| 1 | OsWRKY03, a rice transcriptional activator that functions in defense signaling pathway upstream of OsNPR1显示文摘WRKY family proteins are a class of plant specific transcription factors that involve in many stress response pathways. It has been shown that one Arabidopsis WRKY protein, AtWRKY29/22, is activated by MAP kinase signaling cascade and confers resistance to both bacterial and fungal pathogens. However, little is known about the biological roles of WRKY proteins in rice. In this study, we investigated the expression patterns of rice AtWRKY29/22 homolog, OsWRKY03, under different conditions, and also its possible role involved in plant defense. Our results showed that OsWRKY03 was up-regulated by several defense signaling molecules or different treatments. Further analysis revealed that the expres- sion of OsWRKY03 was light dependent. Transcriptional activation activity of OsWRKY03 was also demonstrated by yeast functional assay. Transient expression of OsWRKY03-GFP fusion protein in onion epidermis cells showed that OsWRKY03 was a nuclear localized protein. OsNPR1 as well as several other pathogenesis-related genes, such as OsPR1b, phenylalanine ammonia-lyase (ZB8) and peroxidase (POX22.3), were induced in OsWRKY03-overexpressing transgenic plants. | 2 | Pre-collision Granites and Post-collision Intrusive Assemblage of the Kelameili-Harlik Orogenic Belt显示文摘The main types of intrusive rocks in the Kelameili-Harlik Hercynian erogenic belt include calc-alkaline granites, diabase dykes, kaligranites and alkaline granites. Investigation in field geology, petrology, mineralogy and geochemistry shows that the calc-alkaline granites belong to the syntexis-type (or I-type) and were formed in a pre-collisional magmatic arc environment. In consideration of the fact that kaligranites have many features of alkaline granites with higher consolidation temperatures than the calc-alkaline granites and show a discontinuity of minor element and REE evolution in respect to the calc-alkaline granites, they could not have been derived by differentiation of magmas for the calc-alkaline granites, but are likely to have been generated in an environment analogous to that for alkaline granites. The triplet of basic dyke swarms, kaligranites and alkaline granites could be regarded as a prominent indication of the initial stage of post-collisional delamination and extension. | 4 | Reversal of multi-drug resistance by pSUPER-shRNA-mdr1 in vivo and in vitro显示文摘AIM:To explore the possibility of reversing multi-drug resistance(MDR) to HepG2/mdr1 in vitro and in vivo with RNA interference(RNAi) . METHODS:HepG2/mdr1 was obtained by cloning the whole gene mdr1 into HepG2 cells.shRNA targeting sequence was designed to be homologous to the P-gp encoding MDR1 mRNA consensus sequence.pSUPERshRNA/mdr1 was constructed using the enzymedigested technique.HepG2/mdr1 cells were transfected with vectors of pSUPER-shRNA/mdr1 to measure their efficacy by real-time PCR for mdr1 mRNA,flow cytometry(FCM) for P-gp expression,and Rhodamine efflux,MTT method for HepG2/mdr1 function,respectively.In vivo,mice tumors were treated by injecting pSUPER-shRNA/mdr1 in situ and into intraabdominal cavity.Tumors were collected to create cell suspension and cryosections after chemothearpy with adiramycin and mytomycin.The cell suspension was incubated in RPMI-1640 supplemented with G418 to screen stable cells for appreciating the reversal of MDR. Cryosections were treated with immunohistochemistry technique to show the effectiveness of transfection and the expression of P-gp. RESULTS:pSUPER-shRNA/mdr1 was successfullyconstructed,which was confirmed by sequencing. The MDR phenotype of HepG2/mdr1 was decreased significantly in vitro transfection.HepG2/mdr1 showing its MDR was reversed notably in P-gp expression(11.0%vs 98.2%,P<0.01) .Real-time PCR showed that mRNA/mdr1 was lower in test groups than in control groups(18.73±1.33 vs 68.03±2.21,P<0.001) . Compared with HepG2,the sensitivity of HepG2/mdr1 and HepG2/mdr1-dsRNA cells to ADM was decreased by 1.64 times and 15.6 times,respectively. The accumulation of DNR in positive groups was decreased evidently.In vivo,the