Tibetan Plateau climate dynamics: recent research progress and outlook显示文摘This paper reviews progress in the study of Tibetan Plateau(TP) climate dynamics over the past decade.Several theoretical frameworks, including thermal adaptation and the TP sensible heat(SH) driving air-pump, have been developed to identify the mechanisms responsible for the circulation anomaly produced by thermal forcing of the TP. Numerical simulations demonstrate that the thermal efects of large-scale orography, including the Tibetan and Iranian Plateaus(TIP), are crucial for the formation of the East Asian and South Asian summer monsoons(SASM) because the surface SH of the TIP is the major driver of the water vapor transport required for the genesis of the north branch of the SASM. he large-scale orography of the TP afects the Asian climate through thermal forcing in spring and summer, and mechanical forcing in winter. he TP forcing can also inluence the Asian summer monsoon(ASM) onset over the Bay of Bengal(BOB) by enhancing the BOB warm pool at the surface and by modulating the South Asian High(SAH) in the upper troposphere. On intra-seasonal timescales, the TP thermal forcing signiicantly modulates spring rainfall in southern China and generates the biweekly oscillation of the SAH in summer. Despite climate warming, the atmospheric heat source over the TP, particularly the spring SH,exhibits a clear weakening trend from the 1980 s to 2000 s. his weakening of the spring SH contributed to the anomalous ‘dry in the north' and ‘wet in the south' rainfall patern observed over East China. Also discussed are challenges to further understanding the mechanism of TP forcing on the multi-scale variability of the ASM. | Guoxiong Wu Anmin Duan Yimin Liu Jiangyu Mao Rongcai Ren Qing Bao Bian He Boqi Liu Wenting Hu | 2015 | National Science Review2015,2,1: | 51

Dynamic alterations in the gut microbiota and metabolome during the development of methionine-choline-deficient diet-induced nonalcoholic steatohepatitis显示文摘AIM To investigate changes in gut microbiota and metabolism during nonalcoholic steatohepatitis(NASH) development in mice fed a methionine-choline-deficient(MCD) diet. METHODS Twenty-four male C57 BL/6 J mice were equally divided into four groups and fed a methionine-choline-sufficient diet for 2 wk(Control 2 w group,n = 6) or 4 wk(Control 4 w group,n = 6) or the MCD diet for 2 wk(MCD 2 w group,n = 6) or 4 wk(MCD 4 w group,n = 6). Liver injury, fibrosis, and intestinal barrier function were evaluated after 2 and 4 wk of feeding. The fecal microbiome and metabolome were studied using 16 s r RNA deep sequencing and gas chromatography-mass spectrometry. RESULTS The mice fed the MCD diet presented with simple hepatic steatosis and slight intestinal barrier deterioration after 2 wk. After 4 wk of feeding with the MCD diet, however, the mice developed prominent NASH with liver fibrosis, and the intestinal barrier was more impaired. Compared with the control diet, the MCD diet induced gradual gut microbiota dysbiosis,as evidenced by a marked decrease in the abundance of Alistipes and the(Eubacterium) coprostanoligenes group(P < 0.001 and P < 0.05,respectively) and a significant increase in Ruminococcaceae UCG 014 abundance(P < 0.05) after 2 wk. At 4 wk,the MCD diet significantly reduced the promising probiotic Bifidobacterium levels and markedly promoted Bacteroides abundance(P < 0.05,and P < 0.01,respectively). The fecal metabolomic profile was also substantially altered by the MCD diet: At 2 wk,arachidic acid, hexadecane, palmitic acid, and tetracosane were selected as potential biomarkers that were significantly different in the corresponding control group, and at 4 wk,cholic acid, cholesterol, arachidic acid, tetracosane, and stearic acid were selected. CONCLUSION The MCD diet induced persistent alterations in the gut microbiota and metabolome. | Jian-Zhong Ye Ya-Ting Li Wen-Rui Wu Ding Shi Dai-Qiong Fang Li-Ya Yang Xiao-Yuan Bian Jing-Jing Wu Qing Wang Xian-Wan Jiang Cong-Gao Peng Wan-Chun Ye Peng-Cheng Xia Lan-Juan Li | 2018 | World Journal of Gastroenterology2018,24,23: | 13

