1Type 1 CD8^+ T Cells are Superior to Type 2 CD8^+ T Cells in Tumor Immunotherapy due to Their Efficient Cytotoxicity,Prolonged Survival and Type 1 Immune Modulation显示文摘CD8+ cytotoxic T(Tc) cells play a crucial role in host immune responses to cancer,and in this context,adoptive CD8+ Tc cell therapy has been studied in numerous animal tumor models. Its antitumor efficacy is,to a large extent,determined by the ability of Tc cells to survive and infiltrate tumors. In clinical trials,such in vitro-activated T cells often die within hours to days,and this greatly limits their therapeutic efficacy. CD8+ Tc cells fall into two subpopulations based upon their differential cytokine secretion. In this study,we in vitro generated that ovalbumin(OVA) -pulsed dendritic cell(DCOVA) -activated CD8+ type 1 Tc(Tc1) cells secreting IFN-γ,and CD8+ type 2 Tc(Tc2) cells secreting IL-4,IL-5 and IL-10,which were derived from OVA-specific T cell receptor(TCR) transgenic OT I mice. We then systemically investigated the in vitro and in vivo effector function and survival of Tc1 and Tc2 cells,and then assessed their survival kinetics after adoptively transferred into C57BL/6 mice,respectively. We demonstrated that,when compared to CD8+ Tc2,Tc1 cells were significantly more effective in perforin-mediated cytotoxicity to tumor cells,had a significantly higher capacity for in vivo survival after the adoptive T cell transfer,and had a significantly stronger therapeutic effect on eradication of well-established tumors expressing OVA in animal models. In addition,CD8+ Tc1 and Tc2 cells skewed the phenotype of CD4+ T cells toward Th1 and Th2 type,respectively. 2Circular RNA circStag1 promotes bone regeneration by interacting with HuR显示文摘Postmenopausal osteoporosis is a common bone metabolic disorder characterized by deterioration of the bone microarchitecture,leading to an increased risk of fractures.Recently,circular RNAs(circ RNAs)have been demonstrated to play pivotal roles in regulating bone metabolism.However,the underlying functions of circ RNAs in bone metabolism in postmenopausal osteoporosis remain obscure.Here,we report that circ Stag1 is a critical osteoporosis-related circ RNA that shows significantly downregulated expression in osteoporotic bone marrow mesenchymal stem cells(BMSCs)and clinical bone tissue samples from patients with osteoporosis.Overexpression of circ Stag1 significantly promoted the osteogenic capability of BMSCs.Mechanistically,we found that circ Stag1 interacts with human antigen R(Hu R),an RNA-binding protein,and promotes the translocation of Hu R into the cytoplasm.A high cytoplasmic level of Hu R led to the activation of the Wnt signaling pathway by stabilizing and enhancing low-density lipoprotein receptor-related protein 5/6(Lrp5/6)andβ-catenin expression,thereby stimulating the osteogenic differentiation of BMSCs.Furthermore,overexpression of circ Stag1 in vivo by circ Stag1-loaded adeno-associated virus(circ Stag1-AAV)promoted new bone formation,thereby preventing bone loss in ovariectomized rats.Collectively,we show that circ Stag1 plays a pivotal role in promoting the regeneration of bone tissue via Hu R/Wnt signaling,which may provide new strategies to prevent bone metabolic disorders such as postmenopausal osteoporosis.Gaoyang Chen Canling Long Shang Wang Zhenmin Wang Xin Chen Wanze Tang Xiaoqin He Zhiteng Bao Baoyu Tan Jin Zhao Yongheng Xie Zhizhong Li Dazhi Yang Guozhi Xiao Songlin Peng 2022Bone Research2022,10,3:7
3New Layered SOA-Based Architecture for Multi-robots Cooperative Online SLAM显示文摘A new layered multi-robots architecture for cooperative online simultaneous localization and mapping based on Service-oriented architecture(SOA) is proposed. This architecture is constructed by the concepts of service-oriented and loosely coupled center scheduling.These methods make robots' underlying function realization be transparent in cooperation and avoid the impact affected by robot heterogeneous characteristics, which will be beneficial to the system construction, expansion, restructuring and maintenance. All data packages follow the SOA unified data format, which can avoid the impact affected by sensor heterogeneous characteristics. The point to point serving model reduces the path of data transmission which can improve the timeliness of the system. All of these features can satisfy the requirements of multi-robots cooperative online SLAM. 7A Novel Dynamic Obstacle Avoidance Algorithm Based on Collision Time Histogram显示文摘Robot path planning in uncertain dynamic environment is a hot issue in the field of Unmanned ground vehicle(UGV).Starting from the practical demands of UGV,we propose a novel dynamic obstacle avoidance algorithm based on Collision time histogram(CTH).Given current steering angle,an effective collision check model,which is called Collision check circles(CCC),is firstly calculated.The local environment information is then combined with CCC to generate the proposed CTH.The nonholonomic nature of the vehicle is embedded in this process.Finally,the proposed algorithm calculates the executing steering angle by considering both the CTH and the target point.Extensive experiments and comparisons are conducted to evaluate the performance of the proposed algorithm.Simulation experiments are firstly conducted to verify its feasibility.Furthermore,real-world experiment is conducted to verify its effectiveness.Experimental results demonstrate the practical value of the proposed algorithm.ZHUGE Chengchen CAI Yunfei TANG Zhenmin 2017Chinese Journal of Electronics2017,26,3:2

Many parallel lines of different direction from different images were extracted, and then were used to optimize the distortion parameters by nonlinear least square. The thought of step by step was added when the optimization method working. 3D world coordination is not need to know, and the method is easy to implement. 14Theory and Application of Multi-robot Service-oriented Architecture显示文摘In order to solve the problem of heterogeneity in multi-robot cooperation, a new service-oriented architecture is proposed for multi-robot cooperation. Service provision and energy consumption are the basic cooperative behaviors. A set of basic concepts of robot service are proposed. A layered multi-robot service-oriented architecture is designed.
