| 6 | Calycosin improves cognitive function in a transgenic mouse model of Alzheimer's disease by activating the protein kinase C pathway显示文摘The major pathological changes in Alzheimer's disease are beta amyloid deposits and cognitive impairment. Calycosin is a typical phytoestrogen derived from radix astragali that binds to estrogen receptors to produce estrogen-like effects. Radix astragali Calycosin has been shown to relieve cognitive impairment induced by diabetes mellitus, suggesting calycosin may improve the cognitive function of Alzheimer's disease patients. The protein kinase C pathway is upstream of the mitogen-activated protein kinase pathway and exerts a neuroprotective effect by regulating Alzheimer's disease-related beta amyloid degradation. We hypothesized that calycosin improves the cognitive function of a transgenic mouse model of Alzheimer's disease by activating the protein kinase C pathway. Various doses of calycosin(10, 20 and 40 mg/kg) were intraperitoneally injected into APP/PS1 transgenic mice that model Alzheimer's disease. Calycosin diminished hippocampal beta amyloid, Tau protein, interleukin-1 beta, tumor necrosis factor-alpha, acetylcholinesterase and malondialdehyde levels in a dose-dependent manner, and increased acetylcholine and glutathione activities. The administration of a protein kinase C inhibitor, calphostin C, abolished the neuroprotective effects of calycosin including improving cognitive ability, and anti-oxidative and anti-inflammatory effects. | 7 | Expenditure and financial burden for the diagnosis and treatment of colorectal cancer in China:a hospital.based,multicenter,cross-sectional survey显示文摘Background: The increasing prevalence of colorectal cancer(CRC) in China and the paucity of information about relevant expenditure highlight the necessity of better understanding the financial burden and effect of CRC diagnosis and treatment. We performed a survey to quantify the direct medical and non-medical expenditure as well as the resulting financial burden of CRC patients in China.Methods: We conducted a multicenter, cross-sectional survey in 37 tertiary hospitals in 13 provinces across China between 2012 and 2014. Each enrolled patient was interviewed using a structured questionnaire. All expenditure data were inflated to the 2014 Chinese Yuan(CNY; 1 CNY = 0.163 USD). We quantified the overall expenditure and financial burden and by subgroup(hospital type, age at diagnosis, sex, education, occupation, insurance type, household income, clinical stage, pathologic type, and therapeutic regimen). We then performed generalized linear modeling to determine the factors associated with overall expenditure.Results: A total of 2356 patients with a mean age of 57.4 years were included, 57.1 % of whom were men; 13.9% of patients had stage I cancer; and the average previous-year household income was 54,525 CNY.The overall average direct expenditure per patient was estimated to be 67,408 CNY, and the expenditures for stage Ⅰ, Ⅱ, Ⅲ, and Ⅳ disease were 56,099 CNY, 59,952 CNY, 67,292 CNY, and 82,729 CNY, respectively. Non-medical expenditure accounted for 8.3%of the overall expenditure. The 1-year out-of-pocket expenditure of a newly diagnosed patient was 32,649 CNY, which accounted for 59.9% of their previous-year household income and caused 75.0% of families to suffer an unmanageable financial burden. Univariate analysis showed that financial burden and overall expenditure differed in almost all subgroups(P < 0.05), except for sex. Multivariate analysis showed that patients who were treated in specialized hospitals and those who were diagnosed with adenocarcinoma or diagnosed at a later stage were likely to spend more,whereas those with a lower household income and those who underwent surgery spent less(all P < 0.05).Conclusions: For patients in China, direct expenditure for the diagnosis and treatment of CRC seemed catastrophic,and non-medical expenditure was non-ignorable. | 8 | Geochronology and geochemistry of the Fangcheng Neoproterozoic alkali-syenites in East Qinling orogen and its geodynamic implications显示文摘Finding of Neoproterozoic syenites at Fangcheng in the northern Qinling region of the East Qinling orogen provides an important constraint on timing of tectonic transformation to extensional regime. The alkaline pluton consists mainly of nepheline syenite,aegirine syenite,and alkali-feldspar syenite. The syenites are of intermediate (SiO2=54%―62%),rich in alkali (K2O+Na2O=12%―15%),aluminum (Al2O3=16.81%―23.26%) and large ion lithophile elements (LILE),without any obvious Nb,Ta,Zr,and Hf anoma-lies. The Fangcheng syenites are geochemically characterized by relative enrichment of LREE,minor differentiation of HREE,significant negative Eu anomalies (Eu=0.13―0.23),less negative εNd(t) values of -1.37 to -3.90,young Nd model ages of 1364 to 1569 Ma,and high zircon saturation temperatures of 915 to 1044℃. The syenitic magmas probably originated from small proportion melting of upper mantle in an extensional regime of intraplate-anorogenic tectonic setting,and have been slightly contaminated by crustal materials during ascending and/or emplacement. LA-ICP-MS zircon U-Pb dating yields ages of 844.3±1.6 Ma (MSWD=0.86),suggesting that the Fangcheng alkaline syenites formed in the early Neo-proterozoic. They are the oldest Neoproterozoic alkaline rocks ever recognized in the Qinling orogen as well as in South China. | 11 | Early Intratracheal Administration of Corticosteroid and Pulmonary Surfactant for Preventing Bronchopulmonary Dysplasia in Preterm Infants with Neonatal Respiratory Distress Syndrome: A Meta-analysis显示文摘There is uncertain result with regard to the use of inhalation or instillation steroids to prevent bronchopulmonary dysplasia in preterm infants. This meta-analysis was designed to evaluate the efficacy and safety of early airway administration (within 2 days after birth) of corticosteroids and pulmonary surfactant (PS) for preventing bronchopulmonary dysplasia (BPD) in premature infants with neonatal respiratory distress syndrome (NRDS). The related studies were retrieved in PubMed, EMBASE, the Cochrane Library, Clinical Trial, CNKI, Wanfang and VIP Database from inception to August 2018. Two reviewers independently screened the studies to ensure that all patients with diagnosis of NRDS were enrolled to studies within 1 day after birth, assessed the quality of included studies by GRADEpro system and extracted the data for review. The meta-analysis was performed by RevMan 5.2 software. A subgroup analysis about inhaled corticosteroid (ICS) delivery method was made between ICS inhalation subgroup [inhalation of ICS by nebulizer or metered dose inhaler (MDI)] and ICS intratracheal instillation subgroup (PS used as a vehicle). Eight randomized controlled trials were enrolled in the meta-analysis, 5 trials of which stated the randomized method, grouping and blinded method, and the follow-up procedures were reported. GRADEpro system showed high quality of 4 trials (5 articles), and the rest 4 trials had moderate quality. Meta-analysis showed that the incidence of BPD was decreased in ICS group, the relative risk (RR) was 0.56 (95% CI: 0.42-0.76), and similar trends were found in ICS inhalation subgroup and ICS intratracheal instillation subgroup, with the corresponding RR being 0.58 (95% CI: 0.41-0.82) and 0.47 (95% CI: 0.24-0.95) respectively. ICS could also significantly reduce the mortality risk as compared with placebo control group (RR: 0.67;95% CI: 0.45-0.99), with RR of ICS inhalation subgroup and ICS intratracheal instillation subgroup being 0.81 (95% CI: 0.34-1.94) and 0.64 (95% CI: 0.41-0.99) respectively. Moreover, the percentage of infants using PS more than one time was lower in ICS group than in the placebo control group, with the RR and 95% CI being 0.55 (95% CI: 0.45-0.67), and that in ICS intratracheal instillation subgroup lower than in ICS inhalation subgroup (RR: 0.56;95% CI: 0.45-0.69, and RR: 0.35;95% CI:0.08-1.52 respectively). There was no significant difference in the incidence of infection or retinopathy of prematurity and neuro-motor system impairment between ICS group and placebo control group, with the corresponding RR being 0.95 (95% CI:0.59-1.52), 0.92 (95% CI: 0.62-1.38) and 1.13 (95% CI: 0.92-1.39), respectively. It was concluded that early administration of ICS and PS is an effective and safe option for preterm infants with NRDS in preventing BPD and reducing mortality, decreasing the additional PS usage, especially for the ICS intratracheal instillation subgroup. | 12 | Influence of High Temperature Stress on Net Photosynthesis, Dry Matter Partitioning and Rice Grain Yield at Flowering and Grain Filling Stages显示文摘Climate change is recognized to increase the frequency and severity of extreme temperature events. At flowering and grain filling stages, risk of high temperature stress (HTS) on rice might increase, and lead to declining grain yields. A regulated cabinet experiment