1Downregulation of serum mi R-17 and mi R-106b levels in gastric cancer and benign gastric diseases显示文摘Altered micro RNA(mi RNA) associated with gastric cancer(GC) development and mi R-17 and mi R-106 b were differentially expressed in GC tissues. This study detected serum levels of mi R-17 and mi R-106 b expression in GC, benign gastric disease(BGD) and healthy controls to assess them as tumor markers for GC. Serum samples from 40 GC, 32 BGD(10 gastric ulcer, 14 gastric polyps, and 8 gastric ulcer with polyps) and 36 healthy individuals were subjected to quantitative reverse transcription polymerase chain reaction(q RT-PCR) analysis of mi R-17 and mi R-106 b expression. The data showed that the serum levels of mi R-17 and mi R-106 b were significantly reduced in healthy individuals and BGD patients compared to GC patients. There was a significant association of mi R-17 and mi R-106 b expression with age, but not with other clinicopathological features, such as gender, tumor differentiation, stage and lymphatic metastasis. Further analysis showed that, in discriminating GC patients from healthy controls, mi R-17 could yield a receiver-operating characteristic(ROC) area under the curve(AUC) of 0.879 with 80.6% sensitivity and 87.5% specificity and mi R-106 b could yield an AUC of 0.856 with 75.0% sensitivity and 92.5% specificity. The combined AUC of mi R-17 and mi R-106 b was 0.913 with 83.3% sensitivity and 87.5% specificity. 2Inhibition of Mcl-1 enhances cell death induced by the Bcl-2-selective inhibitor ABT-199 in acute myeloid leukemia cells显示文摘Acute myeloid leukemia(AML)is a serious disease.The 5-year survival rates remain frustratingly low(65%for children and 26%for adults).Resistance to frontline chemotherapy(usually cytarabine)often develops;therefore a new treatment modality is needed.Bcl-2 family proteins play an important role in balancing cell survival and apoptosis.The antiapoptotic Bcl-2 family proteins have been found to be dysregulated in AML.ABT-199,a BH3 mimetic,was developed to target antiapoptotic protein Bcl-2.Although ABT-199 has demonstrated promising results,resistance occurs.Previous studies in AML show that ABT-199 alone decreases the association of proapoptotic protein Bim with Bcl-2,but this is compensated by increased association of Bim with prosurvival protein Mcl-1,stabilizing Mcl-1,resulting in resistance to ABT-199.In this study,we investigated the antileukemic activity of the Mcl-1-selective inhibitor A-1210477 in combination with ABT-199 in AML cells.We found that A-1210477 synergistically induced apoptosis with ABT-199 in AML cell lines and primary patient samples.The synergistic induction of apoptosis was decreased upon Bak,Bax and Bim knockdown.While A-1210477 treatment alone also increased Mcl-1 protein levels,combination with ABT-199 reduced binding of Bim to Mcl-1.Our results demonstrate that sequestration of Bim by Mcl-1,a mechanism of ABT-199 resistance,can be abrogated by combined treatment with the Mcl-1 inhibitor A-1201477.Daniel A Luedtke Xiaojia Niu Yihang Pan Jianyun Zhao Shuang Liu Holly Edwards Kang Chen Hai Lin Jeffrey W Taub Yubin Ge 2017Signal Transduction and Targeted Therapy2017,2,1:11
3Wireless whispering-gallery-mode sensor for thermal sensing and aerial mapping显示文摘The Internet of Things(IoT)1,2 employs a large number of spatially distributed wireless sensors to monitor physical environments,e.g.,temperature,humidity,and air pressure,and has many applications,including environmental monitoring3,health care monitoring4,smart cities5,and precision agriculture.A wireless sensor can collect,analyze,and transmit measurements of its environment1,2.Currently,wireless sensors used in the IoT are predominately based on electronic devices that may suffer from electromagnetic interference in many circumstances.Being immune to the electromagnetic interference,optical sensors provide a significant advantage in harsh environments6.Furthermore,by introducing optical resonance to enhance light–matter interactions,optical sensors based on resonators exhibit small footprints,extreme sensitivity,and versatile functionalities7,8,which can significantly enhance the capability and flexibility of wireless sensors.Here we provide the first demonstration of a wireless photonic sensor node based on a whisperinggallery-mode(WGM)optical resonator,in which light propagates along the circular rim of such a structure like a sphere,a disk,or a toroid by continuous total internal reflection.The sensor node is controlled via a customized iOS app.Its performance was studied in two practical scenarios:(1)real-time measurement of the air temperature over 12 h and(2)aerial mapping of the temperature distribution using a sensor node mounted on an unmanned drone.Our work demonstrates the capability of WGM optical sensors in practical applications and may pave the way for the large-scale deployment of WGM sensors in the IoT.Xiangyi Xu Weijian Chen Guangming Zhao Yihang Li Chenyang Lu Lan Yang 2018Light(Science & Applications)2018,7,1:8
