1A rapid advice guideline for the diagnosis and treatment of 2019 novel coronavirus(2019-nCoV) infected pneumonia(standard version)显示文摘In December 2019, a new type viral pneumonia cases occurred in Wuhan, Hubei Province;and then named '2019 novel coronavirus(2019-nCoV)' by the World Health Organization(WHO) on 12 January 2020. For it is a never been experienced respiratory disease before and with infection ability widely and quickly, it attracted the world’s attention but without treatment and control manual. For the request from frontline clinicians and public health professionals of 2019-nCoV infected pneumonia management, an evidence-based guideline urgently needs to be developed. Therefore, we drafted this guideline according to the rapid advice guidelines methodology and general rules of WHO guideline development;we also added the first-hand management data of Zhongnan Hospital of Wuhan University. This guideline includes the guideline methodology, epidemiological characteristics, disease screening and population prevention, diagnosis, treatment and control(including traditional Chinese Medicine), nosocomial infection prevention and control, and disease nursing of the 2019-nCoV. Moreover, we also provide a whole process of a successful treatment case of the severe 2019-nCoV infected pneumonia and experience and lessons of hospital rescue for 2019-nCoV infections. 2Targeting PI3K/AKT signaling for treatment of idiopathic pulmonary fibrosis显示文摘Idiopathic pulmonary fibrosis(IPF) is a chronic progressive fibrotic interstitial pneumonia with unknown causes. The incidence rate increases year by year and the prognosis is poor without cure.Recently, phosphatidylinositol 3-kinase(PI3 K)/protein kinase B(PKB/AKT) signaling pathway can be considered as a master regulator for IPF. The contribution of the PI3 K/AKT in fibrotic processes is increasingly prominent, with PI3 K/AKT inhibitors currently under clinical evaluation in IPF. Therefore,PI3 K/AKT represents a critical signaling node during fibrogenesis with potential implications for the development of novel anti-fibrotic strategies. This review epitomizes the progress that is being made in understanding the complex interpretation of the cause of IPF, and demonstrates that PI3 K/AKT can directly participate to the greatest extent in the formation of IPF or cooperate with other pathways to promote the development of fibrosis. 3Expansion and differentiation of human hepatocyte-derived liver progenitor-like cells and their use for the study of hepatotropic pathogens显示文摘The study of pathophysiological mecha nisms in huma n liver disease has bee n constrained by the in ability to expa nd primary hepatocytes in vitro while maintaining proliferative capacity and metabolic function. We and others have previously shown that mouse mature hepatocytes can be con verted to liver progenitor-like cells in vitro with defined chemical factors. Here we describe a protocol achieving efficient conversion of human primary hepatocytes into liver progenitor-like cells (HepLPCs) through delivery of developmentally relevant cues, including NAD +-dependent deacetylase SIRT1 signaling. These HepLPCs could be expanded sign ifica ntly duri ng in vitro passage. The expa nded cells can readily be con verted back into metabolically functional hepatocytes in vitro and upon tran splantati on in vivo. Under three-dime nsional culture conditions, differentiated cells gen erated from HepLPCs regai ned the ability to support infect io n or reactivatio n of hepatitis B virus (HBV). 5Natural products:potential treatments for cisplatin-induced nephrotoxicity显示文摘Cisplatin is a clinically advanced and highly effective anticancer drug used in the treatment of a wide variety of malignancies,such as head and neck,lung,testis,ovary,breast cancer,etc.However,it has only a limited use in clinical practice due to its severe adverse effects,particularly nephrotoxicity;20%-35% of patients develop acute kidney injury(AKI)after cisplatin administration.The nephrotoxic effect of cisplatin is cumulative and dose dependent and often necessitates dose reduction or withdrawal.Recurrent episodes of AKI result in impaired renal tubular function and acute renal failure,chronic kidney disease,uremia,and hypertensive nephropathy.The pathophysiology of cisplatin-induced AKI involves proximal tubular