| 1 | New incompatible pair of TCM:Epimedii Folium combined with Psoraleae Fructus induces idiosyncratic hepatotoxicity under immunological stress conditions显示文摘Epimedii Folium(EF)combined with Psoraleae Fructus(PF)is a common modern preparation,but liver injury caused by Chinese patent medicine preparations containing EF and PF has been frequently reported in recent years.Zhuangguguanjiewan pills(ZGW),which contain EF and PF,could induce immune idiosyncratic liver injury according to clinical case reports and a nonhepatotoxic dose of lipopolysaccharide(LPS)model.This present study evaluated the liver injury induced by EF or PF alone or in combination and investigated the related mechanism by using the LPS model.Liver function indexes and pathological results showed that either EF or PF alone or in combination led to liver injury in normal rats;however,EF or PF alone could lead to liver injury in LPS-treated rats.Moreover,EF combined with PF could induce a greater degree of injury than that caused by EF or PF alone in LPS-treated rats.Furthermore,EF or PF alone or in combination enhanced the LPS-stimulated inflammatory cytokine production,implying that IL-1β,which is processed and released by activating the NLRP3 inflammasome,is a specific indicator of EF-induced immune idiosyncratic hepatotoxicity.Thus,EF may induce liver injury through enhancing the LPS-mediated proinflammatory cytokine production and activating the NLRP3 inflammasome.In addition,the metabolomics analysis results showed that PF affected more metabolites in glycerophospholipid and sphingolipid metabolic pathways compared with EF in LPS model,suggesting that PF increased the responsiveness of the liver to LPS or other inflammatory mediators via modulation of multiple metabolic pathways.Therefore,EF and PF combination indicates traditional Chinese medicine incompatibility,considering that it induces idiosyncratic hepatotoxicity under immunological stress conditions. | Yuan Gao Zhilei Wang Jinfa Tang Xiaoyi Liu Wei Shi Nan Qin Xiaoyan Wang Yu Pang Ruisheng Li Yaming Zhang Jiabo Wang Ming Niu Zhaofang Bai Xiaohe Xiao | 2020 | Frontiers of Medicine2020,14,1: | 32 |

| 3 | Cardamonin from a medicinal herb protects against LPS-induced septic shock by suppressing NLRP3 inflammasome显示文摘Aberrant activation of NLRP3 inflammasome has been implicated in the pathogenesis of diverse inflammation-related diseases, and pharmacological molecules targeting NLRP3 inflammasome are of considerable value to identifying potential therapeutic interventions. Cardamonin(CDN), the major active ingredient of the traditional Chinese medicinal herb Alpinia katsumadai, has exerted an excellent anti-inflammatory activity, but the mechanism underlying this role is not fully understood. Here, we show that CDN blocks canonical and noncanonical NLRP3 inflammasome activation triggered by multiple stimuli. Moreover, the suppression of CDN on inflammasome activation is specific to NLRP3, not to NLRC4 or AIM2 inflammasome. Besides, the inhibitory effect is not dependent on the expression of NF-κB-mediated inflammasome precursor proteins. We also demonstrate that CDN suppresses the NLRP3 inflammasome through blocking ASC oligomerization and speckle formation in a dose-dependent manner.Importantly, CDN improves the survival of mice suffering from lethal septic shock and attenuates IL-1βproduction induced by LPS in vivo, which is shown to be NLRP3 dependent. | 4 | Electrochemical single nucleotide polymorphisms genotyping on surface immobilized three-dimensional branched DNA nanostructure显示文摘An electrochemical assay for single nucleotide polymorphisms (SNPs) genotyping is reported. Although electrochemical method is sensitive for DNA detection on surfaces, the ability of surface assay to precisely recognize DNA hybridization event is sacrificed to some extent due to the crowded confined surfaces environments that disfavor DNA hybridization. In the present study, we employed branched tetrahedron structure probes (TSPs) to replace regular linear single stranded DNA capture probes that were immobilized on solid surfaces. This three-dimensional DNA nanostructure