4Monitoring slope deformation using a 3-D laser image scanning system: a case study显示文摘An ILRIS-36D 3-D laser image scanning system was used to monitor the Anjialing strip mine slope on Pingshuo in Shanxi province. The basic working principles, performance indexes, features and data collection and processing methods are illus-trated. The point cloud results are analyzed in detail. The rescale range analysis method was used to analyze the deformation char-acteristics of the slope. The results show that the trend of slope displacement is stable and that the degree of landslide danger is low. This work indicates that 3-D laser image scanning can supply multi-parameter, high precision real time data over long distances. 6Ultrastable and ultrasensitive pH-switchable carbon dots with high quantum yield for water quality identification,glucose detection,and two starch-based solid-state fluorescence materials显示文摘It is attractive and encouraging to develop new fluorescent carbon dots(CDs)with excellent optical properties and promising applications prospects.Herein,highly-efficient green emissive CDs(m-CDs)with a high quantum yield(QY)of 71.7%in water are prepared through a facile solvothermal method.Interestingly,the m-CDs exhibit excellent fluorescence stability in the pH range of 1–9.However,the fluorescence intensity of the m-CDs is almost completely quenched as the pH is increased from 9 to 10.The mechanism of the unique pH-responsive behavior is discussed in detail and a plausible mechanism is proposed.Owing to the unique pH-responsive behavior,the m-CDs are used as a on-off fluorescent probe for water quality identification.By combining the reversible pH-ultrasensitive optical properties of the m-CDs in the pH range of 9–10 with the glucose oxidase-mimicking(GOx-mimicking)ability of Au nanoparticles(AuNPs),glucose can be quantitatively detected.Finally,two environment-friendly starch-based solid-state fluorescence materials(powder and film)are developed through green preparation routes.Daiyong Chao Jinxing Chen Qing Dong Weiwei Wu Desheng Qi Shaojun Dong 2020Nano Research2020,13,11:7

8Thresholds of geographic environmental elements in sediment yield of drainage basins显示文摘Threshold of environmental elements in drainage basin sediment yield refers to, under effect of climate, underlying surface and human activity, a turning point of abrupt changes in drainage sediment yield related to environmental element characteristics. Previous studies on threshold of sediment yield of relevant drainage basins were mainly concentrated on impact of natural zones with a few researches on impact of other environmental elements. Particularly studies on compound environmental element threshold in drainage basin sediment yield remain blank today. Studies indicate that sediment yield in drainage basins is affected by compound interactions and complex actions. 10Patient-derived non-small cell lung cancer xenograft mirrors complex tumor heterogeneity显示文摘Objective:Patient-derived xenograft(PDX)models have shown great promise in preclinical and translational applications,but their consistency with primary tumors in phenotypic,genetic,and pharmacodynamic heterogeneity has not been well-studied.This study aimed to establish a PDX repository for non-small cell lung cancer(NSCLC)and to further elucidate whether it could preserve the heterogeneity within and between tumors in patients.Methods:A total of 75 surgically resected NSCLC specimens were implanted into immunodeficient NOD/SCID mice.Based on the successful establishment of the NSCLC PDX model,we compared the expressions of vimentin,Ki67,EGFR,and PD-L1 proteins between cancer tissues and PDX models using hematoxylin and eosin staining and immunohistochemical staining.In addition,we detected whole gene expression profiling between primary tumors and PDX generations.We also performed whole exome sequencing(WES)analysis in 17 first generation xenografts to further assess whether PDXs retained the patient heterogeneities.Finally,paclitaxel,cisplatin,doxorubicin,atezolizumab,afatininb,and AZD4547 were used to evaluate the responses of PDX models to the standard-of-care agents.Results:A large collection of serially transplantable PDX models for NSCLC were successfully developed.The histology and pathological immunohistochemistry of PDX xenografts were consistent with the patients’tumor samples.WES and RNA-seq further confirmed that PDX accurately replicated the molecular heterogeneities of primary tumors.Similar to clinical patients,PDX models responded differentially to the standard-of-care treatment,including chemo-,targeted-and immuno-therapeutics.Conclusions:Our established PDX models of NSCLC faithfully reproduced the molecular,histopathological,and therapeutic characteristics,as well as the corresponding tumor heterogeneities,which provides a clinically relevant platform for drug screening,biomarker discovery,and translational research.Xuanming Chen Cheng Shen Zhe Wei Rui Zhang Yongsheng Wang Lili Jiang Ke Chen Shuang Qiu Yuanli Zhang Ting Zhang Bin Chen Yanjun Xu Qiyi Feng Jinxing Huang Zhihui Zhong Hongxia Li Guowei Che Kai Xiao 2021Cancer Biology & Medicine2021,18,1:5