injury,apoptosis,oxidative stress,inflammation,and vascular injury in the kidneys.At present,there are no effective drugs or methods for cisplatin-induced kidney injury.Recent in vitro and in vivo studies show that numerous natural products(flavonoids,saponins,alkaloids,polysaccharide,phenylpropanoids,etc.)have specific antioxidant,anti-inflammatory,and anti-apoptotic properties that regulate the pathways associated with cisplatin-induced kidney damage.In this review we describe the molecular mechanisms of cisplatin-induced nephrotoxicity and summarize recent findings in the field of natural products that undermine these mechanisms to protect against cisplatin-induced kidney damage and provide potential strategies for AKI treatment.Chun-yan Fang Da-yong Lou Li-qin Zhou Jin-cheng Wang Bo Yang Qiao-jun He Jia-jia Wang Qin-jie Weng 2021Acta Pharmacologica Sinica2021,42,12:9
6Autologous transplantation with fewer fibers repairs large peripheral nerve defects显示文摘Peripheral nerve injury is a serious disease and its repair is challenging. A cable-style autologous graft is the gold standard for repairing long peripheral nerve defects; however, ensuring that the minimum number of transplanted nerve attains maximum therapeutic effect remains poorly understood. In this study, a rat model of common peroneal nerve defect was established by resecting a 10-mm long right common peroneal nerve. Rats receiving transplantation of the common peroneal nerve in situ were designated as the in situ graft group. Ipsilateral sural nerves(10–30 mm long) were resected to establish the one sural nerve graft group, two sural nerves cable-style nerve graft group and three sural nerves cable-style nerve graft group. Each bundle of the peroneal nerve was 10 mm long. To reduce the barrier effect due to invasion by surrounding tissue and connective-tissue overgrowth between neural stumps, small gap sleeve suture was used in both proximal and distal terminals to allow repair of the injured common peroneal nerve. At three months postoperatively, recovery of nerve function and morphology was observed using osmium tetroxide staining and functional detection. The results showed that the number of regenerated nerve fibers, common peroneal nerve function index, motor nerve conduction velocity, recovery of myodynamia, and wet weight ratios of tibialis anterior muscle were not significantly different among the one sural nerve graft group, two sural nerves cable-style nerve graft group, and three sural nerves cable-style nerve graft group. These data suggest that the repair effect achieved using one sural nerve graft with a lower number of nerve fibers is the same as that achieved using the two sural nerves cable-style nerve graft and three sural nerves cable-style nerve graft. 10Primary total elbow arthroplasty in complex fractures of the distal humerus显示文摘AIM:To evaluate short-to medium term outcome of total elbow arthroplasty(TEA)in complex fractures of the distal humerus.METHODS:A consecutive series of 24 complex distal humerus fractures operated with TEA in the period2006-2012 was evaluated with the Mayo Elbow Performance score(MEPS),plain radiographs,complications and overall satisfaction.The indications for surgery were 1:AO type B3 or C3 or Sheffield type 3 fracture and age above 65 or 2:fracture and severe rheumatoid arthritis.Mean follow-up time was 21 mo.RESULTS:Twenty patients were followed up.Four patients,of which 3 had died,were lost to follow up.According to the AO classification there were 17 C3,1B2 and 2 A2 fractures.Mean follow-up was 21 months(range 4-54).Mean MEPS was 94(range 65-100).Mean flexion was 114 degrees(range 80-140).According to MEPS there were 15 excellent,4 good and 1 fair result.Patient satisfaction:8 excellent,10 good,2 fair and 1poor.There were two revisions due to infection treated successfully with revision and three months of antibiotics.In two patients the locking split had loosened.One was referred to re-insertion and one chose yearly con-trols.Two patients had persistent dysaesthesia of their5th finger,but were able to discriminate between sharp and blunt.CONCLUSION:Our study suggests that TEA in complex fractures of the distal humerus in elderly patients can result in acceptable short-to medium term outcome.Brian Weng S?rensen Stig Brorson Bo Sanderhoff Olsen 2014World Journal of Orthopedics2014,5,3:5