lowers the density of immobilized DNA probes on confined surfaces, providing a hybridization environment that is similar to homogenous solution. | 6 | Bavachin enhances NLRP3 inflammasome activation induced by ATP or nigericin and causes idiosyncratic hepatotoxicity显示文摘Psoraleae Fructus(PF)is a well-known traditional herbal medicine in China,and it is widely used for osteoporosis,vitiligo,and other diseases in clinical settings.However,liver injury caused by PF and its preparations has been frequently reported in recent years.Our previous studies have demonstrated that PF could cause idiosyncratic drug-induced liver injury(IDILI),but the mechanism underlying its hepatotoxicity remains unclear.This paper reports that bavachin isolated from PF enhances the specific stimuli-induced activation of the NLRP3 inflammasome and leads to hepatotoxicity.Bavachin boosts the secretion of IL-ip and caspase-1 caused by ATP or nigericin but not those induced by poly(I:C),monosodium urate crystal,or intracellular lipopolysaccharide.Bavachin does not affect AIM2 or NLRC4 inflammasome activation.Mechanistically,bavachin specifically increases the production of nigericin-induced mitochondrial reactive oxygen species among the most important upstream events in the activation of the NLRP3 inflammasome.Bavachin increases the levels of aspartate transaminase and alanine aminotransferase in serum and hepatocyte injury accompanied by the secretion of IL-ip via a mouse model of lipopolysaccharide-mediated susceptibility to IDILI.These results suggest that bavachin specifically enhances the ATP-or nigericin-induced activation of the NLRP3 inflammasome.Bavachin also potentially contributes to PF-induced idiosyncratic hepatotoxicity.Moreover,bavachin and PF should be evaded among patients with diseases linked to the ATP-or nigericin-mediated activation of the NLRP3 inflammasome,which may be a dangerous factor for liver injury. | Nan Qin Guang Xu Yan Wang Xiaoyan Zhan Yuan Gao Zhilei Wang Shubin Fu Wei Shi Xiaorong Hou Chunyu Wang Ruisheng Li Yan Liu Jiabo Wang Haiping Zhao Xiaohe Xiao Zhaofang Bai | 2021 | Frontiers of Medicine2021,15,4: | 9 |
| 7 | Provenance of Central Canyon in Qiongdongnan Basin as evidenced by detrital zircon U-Pb study of Upper Miocene sandstones显示文摘Deep-water canyon systems can provide important sandstone reservoirs for deep-water oil and gas exploration in the South China Sea;however,the sedimentary provenance of the Central Canyon in the Qiongdongnan Basin remains controversial.In this work,detrital zircon grains from three drilling sandstones in the Upper Miocene Huangliu Formation in the western part of the Central Canyon were analysed by LA-ICP-MS for U-Pb ages,in order to constrain their provenance.One hundred and ninety-one zircon grains yield concordant U-Pb ages ranging from 28.6 to 3285 Ma.Most of them show oscillatory or linear zoning in CL-images and high Th/U ratios(>0.1),suggesting that they are magmatic zircons.Three major age clusters at about30 Ma(N=6),220–270 Ma(N=29),and 420–440 Ma(N=13),and five minor age clusters at 70–110 Ma(N=7),150–170 Ma(N=4),800–850 Ma(N=11),1800–2000 Ma(N=16),and 2400–2600 Ma(N=7),can be identified in the age spectrum,which are very similar to those of the Upper Miocene sandstones and modern river sands in the Red River area,but different from those of other nearby regions(e.g.,Hainan Island,the Pearl River area,and the Mekong River area)in Southeast Asia.The major age peak at about 30 Ma in our samples is consistent with the timing of tectonothermal events in the Red River Fault Zone.Therefore,we suggest that the provenance of the western part of the Central Canyon,in the Qiongdongnan Basin,was fed dominantly by the Paleo-Red River system during the Late Miocene. | CHEN Hui XIE XiNong GUO JingLiang SU Ming ZONG KeQing SHANG Fei HUANG Wei WANG Wei SHANG ZhiLei | 2015 | Science China Earth Sciences2015,58,8: | 6 |
