| 1 | Origin and phylogenetic analysis of Tibetan Mastiff based on the mitochondrial DNA sequence显示文摘At present,the Tibetan Mastiff is the oldest and most ferocious dog in the world. However,the origin of the Tibetan Mastiff and its phylogenetic relationship with other large breed dogs such as Saint Bernard are unclear. In this study,the primers were designed accord-ing to the mitochondrial genome sequence of the domestic dog,and the 2,525 bp mitochondrial sequence,containing the whole sequence of Cytochrome b,tRNA-Thr,tRNA-Pro,and control region of the Tibetan Mastiff,was obtained. Using grey wolves and coyotes as out-groups,the Tibetan Mastiff and 12 breeds of domestic dogs were analyzed in phylogenesis. Tibetan Mastiff,domestic dog breeds,and grey wolves were clustered into a group and coyotes were clustered in a group separately. This indicated that the Tibetan Mastiff and the other domestic dogs originated from the grey wolf,and the Tibetan Mastiff belonged to Carnivora,Canidae,Canis,Canis lupus,Canis lupus familiaris on the animal taxonomy. In domestic dogs,the middle and small breed dogs were clustered at first; German Sheepdog,Swedish Elkhound,and Black Russian Terrier were clustered into one group,and the Tibetan Mastiff,Old English Sheepdog,Leonberger,and Saint Bernard were clustered in another group. This confirmed the viewpoint that many of the famous large breed dogs worldwide such as Saint Bernard possibly had the blood lineage of the Tibetan Mastiff,based on the molecular data. | 4 | Analysis of IGF2 mRNA expression and its methylation status between cattle yaks and their parents显示文摘Cattle yaks are an F1 hybrid between cattle and yaks and exhibit significant hybrid vigor. However, cattle yaks are sterile males. To study the epigenetic aspects of this reproductive isolation, IGF2 mRNA expression in cattle yaks and their parents and the patterns of the IGF2 differentially methylated regions (DMR) were examined in blood and testes. The results showed that IGF2 expression in cattle yaks' testes was lower than in their parents (P < 0.01). The IGF2 DMR was highly methylated (above 90%) in blood and testes of cattle yaks, yaks and cattle, with the highest level in cattle yaks (P > 0.05). Our study showed that IGF2 plays an important role in bovine spermato-genesis and might be involved in cattle-yak male sterility. | 5 | Zanthoxylum-specific whole genome duplication and recent activity of transposable elements in the highly repetitive paleotetraploid Z.bungeanum genome显示文摘Zanthoxylum bungeanum is an important spice and medicinal plant that is unique for its accumulation of abundant secondary metabolites,which create a characteristic aroma and tingling sensation in the mouth.Owing to the high proportion of repetitive sequences,high heterozygosity,and increased chromosome number of Z.bungeanum,the assembly of its chromosomal pseudomolecules is extremely challenging.Here,we present a genome sequence for Z.bungeanum,with a dramatically expanded size of 4.23 Gb,assembled into 68 chromosomes.This genome is approximately tenfold larger than that of its close relative Citrus sinensis.After the divergence of Zanthoxylum and Citrus,the lineage-specific whole-genome duplication event q-WGD approximately 26.8 million years ago(MYA)and the recent transposable element(TE)burst~6.41 MYA account for the substantial genome expansion in Z.bungeanum.The independent Zanthoxylum-specific WGD event was followed by numerous fusion/fission events that shaped the genomic architecture.Integrative genomic and transcriptomic analyses suggested that prominent speciesspecific gene family expansions and changes in gene expression have shaped the biosynthesis of sanshools,terpenoids,and anthocyanins,which contribute to the special flavor and appearance of Z.bungeanum.In summary,the reference genome provides a valuable model for studying the impact of WGDs with recent TE activity on gene gain and loss and genome reconstruction and provides resources to accelerate Zanthoxylum improvement. | Shijing Feng Zhenshan Liu Jian Cheng Zihe Li Lu Tian Min Liu Tuxi Yang Yulin Liu Yonghong Liu He Dai Zujun Yang Qing Zhang Gang Wang Jisen Zhang Huifeng Jiang Anzhi Wei | 2021 | Horticulture Research2021,8,1: | 5 |