11Therapeutic siRNA:state of the art显示文摘RNA interference(RNAi)is an ancient biological mechanism used to defend against external invasion.It theoretically can silence any disease-related genes in a sequence-specific manner,making small interfering RNA(siRNA)a promising therapeutic modality.After a two-decade journey from its discovery,two approvals of siRNA therapeutics,ONPATTRO®(patisiran)and GIVLAARI™(givosiran),have been achieved by Alnylam Pharmaceuticals.Reviewing the long-term pharmaceutical history of human beings,siRNA therapy currently has set up an extraordinary milestone,as it has already changed and will continue to change the treatment and management of human diseases.It can be administered quarterly,even twice-yearly,to achieve therapeutic effects,which is not the case for small molecules and antibodies.The drug development process was extremely hard,aiming to surmount complex obstacles,such as how to efficiently and safely deliver siRNAs to desired tissues and cells and how to enhance the performance of siRNAs with respect to their activity,stability,specificity and potential off-target effects.In this review,the evolution of siRNA chemical modifications and their biomedical performance are comprehensively reviewed.All clinically explored and commercialized siRNA delivery platforms,including the GalNAc(N-acetylgalactosamine)–siRNA conjugate,and their fundamental design principles are thoroughly discussed.The latest progress in siRNA therapeutic development is also summarized.This review provides a comprehensive view and roadmap for general readers working in the field.Bo Hu Liping Zhong Yuhua Weng Ling Peng Yuanyu Huang Yongxiang Zhao Xing-Jie Liang 2020Signal Transduction and Targeted Therapy2020,5,1:4
12Maxingxiongting mixture attenuates hypoxia pulmonary arterial hypertension to improve right ventricular hypertrophy by inhibiting the rho-kinase signaling pathway显示文摘OBJECTIVE:To explore the mechanism of Maxingxiongting mixture(MXXTM)on pulmonary hypertension in a rat model established by intraperitoneal injection of monocrotaline solution,smoking and forced swimming.METHODS:A total of 30 male Sprague-Dawley rats were randomly divided into five groups:control group,model group,high-dose of MXXTM group(HM),low-dose of MXXTM group(LM),and fasudil group.The mean pulmonary artery pressure(m PAP)was measured by using a miniature catheter.Lung tissue and right ventricular tissue sections were stained with hematoxylin-eosin.The right ventricle(RV)and left ventricle+septum(LV+S)were weighted.RV/(LV+S)was calculated to reflect the degree of right ventricular hypertrophy.Rho/Rho-kinase signaling pathway key proteins(Rho A,ROCKⅠand ROCKⅡ)in rat right ventricular tissue were measured by Western blot analysis.The levels of serum hypoxia-inducible factor-1α(HIF-1α),vascular endothelial growth factor(VEGF)and the levels of plasma renin activity(PRA),angiotensinⅡ(ANG-Ⅱ),aldosterone(ALD)in rat anticoagulated plasma were all measured by enzyme-linked immunosorbent assay.RESULTS:Compared with the control group,the m PAP and RV/(LV+S)in the model group were significantly increased.Administration of fasudil resulted in a significant decrease of m PAP and RV/(LV+S).In the HM group and LM group,m PAP and RV/(LV+S)were significantly lower than the model group.Compared with the control group,the contents of HIF-1α,VEGF,PRA,ANG-Ⅱand ALD in the model group were significantly increased.The administration of fasudil and high-dose MXXTM significantly reduced the contents of HIF-1α,VEGF,PRA,ANG-II and ALD.Compared with the control group,the expression of Rho A,ROCKⅠand ROCKⅡin the right ventricle of the model group were significantly increased.The administration of fasudil and high-dose MXXTM significantly reduced the expression of Rho A and RockⅡproteins.Our results indicated that high-dose of MXXTM had similar effects on reducing pulmonary artery pressure and improving right ventricular remodeling to fasudil.However,MXXTM was unable to restore parameters above to control levels.CONCLUSIONS:MXXTM attenuates hypoxia pulmonary arterial hypertension to improve right ventricular hypertrophy by inhibiting the Rho-kinase signaling pathway.An Xing Li Songtao Weng Xiangwen Wang Xian Wu Hao Zhang Xinyue Gao Jian Yang Renxu Peng Bo 2020Journal of Traditional Chinese Medicine2020,40,6:4

