Antifatigue,Antioxidant and Immunoregulatory Effects of Peptides Hydrolyzed from Manchurian Walnut(Juglans mandshurica Maxim.) on Mice显示文摘The Manchurian walnut(Juglans mandshurica Maxim.) is rich in proteins, whereas this resource has not been used efficiently. The antifatigue, antioxidative and immunoregulatory effects of Manchurian walnut hydrolysate peptides(MWHPs)were evaluated in this study. MWHPs with a degree of hydrolysis of 32.23% were ultrafiltered and divided into three fractions, namely, high(> 10 k Da), medium(3–10 kDa), and low molecular weight(< 3 kDa), and then fed to mice continuously at doses of 200, 400 or 800 mg/(kg·d). The antifatigue, antioxidative, and immunoregulatory effects of the peptides were tested on the second and fourth weeks of MWHP administration. Results showed that low-molecular-weight MWHPs exerted significant antifatigue(prolonging swimming time, elevating liver glycogen contents, and reducing lactic acid contents), antioxidative(enhancing superoxide dismutase(SOD), GSH-Px, and catalase(CAT) activities and reducing malondialdehyde(MDA) content), and immunoregulatory(raising the immune-organ index and promoting T-lymphocyte proliferation and s Ig A secretion in the intestinal tract) effects. This research indicates that MWHPs have potential applications in health care and may be developed as a base for new functional foods. | FANG Li REN Dayong CUI Lingyu LIU Chunlei WANG Ji LIU Wei MIN Weihong LIU Jingsheng | 2018 | Grain & Oil Science and Technology2018,1,1: | 6
Analysis of characteristics and predictive factors of immune checkpoint inhibitor-related adverse events显示文摘Objective:We aimed to retrospectively analyze the toxicity profiles and predictors of immune-related adverse events(irAEs)as well as the correlation between irAEs and the clinical efficacy of multi-type immune checkpoint inhibitors(ICIs)in patients with advanced pan-cancer in a real-world setting. Methods:We retrospectively analyzed data from 105 patients with advanced pan-cancer treated with multi-type ICIs at the First Hospital of Jilin University between January 1,2016 and August 1,2020.We used logistic regression analyses to investigate the associations of irAEs with clinical baseline characteristics, blood count parameters, and biochemical indicators during treatment.Receiver operating characteristic curves were used to determine cutoff values for parameters and area under the curve values.Kaplan–Meier and Cox multivariate regression analyses were performed to estimate the relationships of baseline characteristics and irAEs with progression-free survival(PFS)and overall survival(OS).Results:A lower relative lymphocyte count(cutoff=28.5%),higher albumin level(cutoff=39.05 g/L),and higher absolute eosinophil count(AEC)(cutoff=0.175×10^(9)/L)were significantly associated with the occurrence of irAEs,among which a higher AEC(cutoff=0.205×10^(9)/L)was strongly associated with skin-related irAEs[odds ratios(ORs)=0.163,P=0.004].Moreover,a higher lactate dehydrogenase level(cutoff=237.5 U/L)was an independent predictor of irAEs of grade≥3(OR=0.083,P=0.023).In immune cell subgroup analysis,a lower absolute count of CD8+CD28−suppressor T cells(OR=0.806;95%confidence interval:0.643–1.011;P=0.062),which are regulatory T lymphocytes, was associated with the occurrence of irAEs,although the difference was not statistically significant.Furthermore,a higher percentage of CD19+B cells was associated with the occurrence of irAEs of grade≥3(P=0.02)and grade≥2(P=0.051).In addition, patients with any grade of irAE had a significantly high PFS(8.37 vs.3.77 months, hazard ratios(HR)=2.02,P=0.0038)and OS(24.77 vs.13.83 months,HR=1.84;P=0.024).Conclusions:This retrospective study reports clinical profile data for irAEs in unselected patients in a real-world setting and explored some parameters that may be potential predictive markers of the occurrence, type,or grade of irAEs in clinical practice.Evidence of a correlation between safety and efficacy may facilitate a complete assessment of the risk-benefit ratio for patients treated with ICIs. | Rilan Bai Naifei Chen Xiao Chen Lingyu Li Wei Song Wei Li Yuguang Zhao Yongfei Zhang Fujun Han Zheng Lyu Jiuwei Cui | 2021 | Cancer Biology & Medicine2021,18,4: | 4
