2Generation of B Cell-Deficient Pigs by Highly Efficient CRISPR/Cas9-Mediated Gene Targeting显示文摘Generating B cell-deficient mutant is the first step to produce human antibody repertoires in large animal models. In this study, we applied the clustered regularly interspaced short palindromic repeat(CRISPR)/CRISPR-associated(Cas) system to target the JHregion of the pig Ig M heavy chain gene which is crucial for B cell development and differentiation. Transfection of Ig M-targeting Cas9 plasmid in primary porcine fetal fibroblasts(PFFs) enabled inducing gene knock out(KO) in up to 53.3% of colonies analyzed, a quarter of which harbored biallelic modification, which was much higher than that of the traditional homologous recombination(HR). With the aid of somatic cell nuclear transfer(SCNT) technology, three piglets with the biallelic Ig M heavy chain gene mutation were produced. The piglets showed no antibody-producing B cells which indicated that the biallelic mutation of the Ig M heavy chain gene effectively knocked out the function of the Ig M and resulted in a B cell-deficient phenotype. 3Artificial Intelligence Based Smart Energy Community Management: A Reinforcement Learning Approach显示文摘This paper presents a smart energy community management approach which is capable of implementing P2P trading and managing household energy storage systems.A smart residential community concept is proposed consisting of domestic users and a local energy pool,in which users are free to trade with the local energy pool and enjoy cheap renewable energy while avoiding the installation of new energy generation equipment.The local energy pool could harvest surplus energy from users and renewable resources,at the same time it sells energy at a higher price than Feed-in-Tariff(FIT)but lower than the retail price.In order to encourage the participation in local energy trading,the electricity price of the energy pool is determined by a real-time demand/supply ratio.Under this pricing mechanism,retail price,users and renewable energy could all affect the electricity price which leads to higher consumers’profits and more optimized utilization of renewable energy.The proposed energy trading process was modeled as a Markov Decision Process(MDP)and a reinforcement learning algorithm was adopted to find the optimal decision in the MDP because of its excellent performance in on-going and model-free tasks.In addition,the fuzzy inference system makes it possible to use Q-learning in continuous state-space problems(Fuzzy Q-learning)considering the infinite possibilities in the energy trading process.To evaluate the performance of the proposed demand side management system,a numerical analysis is conducted in a community comparing the electricity costs before and after using the proposed energy management system.Suyang Zhou Zijian Hu Wei Gu Meng Jiang Xiao-Ping Zhang 2019CSEE Journal of Power and Energy Systems2019,5,1:12
4Insight-HXMT observations of the first binary neutron star merger GW170817显示文摘Finding the electromagnetic(EM) counterpart of binary compact star merger, especially the binary neutron star(BNS) merger,is critically important for gravitational wave(GW) astronomy, cosmology and fundamental physics. On Aug. 17, 2017,Advanced LIGO and Fermi/GBM independently triggered the first BNS merger, GW170817, and its high energy EM counterpart,GRB 170817 A, respectively, resulting in a global observation campaign covering gamma-ray, X-ray, UV, optical, IR, radio as well as neutrinos. The High Energy X-ray telescope(HE) onboard Insight-HXMT(Hard X-ray Modulation Telescope) is the unique high-energy gamma-ray telescope that monitored the entire GW localization area and especially the optical counterpart(SSS17 a/AT2017 gfo) with very large collection area(~1000 cm^2) and microsecond time resolution in 0.2-5 MeV. In addition,Insight-HXMT quickly implemented a Target of Opportunity(ToO) observation to scan the GW localization area for potential X-ray emission from the GW source. Although Insight-HXMT did not detect any significant high energy(0.2-5 MeV) radiation from GW170817, its observation helped to confirm the unexpected weak and soft nature of GRB 170817 A. Meanwhile,Insight-HXMT/HE provides one of the most stringent constraints(~10^(-7) to 10^(-6) erg/cm^2/s) for both GRB170817 A and any other possible precursor or extended emissions in 0.2-5 MeV, which help us to better understand the properties of EM radiation from this BNS merger. 