| 1 | Comparison of alternative extraction methods for secretome profiling in human hepatocellular carcinoma cells显示文摘Secreted proteins are important sources for early detection and diagnosis of disease, and as such have received considerable attention. The extraction of low concentration proteins from large volumes of culture media, which are rich in salts and other compounds that interfere with most proteomics techniques, presents a problem for secretome studies. Ultrafiltration, precipitation, and dialysis are three major extraction methods that can be used to overcome this problem. The present study for the first time, compared the merits and shortcomings of these three methods, without bias. Centrifugal ultrafiltration provided the best extraction efficiency, and precipitation provided the highest number of identifiable proteins. The three methods yielded closely related, but different, information on the secretome; thus, they should be considered complementary or, at least, supplementary methods. Three hundred and sixty unique proteins were identified, including 211 potential secreted proteins. Compared with previous studies, this study also identified 42 new secreted proteins. | 2 | Gene Cloning and Expression of the Pyrroline-5-carboxylate Reductase Gene of Perennial Ryegrass(Lolium perenne)显示文摘Salt and drought limit the range of applications of perennial ryegrass(Lolium perenne L.), which is one of the important turf and forage grasses. Previous studies have suggested that pyrroline-5-carboxylate reductase(P5CR) might play a central role in proline accumulation in plants that are responsive to stresses. In the present study, the Lolium perenne L. pyrroline-5-carboxylate reductase(LpP 5CR) gene was cloned from leaves of the cultivar ‘Derby' using the RACE technique. The full-length LpP 5CR gene was 1 047 bp in length, which comprised an open reading frame(ORF) of 840 bp in size. Sequence alignment revealed that the putative Lp P5 CR had a 94.3% similarity to Ta P5 CR. q RT-PCR displayed that the mR NA levels of the LpP 5CR gene were almost the same as that in the roots, stems, and leaves of perennial ryegrass seedlings subjected to normal condition or NaC l treatment for 1 h. Moreover, the transcription level of LpP 5CR was up- or down- regulated with Na Cl, polyethylene glycol(PEG), cold, or abscisic acid(ABA) treatment for 3 to 48 h. In addition, confocal microscopy localized the GFP-Lp P5 CR fusion protein to the cytoplasm of onion epidermal cells. | 3 | Ultra-robust stretchable electrode for e-skin:In situ assembly using a nanofiber scaffold and liquid metal to mimic water-to-net interaction显示文摘The development of stretchable electronics could enhance novel interface structures to solve the stretchability-conductivity dilemma,which remains a major challenge.Herein,we report a nano-liquid metal(LM)-based highly robust stretchable electrode(NHSE)with a self-adaptable interface that mimics water-tonet interaction.Based on the in situ assembly of electrospun elastic nanofiber scaffolds and electrosprayed LM nanoparticles,the NHSE exhibits an extremely low sheet resistance of 52 mΩsq^(-1).It is not only insensitive to a large degree of mechanical stretching(i.e.,350%electrical resistance change upon 570%elongation)but also immune to cyclic deformation(i.e.,5%electrical resistance increases after 330000 stretching cycles with 100%elongation).These key properties are far superior to those of the state-of-the-art reports.Its robustness and stability are verified under diverse circumstances,including long-term exposure to air(420 days),cyclic submersion(30000 times),and resilience against mechanical damages.The combination of conductivity,stretchability,and durability makes the NHSE a promising conductor/electrode solution for flexible/stretchable electronics for applications such as wearable on-body physiological signal detection,human-machine interaction,and heating e-skin. | Jinwei Cao Fei Liang Huayang Li Xin Li Youjun Fan Chao Hu Jing Yu Jin Xu Yiming Yin Fali Li Dan Xu Hanfang Feng Huali Yang Yiwei Liu Xiaodong Chen Guang Zhu Run-Wei Li | 2022 | InfoMat2022,4,4: | 1 |

| 9 | Preparation of nZVI embedded modified mesoporous carbon for catalytic persulfate to degradation of reactive black 5显示文摘High-efficiency and cost-effective catalysts with available strategies for persulfate(PS)activation are critical for the complete mineralization of organic contaminants in the environmental remediation and protection fields.A nanoscale zero-valent iron-embedded modified mesoporous carbon(MCNZVI)with a core-shell structure is synthesized using the hydrothermal synthesis method and high-temperature pyrolysis.The results showed that nZVI could be impregnated within mesoporous carbon frameworks with a comparatively high graphitization degree,rich nitrogen doping content,and a large surface area and pore volume.This material was used as a persulfate activator for the oxidation removal of Reactive Black 5(RB5).The effects of the material dosage,PS concentration,pH,and some inorganic anions(i.e.,Cl^(−),SO_(4)^(2−))on RB5 degradation were then investigated.The highest degradation efficiency(97.3%)of RB5 was achieved via PS(20 mmol/L)activation by the MCNZVI(0.5 g/L).The pseudo-first-order kinetics(k=2.11×10^(−2)min^(−1))in the MCNZVI/PS(0.5 g/L,20 mmol/L)was greater than 100 times than that in the MCNZVI and PS.The reactive oxygen species(ROS),including^(1)O_(2),SO_(4)^(·−),HO^(.),and·O_(2)^(−),were generated by PS activation with the MCNZVI.Singlet oxygen was demonstrated to be the primary ROS responsible for the RB5 degradation.The MCNZVI could be reused and regenerated for recycling.This work provides new insights into PS activation to remove organic contamination. | Zhifei Ma Huali Cao Fengchun Lv Yu Yang Chen Chen Tianxue Yang Haixin Zheng Daishe Wu | 2021 | Frontiers of Environmental Science & Engineering2021,15,5: | 0 |