Plant multiscale networks:charting plant connectivity by multi-level analysis and imaging techniques显示文摘In multicellular and even single-celled organisms, individual components are interconnected at multiscale levels to produce enormously complex biological networks that help these systems maintain homeostasis for development and environmental adaptation.Systems biology studies initially adopted network analysis to explore how relationships between individual components give rise to complex biological processes.Network analysis has been applied to dissect the complex connectivity of mammalian brains across different scales in time and space in The Human Brain Project. In plant science, network analysis has similarly been applied to study the connectivity of plant components at the molecular, subcellular, cellular, organic, and organism levels.Analysis of these multiscale networks contributes to our understanding of how genotype determines phenotype. In this review,we summarized the theoretical framework of plant multiscale networks and introduced studies investigating plant networks by various experimental and computational modalities. We next discussed the currently available analytic methodologies and multi-level imaging techniques used to map multiscale networks in plants. Finally,we highlighted some of the technical challenges and key questions remaining to be addressed in this emerging field. | Xi Zhang Yi Man Xiaohong Zhuang Jinbo Shen Yi Zhang Yaning Cui Meng Yu Jingjing Xing Guangchao Wang Na Lian Zijian Hu Lingyu Ma Weiwei Shen Shunyao Yang Huimin Xu Jiahui Bian Yanping Jing Xiaojuan Li Ruili Li Tonglin Mao Yuling Jiao Sodmergen Haiyun Ren Jinxing Lin | 2021 | Science China(Life Sciences)2021,64,9: | 3
Cholera Caused by a New Clone of Serogroup O1 Vibrio cholerae-Beijing Municipality,China,June 2021显示文摘Several lineages have been identified in the population of serogroup O1 Vibrio cholerae(V.cholerae)(1-3).The strains, which were responsible for the ongoing seventh cholera pandemic, were in Lineage 2.Nearly all the V.cholerae strains in this lineage carried genes coding cholera toxin(ctxAB)(1-2).Lineage 3b consists of strains isolated from different continents and the vast majority of strains in this lineage lack the ctxAB genes. | Hanqiu Yan Bo Pang Xin Lu Zhiyong Gao Pan Lu Xin Zhang Mengyu Wang Lingyu Shen Wenxuan Zhao Jianhong Zhao Weili Liang Lei Jia Haijian Zhou Zhigang Cui Xiaoli Du Biao Kan Quanyi Wang | 2022 | China CDC weekly2022,4,2: | 3
Sliding modes of fault activation under constant normal stiffness conditions显示文摘Fault activation has been the focus of research community for years. However, the studies of fault activation remain immature, such as the fault activation mode and its major factors under constant normal stiffness(CNS)conditions associated with large thickness of fault surrounding rock mass.In this study, the rock friction experiments were conducted to understand the fault activation modes under the CNS conditions. Two major parameters,i.e.the initial normal stress and loading rate, were considered and calibrated in the tests. To reveal the response mechanism of fault activation, the local strains near the fault plane were recorded, and the macroscopic stresses and displacements were analyzed. The testing results show that the effect of displacement-controlled loading rate is more pronounced under the CNS conditions than that under constant normal load(CNL)conditions. Both the normal and shear stresses drop suddenly when the stick-slip occurs. The decrease and increase of the normal stress are synchronous with the shear stress in the regular stick-slip scenario, but mismatch with the shear stress during the chaotic stick-slip process. The results are helpful for understanding the fault sliding mode and the prediction and prevention of fault slip. | Chuanqing Zhang Jie Xu Shengji Jin Guojian Cui Yuhang Guo Lingyu Li | 2023 | Journal of Rock Mechanics and Geotechnical Engineering2023,15,5: | 1
The influence of genetic polymorphisms in drug metabolism enzymes and transporters on the pharmacokinetics of different fluvastatin formulations显示文摘The purpose of the present study was to investigate the impact of genetic polymorphism on fluvastatin pharmacokinetics. In addition,we compared the fluvastatin pharmacokinetics differences between extended-release(ER)80 mg tablet and immediate-release(IR)40 mg capsule in terms of drug metabolism enzyme and transporter genetic polymorphisms. In this open-label,randomized,two-period,two-treatment,crossover study(n=24),effects of ABCG2,SLCO1B1,ABCB1,CYP2C9 and CYP3A5 polymorphisms on the pharmacokinetics of fluvastatin were analyzed. The administration dosage for IR 40 mg and ER 80 mg were twice and once daily, respectively, for total 7 d.Blood samples for pharmacokinetic evaluation were taken on the 1st and 7th d.The lower exposure following ER was observed. For ER tablets,SLCO1B1 T521C genotype correlated with AUC 0-24 of repeat doses(P=0.010).SLCO1B1 T521C genotype had no statistically significant effect on AUC 0-24 of IR capsule of fluvastatin after single or repeated doses. In vitro study demonstrated that when the concentration of fluvastatin was low(<1μmol/l),the uptake of fluvastatin in the HEK293-OATP1B1 with SLCO1B1521TT(K m=0.18μmol/l)was faster than that with SLCO1B1521CC(K m=0.49μmol/l),On the other hand, when concentration reached to higher level(>1μmol/l),transport velocity of fluvastatin by HEK293-OATP1B1 with SLCO1B1521TT(K m=11.4μmol/l)and with SLCO1B1521TCC(K m=15.1μmol/l)tend to be the same.It suggests that the increased effect of SLCO1B1 T521C genotype on ER formulation of fluvastatin was mainly caused by lower blood concentrations. We recommend that formulation should be incorporated into future pharmacogenomics studies. | Qian Xiang Weidang Wu Nan Zhao Chuan Li Junyu Xu Lingyue Ma Xiaodan Zhang Qiufen Xie Zhuo Zhang Jiancheng Wang Weiren Xu Xia Zhao Yimin Cui | 2020 | Asian Journal of Pharmaceutical Sciences2020,15,2: | 1
