1Analysis of COVID-19 Guideline Quality and Change of Recommendations:A Systematic Review显示文摘Background.Hundreds of coronavirus disease 2019(COVID-19)clinical practice guidelines(CPGs)and expert consensus statements have been developed and published since the outbreak of the epidemic.However,these CPGs are of widely variable quality.So,this review is aimed at systematically evaluating the methodological and reporting qualities of COVID-19 CPGs,exploring factors that may influence their quality,and analyzing the change of recommendations in CPGs with evidence published.Methods.We searched five electronic databases and five websites from 1 January to 31 December 2020 to retrieve all COVID-19 CPGs.The assessment of the methodological and reporting qualities of CPGs was performed using the AGREE II instrument and RIGHT checklist.Recommendations and evidence used to make recommendations in the CPGs regarding some treatments for COVID-19(remdesivir,glucocorticoids,hydroxychloroquine/chloroquine,interferon,and lopinavir-ritonavir)were also systematically assessed.And the statistical inference was performed to identify factors associated with the quality of CPGs.Results.We included a total of 92 COVID-19 CPGs developed by 19 countries.Overall,the RIGHT checklist reporting rate of COVID-19 CPGs was 33.0%,and the AGREE II domain score was 30.4%.The overall methodological and reporting qualities of COVID-19 CPGs gradually improved during the year 2020.Factors associated with high methodological and reporting qualities included the evidence-based development process,management of conflicts of interest,and use of established rating systems to assess the quality of evidence and strength of recommendations.The recommendations of only seven(7.6%)CPGs were informed by a systematic review of evidence,and these seven CPGs have relatively high methodological and reporting qualities,in which six of them fully meet the Institute of Medicine(IOM)criteria of guidelines.Besides,a rapid advice CPG developed by the World Health Organization(WHO)of the seven CPGs got the highest overall scores in methodological(72.8%)and reporting qualities(83.8%).Many CPGs covered the same clinical questions(it refers to the clinical questions on the effectiveness of treatments of remdesivir,glucocorticoids,hydroxychloroquine/chloroquine,interferon,and lopinavirritonavir in COVID-19 patients)and were published by different countries or organizations.Although randomized controlled trials and systematic reviews on the effectiveness of treatments of remdesivir,glucocorticoids,hydroxychloroquine/chloroquine,interferon,and lopinavir-ritonavir for patients with COVID-19 have been published,the recommendations on those treatments still varied greatly across COVID-19 CPGs published in different countries or regions,which may suggest that the CPGs do not make sufficient use of the latest evidence.Conclusions.Both the methodological and reporting qualities of COVID-19 CPGs increased over time,but there is still room for further improvement.The lack of effective use of available evidence and management of conflicts of interest were the main reasons for the low quality of the CPGs.The use of formal rating systems for the quality of evidence and strength of recommendations may help to improve the quality of CPGs in the context of the COVID-19 pandemic.During the pandemic,we suggest developing a living guideline of which recommendations are supported by a systematic review for it can facilitate the timely translation of the latest research findings to clinical practice.We also suggest that CPG developers should register the guidelines in a registration platform at the beginning for it can reduce duplication development of guidelines on the same clinical question,increase the transparency of the development process,and promote cooperation among guideline developers all over the world.Since the International Practice Guideline Registry Platform has been created,developers could register guidelines prospectively and internationally on this platform.Siya Zhao Shuya Lu Shouyuan Wu Zijun Wang Qiangqiang Guo Qianling Shi Hairong Zhang Juanjuan Zhang Hui Liu Yunlan Liu Xianzhuo Zhang Ling Wang Mengjuan Ren Ping Wang Hui Lan Qi Zhou Yajia Sun Jin Cao Qinyuan Li Janne Estill Joseph LMathew Hyeong Sik Ahn Myeong Soo Lee Xiaohui Wang Chenyan Zhou Yaolong Chen 2021Health Data Science2021,,1:1
