| 2 | Immune checkpoint signaling and cancer immunotherapy显示文摘Immune checkpoint blockade therapy has become a major weapon in fighting cancer.Antibody drugs,such as anti-PD-1 and antiPD-L1,demonstrate obvious advantages such as broad applicability across cancer types and durable clinical response when treatment is effective.However,the overall response rates are still unsatisfying,especially for cancers with low mutational burden.Moreover,adverse effects,such as autoimmune symptoms and tumor hyperprogression,present a significant downside in some clinical applications.These challenges reflect the urgent need to fully understand the basic biology of immune checkpoints.In this review,we discuss regulation of immune checkpoint signaling at multiple levels to provide an overview of our current understanding of checkpoint biology.Topics include the regulation of surface expression levels for known immune checkpoint proteins via surface delivery,internalization,recycling,and degradation.Upon reaching the surface,checkpoints engage in both conventional trans and also cis interactions with ligands to induce signaling and regulate immune responses.Novel therapeutic strategies targeting these pathways in addition to classical checkpoint blockade have recently emerged and been tested in preclinical models,providing new avenues for developing next-generation immunotherapies. | Xing He Chenqi Xu | 2020 | Cell Research2020,30,8: | 24 |
| 3 | The precise regulation of different COR genes by individual CBF transcription factors in Arabidopsis thaliana显示文摘The transcription factors CBF1/2/3 are reported to play a dominant role in the cold responsive network of Arabidopsis by directly regulating the expression levels of cold responsive(COR) genes.In this study,we obtained CRISPR/Cas9-mediated loss-of-function mutants of cbf1$3.Over 3,000 COR genes identified by RNA-seq analysis showed a slight but significant change in their expression levels in the mutants compared to the wildtype plants after being treated at 4 °C for 12 h.The C-repeat(CRT) motif(50-CCGAC-30) was enriched in promoters of genes that were up-regulated by CBF2 and CBF3 but not in promoters of genes up-regulated by CBF1.These data suggest that CBF2 and CBF3 play a more important role in directing the cold response by regulating different sets of downstream COR genes.More than 2/3 of COR genes were co-regulated by two or three CBFs and were involved mainly in cellular signal transduction and metabolic processes;less than 1/3 of the genes were regulated by one CBF,and those genes up-regulated were enriched in cold-related abiotic stress responses.Our results indicate that CBFs play an important role in the trade-off between cold tolerance and plant growth through the precise regulation of COR genes in the complicated transcriptional network. | Yihao Shi Jiaying Huang Tianshu Sun Xuefei Wang Chenqi Zhu Yuxi Ai Hongya Gu | 2017 | Journal of Integrative Plant Biology2017,59,2: | 7 |

| 5 | Potential-tuned selective electrosynthesis of azoxy-,azoand amino-aromatics over a CoP nanosheet cathode显示文摘Azoxy-, azo-and amino-aromatics are among the most widely used building blocks in materials science pharmaceuticals and synthetic chemistry, but their controllable and green synthesis has not yet been well established. Herein, a facile potential-tuned electrosynthesis of azoxy-, azo-and amino-aromatics via aqueous selective reduction of nitroarene feedstocks over a CoP nanosheet cathode is developed. A series of azoxy-, azo-and amino-compounds with excellent selectivity, good functional group tolerance and high yields are produced by applying different bias input. The synthetically significant and challenging asymmetric azoxy-aromatics can be controllably synthesized in moderate to good yields. The use of water as the hydrogen source makes this strategy remarkably fascinating and promising. In addition, deuterated aromatic amines with a high deuterium content can be readily obtained by using D2O. By pairing with anodic oxidation of aliphatic amines to nitriles, synthetically useful building blocks can be simultaneously produced in a CoP||Ni2 P two-electrode electrolyzer. Only 1.25 V is required to achieve a current density of 20 mA cm-2, which is much lower than that of overall water splitting(1.70 V). The paired oxidation and reduction reactions can also be driven using a 1.5 V battery to synthesize nitrile and azoxybenzene with good yields and selectivity, further emphasizing the flexibility and controllability of our method. | 8 | A novel gatifloxacin-loaded intraocular lens for prophylaxis of postoperative endophthalmitis显示文摘Postoperative