2Potassium Channels: A Potential Therapeutic Target for Parkinson's Disease显示文摘The pathogenesis of the second major neurodegenerative disorder, Parkinson's disease(PD), is closely associated with the dysfunction of potassium(K^+ ) channels. Therefore, PD is also considered to be an ion channel disease or neuronal channelopathy. Mounting evidence has shown that K^+ channels play crucial roles in the regulations of neurotransmitter release, neuronal excitability, and cell volume. Inhibition of K^+ channels enhances the spontaneous firing frequency of nigral dopamine(DA)neurons, induces a transition from tonic firing to burst discharge, and promotes the release of DA in the striatum.Recently, three K^+ channels have been identified to protect DA neurons and to improve the motor and non-motor symptoms in PD animal models: small conductance(SK)channels, A-type K^+ channels, and KV7/KCNQ channels.In this review, we summarize the physiological and pharmacological effects of the three K^+ channels. 3Copper homeostasis and cuproptosis in health and disease显示文摘all cell types.Because the accumulation of intracellular copper can induce oxidative stress and perturbing cellular function,copper homeostasis is tightly regulated.Recent studies identified a novel copper-dependent form of cell death called cuproptosis,which is distinct from all other known pathways underlying cell death.Cuproptosis occurs via copper binding to lipoylated enzymes in the tricarboxylic acid(TCA)cycle,which leads to subsequent protein aggregation,proteotoxic stress,and ultimately cell death.Here,we summarize our current knowledge regarding copper metabolism,copper-related disease,the characteristics of cuproptosis,and the mechanisms that regulate cuproptosis.In addition,we discuss the implications of cuproptosis in the pathogenesis of various disease conditions,including Wilson’s disease,neurodegenerative diseases,and cancer,and we discuss the therapeutic potential of targeting cuproptosis.Liyun Chen Junxia Min Fudi Wang 2022Signal Transduction and Targeted Therapy2022,7,12:10
4Numerical simulation of mould filling process for pressure plate and valve handle in LFC显示文摘In lost foam casting (LFC), the distribution of polymer beads during the bead filling process is not uniform, and the collision between polymer beads determines the distribution of two-phase flow of gas and solid. The interaction between the gas and solid phases reveals as coupling effect of the force that gas exerts on particles or vice versa, or that among particles. The gas-solid flow in filling process is nonlinearity, which makes the coupling effect an essential point to carry out a simulation properly. Therefore, information of each particle's motion is important for acquiring the law of filling process. In bead filling process, compressed air is pressed into mold cavity, and discharged from gas vent, creating a pressure difference between outer and inner space near the gas vent. This pressure difference directly changes the spatial distribution and motion trace of gas and solid phases. In this paper, Discrete Element Method (DEM) and Computational Fluid Dynamics (CFD) are employed to simulate the fluid dynamic character based on Newton's Third Law of Motion. The simulation results of some casting products such as pressure plate and valve handle are compared with the result obtained from practical experiment in order to test the feasibility of DEM. 5Flexible, Porous, and Metal–Heteroatom?Doped Carbon Nanofibers as Efficient ORR Electrocatalysts for Zn–Air Battery显示文摘Developing an e cient and durable oxygen reduction electrocatalyst is critical for clean-energy technology, such as fuel cells and metal–air batteries. In this study, we developed a facile strategy for the preparation of flexible, porous, and well-dispersed metal–heteroatom-doped carbon nanofibers by direct carbonization of electrospun Zn/Co-ZIFs/PAN nanofibers(Zn/Co-ZIFs/PAN). The obtained Zn/Co and N co-doped porous carbon nanofibers carbonized at 800 °C(Zn/Co–N@PCNFs-800) presented a good flexibility, a continuous porous structure, and a superior oxygen reduction reaction(ORR) catalytic activity to that of commercial 20 wt% Pt/C, in terms of its onset potential(0.98 V vs. RHE), half-wave potential(0.89 V vs. RHE), and limiting current density(-5.26 mA cm^(-2)). In addition, we tested the suitability and durability of Zn/Co–N@PCNFs-800 as the oxygen cathode for a rechargeable Zn–air battery. The prepared Zn–air batteries exhibited a higher power density(83.5 mW cm^(-2)), a higher specific capacity(640.3 mAh g^(-1)), an excellent reversibility, and a better cycling life than the commercial 20 wt% Pt/C + RuO_2 catalysts. 