Flexible, stretchable, and transparent InGaN/GaN multiple quantum wells/polyacrylamide hydrogel-based light emitting diodes显示文摘Visualization is a direct, efficient, and simple interface method to realize the interaction between human and machine, whereas the flexible display unit,as the major bottleneck, still deeply hinders the advances of wearable and virtual reality devices. To obtain flexible optoelectronic devices, one of the effective methods is to transfer a high-efficient and long-lifetime inorganic optoelectronic film from its rigid epitaxial substrate to a foreign flexible/soft substrate.Additionally,piezo-phototronic effect is a fundamental theory for guiding the design of flexible optoelectronic devices.Herein,we demonstrate a flexible, stretchable, and transparent InGaN/GaN multiple quantum wells(MQWs)/polyacrylamide(PAAM)hydrogel-based light emitting diode coupling with the piezo-phototronic effect. The quantum well energy band and integrated luminous intensity(increased by more than 31.3%)are significantly modulated by external mechanical stimuli in the device.Benefiting from the small Young's modulus of hydrogel and weak Van der Waals force, the composite film can endure an extreme tensile condition of about 21.1%stretching with negligible tensile strains transmitted to the InGaN/GaN MQWs.And the stable photoluminescence characteristics can be observed. Moreover, the hydrogen-bond adsorption and excellent transparency of the hydrogel substrate greatly facilitate the packaging and luminescence of the optoelectronic device. And thus, such a novel integration scheme of inorganic semiconductor materials and organic hydrogel materials would help to guide the robust stretchable optoelectronic devices, and show great potential in emerging wearable devices and virtual reality applications. | Jiwei Chen Jiangwen Wang Keyu Ji Bing Jiang Xiao Cui Wei Sha Bingjun Wang Xinhuan Dai Qilin Hua Lingyu Wan Weiguo Hu | 2022 | Nano Research2022,15,6: | 1
Genomic characteristics of SARS-CoV-2 in Beijing,China,2021显示文摘At present, severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)spread worldwide, which has emerged multiple variants and brought a threat to global public health. To analyze the genomic characteristics and variations of SARS-CoV-2 imported into Beijing,we collected the respiratory tract specimens of 112 cases of coronavirus disease 2019(COVID-19)from January to September 2021 in Beijing,China, including 40 local cases and 72 imported cases. The whole-genome sequences of the viruses were sequenced by the next-generation sequencing method.Variant markers and phylogenic features of SARS-CoV-2 were analyzed. Our results showed that in all 112 sequences, the mutations were concentrated in spike protein.D614G was found in all sequences, and mutations including L452R,T478K,P681R/H,and D950N in some cases.Furthermore,112 sequences belonged to 23 lineages by phylogenetic analysis.B.1.1.7(Alpha)and B.1.617.2(Delta)lineages were dominant. Our study drew a variation image of SARS-CoV-2 and could help evaluate the potential risk of COVID-19 for pandemic preparedness and response. | Zhaomin Feng Shujuan Cui Bing Lyu Zhichao Liang Fu Li Lingyu Shen Hui Xu Peng Yang Quanyi Wang Daitao Zhang Yang Pan | 2022 | Biosafety and Health2022,4,4: | 0
Advances in novel molecular typing and precise treatment strategies for small cell lung cancer显示文摘Small cell lung cancer(SCLC)is a high-grade neuroendocrine(NE)cancer characterized by high circulating tumor-cell burden and early extensive metastasis.Considering the complexity of SCLC genes and the immune microenvironment, their unique molecular heterogeneity profiles have been continuously explored. The understanding of SCLC subtypes has recently changed from traditional'classical'and'variant'types to'NE'and'non-NE'phenotypes and to the subtypes defined by major transcriptional regulators, which indicates the gradual revelation of high intratumoral heterogeneity and plasticity characteristics of SCLCs.Advances in genomics as well as the development of single-cell sequencing analysis and new preclinical models have helped investigators gain many new insights into SCLCs and the development of targeted therapy and immunotherapy strategies. This article provides an overview of changes in molecular typing, tumor heterogeneity, and plasticity and that of advances in the precise treatment of different subtypes of SCLC. | Rilan Bai Lingyu Li Xiao Chen Yuguang Zhao Wei Song Huimin Tian Jiuwei Cui | 2021 | Chinese Journal of Cancer Research2021,33,4: | 0