4Y_(4)GeO_(8):Er^(3+),Yb^(3+) up-conversion phosphors for optical temperature sensor based on FIR technique显示文摘Herein,we reported novel Y_(4)GeO_(8):Er^(3+),Yb^(3+)phosphors elaborated via conventional solid-state reaction.and we further explored their properties as optical thermometer by using fluorescence intensity ratio(FIR)method complemented by detailed analysis on crystal structure,up-conversion luminescence and energy transfer from Yb^(3+)to Er^(3+).Upon 980 nm laser excitation,Y_(4)GeO_(8):Er^(3+),Yb^(3+)phosphors present525,547 and 659 nm emission bands assigned to the characteristic transitions of Er^(3+).Furthermore,Y_(4)GeO_(8):Er^(3+),Yb^(3+)samples show outstanding temperature sensing performances.To be specific,the minimal temperature resolution is 0.03 K(303 K),and the relative sensitivity of FIR can be up to 1.152%/K(303 K).Hence,Y_(4)GeO_(8):Er^(3+),Yb^(3+)phosphors can be possible candidates for thermometry devices.Yihang Chen Jing Chen Ye Tong Wenna Zhang Xiusha Peng Hai Guo Daxing Huang 2021Journal of Rare Earths2021,39,12:5
5Signaling pathways and therapeutic interventions in gastric cancer显示文摘Gastric cancer(GC)ranks fifth in global cancer diagnosis and fourth in cancer-related death.Despite tremendous progress in diagnosis and therapeutic strategies and significant improvements in patient survival,the low malignancy stage is relatively asymptomatic and many GC cases are diagnosed at advanced stages,which leads to unsatisfactory prognosis and high recurrence rates.With the recent advances in genome analysis,biomarkers have been identified that have clinical importance for GC diagnosis,treatment,and prognosis.Modern molecular classifications have uncovered the vital roles that signaling pathways,including EGFR/HER2,p53,PI3K,immune checkpoint pathways,and cell adhesion signaling molecules,play in GC tumorigenesis,progression,metastasis,and therapeutic responsiveness.These biomarkers and molecular classifications open the way for more precise diagnoses and treatments for GC patients.Nevertheless,the relative significance,temporal activation,interaction with GC risk factors,and crosstalk between these signaling pathways in GC are not well understood.Here,we review the regulatory roles of signaling pathways in GC potential biomarkers,and therapeutic targets with an emphasis on recent discoveries.Current therapies,including signaling-based and immunotherapies exploited in the past decade,and the development of treatment for GC,particularly the challenges in developing precision medications,are discussed.These advances provide a direction for the integration of clinical,molecular,and genomic profiles to improve GC diagnosis and treatments.Zi-Ning Lei Qiu-Xu Teng Qin Tian Wei Chen Yuhao Xie Kaiming Wu Qianlin Zeng Leli Zeng Yihang Pan Zhe-Sheng Chen Yulong He 2022Signal Transduction and Targeted Therapy2022,7,11:4
6Acute toxicity study of Aspidopterys obcordata aqueous extract in Sprague-Dawley rats显示文摘OBJECTIVE: To examine the acute toxicity of an aqueous extract of Aspidopterys obcordata(A. obcordata) in Sprague Dawley rats.METHODS: The rats were orally administered a dose of 5000 mg/kg body weight and observed continuously for 6 h and then daily for 14 days. Control rats were administered distilled water. The effect of the extract on general behavior, body weight, and food and water intake were measured.After 14 days, the rats were sacrificed and their organs(liver, heart, spleen, lungs, kidney, adrenal glands, ovaries, and testes) were removed for macroscopic examination. The body and organ weights in addition to hematology(e.g., hemoglobin and white blood cell counts) and clinical blood biochemistry(e.g., albumin and bilirubin) were also examined.RESULTS: There were no deaths recorded, and the rats treated with A. obcordata showed no signs of toxicity. All measured parameters in rats treated with A. obcordata were unaffected when compared with those in control rats. 7Quasi-two-dimensionalβ-Ga2O3 field effect transistors with large drain current density and low contact resistance via controlled formation of interfacial oxygen vacancies显示文摘Quasi-two-dimensi