The CAS FGOALS-f3-L model, experiment settings, and outputs are all given. In total, there are three ensemble experiments over the period 1979–2014, which are performed with different initial states. The model outputs contain a total of 37 variables and include the required three-hourly mean, six-hourly transient, daily and monthly mean datasets. The baseline performances of the model are validated at different time scales. The preliminary evaluation suggests that the CAS FGOALS-f3-L model can capture the basic patterns of atmospheric circulation and precipitation well, including the propagation of the Madden–Julian Oscillation, activities of tropical cyclones, and the characterization of extreme precipitation. These datasets contribute to the benchmark of current model behaviors for the desired continuity of CMIP. | Bian HE Qing BAO Xiaocong WANG Linjiong ZHOU Xiaofei WU Yimin LIU Guoxiong WU Kangjun CHEN Sicheng HE Wenting HU Jiandong LI Jinxiao LI Guokui NIAN Lei WANG Jing YANG Minghua ZHANG Xiaoqi ZHANG | 2019 | Advances in Atmospheric Sciences2019,36,8: | 12 |

The FGOALS climate system model as a modeling tool for supporting climate sciences:An overview显示文摘Climate system models are useful tools for understanding the interactions among the components of the climate system and predicting/projecting future climate change. The development of climate models has been a central focus of the State Key Laboratory of Numerical Modeling for Atmospheric Sciences and Geophysical Fluid Dynamics, Institute of Atmospheric Physics, Chinese Academy of Sciences(LASG/IAP) since the establishment of the laboratory in 1985. In China, many pioneering component models and fully coupled models of the climate system have been developed by LASG/IAP. The fully coupled climate system developed in the recent decade is named FGOALS(Flexible Global Ocean-Atmosphere-Land System Model). In this paper, an application-oriented review of the LASG/IAP FGOALS model is presented. The improved model performances are demonstrated in the context of cloud-radiation processes, Asian monsoon, ENSO phenomena, Atlantic Meridional Overturning Circulation(AMOC) and sea ice. The FGOALS model has contributed to both CMIP5(Coupled Model Intercomparison Project-phase 5) and IPCC(Intergovernmental Panel on Climate Change) AR5(the Fifth Assessment Report). The release of FGOALS data has supported the publication of nearly 500 papers around the world. The results of FGOALS are cited ~106 times in the IPCC WG1(Working Group 1) AR5. In addition to the traditional long-term simulations and projections, near-term decadal climate prediction is a new set of CMIP experiment, progress of LAGS/IAP in the development of nearterm decadal prediction system is reviewed. The FGOALS model has supported many Chinese national-level research projects and contributed to the national climate change assessment report. The crucial role of FGOALS as a modeling tool for supporting climate sciences is highlighted by demonstrating the model's performances in the simulation of the evolution of Earth's climate from the past to the future. | TianJun Zhou Bin Wang YongQiang Yu YiMin Liu WeiPeng Zheng LiJuan Li Bo Wu PengFei Lin Zhun Guo WenMin Man Qing Bao AnMin Duan HaiLong Liu XiaoLong Chen Bian He JianDong Li LiWei Zou XiaoCong Wang LiXia Zhang Yong Sun WenXia Zhang | 2018 | Earth and Planetary Physics2018,2,4: | 6
Lateral stability region conservativeness estimation and torque distribution for FWIA electric vehicle steering显示文摘Estimation of the lateral stability region and torque distribution on steering is very important to improve stability in lateral handling for all wheel drive electric vehicles.Based on the built-nonlinear vehicle dynamic model, the lateral stability region of the vehicle related to steering is estimated using Lyapunov function. We obtained stable equilibrium points of non-straight driving according to the estimated lateral stability region and also reconstructed the Lyapunov function matrix, which proved that the closed-loop system composed of yaw rate and lateral velocity is satisfied with negative definite property. In addition, the designed controller dynamically allocates the drive torque in terms of the vertical load and slip rate of the four wheels. The simulation results show that the estimated lateral stability region and the designed controller are satisfactory in handling stability performance against different roads and vehicle parameters. | YIN Guo Dong JIN Xian Jian QING Zhi Yong BIAN Chen Tong | 2015 | Science China(Technological Sciences)2015,58,4: | 5