endophthalmitis(POE)has been the most threatening complication after cataract surgery,which perhaps can be solved by the antibiotic-loaded intraocular lens(IOL).However,most drug-loaded IOLs demonstrate insufficient drug quantity,short release time,increased implantation-related difficulties or other noticeable drawbacks.To prevent POE and to address these deficiencies,a drug-loaded copolymer IOL,prepared from poly(urethane acrylate)prepolymer,isobornyl methacrylate(IBOMA),N-vinyl-2-pyrrolidone(NVP),Irgacure 819,RUVA-93,and gatifloxacin(GAT),was rapidly fabricated via photocuring and by using a 3D-printed mold.This composite displayed an outstanding and controllable GAT release behavior in vitro,a high light transmittance,and a moderate refractive index.Also,it demonstrated improved strain stress and elongation compared with the reference commercial acrylic IOL material.In vivo tests demonstrated satisfying released drug concentration at the early treatment stage.In vitro and in vivo studies further confirmed the remarkable bacterial inhibition and prevention of POE by the proposed IOL,which also displayed good biocompatibility.These findings suggested that the GAT-loaded IOL could be a promising implant to prevent and cure POE,also the proposed methods could inspire more designs for various medical applications. | Mengna Li Jing-Wei Xu Jiayong Li Wei Wang Chenqi Luo Haijie Han Zhi-Kang Xu Ke Yao | 2023 | Bioactive Materials2023,,2: | 2 |
| 9 | Tuning charge density of chimeric antigen receptor optimizes tonic signaling and CAR-T cell fitness显示文摘Tonic signaling of chimeric antigen receptor(CAR),i.e.,the spontaneous CAR activation in the absence of tumor antigen stimulation,is considered to be a pivotal event controlling CAR-T efficacy.However,the molecular mechanism underlying the spontaneous CAR signals remains elusive.Here,we unveil that positively charged patches(PCPs)on the surface of the CAR antigenbinding domain mediate CAR clustering and result in CAR tonic signaling.For CARs with high tonic signaling(e.g.,GD2.CAR and CSPG4.CAR),reducing PCPs on CARs or boosting ionic strength in the culture medium during ex vivo CAR-T cell expansion minimizes spontaneous CAR activation and alleviates CAR-T cell exhaustion.In contrast,introducing PCPs into the CAR with weak tonic signaling,such as CD19.CAR,results in improved in vivo persistence and superior antitumor function.These results demonstrate that CAR tonic signaling is induced and maintained by PCP-mediated CAR clustering.Notably,the mutations we generated to alter the PCPs maintain the antigen-binding affinity and specificity of the CAR.Therefore,our findings suggest that the rational tuning of PCPs to optimize tonic signaling and in vivo fitness of CAR-T cells is a promising design strategy for the nextgeneration CAR. | Jian Chen Shizhen Qiu Wentao Li Kun Wang Yu Zhang Han Yang Baichuan Liu Guangfei Li Li Li Min Chen Junjie Lan Jiahua Niu Peijie He Lei Cheng Gaofeng Fan Xin Liu Xianmin Song Chenqi Xu Haitao Wu Haopeng Wang | 2023 | Cell Research2023,33,5: | 2 |
| 10 | SST biases over the Northwest Pacific and possible causes in CMIP5 models显示文摘In this paper, the features and possible causes of sea surface temperature(SST) biases over the Northwest Pacific are investigated based on a mixed-layer heat budget analysis in 21 coupled general circulation models(CGCMs) from phase 5 of the Coupled Model Inter-comparison Project(CMIP5). Most CMIP5 models show cold SST biases throughout the year over the Northwest Pacific. The largest biases appear during summer, and the smallest biases occur during winter. These cold SST biases are seen at the basin scale and are mainly located in the inner region of the low and mid-latitudes. According to the mixed-layer heat budget analysis, overestimation of upward net sea surface heat fluxes associated with atmospheric processes are primarily responsible for the cold SST biases. Among the different components of surface heat fluxes, overestimated upward latent heat fluxes induced by the excessively strong surface winds contribute the most to the cold SST biases during the spring, autumn, and winter seasons. Conversely, during the summer, overestimated upward latent heat fluxes and underestimated downward solar radiations at the sea surface are equally important. Further analysis suggests that the overly strong surface winds over the Northwest Pacific during winter and spring are associated with excessive precipitation over the Maritime Continent region,whereas those occurring during summer and autumn are associated with the excessive northward extension of the