6The transmission link of CAPS navigation and communication system显示文摘The Chinese Area Positioning System (CAPS) is based on communication satellites with integrated capability, which is different from the Global Positioning System (GPS), the International Maritime Satellite Organization (Inmarsat) and so on. CAPS works at C-band, and its navigation information is not directly generated from the satellite, but from the master control station on the ground and transmitted to users via the satellite. The slightly inclined geostationary-satellite orbit (SIGSO) satellites are adopted in CAPS. All of these increase the difficulty in the design of the system and terminals. In this paper, the authors study the CAPS configuration parameters of the navigation master control station, information transmission capability, and the selection of the antenna aperture of the communication center station, as well as the impact of satellite parameters on the whole communication system from the perspective of the transmission link budget. The conclusion of availability of the CAPS navigation system is achieved. The results show that the CAPS inbound communication system forms a new low-data-rate satellite communication system, which can accommodate mass communication terminals with the transmission rate of no more than 1 kbps for every terminal. The communication center station should be configured with a large-aperture antenna (about 10-15 m); spread spectrum com- munication technology should be used with the spreading gain as high as about 40 dB; reduction of the satellite transponder gain attenuation is beneficial to improving the signal-to-noise ratio of the system, with the attenuation value of 0 or 2 dB as the best choice. The fact that the CAPS navigation system has been checked and accepted by the experts and the operation is stable till now clarifies the rationality of the analysis results. 7The zinc transporter Slc39a5 controls glucose sensing and insulin secretion in pancreatic β-cells via Sirt1- and Pgc-1α-mediated regulation of Glut2显示文摘Zinc levels are high in pancreatic β-cells, and zinc is involved in the synthesis, processing and secretion of insulin in these cells. However, precisely how cellular zinc homeostasis is regulated in pancreatic β-cells is poorly understood. By screening the expression of 14 Slc39a metal importer family member genes, we found that the zinc transporter Slc39a5 is significantly downregulated in pancreatic β-cells in diabetic db/db mice, obese ob/ob mice and high-fat diet-fed mice. Moreover,β-cell-specific Slc39a5 knockout mice have impaired insulin secretion. In addition, Slc39a5-deficient pancreatic islets have reduced glucose tolerance accompanied by reduced expression of Pgc-1α and its downstream target gene Glut2. The down-regulation of Glut2 in Slc39a5-deficient islets was rescued using agonists of Sirt1, Pgc-1α and Ppar-γ. At the mechanistic level, we found that Slc39a5-mediated zinc influx induces Glut2 expression via Sirt1-mediated Pgc-1α activation. 8Carbon Isotope Gradient of the Ediacaran Cap Carbonate in the Shennongjia Area and Its Implications for Ocean Stratification and Palaeogeography显示文摘The geochemistry and paleogeography of the Doushantuo succession in the Shennongjia area have been insufficiently studied. Here, we report on the carbon and oxygen isotope compositions of Ediacaran Doushantuo cap carbonates from four sections(Longxi, Muyu, Yazikou and Songluo) in the Shennongjia area. A large C-isotopic gradient(~5‰) between the Longxi and Songluo sections, here identified for the first time, is inferred to have been dynamically maintained by photosynthesis in surface waters and anaerobic oxidation of dissolved organic carbon(DOC) in deep waters. Spatial variation in C-isotope chemostratigraphy among the four sections was related to Marinoan post-glacial sea-level elevation changes. At Longxi, a positive δ^(13)C_(carb) shift below the horizon containing tepee-like structure resulted from intense photosynthesis during early regression. A negative δ^(13)C_(carb) shift within the tepee-horizon was triggered by sulfate reduction and freshwater mixing with ^(13)C-depleted dissolved inorganic carbon(DIC) during late regression. A positive δ^(13)C_(carb) excursion in the uppermost part of the cap carbonate was related to enhance primary productivity and organic matter burial during early transgression. At Muyu, the carbon isotopic variation tendency, which is similar to that at Longxi, may have been mildly influenced by the surface water environment. At Songluo, the positive δ^(13)C_(carb) excursion, up to-4‰ in the lower part of the cap carbonate, was probably associated with methanogenesis in deep waters during late transgression to early regression and subsequently disappeared due to decomposition of methane hydrate during late regression. At Yazikou, the consistently stable δ^(13)C_(carb) values around-4‰ indicate that the cap carbonate may have deposited at intermediate water depths. 9Detection of oil spills in a complex scene of SAR imagery显示文摘We present a method for detecting oil spills in a complex scene of SAR imagery,including segmenting oil spills,and avoiding false alarms.Segmentation is carried out using a multi-time and multi-hierarchical method by dividing the complex sea surface into bright sea and dark sea.Gray-based and edge-based segmentations are done to extract oil spills from bright and dark sea,respectively.The proposed method can extract complete oil spills,obtain better visual results,and increase detection probability more accurately than the traditional method.Based on the surrounding features and the oil spills’features,dark land spots and low contrast dark spots are removed efficiently,thus reducing false alarms.The experimental results demonstrate that the proposed algorithm has fast computation speed,high detection accuracy,and is very useful and effective for detecting oil spills in SAR imagery.FENG Jing CHEN He BI FuKun LI JunXia WEI Hang 2014Science China(Technological Sciences)2014,57,11:4