Pericytes augment glioblastoma cell resistance to temozolomide through CCL5-CCR5 paracrine signaling显示文摘Glioblastoma(GBM)is a prevalent and highly lethal form of glioma, with rapid tumor progression and frequent recurrence.Excessive outgrowth of pericytes in GBM governs the ecology of the perivascular niche, but their function in mediating chemoresistance has not been fully explored.Herein,we uncovered that pericytes potentiate DNA damage repair(DDR)in GBM cells residing in the perivascular niche, which induces temozolomide(TMZ)chemoresistance. We found that increased pericyte proportion correlates with accelerated tumor recurrence and worse prognosis.Genetic depletion of pericytes in GBM xenografts enhances TMZ-induced cytotoxicity and prolongs survival of tumor-bearing mice.Mechanistically,C-C motif chemokine ligand 5(CCL5)secreted by pericytes activates C-C motif chemokine receptor 5(CCR5)on GBM cells to enable DNA-dependent protein kinase catalytic subunit(DNA-PKcs)-mediated DDR upon TMZ treatment.Disrupting CCL5-CCR5 paracrine signaling through the brain-penetrable CCR5 antagonist maraviroc(MVC)potently inhibits pericyte-promoted DDR and effectively improves the chemotherapeutic efficacy of TMZ.GBM patient-derived xenografts with high CCL5 expression benefit from combined treatment with TMZ and MVC.Our study reveals the role of pericytes as an extrinsic stimulator potentiating DDR signaling in GBM cells and suggests that targeting CCL5-CCR5 signaling could be an effective therapeutic strategy to improve chemotherapeutic efficacy against GBM. | Xiao-Ning Zhang Kai-Di Yang Cong Chen Zhi-Cheng He Qiang-Hu Wang Hua Feng Sheng-Qing Lv Yan Wang Min Mao Qing Liu Yao-Yao Tan Wen-Ying Wang Tian-Ran Li Lin-Rong Che Zhong-Yi Qin Ling-Xiang Wu Min Luo Chun-Hua Luo Yu-Qi Liu Wen Yin Chao Wang Hai-Tao Guo Qing-Rui Li Bin Wang Wei Chen Shuang Wang Yu Shi Xiu-Wu Bian Yi-Fang Ping | 2021 | Cell Research2021,31,10: | 5

New strain of Pediococcus pentosaceus alleviates ethanol-induced liver injury by modulating the gut microbiota and short-chain fatty acid metabolism显示文摘BACKGROUND Intestinal dysbiosis has been shown to be associated with the pathogenesis of alcoholic liver disease(ALD), which includes changes in the microbiota composition and bacterial overgrowth, but an effective microbe-based therapy is lacking. Pediococcus pentosaceus(P. pentosaceus) CGMCC 7049 is a newly isolated strain of probiotic that has been shown to be resistant to ethanol and bile salts. However, further studies are needed to determine whether P. pentosaceus exerts a protective effect on ALD and to elucidate the potential mechanism.AIM To evaluate the protective effect of the probiotic P. pentosaceus on ethanol-induced liver injury in mice.METHODS A new ethanol-resistant strain of P. pentosaceus CGMCC 7049 was isolated from healthy adults in our laboratory. The chronic plus binge model of experimental ALD was established to evaluate the protective effects. Twenty-eight C57BL/6 mice were randomly divided into three groups: The control group received a pairfed control diet and oral gavage with sterile phosphate buffered saline, the EtOH group received a ten-day Lieber-DeCarli diet containing 5% ethanol and oral gavage with phosphate buffered saline, and the P. pentosaceus group received a 5% ethanol Lieber-DeCarli diet but was treated with P. pentosaceus. One dose of isocaloric maltose dextrin or ethanol was administered by oral gavage on day 11, and the mice were sacrificed nine hours later. Blood and tissue samples(liver and gut) were harvested to evaluate gut barrier function and liver injury-related parameters. Fresh cecal contents were collected, gas chromatography–mass spectrometry was used to measure short-chain fatty acid(SCFA) concentrations, and the microbiota composition was analyzed using 16S rRNA gene sequencing.RESULTS The P. pentosaceus treatment improved ethanol-induced liver injury, with lower alanine aminotransferase, aspartate transaminase and triglyceride levels and decreased neutrophil infiltration. These changes were accompanied by decreased levels of endotoxin and inflammatory cytokines, including interleukin-5, tumor necrosis factor-α, granulocyte colony-stimulating factor, keratinocyte-derived protein chemokine, macrophage inflammatory protein-1α and monocyte chemoattractant protein-1. Ethanol feeding resulted in intestinal dysbiosis and gut barrier disruption, increased relative abundance of potentially pathogenic Escherichia and Staphylococcus, and the depletion of SCFA-producing bacteria, such as Prevotella, Faecalibacterium, and Clostridium. In contrast, P. pentosaceus administration increased the microbial diversity, restored the relative abundance of Lactobacillus, Pediococcus, Prevotella, Clostridium and Akkermansia and increased propionic acid and butyric acid production by modifying