1Generation of B Cell-Deficient Pigs by Highly Efficient CRISPR/Cas9-Mediated Gene Targeting显示文摘Generating B cell-deficient mutant is the first step to produce human antibody repertoires in large animal models. In this study, we applied the clustered regularly interspaced short palindromic repeat(CRISPR)/CRISPR-associated(Cas) system to target the JHregion of the pig Ig M heavy chain gene which is crucial for B cell development and differentiation. Transfection of Ig M-targeting Cas9 plasmid in primary porcine fetal fibroblasts(PFFs) enabled inducing gene knock out(KO) in up to 53.3% of colonies analyzed, a quarter of which harbored biallelic modification, which was much higher than that of the traditional homologous recombination(HR). With the aid of somatic cell nuclear transfer(SCNT) technology, three piglets with the biallelic Ig M heavy chain gene mutation were produced. The piglets showed no antibody-producing B cells which indicated that the biallelic mutation of the Ig M heavy chain gene effectively knocked out the function of the Ig M and resulted in a B cell-deficient phenotype. 4Gene editing and its applications in biomedicine显示文摘The steady progress in genome editing, especially genome editing based on the use of clustered regularly interspaced short palindromic repeats(CRISPR) and programmable nucleases to make precise modifications to genetic material, has provided enormous opportunities to advance biomedical research and promote human health. The application of these technologies in basic biomedical research has yielded significant advances in identifying and studying key molecular targets relevant to human diseases and their treatment. The clinical translation of genome editing techniques offers unprecedented biomedical engineering capabilities in the diagnosis, prevention, and treatment of disease or disability. Here, we provide a general summary of emerging biomedical applications of genome editing, including open challenges. 5Blown sand motion within the sand-control system in the southern section of the Taklimakan Desert Highway显示文摘Although scientists have performed many studies in the Taklimakan Desert, few of them have reported the blown sand motion along the southern edge of the Taklimakan Desert Highway, which differs significantly from the northern region in terms of aeolian sand geomorphology and formation environment. Based on the field observation data of airflow and aeolian sand transport, continuous monitoring data of erosional and depositional processes between 14 April 2009 and 9 April 2011 and data of surface sand grains from the classical section along the southern edge of the Taklimakan Desert Highway, this paper reported the blown sand motion within the sand-control system of the highway. The main results are as follows: 1) The existing sand-control system is highly effective in preventing and controlling desertification. Wind velocities within the sand-control system were approximately 33%–100% of those for the same height above the mobile sand surface. Aeolian sand fluxes were approximately 0–31.21% of those of the mobile sand surface. Sand grains inside the system, with a mean diameter of 2.89 φ, were finer than those(2.15 φ) outside the system. In addition, wind velocities basically followed a logarithmic law, but the airflow along the classical section was mainly determined by topography and vegetation. 2) There were obvious erosional and depositional phenomena above the surface within the sand-control system, and these phenomena have very consistent patterns for all observation points in the two observed years. The total thicknesses of erosion and deposition ranged from 0.30 to 14.60 cm, with a mean value of 3.67 cm. In contrast, the deposition thicknesses were 1.90–22.10 cm, with a mean value of 7.59 cm, and the erosion thicknesses were 3.51–15.10 cm, with a mean value of 8.75 cm. 8Effectiveness of Catch-Up Vaccinations after COVID-19 Containment--China,2020显示文摘Introduction:In the early stages of the coronavirus disease 2019(COVID-19)epidemic in China,the highest level of public health emergency response(Level 1 Response)was launched in all province-level administrative divisions(PLADs)across the country.Provision of vaccination services was selectively suspended due to the physical distancing and gathering restrictions required of Level 1 Response.The purpose of this paper is to evaluate the impact of COVID-19 on vaccination services and the effectiveness of selective vaccination service suspension and catch-up vaccinations in China.Methods:Vaccine doses administered,routine