14Semen parameters in men recovered from COVID-19显示文摘The novel coronavirus disease(COVID-19)pandemic is emerging as a global health threat and shows a higher risk for men than women.Thus far,the studies on andrological consequences of COVID-19 are limited.To ascertain the consequences of COVID-19 on sperm parameters after recovery,we recruited 41 reproductive-aged male patients who had recovered from COVID-19,and analyzed their semen parameters and serum sex hormones at a median time of 56 days after hospital discharge.For longitudinal analysis,a second sampling was obtained from 22 of the 41 patients after a median time interval of 29 days from first sampling.Compared with controls who had not suffered from COVID-19,the total sperm count,sperm concentration,and percentages of motile and progressively motile spermatozoa in the patients were significantly lower at first sampling,while sperm vitality and morphology were not affected.The total sperm count,sperm concentration,and number of motile spermatozoa per ejaculate were significantly increased and the percentage of morphologically abnormal sperm was reduced at the second sampling compared with those at first in the 22 patients examined.Though there were higher prolactin and lower progesterone levels in patients at first sampling than those in controls,no significant alterations were detected for any sex hormones examined over time following COVID-19 recovery in the 22 patients.Although it should be interpreted carefully,these findings indicate an adverse but potentially reversible consequence of COVID-19 on sperm quality.Tong-Hang Guo Mei-Ying Sang Shun Bai Hui Ma Yang-Yang Wan Xiao-Hua Jiang Yuan-Wei Zhang Bo Xu Hong Chen Xue-Ying Zheng Si-Hui Luo Xue-Feng Xie Chen-Jia Gong Jian-Ping Weng Qing-Hua Shi 2021Asian Journal of Andrology2021,23,5:2
15Osteoblast behaviors on titania nanotube and mesopore layers显示文摘Titania nanotubes and mesopores with different diameter sizes were prepared by electrochemical oxidation of titanium.The responses of osteoblastic cells isolated from Sprague–Dawley rats to the nanotube and mesopore layers were investigated in sequential events of cell adhesion,morphology,actin cytoskeleton,proliferation,differentiation,and mineralization.Nano-structural features,especially diameters of the nanotubes and mesopores,obviously influenced on cell behaviors in the sequential events.The cells showed better proliferation and differentiation abilities on the specimens with the nanotubes and mesopores than on flat titanium disk.Higher levers of calcium mineralization were observed on the nanotube and mesopore layers.The cells adhered much faster onto the nanotubes with about 170nm diameter and the mesopores with about 400nm diameter than onto flat titanium disk and 50nm nanotubes.There is an appropriate range of the tube/pore sizes,and in this present work,titania nantubes with 170nm diameter is the best for enhancing functions of osteoblasts.Yan Zhang Rong Luo Jing Tan Jianxin Wang Xiong Lu Shuxin Qu Jie Weng Bo Feng 2017Regenerative Biomaterials2017,4,2:2
16Effect of Osteoarthritis Patients' Gender on Rehabilitation after Total Knee Arthroplasty显示文摘Objective To investigate the influence of gender on osteoarthritic knee rehabilitation outcome of osteoarthritis patient undergoing total knee arthroplasty(TKA).Methods We prospectively studied thirty male and thirty female knee primary osteoarthritis patients receiving unilateral TKA with posterior stable-fixed plateau prosthesis at our hospital from March 2003 to March 2008.The age and body mass index of male and female patients were matched.The surgical and rehabilitation clinical factors were compared between two groups.Results There was no significant difference in postoperative hospitalization time,surgical tourniquet time,and wound drainage volume between two groups(P>0.05).The extension/flexion degrees of knee joint before operation,1-week,2-week and 1-year after operation for male patients were 6.0 ± 3.3 degrees/ 114.0 ± 10.0 degrees,2.0 ± 1.6 degrees/93.0 ± 7.4 degrees,0.6 ± 0.6 degrees/104.0 ± 9.9 degrees and 0.3 ± 0.5 degrees/125.0 ± 8.8 degrees,for female patients were 7.0 ± 3.4 degrees/112.0 ± 14.0 degrees,2.0 ± 1.3 degrees/89.0 ± 10.9 degrees,0.9 ± 0.8 degrees/101.0 ± 11.8 degrees,0.4 ± 0.5 degrees/124.0 ± 7.1 degrees.The range of motion before operation,1-week,2-week,and 1-year after operation for