| 1 | Overexpression of Sirt6 ameliorates sleep deprivation induced-cognitive impairment by modulating glutamatergic neuron function显示文摘Sleep benefits the restoration of energy metabolism and thereby suppo rts neuronal plasticity and cognitive behaviors.Sirt6 is a NAD+-dependent protein deacetylase that has been recognized as an essential regulator of energy metabolism because it modulates various transcriptional regulators and metabolic enzymes.The aim of this study was to investigate the influence of Sirt6 on cerebral function after chronic sleep deprivation(CSD).We assigned C57BL/6J mice to control or two CSD groups and subjected them to AAV2/9-CMV-EGFP or AAV2/9-CMV-Sirt6-EGFP infection in the prelimbic cortex(PrL).We then assessed cerebral functional connectivity(FC) using resting-state functional MRI,neuron/astrocyte metabolism using a metabolic kinetics analysis;dendritic spine densities using sparse-labeling;and miniature excitato ry postsynaptic currents(mEPSCs) and action potential(AP) firing rates using whole-cell patchclamp recordings.In addition,we evaluated cognition via a comprehensive set of behavioral tests.Compared with controls,Sirt6 was significantly decreased(P<0.05) in the PrL after CSD,accompanied by cognitive deficits and decreased FC between the PrL and accumbens nucleus,piriform cortex,motor co rtex,somatosensory co rtex,olfactory tubercle,insular cortex,and cerebellum.Sirt6 ove rexpression reve rsed CSD-induced cognitive impairment and reduced FC.Our analysis of metabolic kinetics using [1-13C] glucose and [2-13C] acetate showed that CSD reduced neuronal Glu4and GABA2synthesis,which could be fully restored via forced Sirt6 expression.Furthermore,Sirt6 ove rexpression reversed CSD-induced decreases in AP firing rates as well as the frequency and amplitude of mEPSCs in PrL pyramidal neurons.These data indicate that Sirt6 can improve cognitive impairment after CSD by regulating the PrL-associated FC network,neuronal glucose metabolism,and glutamatergic neurotransmission.Thus,Sirt6 activation may have potential as a novel strategy for treating sleep disorder-related diseases. | Jinpiao Zhu Chang Chen Zhen Li Xiaodong Liu Jingang He Ziyue Zhao Mengying He Binbin Nie Zili Liu Yingying Chen Kuanpin Su Xiang Li Juxiang Chen Hongbing Xiang Fuqiang Xu Kangguang Lin Zongze Zhang Jie Wang | 2023 | Neural Regeneration Research2023,18,11: | 1 |

| 3 | New Gene Variants Associated with the Risk of Chronic HBV Infection显示文摘Some patients with chronic hepatitis B virus(HBV)infection failed to clear HBV,even persistently continue to produce antibodies to HBV.Here we performed a two stage genome wide association study in a cohort of Chinese patients designed to discover single nucleotide variants that associate with HBV infection and clearance of HBV.The first stage involved genome wide exome sequencing of 101 cases(HBsAg plus anti-HBs positive)compared with 102 control patients(antiHBs positive,HBsAg negative).Over 80%of individual sequences displayed 209 sequence coverage.Adapters,uncertain bases[10%or low-quality base calls([50%)were filtered and compared to the human reference genome hg19.In the second stage,579 chronic HBV infected cases and 439 HBV clearance controls were sequenced with selected genes from the first stage.Although there were no significant associated gene variants in the first stage,two significant gene associations were discovered when the two stages were assessed in a combined analysis.One association showed rs506121-“T”allele[within the dedicator of cytokinesis 8(DOCK8)gene]was higher in chronic HBV infection group than that in clearance group(P=0.002,OR=0.77,95%CI[0.65,0.91]).The second association involved rs2071676—A allele within the Carbonic anhydrase(CA9)gene that was significantly elevated in chronic HBV infection group compared to the clearance group(P=0.0003,OR=1.35,95%CI[1.15,1.58]).Upon replication these gene associations would suggest the influence of DOCK8 and CA9 as potential risk genetic factors in the persistence of HBV infection. | Mengjie Fan Jing Wang Sa Wang Tengyan Li Hong Pan Hankui Liu Huifang Xu Daria V.Zhernakova Stephen J.O’Brien Zhenru Feng Le Chang Erhei Dai Jianhua Lu Hongli Xi Yanyan Yu Jianguo Zhang Binbin Wang Zheng Zeng | 2020 | Virologica Sinica2020,35,4: | 1 |