vaccination coverage levels,catch-up vaccine doses administered,and coverage levels after the catch-up campaign were determined from individual vaccination records in sampled clinics,standard routine immunization administrative coverage reports,and catch-up vaccination activity reports submitted by PLADs between October 2019 and October 2020.Results:National Immunization Program(NIP)and non-NIP vaccine doses administered in sampled clinics decreased by 80% and 90%,respectively,compared with doses administered before the COVID-19 pandemic.Coverage with the birth dose of hepatitis B vaccine(HepB1)and the Bacillus Calmette-Guérin vaccine(BCG)—two vaccinations recommended to continue throughout the epidemic due to their timecritical nature—were maintained at over 85%,while coverage of other NIP vaccines decreased to below 60%by February 2020.Catch-up vaccination coverage of the 29 PLADs,excluding Xinjiang and Xizang(Tibet),began in April 2020 and exceeded 90%;Hubei catch-up coverage was 95% by October 2020.Conclusions and Implications for Public Health Practice:The COVID-19 pandemic had a great impact on China’s vaccination services.During the epidemic and time of local transmission of the coronavirus,HepB1,BCG,rabies vaccine for postexposure prophylaxis,and tetanus antitoxin(TAT)for wound prophylaxis were maintained at high levels.Of the 69 million vaccine doses postponed during the time of local transmission of the coronavirus,94% were administered in the catch-up campaign.China’s pandemic immunization practices may provide evidence for other countries’immunization programs.Jing Wu Wenzhou Yu Lei Cao Lingsheng Cao Lance Rodewald Jiakai Ye Yifan Song Li Li Xiaoxue Liu Ning Wen Fuzhen Wang Lixin Hao Yixing Li Hui Zheng Keli Li Chao Ma Dan Wu Yanmin Liu Guomin Zhang Zhijie An Huaqing Wang Zundong Yin 2020China CDC weekly2020,2,50:6
9Bone tissue regeneration:The role of finely tuned pore architecture of bioactive scaffolds before clinical translation显示文摘Spatial dimension of pores and interconnection in macroporous scaffolds is of particular importance in facilitating endogenous cell migration and bone tissue ingrowth.However,it is still a challenge to widely tune structure parameters of scaffolds by conventional methods because of inevitable pore geometrical deformation and poor pore interconnectivity.Here,the long-term in vivo biological performances of nonstoichiometric bioceramic scaffolds with different pore dimensions were assessed in critical-size femoral bone defect model.The 6%Mg-substituted wollastonite(CSi-Mg6)powders were prepared via wet-chemical precipitation and the scaffolds elaborately printed by ceramic stereolithography,displaying designed constant pore strut and tailorable pore height(200,320,450,600μm),were investigated thoroughly in the bone regeneration process.Together with detailed structural stability and mechanical properties were collaboratively outlined.BothμCT and histological analyses indicated that bone tissue ingrowth was retarded in 200μm scaffolds in the whole stage(2-16 weeks)but the 320μm scaffolds showed appreciable bone tissue in the center of porous constructs at 6-10 weeks and matured bone tissue were uniformly invaded in the whole pore networks at 16 weeks.Interestingly,the neo-tissue ingrowth was facilitated in the 450μm and 600μm scaffolds after 2 weeks and higher extent of bone regeneration and remodeling at the later stage.These new findings provide critical information on how engineered porous architecture impact bone regeneration in vivo.Simultaneously,this study shows important implications for optimizing the porous scaffolds design by advanced additive manufacture technique to match the clinical translation with high performance.Ronghuan Wu Yifan Li Miaoda Shen Xianyan Yang Lei Zhang Xiurong Ke Guojing Yang Changyou Gao Zhongru Gou Sanzhong Xu 2021Bioactive Materials2021,6,5:5

Specifically, we focus on the nonlinear wave particle interactions and the current understanding of the frequency chirping of rising tone chorus waves. Various other related topics, such as the optimal excitation condition of chorus, the formation of subpackets, and the non-adiabaticity of the nonlinear interaction are also discussed. 