| 6 | Deep eutectic solvent strategy enables an octahedral Ni–Co precursor for creating high-performance NiCo_(2)O_(4) catalyst toward oxygen evolution reaction显示文摘Deep eutectic solvents(DESs)have gained much attention in the fabrication of advanced nanoelectrocatalysts due to their amazing template function.However,their stabilizing function for easily hydrolyzed inorganic nanomaterials is rarely studied.Here,a DES-mediated strategy was reported to synthesize octahedral Ni–Co precursor(NiCo–NH_(3) complex),which could be directly transformed into NiCo_(2)O_(4) nanooctahedrons after thermal decomposition.The NiCo–NH_(3) precursor in octahedral shape was achieved with the DES-mediated crystallization in the choline chloride(ChCl)/glycerol.The ChCl/glycerol DES not only tailored the morphology of the as-prepared precursor by template effect but also inhibited its hydrolysis,ensuring the successful fabrication of octahedral NiCo–NH_(3) complex precursor with high yield.The NiCo–NH_(3) complex precursor was converted to well-defined NiCo_(2)O_(4) nanooctahedrons,where the calcination temperature and time were explored in detail.It revealed that DES could participate in the conversion process to control the morphology of calcination product.The resultant NiCo_(2)O_(4) nanooctahedrons demonstrated excellent electroactivity and remarkable durability for oxygen evolution reaction(OER).The present strategy not only offers an efficient OER electrocatalyst but also enriches the approaches of DESs in designing advanced nanocatalysts. | Chenyun Zhang Bingwei Xin Tingting Chen Hao Ying Zhonghao Li Jingcheng Hao | 2022 | Green Energy & Environment2022,7,6: | 1 |

| 13 | Detection and analysis of anti-latent membrane protein 2A antibodies in the sera of patients with Epstein-Barr virus associated malignancies显示文摘Background Epstein-Barr virus (EBV) associated malignancies with a Type Ⅱ latency gene expression pattern, such as Hodgkin’s disease, and nasopharyngeal carcinoma (NPC), frequently express the EBV antigen latent membrane protein 2A (LMP2A). We expected to establish a highly expressing LMP2A yeast cell strain and get the high quality LMP2A protein, which was used for detection, analysis and characterization of its antibodies in various patients’ sera of EBV associated malignancies.Methods The plasmid pPICZαA-LMP2A containing the full length of LMP2A cDNA was constructed and transformed to Pichia pastoris GS115 to express LMP2A protein. After fermentation and purification, the LMP2A protein was used as an antigen to detect anti-LMP2A antibodies (Abs) in the sera of patients with EBV-associated malignancies in enzyme linked immunosorbent assay (ELISA) or Western-blot.Results LMP2A was expressed successfully with an expected molecular weight of approximately 54 kD and Abs to LMP2A were strikingly specific to NPC. Two-thirds or more sera from NPC patients were positive for anti-LMP2A immunoglobulin G (IgG) Abs. The antibodies were absent from the sera of other EBV-associated diseases except a small fraction of the gastric carcinoma. Comparing anti-viral capsid Ags (VCA) IgA and LMP2A IgA titers in the sera from 76 NPC patients, only 55% were positive for anti-LMP2A IgA Abs while 70% were positive for anti-VCA IgA. | 16 | Cefotetan-bound human RKIP involves in Ras/Raf1/MEK/ERK signaling pathway显示文摘Cefotetan is widely used to treat bacterial infections in the clinic owing to its broad spectrum of antibacterial activity.In the present study,we demonstrate that cefotetan can bind to the conserved ligand-binding pocket of human Raf1 kinase inhibitory protein(hRKIP),which acts as a negative regulator of the Ras/Raf1/MEK/ERK signaling pathway.The cefotetan-bound hRKIP adopts a rigid structure with insufficient space for binding Raf1 kinase,thereby reliving the inhibitory activity of hRKIP in the Ras/Raf1/MEK/ERK signaling pathway and enhancing the phosphorylation level of ERK.Both NMR titration and molecular docking approaches show that several residues(P74,Y81,W84,P111,P112,K113,S142,G143,D144,W173,P178,Y181 and L184)play crucial roles in hRKIP binding cefotetan.NMR dynamics analysis reveals that the binding of cefotetan with hRKIP promotes ps-ns internal motion but reducesμs-ms conformational exchange for residues in the cefotetan-binding pocket of hRKIP.Our results not only disclose the structural basis of cefotetan upregulating the Ras/Raf1/MEK/ERK signaling pathway but also benefit developing novel drugs against diseases caused by the impaired Ras/Raf1/MEK/ERK pathway. | Chenyun Guo Hao Xu Yu Zhou Zhihua Wu Bin Jiang Hanyu Chen Donghai Lin | 2022 | Acta Biochimica et Biophysica Sinica2022,54,12: | 0 |

