1Inhibition of pacemaker activity in interstitial cells of Cajal by LPS via NF-κB and MAP kinase显示文摘AIM:To investigate lipopolysaccharide(LPS) related signal transduction in interstitial cells of Cajal(ICCs) from mouse small intestine.METHODS:For this study,primary culture of ICCs was prepared from the small intestine of the mouse.LPS was treated to the cells prior to measurement of the membrane currents by using whole-cell patch clamp technique.Immunocytochemistry was used to examine the expression of the proteins in ICCs.RESULTS:LPS suppressed the pacemaker currents of ICCs and this could be blocked by AH6809,a prostaglandin E2-EP2 receptor antagonist or NG-Nitro-Larginine Methyl Ester,an inhibitor of nitric oxide(NO) synthase.Toll-like receptor 4,inducible NO synthase or cyclooxygenase-2 immunoreactivity by specific antibodies was detected on ICCs.Catalase(antioxidant agent) had no action on LPS-induced action in ICCs.LPS actions were blocked by nuclear factor kB(NF-kB) inhibitor,actinomycin D(a gene transcription inhibitor),PD 98059(a p42/44 mitogen-activated protein kinases inhibitor) or SB 203580 [a p38 mitogen-activated protein kinases(MAPK) inhibitor].SB 203580 also blocked the prostaglandin E2-induced action on pacemaker currents in ICCs but not NO.CONCLUSION:LPS inhibit the pacemaker currents in ICCs via prostaglandin E2-and NO-dependent mechanism through toll-like receptor 4 and suggest that MAPK and NF-kB are implicated in these actions.Dong Chuan Zuo Seok Choi Pawan Kumar Shahi Man Yoo Kim Chan Guk Park Young Dae Kim Jun Lee In Yeoup Chang Insuk So Jae Yeoul Jun 2013World Journal of Gastroenterology2013,19,8:9
2Anti-parkinsonian effects of octacosanol in 1-methyl-4-phenyl-1,2,3,6 tetrahydropyridine-treated mice显示文摘Our previous research showed that octacosanol exerted its protective effects in 6-hydroxydopamine-induced Parkinsonian rats. The goal of this study was to investigate whether octacosanol would attenuate neurotoxicity in 1-methyl-4-phenyl-1,2,3,6 tetrahydropyridine (MPTP)-treated C57BL/6N mice and its potential mechanism. Behavioral tests, tyrosine hydroxylase immunohistochemistry and western blot were used to investigate the effects of octacosanol in a mouse model of Parkinson's disease. Oral administration of octacosanol (100 mg/kg) significantly improved behavioral impairments in mice treated by MPTP and markedly ameliorated morphological appearances of tyrosine hydroxylase-positive neuronal cells in the substantia nigra. Furthermore, octacosanol blocked MPTP-induced phosphorylation of p38MAPK and JNK, but not ERK1/2. These findings implicated that the protective effects afforded by octacosanol might be mediated by blocking the phosphorylation of p38MAPK and JNK on the signal transduction in vivo. 7The role of herbivores in the grassland carbon budget for Three-Rivers Headwaters region,Qinghai-Tibetan Plateau,China显示文摘Background:An accurate assessment of the carbon budget is a crucial part of projecting future climate change and its impact on ecosystems.Grasslands foster multiple ecological functions including support for wild animals and livestocks.Herbivores intake forage biomass carbon,then digest and metabolize,and finally retain some carbon.The carbon processes have not been well quantified,resulting in uncertainties in the estimation of regional carbon budgets for grassland ecosystems.Methods:An animal metabolic carbon flux model was developed for herbivores in the Three-Rivers Headwaters region of China.The forage intake and metabolic carbon rates were estimated through metabolic body weight and daily digested measures for the main herbivore species.Results:The carbon intake was 5.52 Tg C year−1(45%)from partial aboveground biomass(12.2 Tg C year−1),in which 39.31%was released into the atmosphere by respiration CO_(2),43.77%was returned to the ecosystem as feces and urine,and 16.96%was retained in herbivores for population regeneration or for human well-being.Conclusions:This study,as the first research on this topic,quantified the carbon flux of herbivores and found livestock accounts for a major part of consumed carbon on grasslands,which is important for understanding regional carbon budgets to mitigate and adapt to climate change over grasslands worldwide.Junbang Wang Xinquan Zhao Xihuang Ouyang Liang Zhao Wenying Wang Chan Zuo Zhenhua Zhang Huakun Zhou Alan Watson Yingnian Li 2022Grassland Research2022,1,3:1