| 7 | Mechanical properties,components and decay resistance of Populus davidiana bioincised by Coriolus versicolor显示文摘The experiment was conducted to improve the decay resistance of wood by increasing the wood preservatives uptake and penetration depth by bioincising with Coriolus versicolor.The mechanical properties of bioincised Populus davidiana sapwood were measured.The changes of wood components(lignin,cellulose and hemicellulose)were analyzed by FTIR.The staining treatment using potassium permanganate was to measure the preservatives uptake and penetration depth in wood specimens.The decay resistance of bioincised specimens impregnated with IPBC and CA was assessed against C.versicolor and Gloeophyllum trabeum using bioincised P.davidiana sapwood and untreated controls.The results showed that the mechanical properties of the bioincised specimens were changed with the incubation time and mass loss.The mechanical strength was decreased after 17 days compared to that of the specimens bioincised<13 days.The bioincised specimens demonstrated the deeper penetration depth than the un-bioincised specimens.It was significant correlation(P<0.01)between penetration depth and incubation time.The average value of the penetration depth of the bioincised specimens was deeper than the control groups by 158.7%.By FTIR analysis,the biodegradation ability of C.versicolor to decay cellulose and hemicellulose was weaker than that of lignin.The bioincising treatment significantly increased the wood uptake of IPBC and CA,and reached the maximum uptake value at 17 and 13 days,respectively.Summarily,the bioincising pretreatment can obviously improve the wood decay resistance against C.versicolor and G.trabeum following by introducing the IPBC and CA. | Lulu Chang Binbin Rong Guoqi Xu Qingkai Meng Lihai Wang | 2020 | Journal of Forestry Research2020,31,5: | 0 |
| 8 | Effects of traditional Chinese medicine on treatment outcomes in severe COVID-19 patients:a single-centre study显示文摘As the search for effective treatments for COVID-19 continues,the high mortality rate among critically ill patients in Intensive Care Units(ICU)presents a profound challenge.This study explores the potential benefits of traditional Chinese medicine(TCM)as a supplementary treatment for severe COVID-19.A total of 110 critically ill COVID-19 patients at the Intensive Care Unit(ICU)of Vulcan Hill Hospital between Feb.,2020,and April,2020(Wuhan,China)participated in this observational study.All patients received standard supportive care protocols,with a subset of 81 also receiving TCM as an adjunct treatment.Clinical characteristics during the treatment period and the clinical outcome of each patient were closely monitored and analysed.Our findings indicated that the TCM group exhibited a significantly lower mortality rate compared with the non-TCM group(16 of 81 vs 24 of 29;0.3 vs 2.3 person/month).In the adjusted Cox proportional hazards models,TCM treatment was associated with improved survival odds(P<0.001).Furthermore,the analysis also revealed that TCM treatment could partially mitigate inflammatory responses,as evidenced by the reduced levels of proinflammatory cytokines,and contribute to the recovery of multiple organic functions,thereby potentially increasing the survival rate of critically ill COVID-19 patients. | XIAO Yongjiu LI Binbin LIU Chang HUANG Xiuyu MA Ling QIAN Zhirong ZHANG Xiaopeng ZHANG Qian LI Dunqing CAI Xiaoqing YAN Xiangyong LUO Shuping XIANG Dawei XIAO Kun | 2024 | Chinese Journal of Natural Medicines2024,22,1: | 0 |