p-gp expression in positive groups was significantly lower than that in control groups(65.1%vs 94.1%,P<0.05) .The tumor suppressing rate in test groups was 57.8%. After chemotherapy,the growth rate in test groups was lower than that in control groups(700.14±35.61 vs 1659.70±152.54,P<0.05) .Similar results were also observed under fluorescence microscope,and confirmed by Image-Pro Plus 4.5 analysis. | 6 | Analysis of genetic architecture and favorable allele usage of agronomic traits in a large collection of Chinese rice accessions显示文摘Genotyping and phenotyping large natural populations provide opportunities for population genomic analysis and genome-wide association studies(GWAS). Several rice populations have been re-sequenced in the past decade;however, many major Chinese rice cultivars were not included in these studies. Here, we report large-scale genomic and phenotypic datasets for a collection mainly comprised of 1,275 rice accessions of widely planted cultivars and parental hybrid rice lines from China. The population was divided into three indica/Xian and three japonica/Geng phylogenetic subgroups that correlate strongly with their geographic or breeding origins. We acquired a total of 146 phenotypic datasets for 29 agronomic traits under multi-environments for different subpopulations. With GWAS, we identified a total of 143 significant association loci, including three newly identified candidate genes or alleles that control heading date or amylose content. Our genotypic analysis of agronomically important genes in the population revealed that many favorable alleles are underused in elite accessions, suggesting they may be used to provide improvements in future breeding efforts. | 8 | Multiple linear equation of pore structure and coal–oxygen diffusion on low temperature oxidation process of lignite显示文摘This work aimed at studying the feasibility of calculating the coal–oxygen diffusion properties during the low temperature oxidation process of lignite so as to predict its spontaneous combustion process. Coal samples were oxidized in air ambient under different temperatures. Scanning Electron Microscope was used to indicate the surface morphology changes of oxidization. Then, based on fractal theory and flow characteristics, the fractal dimension of gas diffusion in the pore ways was calculated under different temperature. Considering pore size distribution, connectivity distribution and Fick diffusion mechanisms, the relationship between the gas diffusivity change with pore area fractal dimension and porosity was investigated, and multiple linear equation of the coal–oxygen diffusion coefficients and pore parameters was obtained. Comparison between the experimental data and model prediction verifies the validity of the model. | 9 | Experimental study of mode distortion induced by stimulated Raman scattering in high-power fiber amplifiers显示文摘The experimental investigation of mode distortion induced by stimulated Raman scattering(SRS)in a high-power fiber amplifier,which includes the evolutions of optical spectra,spatial beam profiles,and time-frequency characteristics,has been carried out in detail.Temporal-frequency characteristics have been studied for the first time,to the best of our knowledge,by using a low-speed camera and high-speed photodiode traces,which revealed that temporal-frequency characteristics of SRS-induced mode distortion are different from traditional dynamic mode instability(MI).The experimental results show that the output beam profile remains stable before the mode distortion occurs and fluctuates obviously after the onset of SRS-induced MI but on a time scale of seconds,which is much lower than that of Yb-gain-induced MI featuring millisecond-level beam profile fluctuation.It also shows that the mode distortion became measurable in company with the onset of inter-mode four-wave mixing(IM-FWM)when the ratio of Raman light reaches 3%;further,the beam quality factor M2degrades gradually from 1.4 to 2.1 as the ratio of Raman light increases.The mode distortion is accompanied by an obvious temperature increase of the output passive fiber,which further confirms that the mode distortion originates from SRS.The cause of the mode distortion induced by SRS has been explained in the context of core-pumped SRS effect,and the investigation on the accompanying IM-FWM effect indicates that the main content of the SRSinduced high-order mode is the LP21 mode. | QIUHUI CHU QIANG SHU ZENG CHEN FENGYUN LI DONGLIN YAN CHAO GUO HONGHUAN LIN JIANJUN WANG FENG JING CHUANXIANG TANG RUMAO TAO | 2020 | Photonics Research2020,8,4: | 6 |
