1A rapid advice guideline for the diagnosis and treatment of 2019 novel coronavirus(2019-nCoV) infected pneumonia(standard version)显示文摘In December 2019, a new type viral pneumonia cases occurred in Wuhan, Hubei Province;and then named '2019 novel coronavirus(2019-nCoV)' by the World Health Organization(WHO) on 12 January 2020. For it is a never been experienced respiratory disease before and with infection ability widely and quickly, it attracted the world’s attention but without treatment and control manual. For the request from frontline clinicians and public health professionals of 2019-nCoV infected pneumonia management, an evidence-based guideline urgently needs to be developed. Therefore, we drafted this guideline according to the rapid advice guidelines methodology and general rules of WHO guideline development;we also added the first-hand management data of Zhongnan Hospital of Wuhan University. This guideline includes the guideline methodology, epidemiological characteristics, disease screening and population prevention, diagnosis, treatment and control(including traditional Chinese Medicine), nosocomial infection prevention and control, and disease nursing of the 2019-nCoV. Moreover, we also provide a whole process of a successful treatment case of the severe 2019-nCoV infected pneumonia and experience and lessons of hospital rescue for 2019-nCoV infections. 2High expression of ACE2 receptor of 2019-nCoV on the epithelial cells of oral mucosa显示文摘It has been reported that ACE2 is the main host cell receptor of 2019-nCoV and plays a crucial role in the entry of virus into the cell to cause the final infection. To investigate the potential route of 2019-n Cov infection on the mucosa of oral cavity, bulk RNA-seq profiles from two public databases including The Cancer Genome Atlas(TCGA) and Functional Annotation of The Mammalian Genome Cap Analysis of Gene Expression(FANTOM5 CAGE) dataset were collected. RNA-seq profiling data of 13 organ types with para-carcinoma normal tissues from TCGA and 14 organ types with normal tissues from FANTOM5 CAGE were analyzed in order to explore and validate the expression of ACE2 on the mucosa of oral cavity. Further, single-cell transcriptomes from an independent data generated in-house were used to identify and confirm the ACE2-expressing cell composition and proportion in oral cavity. The results demonstrated that the ACE2 expressed on the mucosa of oral cavity. Interestingly, this receptor was highly enriched in epithelial cells of tongue. 7Effect of Surface Mechanical Attrition Treatment on Corrosion Behavior of 316 Stainless Steel显示文摘The nanocrystalline microstructure of the surface of 316 stainless steel (316SS) induced by surface mechanical attrition treatment (SMAT) was determined by X-ray diffraction (XRD) and scanning electron microscopy (SEM). The technique of hydrogen embrittlement was first used to obtain the information of the brittleness cleavage plane. The effects of SMAT and the following annealing process on the corrosion behavior of 316SS were investigated by potentiodynamic polarization curves and potentiostatic critical pitting temperature measurements. The results demonstrated that the nanocrystalline layer with an average grain size of 19 nm was produced. However, there were lots of cracks on the surface, which led to the degradation in the corrosion resistance of 316SS after SMAT. Nevertheless, after annealing treatment, the corrosion resistance of the nanocrystalline surface had been improved greatly. 11Staging laparoscopy improves treatment decision-makingfor advanced gastric cancer显示文摘AIM To evaluate the clinical value of staging laparoscopyin treatment decision-making for advancedgastric cancer (GC).METHODS: Clinical data of 582 patients with advancedGC were retrospectively analyzed. All patientsunderwent staging laparoscopy. The strength ofagreement between computed tomography (CT) stage,endoscopic ultrasound (EUS) stage, laparoscopic stage,and final stage were determined by weighted Kappastatistic (Kw). The number of patients with treatmentdecision-changes was counted. A χ 2 test was used toanalyze the correlation between peritoneal metastasisor positive cytology and clinical characteristics.RESULTS: Among the 582 patients, the distributions ofpathological T classifications were T2/3 (153, 26.3%),T4a (262, 45.0%), and T4b (167, 28.7%). Treatmentplans for 211 (36.3%) patients were changed after staging laparoscopy was performed. Two (10.5%) of19 patients in M1 regained the opportunity for potentialradical resection by staging laparoscopy. Unnecessarylaparotomy was avoided in 71 (12.2%) patients. Thestrength of agreement between preoperative T stageand final T stage was in almost perfect agreement (Kw= 0.838; 95% confidence interval (CI): 0.803-0.872;P 〈 0.05) for staging laparoscopy; compared with CTand EUS, which was in fair agreement. The strengthof agreement between preoperative M stage andfinal M stage was in almost perfect agreement (Kw= 0.990; 95% CI: 0.977-1.000; P 〈 0.05) for staginglaparoscopy; compared with CT, which was in slightagreement. 