1Cloning of fatty acid elongase1 gene and molecular identification of A and C genome in Brassica species显示文摘The fatty acid elongase 1 (FAE1) genes of Brassic napus were cloned from two cultivars, i.e. Zhong- shuan No. 9 with low erucic acid content, and Zhongyou 821 with high erucic acid content, using the degenerate PCR primers. The sequence analysis showed that there was no intron within the FAE1 genes. The FAE1 genes from Zhongyou 821 contained a coding sequence of 1521 nucleotides, and those cloned from Zhongshuan No. 9 contained a 1517 bp coding sequence. Alignment of the FAE1 sequences from Brassica rapa, B. oleracea and B. napus detected 31 single nucleotide polymorphic sites (2.03%), which resulted in 7 amino-acid substitutions. Further analysis indicated that 19 SNPs were genome-specific, of which, 95% were synonymous mutations. The nucleotide substitution at po- sition 1217 in the FAE1 genes led to a specific site of restricted cleavage. An AvrII cleavage site was present only in the C genome genes and absent in the A genome FAE1 genes. Digestion profile of the FAE1 sequences from B. rapa, B. oleracea and B. napus produced with AvrII confirmed that the FAE1 genes of B. oleracea origin was recognized and digested, while that of B. rapa origin could not. The results indicated that by AvrII cleavage it was possible to distinguish B. rapa from B. oleracea and be- tween the A and C genome of B. napus. 2120 mm Single-crystalline perovskite and wafers: towards viable applications显示文摘As the large single-crystalline silicon wafers have revolutionized many industries including electronics and solar cells, it is envisioned that the availability of large single-crystalline perovskite crystals and wafers will revolutionize its broad applications in photovoltaics, optoelectronics, lasers, photodetectors, light emitting diodes(LEDs), etc. Here we report a method to grow large single-crystalline perovskites including single-halide crystals: CH3NH3PbX3(X=I, Br, Cl), and dual-halide ones:CH3NH3Pb(ClxBr1.x)3 and CH3NH3Pb(BrxI1.x)3, with the largest crystal being 120 mm in length. Meanwhile, we have advanced a process to slice the large perovskite crystals into thin wafers. It is found that the wafers exhibit remarkable features:(1)its trap-state density is a million times smaller than that in the microcrystalline perovskite thin films(MPTF);(2) its carrier mobility is 410 times higher than its most popular organic counterpart P3HT;(3) its optical absorption is expanded to as high as910 nm comparing to 797 nm for the MPTF;(4) while MPTF decomposes at 150 °C, the wafer is stable at high temperature up to270 °C;(5) when exposed to high humidity(75% RH), MPTF decomposes in 5 h while the wafer shows no change for overnight;(6) its photocurrent response is 250 times higher than its MPTF counterpart. A few electronic devices have been fabricated using the crystalline wafers. Among them, the Hall test gives low carrier concentration with high mobility. The trap-state density is measured much lower than common semiconductors. Moreover, the large SC-wafer is found particularly useful for mass production of integrated circuits. By adjusting the halide composition, both the optical absorption and the light emission can be fine-tuned across the entire visible spectrum from 400 nm to 800 nm. It is envisioned that a range of visible lasers and LEDs may be developed using the dual-halide perovskites. 4Streptomyces avermitilis industrial strain as cell factory for Ivermectin B1a production显示文摘Ivermectin,a kind of valuable derivatives of Avermectin,is distinct from Avermectin due to the saturated bond at C22eC23 position.Combinatorial biosynthesis of Ivermectins based on Avermectins biosynthetic gene cluster(ave)has been achieved recently,while the establishment of an Ivermectin homogeneous component producing strain is challenging because of the limited compatibility between the native and heterologous polyketide synthase(PKS)domains.In this study,the PKS module 2 Dehydratase(DH)-Enoylreductase(ER)-Ketoreductase(KR)domain set of Meilingmycin,which is another naturally occurring homologue of Avermectin,was employed to substitute the DH-KR domains of Avermectins PKS module 2 to generate an Ivermectin biosynthetic gene cluster(ive).Ivermectins B1a and A1a were heterologously biosynthesized in a classic actinomyces host Streptomyces lividans.The Avermectin B1a high-producing strain S.avermitilis 3-115 was genetically engineered to give an artificial host cell and Ivermectin B1a single component was effectively produced with a production of 1.25±0.14 g/L.Qian Deng Liqiongzi Xiao Yang Liu Lixin Zhang Zixin Deng Changming Zhao 2019Synthetic and Systems Biotechnology2019,4,1:3