11The heterogeneity of islet autoantibodies and the progression of islet failure in type 1 diabetic patients显示文摘Type 1 diabetes mellitus is heterogeneous in many facets. The patients suffered from type 1 diabetes present several levels of islet function as well as variable number and type of islet-specific autoantibodies. This study was to investigate prevalence and heterogeneity of the islet autoantibodies and clinical phenotypes of type 1 diabetes mellitus; and also discussed the process of islet failure and its risk factors in Chinese type 1 diabetic patients. A total of 1,291 type 1 diabetic patients were enrolled in this study. Demographic information was collected. Laboratory tests including mixed-meal tolerance test, human leukocyte antigen alleles, hemoglobin A1 c, lipids, thyroid function and islet autoantibodies were conducted. The frequency of islet-specific autoantibody in newly diagnosed T1 DM patients(duration shorter than half year) was 73% in East China. According to binary logistic regressions, autoantibody positivity, longer duration and lower Body Mass Index were the risk factors of islet failure. As the disease developed, autoantibodies against glutamic acid decarboxylase declined as well as the other two autoantibodies against zinc transporter 8 and islet antigen 2. The decrease of autoantibodies was positively correlated with aggressive beta cell destruction. Autoantibodies can facilitate the identification of classic T1 DM from other subtypes and predict the progression of islet failure. 14Enhanced production of seed oil with improved fatty acid composition by overexpressing NAD^(+)-dependent glycerol-3-phosphate dehydrogenase in soybean显示文摘There is growing interest in expanding the production of soybean oils(mainly triacylglycerol, or TAG) to meet rising feed demand and address global energy concerns. We report that a plastidlocalized glycerol-3-phosphate dehydrogenase(GPDH), encoded by GmGPDHp1 gene, catalyzes the formation of glycerol-3-phosphate(G3 P), an obligate substrate required for TAG biosynthesis.Overexpression of GmGPDHp1 increases soybean seed oil content with high levels of unsaturated fatty acids(FAs), especially oleic acid(C18:1), without detectably affecting growth or seed protein content or seed weight. Based on the lipidomic analyses, we found that the increase in G3 P content led to an elevated diacylglycerol(DAG) pool, in which the Kennedy pathwayderived DAG was mostly increased, followed by PC-derived DAG, thereby promoting the synthesis of TAG containing relatively high proportion of C18:1. The increased G3 P levels induced several transcriptional alterations of genes involved in the glycerolipid pathways. In particular, genes encoding the enzymes responsible for de novo glycerolipid synthesis were largely upregulated in the transgenic lines, in-line with the identified biochemical phenotype. However,their shortcomings such as great release shock, poor safety, unrepeatability and other prominent defects make them unsuitable for new generation spacecraft such as microsatellites to separate at low shock or lock repeatedly, etc. Therefore, it is necessary to develop space non-pyrotechnic low-shock connection and separation devices(SNLD) which are required for advanced aerospace missions. In this paper, the progress of the research on space non-pyrotechnic low-shock connection and separation technology(SNLT) is summarized and reviewed. Proceed from the principle of reducing shock for non-pyrotechnic devices, present studies are classified from the perspective of actuating technology and systematic designing methods. For non-pyrotechnic actuating techniques,according to different driving sources, the separation devices are classified into several main categories: electric, magnetic, gas and thermal actuating devices. The actuation principle and application prospect of separation techniques are introduced and the working process, dimension and mechanical properties of typical devices are compared and evaluated. For the systematic designing method, the common mechanism types of SNLDs are summarized according to the designing concept of reducing shock. Then connection configurations are classified according to the structural forms of connection devices, of which the principles, bearing capacities and general applications are discussed. 