| 6 | Molecular Fingerprints of Soil Organic Matter in a Typical Freshwater Wetland in Northeast China显示文摘Natural wetlands are known to store huge amounts of organic carbon in their soils. Despite the importance of this storage,uncertainties remain about the molecular characteristics of soil organic matter(SOM), a key factor governing the stability of soil organic carbon(SOC). In this study, the molecular fingerprints of SOM in a typical freshwater wetland in Northeast China were investigated using pyrolysis gas-chromatography/mass-spectrometry technology(Py-GC/MS). Results indicated that the SOC, total nitrogen(TN),and total sulfur contents of the cores varied between 16.88% and 45.83%, 0.93% and 2.82%, and 1.09% and 3.79%, respectively. The bulk δ^13C and δ^15N varied over a range of 9.85‰, between –26.85‰ and –17.00‰, and between –0.126‰ and 1.002‰, respectively. A total of 134 different pyrolytic products were identified, and they were grouped into alkyl(including n-alkanes(C:0) and n-alkenes(C:1),aliphatics(Al), aromatics(Ar), lignin(Lg), nitrogen-containing compounds(Nc), polycyclic aromatic hydrocarbons(PAHs), phenols(Phs), polysaccharides(Ps), and sulfur-containing compounds(Sc). On average, Phs moieties accounted for roughly 24.11% peak areas of the total pyrolysis products, followed by Lg(19.27%), alkyl(18.96%), other aliphatics(12.39%), Nc compounds(8.08%), Ps(6.49%), aromatics(6.32%), Sc(3.26%), and PAHs(1.12%). Soil organic matter from wetlands had more Phs and Lg and less Nc moieties in pyrolytic products than soil organic matters from forests, lake sediments, pastures, and farmland.δ^13 C distribution patterns implied more C3 plant-derived soil organic matter, but the vegetation was in succession to C4 plant from C3 plant. Significant negative correlations between Lg or Ps proportions and C3 plant proportions were observed. Multiple linear analyses implied that the Ar and Al components had negative effects on SOC. Alkyl and Ar could facilitate ratios between SOC and total nitrogen(C/N), while Al plays the opposite role. Al was positively related to the ratio of dissolved organic carbon(DOC) to SOC. In summary, SOM of wetlands might characterize by more Phs and lignin and less Nc moieties in pyrolytic products. | 10 | Characteristic of solid product layer of MgSO in the reaction of MgO with SO2显示文摘The microstructure, nucleation and growth of MgSO4 product layer during the reaction of MgO single crystal with SO2 and O2 were investigated with thermo gravity analyzer (TGA) and atomic force microscopy (AFM). The AFM images indicated that three dimensional islands with different sizes were formed during the initial reaction stage. At the initial stage, cone-shaped islands were formed, and most of them appeared at the position with terrace-step-kink. With the reaction time increasing, small islands would grow to large islands, and the coalescent would happen during this growth stage. During the product layer growth stage, the space and surface between islands would be occupied by islands, and continuum islands were formed. | 11 | The spatiotemporal changes of marshland and the driving forces in the Sanjiang Plain, Northeast China from 1980 to 2016显示文摘Background:Wetland loss is a global concern due to its enormous ecosystem services.Marshland,a typical natural wetland,which is concentrated in the Sanjiang Plain,has undergone dramatic loss in the last several decades.The spatiotemporal changes in marshland were studied based on Landsat images of the Sanjiang Plain from 1980 to 2016 with the land use maps in 1980,1995,2000,2005,2010,and 2016 using land use dynamic degree and landscape indices.The driving forces of marshland loss,including biophysical factors,socio-economic factors,and land management,were analyzed with boosted regression trees(BRTs)methods.Results:The area of marshland loss was 7372 km^(2),which accounted for 65.7%of the area of marshland in 1980;however,the paddy field area was expanded by 22,313 km^(2).The lost marshland was mainly converted to dry farmland(47.5%)and paddy field(47.2%)during 1980-2016.Both the landscape pattern of the study area and marshland became increasingly fragmented.The relatively important factors responsible for marshland loss were biophysical factors,socio-economic factors,and land management,which accounted for 65.2%,25.5%,and 8.4%,respectively.The most important driving forces with high“relative influence”were“distance to river,”which accounted for 20.6%of the total variance explained.The“relative influence”of potential crop yield and ditch density reached 20.2%and 8.4%,respectively.Conclusion:Significant land use changes have occurred in the Sanjiang Plain over the past 37 years,with characteristics of rapid paddy field expansion and drastic marshland loss.Meanwhile,marshland fragmentation continued to increase.Reclamation was the main reason for the large-scale marshland loss.Biophysical factors influenced the decisions regarding the locations of marshland loss.Moreover,climate factors(i.e.,average annual precipitation and average annual temperature)also played an important role in marshland loss.These results can provide helpful knowledge for understanding the patterns and reasons for marshland loss and protecting and managing strategies for wetlands restoration. | Zhen Li Miao Liu Yuanman Hu Zhenshan Xue Jinling Sui | 2020 | Ecological Processes2020,9,1: | 2 |
