1Circular RNAs and human glioma显示文摘As a newly discovered type of RNA, circular RNAs(circRNAs) are widespread throughout the eukaryotic genome. The expression of circRNAs is regulated by both cis-elements and trans-factors, and the expression pattern of circRNAs is cell type-and diseasespecific. Similar to other types of non-coding RNAs, functions of circRNAs are also versatile. CircRNAs have been reported previously to function as microRNA(miRNA) sponges, protein sponges, coding RNAs or scaffolds for protein complexes.Recently, several circRNAs have been reported to play important roles in human malignancies, including glioma. Here, we reviewed several reports related to circRNAs and glioma, as well as the potential diagnostic and therapeutic applications of circRNAs in brain cancer. In general, some circRNAs, such as circSMARCA5 and circCFH, are found to be expressed in a gliomaspecific pattern, these circRNAs may be used as tumor biomarkers. In addition, some circRNAs have been found to play oncogenic roles in glioma(e.g., circNFIX and circNT5E), whereas others have been reported to function as tumor suppressors(e.g.,circFBXW7 and circSHPRH). Furthermore, circRNA is a good tool for protein expression because of its higher stability compared to linear RNAs. Thus, circRNAs may also be an ideal choice for gene/protein delivery in future brain cancer therapies. There are some challenges in circRNA research in glioma and other diseases. 5Successful treatment of pyogenic ventriculitis caused by extensively drug-resistant Acinetobacter baumannii with multi-route tigecycline: A case report显示文摘BACKGROUND Pyogenic ventriculitis caused by extensively drug-resistant Acinetobacter baumannii(A.baumannii)is one of the most severe complications associated with craniotomy.However,limited therapeutic options exist for the treatment of A.baumannii ventriculitis due to the poor penetration rate of most antibiotics through the blood-brain barrier.CASE SUMMARY A 68-year-old male patient with severe traumatic brain injury developed pyogenic ventriculitis on postoperative day 24 caused by extensively drug-resistant A.baumannii susceptible to tigecycline only.Successful treatment was accomplished through multi-route administration of tigecycline,including intravenous combined with continuous ventricular irrigation plus intraventricular administration.The pus was cleared on the 3rd day post-irrigation,and cerebrospinal fluid cultures were negative after 12 d.CONCLUSION Our findings suggest that multi-route administration of tigecycline can be a therapeutic option against pyogenic ventriculitis caused by extensively drugresistant A.baumannii.Wei Li Dan-Dong Li Bo Yin Dong-Dong Lin Han-Song Sheng Nu Zhang 2021World Journal of Clinical Cases2021,9,3:2

8A novel protein RASON encoded by a lncRNA controls oncogenic RAS signaling in KRAS mutant cancers显示文摘Mutations of the RAS oncogene are found in around 30%of all human cancers yet direct targeting of RAS is still considered clinically impractical except for the KRAS^(G12C)mutant.Here we report that RAS-ON(RASON),a novel protein encoded by the long intergenic non-protein coding RNA 00673(LINC00673),is a positive regulator of oncogenic RAS signaling.RASON is aberrantly overexpressed in pancreatic ductal adenocarcinoma(PDAC)patients,and it promotes proliferation of human PDAC cell lines in vitro and tumor growth in vivo.CRISPR/Cas9-mediated knockout of Rason in mouse embryonic fibroblasts inhibits KRAS-mediated tumor transformation.Genetic deletion of Rason abolishes oncogenic KRAS-driven pancreatic and lung cancer tumorigenesis in LSL Kras^(G12D);Trp53R172H/+mice.Mechanistically,RASON directly binds to KRAS^(G12D)/V and inhibits both intrinsic and GTPase activating protein(GAP)-mediated GTP hydrolysis,thus sustaining KRAS^(G12D/V)in the GTP-bound hyperactive state.Therapeutically,deprivation of RASON sensitizes KRAS mutant pancreatic cancer cells and patient-derived organoids to EGFR inhibitors.Our findings identify RASON as a critical regulator of oncogenic KRAS signaling and a promising therapeutic target for KRAS mutant cancers.Rongjie Cheng Fanying Li Maolei Zhang Xin Xia Jianzhuang Wu Xinya Gao Huangkai Zhou Zhi Zhang Nunu Huang Xuesong Yang Yaliang Zhang Shunli Shen Tiebang Kang Zexian Liu Feizhe Xiao Hongwei Yao Jianbo Xu Chao Yan Nu Zhang 2023Cell Research2023,33,1:1