SCFA-producing bacteria. Furthermore, the levels of the tight junction protein ZO-1, mucin proteins(mucin [MUC]-1, MUC-2 and MUC-4) and the antimicrobial peptide Reg3β were increased after probiotic supplementation.CONCLUSION Based on these results, the new strain of P. pentosaceus alleviated ethanol-induced liver injury by reversing gut microbiota dysbiosis, regulating intestinal SCFA metabolism, improving intestinal barrier function, and reducing circulating levels of endotoxin and proinflammatory cytokines and chemokines. Thus, this strain is a potential probiotic treatment for ALD. | Xian-Wan Jiang Ya-Ting Li Jian-Zhong Ye Long-Xian Lv Li-Ya Yang Xiao-Yuan Bian Wen-Rui Wu Jing-Jing Wu Ding Shi Qing Wang Dai-Qiong Fang Kai-Cen Wang Qiang-Qiang Wang Yan-Meng Lu Jiao-Jiao Xie Lan-Juan Li | 2020 | World Journal of Gastroenterology2020,26,40: | 4
Acute carbon monoxide poisoning in Shandong, China: an observational study显示文摘Background: Carbon monoxide (CO) poisoning remains a major cause of accidental injuries and multiple studies have indicated that CO is also associated with significantly severe or long-term toxicity to the central nervous system. Given that CO poisoning causes serious morbidity and mortality, a better understanding of epidemiological features and clinical characteristics of acute CO poisoning in China is crucial. Methods: We collected the clinical data of acute CO poisoning in patients between November 2019 and April 2020 across Shandong province, China and analyzed its characteristics focusing on the weekly amount and the severity of the confirmed cases. Results: A total number of 21,088 acute CO poisoning cases were diagnosed. The overall incidence of acute CO poisoning was approximately 0.021%. On severity rankings, 63% of confirmed cases (n=13,378) were mild, 27%(n=5635) were moderate, and 10% (n=2075) were severe. Interestingly, the coastal cities had more confirmed cases than the inland/suburban areas in Shandong. Meanwhile, the number of confirmed cases was negatively correlated with the local mean daily temperature (P=0.0167).Conclusions: Mild acute CO poisoning cases accounted for the majority of all confirmed cases during the winter of 2019. In Shandong province, which is located in east China, residents of the coastal cities are more susceptible to CO poisoning than residents of inland cities. | Lina Zhang Di Wu Mingyue Xu Yonghui Bian Youcun Wang Guangkai Gao Qing Sun | 2022 | Chinese Medical Journal2022,,13: | 4
A proof of concept for neutralizing antibody-guided vaccine design against SARS-CoV-2显示文摘Mutations and transient conformational movements of the receptor binding domain(RBD)that make neutralizing epitopes momentarily unavailable present immune escape routes for severe acute respiratory syndrome coronavirus 2(SARS-CoV-2).To mitigate viral escape,we developed a cocktail of neutralizing antibodies(NAbs)targeting epitopes located on different domains of spike(S)protein.Screening of a library of monoclonal antibodies generated from peripheral blood mononuclear cells of COVID-19 convalescent patients yielded potent NAbs,targeting the N-terminal domain(NTD)and RBD domain of S,effective at n M concentrations.Remarkably,a combination of RBD-targeting NAbs and NTD-binding NAbs,FC05,enhanced the neutralization potency in cell-based assays and an animal model. Results of competitive surface plasmon resonance assays and cryo-electron microscopy(cryo-EM)structures of antigen-binding fragments bound to S unveil determinants of immunogenicity.Combinations of immunogens, identified in the NTD and RBD of S,when immunized in rabbits and macaques, elicited potent protective immune responses against SARS-CoV-2.More importantly, two immunizations of this combination of NTD and RBD immunogens provided complete protection in macaques against a SARS-CoV-2 challenge, without observable antibody-dependent enhancement of infection. These results provide a proof of concept for neutralization-based immunogen design targeting SARS-CoV-2 NTD and RBD. | Li Zhang Lei Cao Xing-Su Gao Bin-Yang Zheng Yong-Qiang Deng Jing-Xin Li Rui Feng Qian Bian Xi-Ling Guo Nan Wang Hong-Ying Qiu Lei Wang Zhen Cui Qing Ye Geng Chen Kui-Kui Lu Yin Chen Yu-Tao Chen Hong-Xing Pan Jiaping Yu Wenrong Yao Bao-Li Zhu Jianping Chen Yong Liu Cheng-Feng Qin Xiangxi Wang Feng-Cai Zhu | 2021 | National Science Review2021,8,8: | 4

