Sichuan pepper(Zanthoxylum armatum)显示文摘As an important spice species in Rutaceae, the Sichuan pepper (Zanthoxylum armatum) can provide pungent and numbing taste, as well as aroma in its mature fruit. Here we assembled a chromosome-level genome of green prickly ash which was widely cultivated in a major production area including Chongqing and Sichuan province, China. We generated 712 Gb (~112×) PacBio long reads and 511 Gb (~82×) Hi-C data, and yielded an assembly of 99 pseudochromosomes with total size of 5.32 Gb and contig N50 of 796 kb. The genomic analyses and cytogenetic experiments both indicated that the cultivarZhuye Huajiao’ was a triploid. We identified a Zanthoxylum-specific whole genome duplication event emerging about 24.8 million years ago (Mya). We also detected a transposition burst event (0.3-0.4 Mya) responsible for the large genome size of Z. armatum. Metabolomic analysis of the Zanthoxylum fruits during development stages revealed profiles of39 volatile aroma compounds and 528 secondary metabolites, from which six types of sanshools were identified. Based on metabolomic and transcriptomic network analysis, we screened candidate genes encoding long chain acyl-CoA synthetase, fatty acid desaturase,branched-chain amino acid aminotransferase involved in sanshool biosynthesis and three genes encoding terpene synthase during fruit development.