5Relevant experience learning:A deep reinforcement learning method for UAV autonomous motion planning in complex unknown environments显示文摘Unmanned Aerial Vehicles(UAVs)play a vital role in military warfare.In a variety of battlefield mission scenarios,UAVs are required to safely fly to designated locations without human intervention.Therefore,finding a suitable method to solve the UAV Autonomous Motion Planning(AMP)problem can improve the success rate of UAV missions to a certain extent.In recent years,many studies have used Deep Reinforcement Learning(DRL)methods to address the AMP problem and have achieved good results.From the perspective of sampling,this paper designs a sampling method with double-screening,combines it with the Deep Deterministic Policy Gradient(DDPG)algorithm,and proposes the Relevant Experience Learning-DDPG(REL-DDPG)algorithm.The REL-DDPG algorithm uses a Prioritized Experience Replay(PER)mechanism to break the correlation of continuous experiences in the experience pool,finds the experiences most similar to the current state to learn according to the theory in human education,and expands the influence of the learning process on action selection at the current state.All experiments are applied in a complex unknown simulation environment constructed based on the parameters of a real UAV.The training experiments show that REL-DDPG improves the convergence speed and the convergence result compared to the state-of-the-art DDPG algorithm,while the testing experiments show the applicability of the algorithm and investigate the performance under different parameter conditions.Zijian HU Xiaoguang GAO Kaifang WAN Yiwei ZHAI Qianglong WANG 2021Chinese Journal of Aeronautics2021,34,12:10
6DEVELOPMENT OF EXPERIMENTAL METHODS FOR IMPACT TESTING BY COMBINING HOPKINSON PRESSURE BAR WITH OTHER TECHNIQUES显示文摘The split Hopkinson pressure bar(SHPB) technique and the wave propagation inverse analysis(WPIA) technique are both extensively used to experimentally investigate the impact behavior of materials, although neither of them alone provides a fully satisfactory analysis. In the present paper, attention is given to new experimental techniques by incorporating a damagemodified constitutive model into the SHPB technique and combining the Hopkinson pressure bar(HPB) technique with WPIA. First, to distinguish the response due to dynamic constitutive behavior and the response due to dynamic damage evolution, the SHPB method incorporating a damage-modified constitutive model is developed, including an explicit damage-modified Zhu–Wang–Tang model and an implicit damage-modified constitutive model. Second, when the SHPB results become invalid, a method of combining new Lagrange inverse analyses with the HPB technique is developed, including cases of the HPB arranged in front of a long specimen and behind the specimen. 9Experimental and modeling study of NO emission under high CO_2 concentration显示文摘An experimental and numerical study of the NOx formation and reduction process in a designed coal combustion furnace under both traditional air atmosphere and O2/CO2 atmosphere was conducted, in an attempt to explore the chemistry mechanism of the experimentally observed NOx suppression under high CO2 concentration atmospheres. A simplified ‘chemically oriented’ approach, computational fluid dynamics (CFD)-chemical kinetics modeling method, was validated and used to model the experimental process. The high CO2 concentration’s chemical effect on NO reduction has been studied, and the differences in NOx reaction behaviors between O2/CO2 atmosphere and air atmosphere were analyzed by detailed chemical kinetic model. On the basis of investigations through elementary chemical reactions, it can be concluded that high CO2 concentration plays an important role on NOx conversion process during oxy-fuel combustion. Moreover, the dominant reaction steps and the most important reactions for NO conversion under different atmospheres were discussed. 11Pathological matrix stiffness promotes cardiac fibroblast differentiation through the POU2F1 signaling pathway显示文摘Cardiac fibroblast(CF)differentiation into myofibroblasts is a crucial cause of cardiac fibrosis,which increases in the extracellular matrix(ECM)stiffness.The increased stiffness further promotes CF differentiation and fibrosis.However,the molecular mechanism is still unclear.We used bioinformatics analysis to find new candidates that regulate the genes involved in stiffnessinduced CF differentiation,and found that there were binding sites for the POU-domain transcription factor,POU2F1(also known as Oct-1),in the promoters of 50 differentially expressed genes(DEGs)in CFs on the stiffer substrate.Immunofluorescent staining and Western blotting revealed that pathological