was carried out to investigate effects of high temperature stress on rice growth at flowering and grain-filling stages. Results showed that no obvious decrease pattern in net photosynthesis appeared along with the temperature rising, but the dry matter allocation in leaf, leaf sheath, culm, and panicle all changed. Dry weight of panicle decreased, and ratio of straw to total above ground crop dry weight increased 6-34% from CK, which might have great effects on carbon cycling and green house gas emission. Grain yield decreased significantly across all treatments on average from 15 to 73%. Occurrence of HTS at flowering stage showed more serious influence on grain yield than at grain filling stage. High temperature stress showed negative effects on harvest index. | 13 | Spatial Variability of Soil Organic Carbon Under Maize Monoculture in the Song-Nen Plain, Northeast China显示文摘Soil organic carbon (SOC) and its relationship with landscape attributes are important for evaluating current regional, continental, and global carbon stores. Data of SOC in surface soils (0–20 cm) of four main soils, Cambisol, Arenosol, Phaeozem, and Chernozem, were collected at 451 locations in Nongan County under maize monoculture in the Song-Nen Plain, Northeast China. The spatial characteristics of soil organic carbon were studied, using geographic information systems (GIS) and geostatistics. Effects of other soil physical and chemical properties, elevation, slope, and soil type on SOC were explored. SOC concentrations followed a normal distribution, with an arithmetic mean of 14.91 g kg-1 . The experimental variogram of SOC was fitted with a spherical model. There were significant correlations between soil organic carbon and bulk density (r =-0.374**), pH (r = 0.549**), total nitrogen (r = 0.781**), extractable phosphorus (r =-0.109*), exchangeable potassium (r = 0.565**), and cation exchange capacity (r = 0.313**). Generally, lower SOC concentrations were significantly associated with high elevation (r =-0.429**). Soil organic carbon was significantly negatively correlated with slope gradient (r =-0.195**). Quantitative assessment of the extent, spatial distribution, and dynamics of aquaculture ponds is of utmost importance for sustainable economic development and scientific management of land and water resources in the coastal area. An object-oriented classification approach was applied to Landsat images acquired over three decades to investigate the long-term change of aquaculture ponds in the coastal region of the Yellow River Delta. The results indicated that the aquaculture ponds in the study area undergone a sharp expansion from 40.38 km^2 in 1983 to 1406.89 km^2 in 2015, and the fast expansion occurred during the period of 2010–2015 and 1990–2000. Natural wetlands, especially mudflat, and cropland were main land use types contributing to the increase of aquaculture ponds. The patches of aquaculture ponds were consequently prevalence in the north of the Yellow River Estuary and landscape metrics indicated an increase of the aquaculture ponds of the study area in the quantity and complexity. | 16 | Methylation of Dickkopf-3 as a prognostic factor in cirrhosis-related hepatocellular carcinoma显示文摘AIM:To investigate the prevalence and time of Dickkopf(DKK) family methylation and its clinical significance in hepatocarcinogenesis. METHODS:Methylation of DKK family genes was quantitatively analyzed in 115 liver tissue samples,including 50 pairs of primary hepatocellular carcinoma(HCC) and matched noncancerous cirrhotic tissue samples,as well as 15 liver cirrhosis biopsy samples. RESULTS:The methylation level of DKK3 was significantly higher in HCC tissue samples than in matched noncancerous cirrhotic tissue samples(P<0.0001) orin liver cirrhosis biopsy samples(P=0.0139) .Receiver operator characteristic curve analysis confirmed that the percent of methylated reference(PMR) values of DKK3 could effectively discriminate HCC tissue samples from noncancerous tissue samples(AUC=0.8146) or liver cirrhosis biopsy samples(AUC=0.7093) .Kaplan-Meier survival curves revealed that the progression-free survival time of patients with a higher DKK3 methylation level(PMR >1%) was significantly shorter than that of those with a lower DKK3 methylation level(PMR≤1%)(P=0.0255) . Multivariate Cox analysis indicated that methylated DKK3 was significantly and independently related with a shorter survival time(relative risk=2.527,95%CI:1.063-6.008,P=0.036) of HCC patients.