17Inhibitive effect on apoptosis in splenic lymphocytes of mice pretreated with Lingzhi (GanodermaLucidum) spores显示文摘OBJECTIVE: To investigate how the pretreatment of mice with Ganoderma spores affected the apoptosis of their splenic lymphocytes induced by dexamethasone after 19 days treatment.METHODS: Sixty Kunming mice were randomly divided into six groups: blank control groupdrenched with normal saline; a drug control group drenched with 150 mg/mL Ganoderma spores; a model group treated with saline; a low dose group with50 mg/mL Ganoderma spores; a moderate dose group with 100 mg/mL Ganoderma spores; and a high dose group with 150 mg/mL Ganoderma spores. The effect of Ganoderma spores on apoptosis in spleen lymphocytes was analyzed. All groups were treated for 19 days. On day 20, the model group and the 3 treatment groups were intraperitoneally injected dexamethasone to induce apoptosis. Splenic index and apoptosis indes were employed to measure cell apoptosis.RESULTS: The results showed that Ganoderma spores reduced the splenic index to different degrees in each group and the best effect was seen in the high dose group(P<0.05).Terminal dexynucleotidyl transferase(TdT)-mediated 2'-Deoxyuridine5'-Triphosphate nick end labeling staining revealed that the apoptotic index in all groups administered Ganoderma spores differed significantly from the model group, and a dose-response was observed.Flow cytometric analysis indicated that spleen lymphocyte apoptosis in the model group was extensive. Each dose of Ganoderma spores inhibited dexamethasone-induced apoptosis in spleen lymphocytes, and a dose-response was observed as well. 18Tailoring the Meso-Structure of Gold Nanoparticles in Keratin-Based Activated Carbon Toward High-Performance Flexible Sensor显示文摘Flexible biosensors with high accuracy and reliable operation in detecting pH and uric acid levels in body fluids are fabricated using well-engineered metaldoped porous carbon as electrode material.The gold nanoparticles@N-doped carbon in situ are prepared using wool keratin as both a novel carbon precursor and a stabilizer.The conducting electrode material is fabricated at 500℃ under customized parameters,which mimics A-B type(two different repeating units) polymeric material and displays excellent deprotonation performance(pH sensitivity).The obtained pH sensor exhibits high pH sensitivity of 57 mV/pH unit and insignificant relative standard deviation of 0.088%.Conversely,the composite carbon material with sp^2 structure prepared at 700℃ is doped with nitrogen and gold nanoparticles,which exhibits good conductivity and electrocatalytic activity for uric acid oxidation.The uric acid sensor has linear response over a range of 1-150 μM and a limit of detection 0.1 μM.These results will provide new avenues where biological material will be the best start,which can be useful to target contradictory applications through molecular engineering at mesoscale.Aniruddha BPatil Zhaohui Meng Ronghui Wu Liyun Ma Zijie Xu Chenyang Shi Wu Qiu Qiang Liu Yifan Zhang Youhui Lin Naibo Lin Xiang Yang Liu 2020Nano-Micro Letters2020,12,9:3

20Label-free visualization of lignin deposition in loquats using complementary stimulated and spontaneous Raman microscopy显示文摘The lignification triggered by biotic or abiotic stresses hardens fruits and vegetables and eventually influences their consumer appeal.Extensive prior efforts have been made to unveil the underlying mechanism of flesh lignification,primarily focused on its physicochemical and molecular biological properties.Nevertheless,most of these studies used destroyed and homogenized bulk tissues as analytes;as a result,potentially valuable spatial information was lost.In this study,the deposition of lignin in loquat flesh during lignification was visualized from the tissue level to the singlecell level by combining the advantages of stimulated Raman scattering(SRS)and spontaneous Raman microscopy using label-free in situ molecular imaging.SRS has the advantages of being fast and providing large-area chemical imaging to reveal the spatial heterogeneity of lignin and cell wall polysaccharide distribution in loquat flesh.After 2 days of storage at 0℃,increased lignins were observed by large-area SRS imaging.In addition,microscopic SRS images of the flesh cells indicated that the increased lignins were trapped in the cell corner(CC)and middle lamella(ML).Furthermore,the compositional and structural features of lignified cells(LCs),CC and ML of loquat flesh were investigated by spontaneous Raman microscopy,and the results showed that the LCs were a combination of lignin,cellulose,and hemicellulose,whereas CC and ML showed only deposited lignin and pectin without cross-linked cellulose and hemicellulose.This result further suggests that the lignins in the CC and ML regions of loquats were later synthesized alone during postharvest storage.This innovative combination of SRS and spontaneous Raman microscopy allows the label-free macroscale and fine chemical imaging of plant cell walls and will enhance our fundamental understanding of the structures and functions of the plant cell wall.Nan Zhu Yifan Yang Minbiao Ji Di Wu Kunsong Chen 2019Horticulture Research2019,6,1:3