| 8 | Phytochrome B interacts with SWC6 and ARP6 to regulate H2A.Z deposition and photomorphogensis in Arabidopsis显示文摘Light serves as a crucial environmental cue which modulates plant growth and development, and which is controlled by multiple photoreceptors including the primary red light photoreceptor,phytochrome B(phyB). The signaling mechanism of phyB involves direct interactions with a group of basic helix-loop-helix(bHLH) transcription factors, PHYTOCHROME-INTERACTING FACTORS(PIFs), and the negative regulators of photomorphogenesis, COP1 and SPAs. H2 A.Z is an evolutionarily conserved H2 A variant which plays essential roles in transcriptional regulation. The replacement of H2 A with H2 A.Z is catalyzed by the SWR1 complex. Here, we show that the Pfr form of phyB physically interacts with the SWR1 complex subunits SWC6 and ARP6. phyB and ARP6 coregulate numerous genes in the same direction,some of which are associated with auxin biosynthesis and response including YUC9, which encodes a rate-limiting enzyme in the tryptophandependent auxin biosynthesis pathway. Moreover,phyB and HY5/HYH act to inhibit hypocotyl elongation partially through repression of auxin biosynthesis. | 9 | Machine learning-based predictions of fatigue life and fatigue limit for steels显示文摘To predict the fatigue life for oblique hyperbola-and bilinear-mode S-N curves of metallic materials with various strengths,a machine-learning approach for direct analysis was employed.Additionally,to determine the fatigue limit of the utilized materials(AISI 316,AISI 4140 and CA6 NM series)with different S-N curve modes using finite-fatigue life data,a Bayesian optimization-based inverse analysis was performed.The results indicated that predictions of the fatigue life for the utilized datasets via the random forest(RF)algo rithm for AISI 4140 and CA6 NM,and artificial neural network(ANN)for AISI 316,distribute within 2 factor error lines for most data.In the Bayesian optimization-based inverse analysis,the specific explanatory variables corresponding to the optimized maximum fatigue life were treated as the fatigue limits.The predicted fatigue limits either approximated to or slightly underestimated the experimental results,except for several cases with large errors.Using the inverse analysis to predict the fatigue limit for both S-N curve modes is applicable for current employed data-set.However,the explored maximum fatigue lives via BO corresponding to the predicted fatigue limit were underestimated for AISI 4140 and CA6 NM,and was overestimated for AISI 316 because of effect of shape of S-N curves.By combining the ANN or RF direct and BO inverse algorithms,whole S-N curves(including the fatigue limit)were evaluated for the S-N curve shapes of the oblique hyperbola and bilinear modes. | steelsLei He ZhiLei Wang Hiroyuki Akebono Atsushi Sugeta | 2021 | Journal of Materials Science & Technology2021,,31: | 5 |
| 10 | Numerical simulation for propagation characteristics of shock wave and gas flow induced by outburst intensity显示文摘In order to analyze the propagation characteristics of shock wave and gas flow induced by outburst intensity, the governing equations of shock wave and gas flow propagation were put forward, and the numerical simulation boundary condition was obtained based on outburst characteristics. The propagation characteristics of shock wave and gas flow were simulated by Fluent software, and the simulation results were verified by experiments. The results show that air shock wave is formed due to air medium compressed by the transient high pressure gas which rapidly expands in the roadway; the shock wave and gas flow with high velocity are formed behind the shock wave front, which significantly decays due to limiting effect of the roadway wall. | 13 | Growth model of a hydrothermal low-temperature Si-rich chimney:Example from the CDE hydrothermal field,Lau Basin显示文摘The CDE hydrothermal field was first discovered during a Chinese cruise to the East Lau Basin Spreading Centre in 2007.Apart from significant amounts of loose Fe-Si-Mn(oxyhydr) oxide(referred to as oxide below) precipitates,a small Si-rich oxide chimney was also recovered on this cruise.In this study,we report on the mineralogical and geochemical analyses of this chimney and a model for its growth that has been developed.Based on the mineralogy and O isotope results,the chimney