male patients was 108.0 ± 9.5 degrees,91.0 ± 7.1 degrees,103.0 ± 9.9 degrees,and 125.0 ± 8.9 degrees,for female patients was 105.0 ± 14.1 degrees,87.0 ± 11.4 degrees,100.0±11.9 degrees,and 124.0 ± 7.0 degrees.The preoperative and 1-year postoperative HSS scores were 55.8 ± 13.3 and 89.6 ± 6.7 for males and 54.5 ± 13.8 and 89.2 ± 4.1 for females.No significant statistical difference was observed between two gender groups in regards to degree of extension and flexion,range of motion and HSS score(P>0.05).Conclusions Gender does not influence postoperative knee function of osteoarthritis patients.TKA significantly improves knee joint function and relieve osteoarthritis-related pain.Jin Lin Bo Yang Xi-sheng Weng Jin Jin Qing Zhao Gui-xing Qiu 2009Chinese Medical Sciences Journal2009,24,2:2

18Ionizable lipid-assisted efficient hepatic delivery of gene editing elements for oncotherapy显示文摘CRISPR/Cas9-based gene editing has emerged as a powerful biotechnological tool,that relies on Cas9 protein and single guided RNA(sgRNA)to edit target DNA.However,the lack of safe and efficient delivery carrier is one of the crucial factors restricting its clinical transformation.Here,we report an ionizable lipid nanoparticle(iLP181,pKa=6.43)based on iLY1809 lipid enabling robust gene editing in vitro and in vivo.The iLP181 effectively encapsulate psgPLK1,the best-performing plasmid expressing for both Cas9 protein and sgRNA targeting Polo-like kinase 1(PLK1).The iLP181/psgPLK1 nanoformulation showed uniformity in size,regular nanostructure and nearly neutral zeta potential at pH 7.4.The nanoformulation effectively triggered editing of PLK1 gene with more than 30%efficiency in HepG2-Luc cells.iLP181/psgPLK1 significantly accumulated in the tumor for more than 5 days after a single intravenous injection.In addition,it also achieved excellent tumor growth suppression compared to other nucleic acid modalities such as siRNA,without inducing adverse effects to the main organs including the liver and kidneys.This study not only provides a clinically-applicable lipid nanocarrier for delivering CRISPR/Cas system(even other bioactive molecules),but also constitutes a potential cancer treatment regimen base on DNA editing of oncogenes.Chunhui Li Tongren Yang Yuhua Weng Mengjie Zhang Deyao Zhao Shuai Guo Bo Hu Wanxuan Shao Xiaoxia Wang Abid Hussain Xing-Jie Liang Yuanyu Huang 2022Bioactive Materials2022,7,3:2
19Sustainable routes for quantitative green selenocyanation of activated alkynes显示文摘Most organic reactions require the usage of volatile organic compounds in the synthesis, work-up and purification processes, thus resulting in major environmental pollution and high manufacturing cost. By using cheap biomass lactic acid as the reaction media and catalyst, a sustainable protocol for the synthesis of Z-3-selenocyanatoacrylates and analogues through green selenocyanation of activated alkynes has been achieved. 20Trace tritium recovery from the residue of liquid Li_(17)Pb_(83) alloy显示文摘The liquid Li17Pb83 alloy is a prominent breeder material for use in a fusion reactor.In the design of an effective tritium extraction system for the liquid lithium lead bubbler of the test blanket module of such a reactor,finding ways to strictly limit the losses of tritium and to minimize radioactive risks is very important.For this purpose,the isotope exchange process has been investigated as a means of trace tritium recovery from a model of the residue from Li17Pb83 alloy.The results indicate that the isotope exchange process is an effective means of tritium recovery from the residue of Li17Pb83 alloy,and the optimum composition of the exchange carrier gas is He + 0.1% D2.The exchange temperature and number of exchange steps are the main factors influencing the efficiency of tritium recovery from the residue.Trace tritium recovery efficiency increases with increasing exchange temperature and number of times of exchange.Tritium recovery efficiency can approach 80% when the residue is treated six times at 823 K.A gas-liquid two-phase contact model to describe the proceeding of tritium release from the liquid Li17Pb83 alloy has been derived on the basis of this experiment.XIE Bo,YANG TongZai & WENG KuiPing Institute of Nuclear Physics and Chemistry,China Academy of Engineering Physics,Mianyang 621900,China 2010Science China Chemistry2010,53,8:2