on al(2D)β-Ga2O3 is a rediscovered metal-oxide semiconductor with an ultra-wide bandgap of 4.6-4.9eV.It has been reported to be a promising material for next-generation power and radio frequency electronics.Field effect transistors(FETs)that can switch at high voltage are key compone nts in power and radio frequency devices,and reliable Ohmic con tacts are essential for high FET performance.However,obtaining low contact resistance onβ-Ga2O3 FETs is difficult since reactions betweenβ-Ga2O3 and metal contacts are not fully understood.Herein,we experimentally demonstrate the importanee of reactions at the metal/β-Ga2O3 interface and the corresponding effects of these reactions on FET performance.When Ti is employed as the metal contact,annealing ofβ-Ga2O3 FETs in argon can effectively transform Schottky contacts into Ohmic contacts and permit a large drain current density of-3.1 mA//μm.The contact resistance(Rcontact)between the Ti electrodes andβ-Ga2O3 decreased from^430 to^0.387Ω·mm after annealing.X-ray photoelectron spectroscopy(XPS)confirmed the formation of oxygen vacancies at the Ti/β-Ga2O3 interface after annealing,which is believed to cause the improved FET performance.The results of this study pave the way for greater application ofβ-Ga2O3 in electronics.Zhen Li Yihang Liu Anyi Zhang Qingzhou Liu Chenfei Shen Fanqi Wu Chi Xu Mingrui Chen Hongyu Fu Chongwu Zhou 2019Nano Research2019,12,1:2
8Effect of Colored Sticky Cards on Non-target Insects显示文摘Field experiments to evaluate four different colored sticky cards for trapping non-target insects were conducted in an organic maize field in the Heinigou region of China. Yellow, blue, green, and red sticky cards were used to trap insects in the field. The total number of insects species caught was 54, with 3,862 individuals recorded. Over half of the specimens caught were non-target insects, including phytophagous insects, particularly dipteran species(including many mosquitoes)(50.3%), followed by target pests(37.0%), and beneficial insects(12.7%). Statistical analysis revealed a significant difference in attraction to target pests, non-target pests, and beneficial insects among treatment groups. The results showed that higher numbers of target pests(Myzus persicae Sulzer, Empoasca flavescens Fabricius, Nysius ericaecshinly Schilling) were caught on yellow sticky card traps compared with blue, green, or red sticky card traps, indicating that yellow was the best trap color for target pests, with green and blue being progressively less attractive. For non-target insects, including phytophagous insects, flies, and mosquitoes,higher numbers of were caught on blue sticky card traps compared with yellow,green, or red sticky card traps. Our study indicated that blue was the most attractive color for flies, especially for the housefly, Musca domestica Linnaeus. Our study also showed that most beneficial insects exhibited preferences to particular trap color characteristics: yellow was the most attractive color for parasitic wasps and lady beetles; blue was the most attractive color for hoverflies and honeybees. 9Robust structured total least squares algorithm for passive location显示文摘A new approach called the robust structured total least squares(RSTLS) algorithm is described for solving location inaccuracy caused by outliers in the single-observer passive location. It is built within the weighted structured total least squares(WSTLS)framework and improved based on the robust estimation theory.Moreover, the improved Danish weight function is proposed according to the robust extremal function of the WSTLS, so that the new algorithm can detect outliers based on residuals and reduce the weights of outliers automatically. Finally, the inverse iteration method is discussed to deal with the RSTLS problem. 10Synergistic effects of autophagy/mitophagy inhibitors and magnolol promote apoptosis and antitumor efficacy显示文摘Mitochondria as a signaling platform play crucial roles in deciding cell fate.Many classic anticancer agents are known to trigger cell death through induction of mitochondrial damage.Mitophagy,one selective autophagy,is the key mitochondrial quality control that effectively removes damaged mitochondria.However,the precise roles of mitophagy in tumorigenesis and anticancer agent treatment remain largely unclear.Here,we examined the functional implication of mitophagy in the anticancer properties of magnolol,a natural product isolated from herbal Magnolia officinalis.First,we found that magnolol induces mitochondrial depolarization,causes excessive mitochondrial