11Atomic force microscopic observation on substructure of pollen exine in Cedrus deodara and Metasequoia glyptostroboides显示文摘The substructure of pollen exine in Cedrus deodara (Roxb.) Loud, and Metasequoia glyptostroboides Hu et Cheng has been examined with an atomic force microscope (AFM). The results indicate that the exine substructure units containing sporopollenin in two species are similar in shape, which are granular, but slightly different in size. In Cedrus the substructure unit of pollen exine appears to be 56-99 nm long and 42-74 nm wide, while in Metasequoia it appears to be 81-118 nm long and 43-98 nm wide. It has been observed that the subunits of pollen exine in Cedrus arranged tightly to form short-rod-like or spheroidal pollen exine units, several or more than ten of which formed an island-like structure. There are various spaces among these island-like structures which are interconnected to occupy the entire pollen exine. In Metasequoia, the subunits of pollen exine also arranged tightly with a distribution tendency of cluster of 3-10, however, no obvious boundary exists among these clusters. 12Transient expression of a TaGRF4-TaGIF1 complex stimulates wheat regeneration and improves genome editing显示文摘Genome editing is an unprecedented technological breakthrough but low plant regeneration frequencies and genotype dependence hinder its implementation for crop improvement. Here, we found that transient expression of a complex of the growth regulators TaGRF4 and TaGIF1(TaGRF4-TaGIF1) increased regeneration and genome editing frequency in wheat. When we introduced synonymous mutation in the miR396 target site of TaGRF4, the resulting complex(mTaGRF4-TaGIF1) performed better than original TaGRF4-TaGIF1. Use of m TaGRF4-TaGIF1 together with a cytosine base editor targeting TaALS resulted in 2-9-fold increases in regeneration and transgene-free genome editing in 11 elite common wheat cultivars. 13Optimized pre-treatment of high strength food waste digestate by high content aluminum-nanocluster based magnetic coagulation显示文摘Coagulation-based pre-treatment efficiency of high strength digestate of food waste(HSDFW) anaerobic digestion is negated by organic ligand-catalyzed decomposition of coagulants. In this study, an efficient HSDFW pre-treatment method, magnetic seeds(MS) coagulation, was employed by using highly stable Keggin Al_(30) nanocluster(PAC_(30)), MS and polyacrylamide(PAM), and its operation was optimized by evaluating the performance of removing turbidity, total suspended solids(TSS), chemical oxygen demand(COD), and total phosphorous(TP) phosphate. Results showed that at the optimum dosage of 4.82 g/L, PAC_(30) demonstrated excellent removals as high as 98.93% ± 0.1% of turbidity, 98.04% ± 0.1% of TSS, 58.28% ± 0.3% of total COD, 99.98% ± 0.01% of TP and 99.50% ± 0.01% of dissolved phosphate, respectively. Apparent molecular weight(AMW) and three-dimensional excitation-emission matrix(3 D-EEM) fluorescence spectroscopy analyses demonstrated more efficient removal of dissolved organic matter(DOM), particularly non-biodegradable and hydrophobic components by PAC_(30) than commercial coagulant. The sedimentation was much improved from 40 min by coagulation/flocculation to about 5 min settling by MS coagulation. 15Progress on the ultrasonic testing and laser thermography techniques for NDT of tokamak plasma-facing components显示文摘During manufacturing and operation, different kinds of defects, e.g., delamination or surface cracks, may be generated in the plasma-facing components (PFCs) of a Tokamak device. To ensure the safety of the PFCs, various kinds of nondestructive testing (NDT) techniques are needed for different defect and failure mode. This paper gives a review of the recently developed ultrasonic testing (UT) and laser thermography methods for inspection of the delamination and surface cracks in PFCs. For monoblock W/Cu PFCs of divertor, the bonding quality at both W-Cu and Cu- CuCrZr interfaces was qualified by using UT with a focus probe during manufacturing. A noncontact, coupling-free and flexible ultrasonic scanning testing system with use of an electromagnetic acoustic transducer and a robotic inspection manipulator was introduced then for the in-vessel inspection of delamination defect in first wall (FW). A laser infrared thermography testing method is highlighted for the on-line inspection of delamination defect in FW through the vacuum vessel window of the Tokamak reactor. 20NIR-to-NIR UCL/T_1-weighted MR/CT multimodal imaging by NaYbF_4:Tm@NaGdF_4:Yb-PVP upconversion nanoparticles显示文摘Multimodal imaging nanoprobes are urgently sought because they can integrate different imaging function into individual nanoplatform and provide more comprehensive and accurate information for the diagnosis of early-stage tumor.Lanthanide-based upconversion nanoparticles(UCNPs) are regarded as promising nanoplatforms to fabricate these probes.Herein,we firstly developed the active core-active shell structured NaYbF_4:Tm@NaGdF_4:Yb-PVP UCNPs with the average diameter of 13.23±0.96 nm as multimodal imaging probes.These water-dispersible nanoprobes presented excellent near-infrared to near-infrared(NIR-to-NIR) upconversion luminescence(UCL) performance,which is favorable for optical bioimaging due to deeper tissue penetration and autofluorescence reduction.After coated with the NaGdF_4:Yb active shell,the UCL emission intensity at 800 nm increased by 7.2 times.These nanoprobes exhibited a desirable longitudinal relaxivity(r_1=3.58 L/(mmol s)) and strong X-ray attenuation property(58.84 HU L/g).The cytotoxicity assessment,histology analysis and biodistribution study revealed that NaYbF_4:Tm@NaGdF_4:Yb-PVP UCNPs had relatively low cytotoxicity and negligible organ toxicity.These UCNPs were applied for NIR-to-NIR UCL imaging in vivo.More importantly,the detection of small tumor was successfully achieved under T_1-weighted MRI and CT imaging modalities after intravenous injection of these UCNPs.These results revealed that NaYbF_4:Tm@NaGdF_4:Yb-PVP UCNPs could serve as promising NIR-to-NIR UCL/MRI/CT trimodal imaging probes.Tao Wang Min Yang Jiahui Huang Yanzhi Zhao Honglei Wang Shuang Leng Jinxing Chen Guoying Sun Jianhua Liu 2017Science Bulletin2017,62,13:2