13Microstructure Evolution and Mechanical Behavior of Laser Melting Deposited TA15 Alloy at 500℃ under In-Situ Tension in SEM显示文摘TA15 alloy fabricated by laser melting deposition was investigated at 500℃ under tensile deformation. The damage behavior of microstructure was analyzed by the real time observation of the microstructure evolution, microcracks initiation and propagation using in-situ tensile equipment fitted in the SEM chamber. Finally, the mechanism of fracture was discussed. The result showed anisotropic mechanical properties in X-and Z-direction. The existence of columnar β grains and its orientation to the tensile direction were the major factors inducing the anisotropic mechanical properties. As compared to Z-direction specimen, high tensile strength was observed in X-direction specimen due to the resistance in slips propagation provided by the prior-β grain boundaries( β GBs). Accumulation of the cracks at prior β GB caused the shear fracture. In case of Z-direction specimen, parallel orientation of prior β GB and GB α with the tensile direction resulted in a homogeneous deformation. 14The novel ER stress inducer Sec C triggers apoptosis by sulfating ER cysteine residues and degrading YAP via ER stress in pancreatic cancer cells显示文摘Pancreatic adenocarcinoma(PAAD) is one of the most lethal malignancies. Although gemcitabine(GEM) is a standard treatment for PAAD, resistance limits its application and therapy. Secoemestrin C(Sec C) is a natural compound from the endophytic fungus Emericella, and its anticancer activity has not been investigated since it was isolated. Our research is the first to indicate that Sec C is a broad-spectrum anticancer agent and could exhibit potently similar anticancer activity both in GEM-resistant and GEM-sensitive PAAD cells. Interestingly, Sec C exerted a rapid growth-inhibiting effect(80% death at 6 h), which might be beneficial for patients who need rapid tumor shrinkage before surgery. Liquid chromatography/mass spectrometry and N-acetyl-L-cysteine(NAC) reverse assays show that Sec C sulfates cysteines to disrupt disulfide-bonds formation in endoplasmic reticulum(ER) proteins to cause protein misfolding, leading to ER stress and disorder of lipid biosynthesis. Microarray data and subsequent assays show that ER stress-mediated ER-associated degradation(ERAD) ubiquitinates anddownregulates YAP to enhance ER stress via destruction complex(YAP-Axin-GSK-βTr CP), which also elucidates a unique degrading style for YAP. 15Susceptibility of patients with chronic obstructive pulmonary disease to heart rate difference associated with the short-term exposure to metals in ambient fine particles:A panel study in Beijing,China显示文摘Susceptibility of patients with chronic obstructive pulmonary disease(COPD)to cardiovascular autonomic dysfunction asso-ciated with exposure to metals in ambient fine particles(PM2.5,particulate matter with aerodynamic diameter≤2.5µm)remains poorly evidenced.Based on the COPDB(COPD in Beijing)panel study,we aimed to compare the associations of heart rate(HR,an indicator of cardiovascular autonomic function)and exposure to metals in PM2.5 between 53 patients with COPD and 82 healthy controls by using linear mixed-effects models.In all participants,the HR levels were significantly associated with interquartile range increases in the average concentrations of Cr,Zn,and Pb,but the strength of the associations differed by exposure time(from 1.4%for an average 9 days(d)Cr exposure to 3.5%for an average 9 d Zn exposure).HR was positively associated with the average concentrations of PM2.5 and certain metals only in patients with COPD.Associations between HR and exposure to PM2.5,K,Cr,Mn,Ni,Cu,Zn,As,and Se in patients with COPD significantly differed from those in health controls.Furthermore,association between HR and Cr exposure was robust in COPD patients.In conclusion,our findings indicate that COPD could exacerbate difference in HR following exposure to metals in PM2.5.Ke Gao Xi Chen Xiaoying Li Hanxiyue Zhang Mengxiao Luan Yuan Yao Yifan Xu Teng Wang Yiqun Han Tao Xue Junxia Wang Mei Zheng Xinghua Qiu Tong Zhu 2022Science China(Life Sciences)2022,65,2:2
16Improved EEMD-based standardization method for developing long tree-ring chronologies显示文摘Long tree-ring chronologies can be developed by overlapping data from living trees with data from fossil trees through cross-dating.However,low-frequency climate signals are lost when standardizing tree-ring series due to the'segment length curse'.To alleviate the segment length curse and thus improve the standardization method for developing long tree-ring chronologies,here we first calculated a mean value for all the tree ring series by overlapping all of the tree ring series.The growth trend of the mean tree ring width(i.e.,cumulated average growth trend of all the series)was determined using ensemble empirical mode decomposition.Then the chronology was developed by dividing the mean value by the growth trend of the mean value.Our improved method alleviated the problem of trend distortion.Long-term signals were better preserved using the improved method than in previous detrending methods.The chronologies developed using the improved method were better correlated with climate than those developed using conservative methods.The improved standardization method alleviates trend distortion and retains more of the low-frequency climate signals.Xianliang Zhang Junxia Li Xiaobo Liu Zhenju Chen 2020Journal of Forestry Research2020,31,6:2