2Classification of human movements with and without spinal orthosis based on surface electromyogram signals显示文摘Spinal orthoses were designed to correct poor posture;however,they may restrict trunk movements at all times,making daily activities difficult.Detecting trunk movements can provide instructions for adjusting the stiffness of the spinal orthosis.This study evaluated the feasibility of identifying movements based on surface electromyography(sEMG)signals.Ten participants were tested for different movements with two different modalities:motion without the spinal orthosis(Normal)and with the spinal orthosis(Spinal orthosis).The sEMG signals were collected from eight muscles using surface electrodes during four movements[flexion-extension,lateral bending,axial rotation,and stand to sit to stand].Four time domain features were extracted,with a total of 32 feature vectors.The principal component analysis(PCA)method was adopted to feature selection,and it was found that eight feature dimensions can make cumulative explained variance exceed 95%.The results showed that machine learning algorithms could not only identify Normal and Spinal orthosis movement modalities,but also distinguish four daily movements.Moreover,the classification performance of Random Forest(RF),k-Nearest Neighbor(kNN),and Support Vector Machine(SVM)algorithms were also compared.The results showed that all three machine algorithms have high classification accuracy.The machine learning methods can accurately identify movement patterns by considering sEMG signals,which may provide instructions for adjusting the stiffness of the spinal orthosis.In the future,the spinal orthosis with adjustable stiffness controlled by sEMG signals could help correct poor posture,and permit the wearer to achieve free movement when needed.Chenyan Wang Xiaona Li Yuan Guo Ruixuan Zhang Weiyi Chen 2022Medicine in Novel Technology and Devices2022,,4:0
3Clock component OsPRR59 delays heading date by repressing transcription of Ehd3 in rice显示文摘Heading date(or flowering time),an important agronomic trait in crop species,is closely associated with regional adaptation and yield.Members of the Pseudo-Response Regulator(PRR)family play key roles in regulating flowering.However,their role and molecular mechanism controlling heading date in rice is not very clear.Here,we identified rice OsPRR protein,OsPRR59,which delayed heading under longday conditions.OsPRR59 positively regulates yield by affecting plant height,secondary branches number per panicle,grain number per panicle,seed setting rate,and grain weight per plant.OsPRR59 is expressed in most tissues and its protein is localized to the nucleus.We also found that OsPRR59 directly binds to the promoter of Ehd3 to inhibit its expression.Compared with the WT,osprr59 ehd3 showed a significantly delayed heading phenotype,as did the ehd3 mutant.This was opposite to the phenotype of the osprr59 mutant,confirming that Ehd3 acted downstream of OsPRR59 in regulating rice flowering.Our results identified a direct regulator of Ehd3,and revealed a novel molecular mechanism of clock component OsPRR proteins in regulating heading date and provide a new genetic resource for fine-tuning heading date in rice.Yupeng Wang Fuqing Wu Shirong Zhou Weiwei Chen Chenyan Li Erchao Duan Jiachang Wang Zhijun Cheng Xin Zhang Qibing Lin Yulong Ren Cailin Lei Xiuping Guo Ziming Wu Shanshan Zhu Jianmin Wan 2022The Crop Journal2022,10,6:0
4Identification of a natural PLA2 inhibitor from the marine fungus Aspergillus sp.c1 for MAFLD treatment that suppressed lipotoxicity by inhibiting the IRE-1a/XBP-1s axis and JNK signaling显示文摘Lipotoxicity is a pivotal factor that initiates and exacerbates liver injury and is involved in the development of metabolic-associated fatty liver disease(MAFLD).However,there are few reported lipotoxicity inhibitors.Here,we identified a natural anti-lipotoxicity candidate,HN-001,from the marine fungus Aspergillus sp.C1.HN-001 dose-and time-dependently reversed palmitic acid(PA)-induced hepatocyte death.This protection was associated with IRE-1a-mediated XBP-1 splicing inhibition,which resulted in suppression of XBP-1s nuclear translocation and transcriptional regulation.Knockdown of XBP-1s attenuated lipotoxicity,but no additional ameliorative effect of HN-001 on lipotoxicity was observed in XBP-1s knockdown hepatocytes.Notably,the ER stress and lipotoxicity amelioration was associated with PLA2.Both HN-001 and the PLA2 inhibitor MAFP inhibited PLA2 activity,reduced lysophosphatidylcholine(LPC)level,subsequently ameliorated lipotoxicity.In contrast,overexpression of PLA2 caused exacerbation of lipotoxicity and weakened the anti-lipotoxic effects of HN-001.Additionally,HN-001 treatment suppressed the downstream pro-apoptotic JNK pathway.In vivo,chronic administration of HN-001(i.p.)in mice alleviated all manifestations of MAFLD,including hepatic steatosis,liver injury,inflammation,and fibrogenesis.These effects were correlated with PLA2/IRE-1a/XBP-1s axis and JNK signaling suppression.These data indicate that HN-001 has therapeutic potential for MAFLD because it suppresses lipotoxicity,and provide a natural structural basis for developing anti-MAFLD candidates.Yong Rao Rui Su Chenyan Wu Xingxing Chai Jinjian Li Guanyu Yang Junjie Wu Tingting Fu Zhongping Jiang Zhikai Guo Congjun Xu Ling Huang 2024Acta Pharmaceutica Sinica B2024,14,1:0