walls can be divided into four growth generations(layers) from the inner to the outer layers:amorphous opal and barite layer(precipitation temperature 68.5°C based on oxygen isotope determinations),a rod-like amorphous layer(precipitation temperature 39.6°C),a filamentous Fe-Si oxide layer,and an outer Fe-Mn oxide layer.Investigations based on SEM and EDS showed that neutrophilic Fe-oxidizing bacteria play an important role in the formation of this chimney,particularly in the outer two generations.In the first stage,the metabolic activity of the microbes results in the pervasive precipitation of the filamentous Fe-rich oxides inside a ring formed by some amorphous opal and barite;therefore,a loose porous layer forms.In the second stage,amorphous opal then precipitates inside this wall as a result of conductive cooling and gradually controls the mixing between the hydrothermal fluids and ambient seawaters.In the third stage,barite and some amorphous opal form from the higher temperature fluids at the summit of the chimney growth history.In the last stage,the chimney wall becomes thicker and denser and the exchange of hydrothermal fluids and seawater ceases.As a result,a Fe-Mn oxide layer precipitates onto the outer surface of the chimney wall as neutrophilic Fe-oxidizing bacteria reoccupy the surface of the chimney.This mineral sequence and the resultant growth generations are confirmed by the chemical characteristics of the chimney wall.Sr isotopes extracted from the Fe oxides of the four-generation wall generally show a decreasing trend of the 87 Sr/86 Sr ratios from the second layer to the inner layer(from 0.707008 to 0.705877) except for the outer layer(0.706502).The Sr isotope and chondrite normalized REE patterns of the corresponding bulk samples from the chimney wall also display a similar trend.Our study shows that the biogenic filament network plays a key role in the formation of the chimney in contrast to previous growth models of higher temperature chimneys,which often ignore the influence of biogenic factors. | SUN ZhiLei ZHOU HuaiYang YANG QunHui YIN XiJie WANG Hu YAO HuiQiang DONG CongFang | 2012 | Science China Earth Sciences2012,55,10: | 4 |

| 16 | Asari Radix et Rhizoma consumption lacks relevance for hepatocellular carcinoma in patients:A retrospective cohort study显示文摘Objective:Although some studies have linked Asari Radix et Rhizoma(Asari Radix)administration to hepatocellular carcinoma(HCC),few studies have examined the association between the development of HCC and use of Asari Radix among patients in China's Mainland.This study aimed to evaluate the realworld association between Asari Radix and HCC in patients to strengthen the understanding of Asari Radix safety.Methods:A retrospective cohort study among hepatitis B virus(HBV)-monoinfected patients and nonHBV-monoinfected patients were performed.Patients over 18 years of age were eligible for inclusion.Prescription records of inpatients and outpatients were inquired to distinguish Asari Radix users and nonusers.The risk of developing HCC among Asari Radix users and nonusers in the HBV cohort and the non-HBV cohort was analyzed.Results:There were 49500 HBV and 133148 non-HBV patients involved in the two cohorts.Among HBV patients(2901 users;46599 nonusers),the prevalence of HCC in Asari Radix users was lower than that in nonusers(145.70 vs.265.43 per 105).Among non-HBV patients(5042 users;128106 nonusers),the prevalence of HCC in Asari Radix users was lower than that in nonusers(81.62 vs.134.11 per 105).None of the hazard ratios(HRs)of Asari Radix exposure ranging from 1 g to 200 g in the two cohorts showed correlation between Asari Radix exposure and hepatocarcinogenesis.Conclusion:An obvious irrelevancy was found between the consumption of Asari Radix and HCC development both in patients with and in those without HBV infection.Use of Asari Radix under 200 g appears safe in terms of HCC risk in the Chinese population;further prospective studies are needed to confirm our results. | Zhi-e Fang Yuming Guo Zhilei Wang Tingting He Jiabo Wang Zhaofang Bai Xiaohe Xiao | 2022 | Chinese Herbal Medicines2022,14,3: | 2 |