stiffness upregulated POU2F1 expression and increased CF differentiation on polyacrylamide hydrogel substrates and in mouse myocardial infarction tissue.A chromatin immunoprecipitation assay showed that POU2F1 bound to the promoters of fibrosis repressors IL1R2,CD69,and TGIF2.The expression of these fibrosis repressors was inhibited on pathological substrate stiffness.Knockdown of POU2F1 upregulated these repressors and attenuated CF differentiation on pathological substrate stiffness(35 kPa).Whereas,overexpression of POU2F1 downregulated these repressors and enhanced CF differentiation.In conclusion,pathological stiffness upregulates the transcription factor POU2F1 to promote CF differentiation by inhibiting fibrosis repressors.Our work elucidated the crosstalk between CF differentiation and the ECM and provided a potential target for cardiac fibrosis treatment.Mingzhe Li Jimin Wu Guomin Hu Yao Song Jing Shen Junzhou Xin Zijian Li Wei Liu Erdan Dong Ming Xu Youyi Zhang Han Xiao 2021Science China(Life Sciences)2021,64,2:3
12A learning-based flexible autonomous motion control method for UAV in dynamic unknown environments显示文摘This paper presents a deep reinforcement learning(DRL)-based motion control method to provide unmanned aerial vehicles(UAVs)with additional flexibility while flying across dynamic unknown environments autonomously.This method is applicable in both military and civilian fields such as penetration and rescue.The autonomous motion control problem is addressed through motion planning,action interpretation,trajectory tracking,and vehicle movement within the DRL framework.Novel DRL algorithms are presented by combining two difference-amplifying approaches with traditional DRL methods and are used for solving the motion planning problem.An improved Lyapunov guidance vector field(LGVF)method is used to handle the trajectory-tracking problem and provide guidance control commands for the UAV.In contrast to conventional motion-control approaches,the proposed methods directly map the sensorbased detections and measurements into control signals for the inner loop of the UAV,i.e.,an end-to-end control.The training experiment results show that the novel DRL algorithms provide more than a 20%performance improvement over the state-ofthe-art DRL algorithms.The testing experiment results demonstrate that the controller based on the novel DRL and LGVF,which is only trained once in a static environment,enables the UAV to fly autonomously in various dynamic unknown environments.Thus,the proposed technique provides strong flexibility for the controller.WAN Kaifang LI Bo GAO Xiaoguang HU Zijian YANG Zhipeng 2021Journal of Systems Engineering and Electronics2021,32,6:2
13Overview to the Hard X-ray Modulation Telescope (Insight-HXMT) Satellite显示文摘As China’s first X-ray astronomical satellite, the Hard X-ray Modulation Telescope (HXMT), which was dubbed as Insight-HXMT after the launch on June 15, 2017, is a wide-band(1-250 ke V) slat-collimator-based X-ray astronomy satellite with the capability of all-sky monitoring in 0.2-3 Me V. 15HY5-HDA9 orchestrates the transcription of HsfA2 to modulate salt stress response in Arabidopsis显示文摘Integration of light signaling and diverse abiotic stress responses contribute to plant survival in a changing environment.Some reports have indicated that light signals contribute a plant’s ability to deal with heat,cold,and stress.However,the molecular link between light signaling and the saltresponse pathways remains unclear.We demonstrate here that increasing light intensity elevates the salt stress tolerance of plants.Depletion of HY5,a key component of light signaling,causes Arabidopsis thaliana to become salinity sensitive.Interestingly,the small heat shock protein(sHsp)family genes are upregulated in hy5-215 mutant plants,and HsfA 2 is commonly involved in the regulation of these sH sps.We found that HY5directly binds to the G-box motifs in the HsfA2promoter,with the cooperation of HISTONE DEACETYLASE 9(HDA9),to repress its expression.Furthermore,the accumulation of HDA9 and the interaction between HY5 and HDA9 are significantly enhanced by salt stress.On the contrary,high temperature triggers HY5 and HDA9 degradation,which leads to dissociation of HY5-HDA9from the HsfA2 promoter,thereby reducing salt tolerance.Under salt and heat stress conditions,fine tuning of protein accumulation and an interaction between HY5 and HDA9 regulate HsfA2 expression.This implies that HY5,HDA9,and HsfA2play important roles in the integration of light signaling with salt stress and heat shock response.Jiaheng Yang Xiao Qu Tao Li Yixiang Gao Haonan Du Lanjie Zheng Manchun Ji Paifeng Zhang Yan Zhang Jinxin Hu Liangyu Liu Zefu Lu Zijian Yang Huiyong Zhang Jianping Yang Yongqing Jiao Xu Zheng 2023Journal of Integrative Plant Biology2023,65,1:1
