1Comparison of alternative extraction methods for secretome profiling in human hepatocellular carcinoma cells显示文摘Secreted proteins are important sources for early detection and diagnosis of disease, and as such have received considerable attention. The extraction of low concentration proteins from large volumes of culture media, which are rich in salts and other compounds that interfere with most proteomics techniques, presents a problem for secretome studies. Ultrafiltration, precipitation, and dialysis are three major extraction methods that can be used to overcome this problem. The present study for the first time, compared the merits and shortcomings of these three methods, without bias. Centrifugal ultrafiltration provided the best extraction efficiency, and precipitation provided the highest number of identifiable proteins. The three methods yielded closely related, but different, information on the secretome; thus, they should be considered complementary or, at least, supplementary methods. Three hundred and sixty unique proteins were identified, including 211 potential secreted proteins. Compared with previous studies, this study also identified 42 new secreted proteins. 12The impact of body weight trajectory from childhood on chronic inflammation in adulthood: The Bogalusa Heart Study显示文摘Importance:The impact of long-term burden of excessive body weight,beginning in childhood,on inflammatory status in adulthood has been poorly described.Objective:To characterize the longitudinal body mass index(BMI)trajectory from childhood and examine its relationship with inflammatory status in adulthood.Methods:We included 1285 adults who had 4-15 repeat measurements of BMI from childhood to adulthood.The area under the curve(AUC)of growth curves was calculated to characterize long-term burden(total AUC)and trends(incremental AUC)of BMI.Results:After adjusting for covariates,higher values of BMI in terms of childhood and adulthood,as well as total and incremental AUC,were strongly associated with elevated levels of adult C-reactive protein(CRP)in the four race-sex groups.There were significant differences in linear and nonlinear curve parameters between the normal and high CRP groups for all race-sex groups(P<0.01).Compared with participants who had consistently low BMI in both childhood and adulthood,participants with high BMI in adulthood had higher CRP levels(P<0.001),irrespective of their childhood BMI status;participants with high BMI in childhood but low BMI in adulthood had similar adult CRP levels.Interpretation:The impact of excessive body weight on inflammation is cumulative and exacerbated over time.The influence of childhood overweight/obesity on inflammatory status in adulthood can be alleviated by reducing adiposity in adulthood.Yinkun Yan Shengxu Li Yang Liu Lydia Bazzano Jiang He Jie Mi Wei Chen 2021Pediatric Investigation2021,5,1:0
13Characteristics of pediatric inpatients with primary and secondary hypertension显示文摘Importance:Extensive population-based studies have explored the prevalence of primary hypertension(HTN)in children and adolescents.However,there is little published data on the characteristics of different types of pediatric HTN and the causes of secondary HTN.Objective:To investigate the characteristics of different types of pediatric HTN and the causes of secondary HTN in a hospital setting.Methods:The study cohort comprised pediatric inpatients(<18 years of age)discharged with a diagnosis of HTN from Beijing Children's Hospital during 2015-2020.Pediatric patients with HTN were allocated to secondary and primary HTN groups on the basis of comprehensive analyses of their diagnoses,family history of HTN,and findings on physical examination,as documented in their medical records.The Mann-Whitney U test,χ^(2) and Fisher's exact test were used to assess differences in characteristics of patients with different HTN types and causes of secondary HTN.Results:Data of 1470 inpatients with HTN from 18 clinical departments were included in the analysis.Among them,458(31.2%)had primary HTN,and 1012(68.8%)had secondary HTN.Compared with patients had primary HTN,children with secondary HTN were younger and had lower body mass indexes and longer lengths of stay.Moreover,children with primary HTN had mostly been managed by the Endocrinology and Cardiology Departments,75.8%of them having obesity-related comorbidities.In contrast,most patients with secondary HTN had been managed by the Nephrology Department,renal diseases being the leading cause of their HTN(46.3%).Interpretation:Secondary HTN is more common than primary HTN in pediatric clinical settings,renal diseases being the leading cause of secondary HTN.Yijun Chen Peiyu Ye Xiaorong Liu Chunxiu Gong Caifeng Li Yue Yuan Huyong Zheng Xin Xu Hongbo Dong Qin Kong Yinkun Yan Jie Mi 2021Pediatric Investigation2021,5,1:0
14Reliability and consistency assessment of land cover products atmacro and local scales in typical cities显示文摘Urban areas have higher heterogeneity compared to natural areas,it is crucial to assessfine-resolution land cover products and discover how they differ in urban areas so that they can be efficiently used for various application scenarios.In this study,five typical cities in China were chosen as study areas to evaluate four commonly used 30 m land cover products:GLC_FCS30-2020,FROM-GLC30-2017,Globeland30-2020,and CLCD-2019.We analyzed the reliability of these four products using validation samples as well as by examining their area and spatial pattern consistency.Given the limitations of traditional accuracy assessments at the macro level,we added a local area evaluation to further examine the classification details in these products.The macro results indicated that four land cover products within urban areas have a similar overall accuracy,surpassing 76%,but there was a low consistency among them,ranging from 42.21%to 61.13%.The local accuracy assessment illustrated that GLC_FCS30-2020 and FROM-GLC30-2017 performed well in reflecting the intricate details of the city,however,the four products exhibited varying degrees of misclassifications and omissions.These phenomena suggest that more sophisticated algorithms are needed to consider urban particularities sincefine-resolution land cover products may fail to capture complex urban details.Wenxi Shi Xiang Zhao Jiacheng Zhao Siqing Zhao Yinkun Guo Naijing Liu Na Sun Xiaozheng Du Mengmeng Sun 2023International Journal of Digital Earth2023,16,1:0
15Effects of galectin-3 inhibition on endometrial cell cycle and adhesion显示文摘Galectin-3(gal-3)and its ligands have been implicated in cell transformation and cancer metastasis.Gal-3 protein has been found in uterine epithelial cells adjacent to implanting blastocysts in different cell types.In order to investigate the role of gal-3 in the establishment of human endometrial receptivity,the expression of gal-3 in human endometrial cell line RL95-2 was silenced by RNA inter-ference technology using gal-3 specific small RNA.The expression of gal-3 was detected by the reverse transcriptase-polymerase chain reaction and Western blot analysis.After the suppression of gal-3,cell cycle changes and the expres-sion of integrin b1 were detected by flow cytometry.The adhesive ability of RL95-2 cells was analyzed by the adhesion test.Gal-3 siRNA transfection efficiency reached 70%–90%.The expression of gal-3 mRNA and protein in RL95-2 cells was strongly inhibited by 70%–90%after RNA interference.Inhibition of gal-3 expression decreased S-phase but increased G1 phase cells.Integrin b1 expression was down-regulated,and the adhesive ability of RL95-2 cells to fibronectin(FN)was significantly reduced.Gal-3 may be involved in the establishment of endometrial receptivity by regulating the proliferation and adhesion of endometrial cells.The influence on adhesion may be related with the integrin modulation.LEI Caixia ZHANG Wei SUN Xiaowei DU Guoping WANG Li LIU Yinkun 2007Frontiers of Medicine2007,1,4:0