intertropical convergence zone(ITCZ). The excessive precipitation over the Maritime Continent region and the biases in the simulated ITCZ induce anomalous northeasterlies, which are in favor of enhancing low-level winds over the North Pacific. The enhanced surface wind increases the sea surface evaporation, which contributes to the excessive upward latent heat fluxes. | 11 | Experimental research on oil fi lm thickness and its microwave scattering during emulsifi cation显示文摘Synthetic Aperture Radar(SAR) plays a major role in identifying oil spills on the sea surface.However,obtaining information of oil spill thickness(volume) is still a challenge.Emulsification is an important process affecting the thickness and normalized radar cross section(NRCS) of oil film.Experiments of crude oil emulsification with C-band fully-polarized scatterometer were conducted combining airborne hyperspectral imaging spectrometer and 3 D laser scanner observation data,to provide experimental parameters and method to support accurate remote sensing monitoring on marine oil spill.It is further proved that through quantitative homogeneous emulsified oil spill experiments,to a certain extent,the NRCS of oil film increased during the emulsification process of crude oil.The backscattering mechanism of crude oil emulsification was explored using a semi-empirical model(SEM);the change of oil film NRCS was modulated by its dielectric constant and surface roughness,in which the dielectric constant showed a dominant effect.The relationship between thickness and NRCS of oil film was studied under two experimental conditions.The differences of NRCS between oil film and adjacent seawater(Δσ~0) and the damping ratio(DR) were found to have a linear relationship with oil thickness,which were best in the vertical polarization mode(VV) at 45° incident angle during the quantitative crude oil homogeneous emulsification process.In the natural emulsification process of continuous oil spill in which oil film was mixed with both crude oil and emulsified oil,an empirical equation of oil film thickness is preliminarily established.The Δσ~0,DR,and the empirical equation of oil film thickness were applied to the marine continuous oil spill incident on a 19-3 oil platform with spaceborne SAR image and successfully explained the distribution of the relative thickness of the oil film. | Jie GUO Chenqi XU Genwang LIU Xi ZHANG Junmin MENG Guangbo REN | 2022 | Journal of Oceanology and Limnology2022,40,4: | 2 |

| 14 | A new oil spill detection algorithm based on Dempster-Shafer evidence theory显示文摘Features of oil spills and look-alikes in polarimetric synthetic aperture radar(SAR)images always play an important role in oil spill detection.Many oil spill detection algorithms have been implemented based on these features.Although environmental factors such as wind speed are important to distinguish oil spills and look-alikes,some oil spill detection algorithms do not consider the environmental factors.To distinguish oil spills and look-alikes more accurately based on environmental factors and image features,a new oil spill detection algorithm based on Dempster-Shafer evidence theory was proposed.The process of oil spill detection taking account of environmental factors was modeled using the subjective Bayesian model.The Faster-region convolutional neural networks(RCNN)model was used for oil spill detection based on the convolution features.The detection results of the two models were fused at decision level using Dempster-Shafer evidence theory.The establishment and test of the proposed algorithm were completed based on our oil spill and look-alike sample database that contains 1798 image samples and environmental information records related to the image samples.The analysis and evaluation of the proposed algorithm shows a good ability to detect oil spills at a higher detection rate,with an identifi cation rate greater than 75%and a false alarm rate lower than 19%from experiments.A total of 12 oil spill SAR images were collected for the validation and evaluation of the proposed algorithm.The evaluation result shows that the proposed algorithm has a good performance on detecting oil spills with an overall detection rate greater than 70%. | Tianlong ZHANG Jie GUO Chenqi XU Xi ZHANG Chuanyuan WANG Baoquan LI | 2022 | Journal of Oceanology and Limnology2022,40,2: | 1 |

Results of statistics, spectral estimate and correlation analysis of the low-passed velocity data show that time scales of low-frequency components of the near-bottom currents are 25~120 days, in which 51-day period dominates the lower band of the frequency domain.