12Mature Archean continental crust in the Yangtze craton: Evidence from petrology, geochronology and geochemistry显示文摘Previous studies have shown that the Archean basement is widely distributed throughout the Yangtze craton. To date, however, Archean basement terrains have not been found, except for a few Kongling high-grade metamorphic terrains in the Huangling dome that have been confirmed to be of Archean age. To further understand the basement component and crustal evolution of the Yangtze craton, we carried out a petrological, geochronological and geochemical study of the Jinshan K-feldspar granite emplaced within the Yangpo Group, located in Huji Town, Zhongxiang City, Hubei Province. Results indicate that the zircon SHRIMP U-Pb age of the Jinshan granite is 2655±9 Ma, placing it within the middle Neoarchean. Chemically, this pluton yields abundant silica and alkalis and is depleted in Ca, Mg and Ti. Furthermore, it is enriched in Rb, Th, Ga, Y and Zr, depleted in Sr, Ba, Nb and Ta, and especially lacking in Eu. High ratios of FeO*/MgO (32.0 to 58.7) and 104 ×Ga/Al (3.19 to 3.41) were also found. The pluton exhibits characteristics typical of A-type granites with crustal source magmas. Moreover, the meta-sedimentary rock association of the Yangpo Group, into which the pluton intruded, clearly shows relatively stable depositional environments of a shallow shelf sequence. Therefore, before the middle Neoarchean, the Yangtze craton contained mature continental crust. 14Calcium channel blockers reduce severe fever with thrombocytopenia syndrome virus (SFTSV) related fatality显示文摘Severe fever with thrombocytopenia syndrome (SFTS),an emerging tick-borne infectious disease caused by a novel phlebovirus (SFTS virus, SFTSV), was listed among the top 10 priority infectious diseases by the World Health Organization due to its high fatality of 12%-50% and possibility of pandemic transmission. Currently, effective anti-SFTSV intervention remains unavailable. Here, by screening a library of FDA-approved drugs, we found that benidipine hydrochloride, a calcium channel blocker (CCB), inhibited SFTSV replication in vitro. Benidipine hydrochloride was revealed to inhibit virus infection through impairing virus internalization and genome replication. Further experiments showed that a broad panel of CCBs, including nifedipine, inhibited SFTSV infection. The anti-SFTSV effect of these two CCBs was further an a lyzed in a humanized mouse model in which CCB treatment resulted in reduced viral load and decreased fatality rate. Importantly, by performing a retrospective clinical investigation on a large cohort of 2087 SFTS patients, we revealed that nifedipine administration enhaneed virus clearance, improved clinical recovery, and remarkably reduced the case fatality rate by >5-fold. 15Prognostic value of number of examined lymph nodes in patients with node-negative gastric cancer显示文摘AIM:To elucidate the potential impact of examined lymph nodes(eLNs)on long-term survival of nodenegative gastric cancer patients after curative surgery.METHODS:A total of 497 node-negative gastric cancer patients who underwent curative gastrectomy between January 2000 and December 2008 in our center were enrolled in this study.Patients were divided into4 groups according to eLNs through cut-point analysis.Clinicopathological features were compared between≤15 eLNs group and>15 eLNs group and potential prognostic factors were analyzed.The Log-rank test was used to assess statistical differences between the groups.Independent prognostic factors were identifiedusing the Cox proportional hazards regression model.Stratified analysis was performed to investigate the impact of eLNs on patient survival in each stage.Overall survival was also compared among the four groups.Finally,we explored the recurrent sites associated with eLNs.RESULTS:Patients with eLNs>15 had a better survival compared with those with eLNs≤15 for the entire cohort.By the multivariate survival analysis,we found that the depth of invasion and the number of eLNs were the independent predictors of overall survival(OS)of patients with node-negative gastric cancer.According to the cut-point analysis,T2-T4 patients with 11-15 eLNs had a significantly longer mean OS than those with 4-10 eLNs or 1-3 eLNs.Patients with≤15 eLNs were more likely to experience locoregional and peritoneal recurrence than those with>15eLNs.CONCLUSION:Number of eLNs could predict the prognosis of node-negative gastric cancer,and dissection of>15 eLNs is recommended during lymphadenectomy so as to improve the long-term survival.Xu-Guang Jiao Jing-Yu Deng Ru-Peng Zhang Liang-Liang Wu Li Wang Hong-Gen Liu Xi-Shan Hao Han Liang 2014World Journal of Gastroenterology2014,20,13:15