| 10 | miR-29a up-regulation in AR42J cells contributes to apoptosis via targeting TNFRSF1A gene显示文摘AIM: To investigate the expression of mi R-29 a in rat acute pancreatitis and its functional role in AR42 J cell apoptosis.METHODS: Twelve SD rats were divided into a control group and an acute edematous pancreatitis(AEP) group randomly. AEP was induced by intraperitoneal injection of L-arginine(150 mg/kg) in the AEP group and equal volume of 0.9% Na Cl was injected in the control group. The apoptosis of acinar cells in pancreatic tissue was determined by TUNEL assay. mi RNA chip assay was performed to examine the expression of mi RNAs in two groups. Besides, to further explore the role of mi R-29 a in apoptosis in vitro, recombinant rat TNF-α(50 ng/m L) was administered to treat the rat pancreatic acinar cell line AR42 J for inducing AR42 J cell apoptosis. Quantitative real-time PCR(q RT-PCR) was adopted to measure mi R-29 a expression. Then, mi RNA mimic, mi RNA antisense oligonucleotide(AMO) and control vector were used to transfect AR42 J cells. The expression of mi R-29 a was confirmed by q RT-PCR andthe apoptosis rate of AR42 J cells was detected by flow cytometry analysis. Western blot was used to detect the expression of activated caspase3. Moreover, we used bioinformatics software and luciferase assay to test whether TNFRSF1 A was the target gene of mi R-29 a. After transfection, q RT-PCR and Western blot was used to detect the expression of TNFRSF1 A in AR42 J cells after transfection.RESULTS: The expression of mi R-29 a was much higher in the AEP group compared with the control group as displayed by the mi RNA chip assay. After inducing apoptosis of AR42 J cells in vitro, the expression of mi R-29 a was significantly increased by 1.49 ± 0.04 times in comparison with the control group. As revealed by q RT-PCR assay, the expression of mi R-29 a was 2.68 ± 0.56 times higher in the mi R-29 a mimic group relative to the control vector group, accompanied with an obviously increased acinar cell apoptosis rate(42.83 ± 1.25 vs 24.97 ± 0.15, P < 0.05). Moreover, the expression of mi R-29 a in the mi RNA AMO group was 0.46 ± 0.05 times lower than the control vector group, and the cell apoptosis rate was much lower accordingly(17.27 ± 1.36 vs 24.97 ± 0.15, P < 0.05). The results of bioinformatics software and luciferase assay showed that TNFRSF1 A might be a target gene of mi R-29 a. | 11 | Fruit and vegetable consumption and its relation to risk of asthenopia among Chinese college students显示文摘AIM: To investigate the associations between fruit and vegetable consumption and risk of asthenopia among Chinese college students.METHODS: A total of 1022 students were selected from five universities by a multi-stage stratified cluster sampling method. They were surveyed via a self-administered questionnaire including socio-demographic features,dietary and lifestyle habits, eye-related symptoms, eye care habits and history of diseases. Ascertainment of asthenopia was based on participants' subjectively reported symptoms. The associations between fruit and vegetable intake with asthenopia risk were assessed using multivariate logistic regression analysis.RESULTS: There were no significant associations between total fruit and vegetable, total vegetable, or fruit and the risk of asthenopia. Higher intake of dark-green leafy vegetable was likely to be inversely associated with asthenopia risk [odd ratio(OR): 0.60; 95%CI: 0.37-0.97;P_(trend)=0.21] after controlling for nondietary and dietary risk factors. | 13 | Development of a Novel PmpD-N ELISA for Chlamydia psittaci Infection显示文摘Objective Chlamydia psittaci is an avian respiratory pathogen and zoonotic agent. The wide prevalence of C. psittaci