| 1 | Study of-amyloid adsorption and aggregation on graphite by STM and AFM显示文摘The scanning tunneling microscopy (STM) and the atomic force microscopy (AFM) have been applied to the direct study of the adsorption and aggregation of -amyloid(1-42)(A42) on the hydrophobic graphite surface. It was found that A42 were preferentially adsorbed on graphite defects such as the edges. A42 peptides self-assembled into intermediate protofibrils, which in turn self-associated to form fibrils. Usually, two or more fibrils intertwined to form the helical structure. | 2 | Formation of domain structure of erythrocyte membrane in Wistar rat fed with CeCl_3per os显示文摘To explore the possibility of absorption of lanthanides via digestive duct and their effects on one membrane structure and permeability of erythrocytes, the fine structure of erythrocyte membrane from Wistar rats, fed for 70 days of daily administration per os with 20 mg CeCl3/kg weight, was imaged by means of atomic force microscopy and FT-IR deconvolution spectra. The results show that, although the erythrocytes maintain the intact shape, the change OT secondary structure, aggregation and crosslinking of the protein particles of membrane surface and the enlarged lipid regions lead to the domain structure formation. | 4 | Circular dichroism spectroscopic studies on structures formed by telomeric DNA sequences in vitro显示文摘Telomere plays an important role in cellular processes, such as cell aging, death and carcinogenisis. Having special sequences, it can form quadrupiex structure in vitro. Circular dichroism (CD) spectroscopic studies show that TTAGGG, (TTAGGG)2 and (TTAGGG)4 can all form quadrupiex in vitro and exist mainly as parallel quadrupiex without metal ions. Both K+ and Na+ can stabilize the tetrameric structure and facilitate the forming of anti-parallel conformation. | 6 | Ordered arrays of semi-crown ligands on an Au(111) electrode surface: in situ STM study显示文摘In situ scanning tunneling microscopy (STM) and cyclic voltammetry were employed to investigate the adsorption structures of three semi-crown ligands on an Au(111) surface under the potential control. It is found that all the molecules formed ordered arrays in 0.1 mol/L HClO4 solution, although their geometric structures are complex and asymmetric. The driving force was supposed to come from the balance between intermolecular and molecule-substrate interactions. High resolution STM images revealed internal molecular structures, orientations and packing arrangements in the ordered adlayers. | 8 | Oral nano-antioxidants improve sleep by restoring intestinal barrier integrity and preventing systemic inflammation显示文摘Sleep deprivation(SD)is a severe public health threat that can cause systemic inflammation and nerve damage.Few effective and side-effect-free drugs are available to address SD.However,the bidirectional communications between the brain and gut provide new strategies for anti-SD therapeutics.Here we explored oral delivery of fullerene nano-antioxidants(FNAO)in the SD model to improve sleep by regulating abnormal intestinal barrier and systemic inflammation via the brain-gut axis.SD caused excessive reactive oxygen species(ROS)production and hyperactive inflammatory responses in the intestines of zebrafish and mouse models,leading to disturbed sleep patterns and reduced brain nerve activity.Of note,based on the property of the conjugatedπbond of the C_(60)structure to absorb unpaired electrons,oral FNAO efficiently reduced the excessive ROS in the intestines,maintained redox homeostasis and intestinal barrier integrity,and ameliorated intestinal and systemic inflammation,resulting in superior sleep improvement.Our findings suggest that maintaining intestinal homeostasis may be a promising avenue for SD-related nerve injury therapy. | Zhanfeng Wu Lei Liu Lei Li Xinran Cao Wang Jia Xiaodan Liao Zhongpu Zhao Hedong Qi Guoqiang Fan Huiqiang Lu Chunying Shu Mingming Zhen Chunru Wang Chunli Bai | 2023 | National Science Review2023,10,12: | 1 |

| 12 | Development of a novel platform for recombinant protein production in Corynebacterium glutamicum on ethanol显示文摘Corynebacterium glutamicum represents an emerging recombinant protein expression factory due to its ideal features for protein secretion,but its applicability is harmed by the lack of an autoinduction system with tight regulation and high yield.Here,we propose a new recombinant protein manufacturing platform that leverages ethanol as both a delayed carbon source and an inducer.First,we reanalysed the native inducible promoter PICL from the acetate uptake operon and found that its limited capacity is the result of the inadequate translation initial architecture.The two strategies of bicistronic design and ribozyme-based insulator can ensure the high activity of this promoter.Next,through transcriptional engineering that alters transcription factor binding sites(TFBSs)and the first transcribed sequence,the truncated promoter PA256 with a dramatically higher transcription level was generated.When producing the superfolder green fluorescent protein(sfGFP)under 1%ethanol conditions,PA256 exhibited substantially lower protein accumulation in prophase but an approximately 2.5-fold greater final yield than the strong promoter PH36.This superior expression mode was further validated using two secreted proteins,camelid antibody fragment(VHH)and endoxylanase(XynA).Furthermore,utilizing CRISPRi technology,ethanol utilization blocking strains were created,and PA256 was shown to be impaired in the phosphotransacetylase(PTA)knockdown strains,indicating that ethanol metabolism into the tricarboxylic acid cycle is required for PA256 upregulation.Finally,this platform was applied to produce the“de novo design”protein NEO-2/15,and by introducing the N-propeptide of CspB,NEO-2/15 was effectively secreted with the accumulation 281 mg/L obtained after 24 h of shake-flask fermentation.To the best of our knowledge,this is the first report of NEO-2/15 secretory overexpression. | Xinyu Yu Xiuxia Liu Xiong Gao Xunxun Luo Yankun Yang Ye Li Chunli Liu Chong Zhang Zhonghu Bai | 2022 | Synthetic and Systems Biotechnology2022,7,2: | 1 |