5The Effects of Fertilizers on Rabbiteye Blueberry(Vaccinium ashei Reade.)Root Distribution and Fruit Yield显示文摘The root system plays an important role in the growth and development of blueberry.The aim of this study was to assess the impacts of different fertilizers on the root growth and root–yield relationship of blueberry to provide insight into the regulation of root growth and fruit yield by fertilizing from the perspective of the root system.Rabbiteye blueberry variety‘Britewell’as the test material,and six fertilizers,including BF,OR,CF,SF,HF,and RT were used in single-factor fertilization experiments to analyze the effects of different fertilizer treatments on the root morphology,root distribution,and fruit yield of blueberry.Fertilization overall increased the root length density and root surface area in most soil layers,and the RT treatment significantly increased the total root length density and total root surface area 98.6%and 98.5%,respectively,compared with a control lacking fertilizer.In addition,the effect of fertilization on the blueberry root system was mainly observed in the 0–20 cm layer.Fruit yield was positively correlated with total root length density and total root surface area,and negatively correlated with average root diameter.In summary,the SF and RT treatments increased the morphological indexes of the root system,particularly in the shallow soil layers,leading to an increase in blueberry fruit yield.Xiaolan Guo Chenyan Liu Muhammad Shakeel Delu Wang 2022Phyton-International Journal of Experimental Botany2022,91,6:0
6Study on Infrared Spectra of Citrus grandis Peels from Guangxi Zhuang Autonomous Region显示文摘[Objective] This study aimed to establish the infrared (IR) spectra fingerprints for peels of Citrus grandis from Guangxi Zhuang Autonomous Region. [Method] The fingerprint was established by fourier transform infrared spectroscopy (FT-IR), and then were analyzed by Omic software. [Result] IR spectra fingerprint of C. grandis were successfully established. There were six common peaks in the characteristic spectra of 12 samples. 7Interactions of Indoleamine 2,3-dioxygenase-expressing LAMP3+dendritic cells with CD4+regulatory T cells and CD8+exhausted T cells:synergistically remodeling of the immunosuppressive microenvironment in cervical cancer and therapeutic implications显示文摘Background Cervical cancer(CC)is the fourth most common cancer in women worldwide.Although immunotherapy has been applied in clinical practice,its therapeutic efficacy remains far from satisfactory,necessitating further investigation of the mechanism of CC immune remodeling and exploration of novel treatment targets.This study aimed to investigate the mechanism of CC immune remodeling and explore potential therapeutic targets.Methods We conducted single-cell RNA sequencing on a total of 17 clinical specimens,including normal cervical tissues,high-grade squamous intraepithelial lesions,and CC tissues.To validate our findings,we conducted multicolor immunohistochemical staining of CC tissues and constructed a subcutaneous tumorigenesis model in C57BL/6 mice using murine CC cell lines(TC1)to evaluate the effectiveness of combination therapy involving indoleamine 2,3-dioxygenase 1(IDO1)inhibition and immune checkpoint blockade(ICB).We used the unpaired two-tailed Student's t-test,Mann-Whitney test,or Kruskal-Wallis test to compare continuous data between two groups and one-way ANOVA with Tukey's post hoc test to compare data between multiple groups.Results Malignant cervical epithelial cells did not manifest noticeable signs of tumor escape,whereas lysosomal-associated membrane protein 3-positive(LAMP3+)dendritic cells(DCs)in a mature state with immunoregulatory roles were found to express IDO1 and affect tryptophan metabolism.These cells interacted with both tumor-reactive exhausted CD8+T cells and CD4+regulatory T cells,synergistically forming a vicious immunosuppressive cycle and mediating CC immune escape.Further validation through multicolor immunohistochemical staining showed co-localization of neoantigen-reactive T cells(CD3+,CD4+/CD8+,and PD-1+)and LAMP3+DCs(CD80+and PD-L1+).Additionally,a combination of the IDO1 inhibitor with an ICB agent significantly reduced tumor volume in the mouse model of CC compared with an ICB agent alone.Conclusions Our study suggested that a combination treatment consisting of targeting IDO1 and ICB agent could improve the therapeutic efficacy of current CC immunotherapies.Additionally,our results provided crucial insights for designing drugs and conducting future clinical trials for CC.Xinyu Qu Yumeng Wang Qian Jiang Tingting Ren Chenyan Guo Keqin Hua Junjun Qiu 2023Cancer Communications2023,43,11:0