Safety and efficacy of meplazumab in healthy volunteers and COVID-19 patients: a randomized phase 1 and an exploratory phase 2 trial显示文摘Recent evidence suggests that CD147 serves as a novel receptor for severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)infection.Blocking CD147 via anti-CD147 antibody could suppress the in vitro SARS-CoV-2 replication.Meplazumab is a humanized anti-CD147 IgG_(2) monoclonal antibody, which may effectively prevent SARS-CoV-2 infection in coronavirus disease 2019(COVID-19)patients.Here,we conducted a randomized,double-blinded,placebo-controlled phase 1 trial to evaluate the safety, tolerability, and pharmacokinetics of meplazumab in healthy subjects, and an open-labeled,concurrent controlled add-on exploratory phase 2 study to determine the efficacy in COVID-19 patients. In phase 1 study,59 subjects were enrolled and assigned to eight cohorts, and no serious treatment-emergent adverse event(TEAE)or TEAE grade≥3 was observed. The serum and peripheral blood Cmax and area under the curve showed non-linear pharmacokinetic characteristics. No obvious relation between the incidence or titer of positive anti-drug antibody and dosage was observed in each cohort. The biodistribution study indicated that meplazumab reached lung tissue and maintained>14 days stable with the lung tissue/cardiac blood-pool ratio ranging from 0.41 to 0.32.In the exploratory phase 2 study,17 COVID-19 patients were enrolled, and 11 hospitalized patients were involved as concurrent control. The meplazumab treatment significantly improved the discharged(P=0.005)and case severity(P=0.021),and reduced the time to virus negative(P=0.045)in comparison to the control group. These results show a sound safety and tolerance of meplazumab in healthy volunteers and suggest that meplazumab could accelerate the recovery of patients from COVID-19 pneumonia with a favorable safety profile. | Huijie Bian Zhao-Hui Zheng Ding Wei Aidong Wen Zheng Zhang Jian-Qi Lian Wen-Zhen Kang Chun-Qiu Hao Jing Wang Rong-Hua Xie Ke Dong Jie-Lai Xia Jin-Lin Miao Wen Kang Guoquan Li Di Zhang Mingru Zhang Xiu-Xuan Sun Likun Ding Kui Zhang Junfeng Jia Jin Ding Zhiqin Li Yanyan Jia Lin-Na Liu Zhe Zhang Zhao-Wei Gao Hong Du Na Yao Qing Wang Ke Wang Jie-Jie Geng Bin Wang Ting Guo Ruo Chen Yu-Meng Zhu Li-Juan Wang Qian He Rui-Rui Yao Ying Shi Xiang-Min Yang Jian-Sheng Zhou Yi-Nan Ma Ya-Tao Wang Xue Liang Fei Huo Zhe Wang Yang Zhang Xu Yang Ye Zhang Lu-Hua Gao Ling Wang Xiao-Chun Chen Hao Tang Shuang-Shuang Liu Qing-Yi Wang Zhi-Nan Chen Ping Zhu | 2021 | Signal Transduction and Targeted Therapy2021,6,6: | 2
A four-gene signature-derived risk score for glioblastoma:prospects for prognostic and response predictive analyses显示文摘Objective: Glioblastoma(GBM) is the most common primary malignant brain tumor regulated by numerous genes, with poor survival outcomes and unsatisfactory response to therapy. Therefore, a robust, multi-gene signature-derived model is required to predict the prognosis and treatment response in GBM.Methods: Gene expression data of GBM from TCGA and GEO datasets were used to identify differentially expressed genes(DEGs)through DESeq2 or LIMMA methods. The DEGs were then overlapped and used for survival analysis by univariate and multivariate COX regression.Based on the gene signature of multiple survival-associated DEGs, a risk score model was established, and its prognostic and predictive role was estimated through Kaplan–Meier analysis and log-rank test.Gene set enrichment analysis(GSEA) was conducted to explore high-risk score-associated pathways.Western blot was used for protein detection. Results: Four survival-associated DEGs of GBM were identified: OSMR, HOXC10, SCARA3, and SLC39A10.The four-gene signature-derived risk score was higher in GBM than in normal brain tissues.GBM patients with a high-risk score had poor survival outcomes. The high-risk group treated with temozolomide chemotherapy or radiotherapy survived for a shorter duration than the low-risk group.GSEA showed that the high-risk score was enriched with pathways such as vasculature development and cell adhesion.Western blot confirmed that the proteins of these four genes were differentially expressed in GBM cells. Conclusions: The four-gene signature-derived risk score functions well in predicting the prognosis and treatment response in GBM and will be useful for guiding therapeutic strategies for GBM patients. | Mianfu Cao Juan Cai Ye Yuan Yu Shi Hong Wu Qing Liu Yueliang Yao Lu Chen Weiqi Dang XiangZhang Jingfang Xiao Kaidi Yang Zhicheng He Xiaohong Yao Yonghong Cui Xia Zhang Xiuwu Bian | 2019 | Cancer Biology & Medicine2019,16,3: | 2