5Three Dimensional Numerical Investigation on the Effect of Wedge Angle of a Passage with Pin-fin Arrays显示文摘Heat transfer in passage with pin-fin arrays for cooling blade trailing edge was studied numerically. Three-dimensional numerical simulations were carried out for steady laminar flow in passages with different wedge angles between pressure surface and suction surface of cooling blade trailing edge to study the effect of different wedge angles (from 0°to 30°) on heat transfer and pressure losses. Research was carried out for both in-line array and staggered array. 12Photoisomerization,assembling and fluorescence photoswitching behaviors of a water-soluble stiff-stilbene with cucurbit[7]uril显示文摘Photoresponsive supramolecular systems have merited extensive attention for their applications in materials science and life science.Here,we synthesized a water-soluble stiff-stilbene molecular photoswitch,exhibiting outstanding photoisomerization reaction between its(E)-and(Z)-configurations upon irradiation at distinct light.The photoswitch can assemble with cucurbit[7]uril(CB[7])to form a superior fluorescent supramolecular complex(compared to the only guest)with excellent water solubility,which manifested dramatic photoluminescence enhancement caused by restriction of intramolecular rotation and remained good photochromic characteristics.Furthermore,introduction of CB[7]influence photoreaction quantum yield(Φ)of the stiff-stilbene,leading to reduction ofΦ_(E→Z) and increase ofΦ_(Z→E) of the stiff-stilbene.Importantly,the photoadjustable supramolecular assembly can act as a fluorescence switch,and the phototunable guest further selectively modulate G-quadruplex structure of Tel22 upon light irradiation or with addition of CB[7].The study provides a new simple way for accurately regulating photochromic properties of molecular switches and developing smart materials with potential applications in controlled modulation of G-quadruplex,targeted biological imaging and so on.Liangru Yang Yixin Li Haifan Zhang Changming Tian Qiaohong Cao Yongmei Xiao Libo Yuan Guoxing Liu 2023Chinese Chemical Letters2023,34,8:0
13Parallelization and I/O Performance Optimization of a Global Nonhydrostatic Dynamical Core Using MPI显示文摘The Global-Regional Integrated forecast System(GRIST)is the next-generation weather and climate integrated model dynamic framework developed by Chinese Academy of Meteorological Sciences.In this paper,we present several changes made to the global nonhydrostatic dynamical(GND)core,which is part of the ongoing prototype of GRIST.The changes leveraging MPI and PnetCDF techniques were targeted at the parallelization and performance optimization to the original serial GND core.Meanwhile,some sophisticated data structures and interfaces were designed to adjust flexibly the size of boundary and halo domains according to the variable accuracy in parallel context.In addition,the I/O performance of PnetCDF decreases as the number of MPI processes increases in our experimental environment.Especially when the number exceeds 6000,it caused system-wide outages(SWO).Thus,a grouping solution was proposed to overcome that issue.Several experiments were carried out on the supercomputing platform based on Intel x86 CPUs in the National Supercomputing Center in Wuxi.The results demonstrated that the parallel GND core based on grouping solution achieves good strong scalability and improves the performance significantly,as well as avoiding the SWOs.Tiejun Wang Liu Zhuang Julian MKunkel Shu Xiao Changming Zhao 2020Computers, Materials & Continua2020,,6:0

15Genome-Wide Identification, Evolution and Expression Analyses of GA2ox Gene Family in Brassica napus L.显示文摘Gibberellin 2-oxidases(GA2ox)are important enzymes that maintain the balance of bioactive GAs in plants.GA2ox genes have been identified and characterized in many plants,but these genes were not investigated in Brassica napus.Here,we identified 31 GA2ox genes in B.napus and 15 of these BnaGA2ox genes were distributed in the A and C subgenomes.Subcellular localization predictions suggested that all BnaGA2ox proteins were localized in the cytoplasm,and gene structure analysis showed that the BnaGA2ox genes contained 2–4 exons.Phylogenetic analysis indicated that BnGA2ox family proteins in monocotyledons and dicotyledons can be divided into four groups,including two C_(19)-GA2ox and two C_(20)-GA2ox clades.Group 4 is a C_(20)-GA2ox Class discovered recently.Most BnaGA2ox genes had a syntenic relationship with AtGA2ox genes.BnaGA2ox genes in the C subgenome had experienced stronger selection pressure than genes in the A subgenome.BnaGA2ox genes were highly expressed in specific tissues such as those involved in growth and development,and most of them were mainly involved in abiotic responses,regulation of phytohormones and growth and development.Our study provided a valuable evolutionary analysis of GA2ox genes in monocotyledons and dicotyledons,as well as an insight into the biological functions of GA2ox family genes in B.napus.Yanhua Li Hualei Huang Youming Shi Shuqin Huang Tao Liu Changming Xiao Xiaoqing Tian Ping Zhao Xiaoyan Dai Taocui Huang Yan Zhou 2023Phyton-International Journal of Experimental Botany2023,92,3:0
16Five-point Element Scheme of Finite Analytic Method for Unsteady Groundwater Flow显示文摘In order to improve the finite analytic method’s adaptability for irregular unit, by using coordinates rotation technique this paper establishes a five-point element scheme of finite analytic method. It not only solves unsteady groundwater flow equation but also gives the boundary condition. This method can be used to calculate the three typical questions of groundwater.