poses a threat to the poultry industry and its employees. However, few commercial kits are available for detecting avian antibodies excluding the in-house ELISA kit. In this study, we developed a novel ELISA kit for detecting antibodies against C. psittaci based on the N-terminal fragment of polymorphic outer membrane protein D(Pmp D-N) as the coating antigen. Methods The antigen concentrations, primary antibody, and cut-off value were determined and optimized. The ELISA, designated Pmp D-N ELISA, was assessed for sensitivity, specificity, and concordance using sera samples from 48 experimentally infected and 168 uninfected SPF chickens. Results The sensitivity and specificity of Pmp D-N ELISA were 97.9%, 100%, respectively, while the concordance was 98.1% as compared to that of MOMP-ELISA. No cross-reaction with positive sera for other avian pathogens was found. Using Pmp D-N ELISA, 799/836 clinical samples were positive, including 93.0% and 98.1% positivity in layers and broilers, respectively. | 17 | Displacement characteristic of landslides reinforced with flexible piles: field and physical model test显示文摘A field monitoring system was established in an active river bank landslide in the Three Gorges area, China, and a consecutive monitoring for about 5 years were conducted to understand the displacement characteristics of flexible piles and the surrounding soil. It was found that piles deformed elastically under reservoir operation, and the soil in front of piles was gradually separated from piles. The movement of the pile heads exceeded that of the soil between and behind piles. This phenomenon was further studied by a large-scale physical model test to gain insights into the pile-soil interaction. The displacement relationship between pile heads and the surrounding soil is in good agreement with the field data. The physical model test shows that the deformation process of pile-reinforced landslides can be divided into two stages: firstly, when the piles head movement exceeds soil movement, the soil arching is mainly affected by the deflection of the piles, the arches between and behind piles bent upwards;but when the soil movement exceeds piles head movement, the arches near the upslope and downslope bent downwards and upwards, respectively. | 20 | TGF-β1-supplemented decellularized annulus fibrosus matrix hydrogels promote annulus fibrosus repair显示文摘Annulus fibrosus(AF)repair remains a challenge because of its limited self-healing ability.Endogenous repair strategies combining scaffolds and growth factors show great promise in AF repair.Although the unique and beneficial characteristics of decellularized extracellular matrix(ECM)in tissue repair have been demonstrated,the poor mechanical property of ECM hydrogels largely hinders their applications in tissue regeneration.In the present study,we combined polyethylene glycol diacrylate(PEGDA)and decellularized annulus fibrosus matrix(DAFM)to develop an injectable,photocurable hydrogel for AF repair.We found that the addition of PEGDA markedly improved the mechanical strength of DAFM hydrogels while maintaining their porous structure.Transforming growth factor-β1(TGF-β1)was further incorporated into PEGDA/DAFM hydrogels,and it could be continuously released from the hydrogel.The in vitro experiments showed that TGF-β1 facilitated the migration of AF cells.Furthermore,PEGDA/DAFM/TGF-β1 hydrogels supported the adhesion,proliferation,and increased ECM production of AF cells.In vivo repair performance of the hydrogels was assessed using a rat AF defect model.The results showed that the implantation of PEGDA/DAFM/TGF-β1 hydrogels effectively sealed the AF defect,prevented nucleus pulposus atrophy,retained disc height,and partially restored the biomechanical properties of disc.In addition,the implanted hydrogel was infiltrated by cells resembling AF cells and well integrated with adjacent AF tissue.In summary,findings from this study indicate that TGF-β1-supplemented DAFM hydrogels hold promise for AF repair. | Qiang Wei Dachuan Liu Genglei Chu Qifan Yu Zhao Liu Jiaying Li Qingchen Meng Weishan Wang Fengxuan Han Bin Li | 2023 | Bioactive Materials2023,,1: | 3 |