| 18 | Biophysical and functional characterizations of recombinant RimI acetyltransferase from Mycobacterium tuberculosis显示文摘Nα-acetylation is a universal protein modification related to a wide range of physiological processes in eukaryotes and prokaryotes. RimI, an Nα-acetyltransferase in Mycobacterium tuberculosis, is responsible for the acetylation of the α-amino group of the N-terminal residue in the ribosomal protein S18. Despite growing evidence that protein acetylation may be correlated with the pathogenesis of tuberculosis, no structural information is yet available for mechanistically understanding the MtRimI acetylation. To enable structural studies for MtRimI, we constructed a serial of recombinant MtRimI proteins and assessed their biochemical properties. We then chose an optimal construct MtRimIC21A4-153 and expressed and purified the truncated high-quality protein for further biophysical and functional characterizations. The 2D 1H-15N heteronuclear single quantum coherence spectrum of MtRimIC21A4-153 exhibits wider chemical shift dispersion and favorable peak isolation, indicating that MtRimIC21A4-153 is amendable for further structural determination. Moreover, bio-layer interferometry experiments showed that MtRimIC21A4-153 possessed similar micromolar affinity to full-length MtRimI for binding the hexapeptide substrate Ala-Arg-Tyr-Phe-Arg-Arg. Enzyme kinetic assays also exhibited that MtRimIC21A4-153 had almost identical enzymatic activity to MtRimI, indicating insignificant influence of the recombinant variations on enzymatic functions. Furthermore, binding sites of the peptide were predicted by molecular docking approach, suggesting that this substrate binds to MtRimI primarily through electrostatic and hydrogen bonding interactions. | 19 | Study on the correlation between bandage bandage and pressure therapy of lymphedema显示文摘To explore the effects of bandage technology and pressure therapy in breast cancer with lymphedema.Methods A total of 91 patients,with breast cancer-related lymphedema and hospitalized in a third-class hospital in Jiangsu Province from May to December,2018,were selected and sequenced in class“ABC”in the order of entry.The patients in class“A”were enrolled in“8”shaped bandaging group.The patients in class“B”were enrolled in“5”spiral bandaging group.The patients in class“C”were enrolled in combined bandage group.The primary outcome was the degree of limb swelling used by multiple-frequency bioelectrical impedance analysis.Secondary outcomes were the amount of bandage losses,counting 24-hour bandage loose occurrences and patients satisfaction.All outcomes were evaluated both before and after treatment.Results After treatment,there was a significant difference in the regression of limb swelling in all three groups(P<0.05).However,the largest amplitude of regression was observed in combined bandage group(limb circumference ratio:53%and tissue water ratio:68%).Although“8”bandaging group had large material,bandage cost and minimum comfort,the bandage has good stability and no case of 24-hour bandage loose occurrences was found in this group.The“5”partial binding group had an advantage in the amount of bandage losses and comfort level,but among which 3 patients had the phenomenon of loose bandage at the joint.The combined bandage group had the best therapeutic effect in limb swelling and patients satisfaction(P<0.05).Conclusion The combined bandage group significantly decreased the degree of limb swelling and 24-hour bandage loose occurrences.Importantly,this treatment improved the degree of patients’satisfaction.The treatment of“8”shaped bandaging combined with spiral bandaging is worthy of clinical promotion. | WANG Bei Li Haihong LOU Qingqing LI Jianing WANG Xiaoyuan XU Chenyun | 2020 | 中西医结合护理(中英文)2020,6,10: | 0 |
| 20 | Recombinant expression, biophysical and functional characterization of ClpS from Mycobacterium tuberculosis显示文摘Intracellular proteolysis is attracting more and more attention for its unique and important character in Mycobacterium tuberculosis(Mt).The ClpS protein from Mt(MtClpS)plays a critical role in intracellular proteolysis by recognizing N-end rule substrates,which makes it become a potential target for antibacterial drugs.However,the molecular mechanism of MtClpS recognizing N-end rule substrates remains unclear.Preparation of highly concentrated and pure MtClpS protein is a prerequisite for further structural and functional studies.In the present work,we tried several fusion tags and various expression conditions to maximize the production of MtClpS in Escherichia coli.We established an efficient approach for preparing the MtClpS protein with a high yield of 24.7 mg/l and a high purity of 98%.After buffer screening,we obtained a stable MtClpS protein sample concentrated at 0.63 mM in the presence of glycerol,l-Arginine,and l-Glutamate.Moreover,circular dichroism characterization indicated that the secondary structure of MtClpS consists of 38%α-helix and 24%β-sheet.The 2D 1H-15N HSQC nuclear magnetic resonance spectrum showed a good dispersion of resonance peaks with uniform intensity,indicating that the purified MtClpS protein was well folded and conformationally homogeneous.Isothermal titration calorimetry experiments revealed significant interactions of MtClpS with N-end rule peptides beginning with Leu,Tyr,Trp,or Phe.Furthermore,residues D34,D35,and H66 were confirmed as key residues for MtClpS recognizing the N-end rule peptide.The successful expression and biophysical characterization of MtClpS enabled us to gain insight into the molecular mechanism of MtClpS recognizing N-end rule substrates.The obtained stable and pure recombinant MtClpS will enable future inhibitor screening experiments. | Chenyun Guo Yihang Xiao Fangkai Bs Weiliang Lin Huilin Wang Hongwei Yao Donghai Lin | 2019 | Acta Biochimica et Biophysica Sinica2019,51,11: | 0 |