fragmentation,and increases mitochondrial reactive oxygen species(mtROS).Second,magnolol induces PTEN-induced putative kinase protein 1(PINK1)-Parkin-mediated mitophagy through regulating two positive feedforward amplification loops.Third,magnolol triggers cancer cell death and inhibits neuroblastoma tumor growth via the intrinsic apoptosis pathway.Moreover,magnolol prolongs the survival time of tumor-bearing mice.Finally,inhibition of mitophagy by PINK1/Parkin knockdown or using inhibitors targeting different autophagy/mitophagy stages significantly promotes magnolol-induced cell death and enhances magnolol's anticancer efficacy,both in vitro and in vivo.Altogether,our study demonstrates that magnolol can induce autophagy/mitophagy and apoptosis,whereas blockage of autophagy/mitophagy remarkably enhances the anticancer efficacy of magnolol,suggesting that targeting mitophagy may be a promising strategy to overcome chemoresistance and improve anticancer therapy.Yancheng Tang Liming Wang Tao Yi Jun Xu Jigang Wang Jiang-Jiang Qin Qilei Chen Ka-Man Yip Yihang Pan Peng Hong Yingying Lu Han-Ming Shen Hu-Biao Chen 2021Acta Pharmaceutica Sinica B2021,11,12:2
11Modulating the function of ABCB1: in vitro and in vivo characterization of sitravatinib, a tyrosine kinase inhibitor显示文摘Background:Overexpression of ATP-binding cassette(ABC)transporter is a major contributor to multidrug resistance(MDR),in which cancer cells acquire resistance to a wide spectrum of chemotherapeutic drugs.In this work,we evaluated the sensitizing effect of sitravatinib,a broad-spectrum tyrosine kinase inhibitor(TKI),on ATP-binding cassette subfamily B member 1(ABCB1)-and ATP-binding cassette subfamily C member 10(ABCC10)-mediated MDR.Methods:MTT assay was conducted to examine cytotoxicity and evaluate the sensitizing effect of sitravatinib at non-toxic concentrations.Tritium-labeled paclitaxel transportation,Western blotting,immunofluorescence analysis,and ATPase assay were carried out to elucidate the mechanism of sitravatinib-induced chemosensitization.The in vitro findings were translated into preclinical evaluation with the establishment of xenograft models.Results:Sitravatinib considerably reversed MDR mediated by ABCB1 and partially antagonized ABCC10-mediated MDR.Our in silico docking simulation analysis indicated that sitravatinib strongly and stably bound to the transmembrane domain of ABCB1 human-mouse chimeric model.Furthermore,sitravatinib inhibited hydrolysis of ATP and synchronously decreased the efflux function of ABCB1.Thus,sitravatinib could considerably enhance the intracellular concentration of anticancer drugs.Interestingly,no significant alterations of both expression level and localization of ABCB1 were observed.More importantly,sitravatinib could remarkably restore the antitumor activity of vincristine in ABCB1-mediated xenograft model without observable toxic effect.Conclusions:The findings in this study suggest that the combination of sitrvatinib and substrate antineoplastic drugs of ABCB1 could attenuate the MDR mediated by the overexpression of ABCB1.Yuqi Yang Ning Ji Chao-Yun Cai Jing-Quan Wang Zi-Ning Lei Qiu-Xu Teng Zhuo-Xun Wu Qingbin Cui Yihang Pan Zhe-Sheng Chen 2020Cancer Communications2020,40,7:2

17Gold-vapor-assisted chemical vapor deposition of aligned monolayer WSe2 with large domain size and fast growth rate显示文摘Orientation-controlled growth of two-dimensional(2D)transition metal dichalcogenides(TMDCs)may enable many new electronic and optical applications.However,previous studies reporting aligned growth of WSe2 usually yielded very small domain sizes.Herein,we introduced gold vapor into the chemical vapor deposition(CVD)process as a catalyst to assist the growth of WSe2 and successfully achieved highly aligned monolayer WSe2 triangular flakes grown on c-plane sapphire with large domain sizes(130μm)and fast growth rate(4.3μm·s^−1).When the aligned WSe2 domains merged together,a continuous monolayer WSe2 was formed with good uniformity.After transferring to Si/SiO2 substrates,field effect transistors were fabricated on the continuous monolayer WSe2,and an average mobility of 12 cm^2·V^−1·s−1 was achieved,demonstrating the good quality of the material.This report paves the way to study the effect of catalytic metal vapor in the CVD process of TMDCs and contributes a novel approach to realize the growth of aligned TMDC flakes.Mingrui Chen Anyi Zhang Yihang Liu Dingzhou Cui Zhen Li Yu-Han Chung Sai Praneetha Mutyala Matthew Mecklenburg Xiao Nie Chi Xu Fanqi Wu Qingzhou Liu Chongwu Zhou Mork 2020Nano Research2020,13,10:1