16Combining Residual Attention Mechanisms and Generative Adversarial Networks for Hippocampus Segmentation显示文摘This research discussed a deep learning method based on an improved generative adversarial network to segment the hippocampus.Different convolutional configurations were proposed to capture information obtained by a segmentation network.In addition,a generative adversarial network based on Pixel2Pixel was proposed.The generator was a codec structure combining a residual network and an attention mechanism to capture detailed information.The discriminator used a convolutional neural network to discriminate the segmentation results of the generated model and that of the expert.Through the continuously transmitted losses of the generator and discriminator,the generator reached the optimal state of hippocampus segmentation.T1-weighted magnetic resonance imaging scans and related hippocampus labels of 130 healthy subjects from the Alzheimer’s disease Neuroimaging Initiative dataset were used as training and test data;similarity coefficient,sensitivity,and positive predictive value were used as evaluation indicators.Results showed that the network model could achieve an efficient automatic segmentation of the hippocampus and thus has practical relevance for the correct diagnosis of diseases,such as Alzheimer’s disease.Hongxia Deng Yuefang Zhang Ran Li Chunxiang Hu Zijian Feng Haifang Li 2022Tsinghua Science and Technology2022,27,1:1
17Effect of moxibustion on autophagy and the inflammatory response of synovial cells in rheumatoid arthritis model rats显示文摘OBJECTIVE:To investigate the effect of moxibustion on synovitis and the autophagy of synoviocytes in rheumatoid arthritis(RA).METHODS:Forty Sprague-Dawley rats were randomly divided into a normal group,model group,moxibustion group,cigarette moxibustion group,and medicine group,with eight rats included in each group.The RA model was established by subcutaneous injection of complete Freund's adjuvant into the left posterior toe.Rats in the model group were not interfered with.In the moxibustion group,rats were treated by moxibustion,where a 1-cm diameter moxa stick was applied at the left Zusanli(ST 36)point.The distance of the moxa stick to the skin was 2 cm and moxibustion was completed for 20 min daily for 15 d total.In the cigarette moxibustion group,the moxa stick was replaced by a common cigarette.In the medicine group,rats were treated with a tripterygium glycoside suspension(8 mg/kg)once a day for 15 d total.In each group,the left hind limb toe volume was measured with a toe volume meter;the synovial cells were observed by hematoxylin and eosin staining;the interleukin(IL)-4,IL-6,IL-10,IL-1β,IL-23,IL-17,and tumor necrosis factor(TNF)-αlevels in serum were measured by enzyme-linked immunosorbent assay;the erythrocyte sedimentation rate(ESR)were detected by Westergren sedimentation rate testing;the C-reactive protein(CRP)and rheumatoid factor(RF)levels in serum were detected by rate nephelometry;the expression levels of ULK1,autophagy-associated protein(Atg)3,Atg5,and Atg12 messenger RNA(m RNA)in synovium were detected by real time-quantitative polymerase chain reaction(RT-q PCR);and the protein expression levels of phosphatidylinositol-3-kinase(PI3K),protein kinase B(Akt),mammalian target of rapamycin(mTOR),LC3-Ⅱ,beclin-1,phosphorylated-PI3K(p-PI3K),p-Akt,p-mTOR in synovium were detected by Western blotting.RESULTS:Among the RA model rats,joint swelling,an inflammatory reaction,and the proliferation of synovial tissue were obvious and the signal of the PI3K/Akt/mTOR pathway was active,while autophagy was inhibited.Moxibustion at Zusanli(ST36)or intragastric administration of Tripterygium wilfordii glycosides could alleviate the inflammatory reaction of RA rats;relieve the swelling of the toes;downregulate the levels of ESR,CRF,RF;lower the levels of IL-6,IL-1β,TNF-α,and IL-17;and increase the IL-4 and IL-10.At the same time,the m RNA expression levels of ULK1,Atg3,Atg5,and Atg12 and those of LC3-Ⅱand beclin-1 were increased,while the PI3K,Akt,mTOR,p-PI3K,p-Akt,p-mTOR were decreased.Cigarette moxibustion did not significantly reduce the swelling of the toe joint in RA rats,and was not as good as that of moxibustion or Tripterygium wilfordii polyglycosides in the effects of inflammation relief and the influences of the levels of ESR,CRF,RF.While cigarette moxibustion has a weak effect to affect the expression