16Identification of mecciRNAs and their roles in the mitochondrial entry of proteins显示文摘Mammalian mitochondria have small genomes encoding very limited numbers of proteins.Over one thousand proteins and noncoding RNAs encoded by the nuclear genome must be imported from the cytosol into the mitochondria.Here,we report the identification of hundreds of circular RNAs(mecciRNAs)encoded by the mitochondrial genome.We provide both in vitro and in vivo evidence to show that mecciRNAs facilitate the mitochondrial entry of nuclear-encoded proteins by serving as molecular chaperones in the folding of imported proteins.Known components involved in mitochondrial protein and RNA importation,such as TOM40 and PNPASE,interact with mecciRNAs and regulate protein entry.The expression of mecciRNAs is regulated,and these transcripts are critical for the adaption of mitochondria to physiological conditions and diseases such as stresses and cancers by modulating mitochondrial protein importation.mecciRNAs and their associated physiological roles add categories and functions to the known eukaryotic circular RNAs and shed novel light on the communication between mitochondria and the nucleus.Xu Liu Xiaolin Wang Jingxin Li Shanshan Hu Yuqi Deng Hao Yin Xichen Bao Qiangfeng Cliff Zhang Geng Wang Baolong Wang Qinghua Shi Ge Shan 2020Science China(Life Sciences)2020,63,10:15

18Hyperparameter Optimization for Machine Learning Models Based on Bayesian Optimization显示文摘Hyperparameters are important for machine learning algorithms since they directly control the behaviors of training algorithms and have a significant effect on the performance of machine learning models. Several techniques have been developed and successfully applied for certain application domains. However, this work demands professional knowledge and expert experience. And sometimes it has to resort to the brute-force search.Therefore, if an efficient hyperparameter optimization algorithm can be developed to optimize any given machine learning method, it will greatly improve the efficiency of machine learning. In this paper, we consider building the relationship between the performance of the machine learning models and their hyperparameters by Gaussian processes. In this way, the hyperparameter tuning problem can be abstracted as an optimization problem and Bayesian optimization is used to solve the problem. Bayesian optimization is based on the Bayesian theorem. It sets a prior over the optimization function and gathers the information from the previous sample to update the posterior of the optimization function. A utility function selects the next sample point to maximize the optimization function.Several experiments were conducted on standard test datasets. 20Ciliary parathyroid hormone signaling activates transforming growth factor-βto maintain intervertebral disc homeostasis during aging显示文摘Degenerative disc disease(DDD) is associated with intervertebral disc degeneration of spinal instability. Here, we report that the cilia of nucleus pulposus(NP) cells mediate mechanotransduction to maintain anabolic activity in the discs. We found that mechanical stress promotes transport of parathyroid hormone 1 receptor(PTH1 R) to the cilia and enhances parathyroid hormone(PTH) signaling in NP cells. PTH induces transcription of integrin α_vβ_6 to activate the transforming growth factor(TGF)-β-connective tissue growth factor(CCN2)-matrix proteins signaling cascade. Intermittent injection of PTH(iPTH) effectively attenuates disc degeneration of aged mice by direct signaling through NP cells, specifically improving intervertebral disc height and volume by increasing levels of TGF-β activity, CCN2, and aggrecan. PTH1 R is expressed in both mouse and human NP cells. Importantly,knockout PTH1 R or cilia in the NP cells results in significant disc degeneration and blunts the effect of PTH on attenuation of aged discs.