of corresponding molecules in autophages and the expression level of the autophagy biomaker in synovial tissue.Moxibustion and tripterygium glycosides can significantly reduce the joint swelling,relieve synovitis and synovial hyperplasia,and inhibit the PI3K/Akt/mTOR signaling pathway to increase autophagy in a manner superior to cigarette moxibustion.CONCLUSION:Moxibustion can limit the proliferation of synoviocytes in RA rats by inhibiting the PI3K/Akt/mTOR signaling pathway,promoting autophagy,effectively reducing synovitis,and alleviating joint swelling.HAO Feng WANG Qiang LIU Lei WU Libin CAI Ronglin SANG Jiajia HU Jun WANG Jie YU Qing HE Lu SHEN Yingchao MIAO Yiming HU Ling WU Zijian 2022Journal of Traditional Chinese Medicine2022,42,1:1
18Identification of A-to-I RNA editing profiles and their clinical relevance in lung adenocarcinoma显示文摘Adenosine-to-inosine(A-to-I)RNA editing is a widespread posttranscriptional modification that has been shown to play an important role in tumorigenesis.Here,we evaluated a total of 19,316 RNA editing sites in the tissues of 80 lung adenocarcinoma(LUAD)patients from our Nanjing Lung Cancer Cohort(NJLCC)and 486 LUAD patients from the TCGA database.The global RNA editing level was significantly increased in tumor tissues and was highly heterogeneous across patients.The high RNA editing level in tumors was attributed to both RNA(ADAR1 expression)and DNA alterations(mutation load).Consensus clustering on RNA editing sites revealed a new molecular subtype(EC3)that was associated with the poorest prognosis of LUAD patients.Importantly,the new classification was independent of classic molecular subtypes based on gene expression or DNA methylation.We further proposed a simplified model including eight RNA editing sites to accurately distinguish the EC3 subtype in our patients.The model was further validated in the TCGA dataset and had an area under the curve(AUC)of the receiver operating characteristic curve of 0.93(95%CI:0.91-0.95).In addition,we found that LUAD cell lines with the EC3 subtype were sensitive to four chemotherapy drugs.These findings highlighted the importance of RNA editing events in the tumorigenesis of LUAD and provided insight into the application of RNA editing in the molecular subtyping and clinical treatment of cancer.Cheng Wang Mingtao Huang Congcong Chen Yuancheng Li Na Qin Zijian Ma Jingyi Fan Linnan Gong Hui Zeng Liu Yang Xianfeng Xu Jun Zhou Juncheng Dai Guangfu Jin Zhibin Hu Hongxia Ma Fengwei Tan Hongbing Shen 2022Science China(Life Sciences)2022,65,1:1
19Optimization and Evaluation of Sputtering Barrier/Seed Layer in Through Silicon Via for 3-D Integration显示文摘The barrier/seed layer is a key issue in Through Silicon Via(TSV) technology for 3-D integration.Sputtering is an important deposition method for via metallization in semiconductor process. However,due to the limitation of sputtering and a 'scallop' profile inside vias,poor step coverage of the barrier/seed layer always occurs in the via metallization process. In this paper,the effects of several sputter parameters(DC power,Ar pressure,deposition time,and substrate temperature) on thin film coverage for TSV applications are investigated.Robust TSVs with aspect ratio 5 1 were obtained with optimized magnetron sputter parameters. 20Far-infrared transparent conductors显示文摘The long-standing challenge in designing far-infrared transparent conductors(FIRTC)is the combination of high plasma absorption edge(λ_(p))and high conductivity(σ).These competing requirements are commonly met by tuning carrier concentration or/and effective carrier mass in a metal oxide/oxonate with low optical dielectric constant(ε_(opt)=2-7).However,despite the highσ,the transparent band is limited to mid-infrared(λ_(p)<5μm).In this paper,we break the trade-off between highσandλ_(p)by increasing the“so-called constant”ε_(opt)that has been neglected,and successfully develop the material family of FIRTC withε_(opt)>15 andλ_(p)>15μm.These FIRTC crystals are mainly octahedrally-coordinated heavy-metal chalcogenides and their solid solutions with shallow-level defects.Their highε_(opt)relies on the formation of electron-deficiency multicenter bonds resulting in the great electron-polarization effect.The new FIRTC enables us to develop the first“continuous film”type far-infrared electromagnetic shielder that is unattainable using traditional materials.Therefore,this study may inaugurate a new era in far-infrared optoelectronics.Chaoquan Hu Zijian Zhou Xiaoyu Zhang Kaiyu Guo Can Cui Yuankai Li Zhiqing Gu Wei Zhang Liang Shen Jiaqi Zhu 2023Light(Science & Applications)2023,12,6:1
