2Treatment outcomes for different subgroups of nasopharyngeal carcinoma patients treated with intensity-modulated radiation therapy显示文摘Although many studies have investigated intensity-modulated radiation therapy (IMRT) for nasopharyngeal carcinoma (NPC),sample sizes in the reported studies are usually small and different in outcomes in different T and N subgroups are seldom analyzed.Herein,we evaluated the outcomes of NPC patients treated with IMRT and further explored treatment strategy to improve such outcome.We collected clinical data of 865 NPC patients treated with IMRT alone or in combination with chemotherapy,and classified all cases into the following prognostic categories according to different TNM stages:early stage group (T1-2N0-1M0),advanced local disease group (T3-4N0-1M0),advanced nodal disease group (T1-2N2-3M0),and advanced locoregional disease group (T3-4N2-3M0).The 5-year overall survival (OS),local relapse-free survival (LRFS),and distant metastases-free survival (DMFS) were 83.0%,90.4%,and 84.0%,respectively.The early disease group had the lowest treatment failure rate,with a 5-year OS of 95.6%.The advanced local disease group and advanced nodal disease group had similar failure pattern and treatment outcomes as well as similar hazard ratios for death (4.230 and 4.625,respectively).The advanced locoregional disease group had the highest incidence of relapse and death,with a 5-year DMFS and OS of 62.3% and 62.2%,respectively,and a hazard ratio for death of 10.402.Comparing with IMRT alone,IMRT in combination with chemotherapy provided no significant benefit to locoregionally advanced NPC.Our results suggest that the decision of treatment strategy for NPC patients should consider combinations of T and N stages,and that IMRT alone for early stage NPC patients can produce satisfactory results.However,for advanced local,nodal,and locoregional disease groups,a combination of chemotherapy and radiotherapy is recommended.Sheng-Fa Su 1,2,3,Fei Han 1,2,Chong Zhao 1,2,Ying Huang 1,2,Chun-Yan Chen 1,2,Wei-Wei Xiao 1,2,Jia-Xin Li 1,2 and Tai-Xiang Lu 1,2 1 State Key Laboratory of Oncology in South China,Guangzhou,Guangdong 510060,P.R.China 2 Department of Radiation Oncology,Sun Yat-sen University Cancer Center,Guangzhou,Guangdong 510060,P.R.China 3 Department of Oncology,Guiyang Medical College Hospital,Tumor Hospital of Guizhou,Guiyang,Guizhou 550003,P.R.China. 2011Chinese Journal of Cancer2011,30,8:40
3Nutritional and medicinal characteristics of Chinese giant salamander (Andrias davidianus) for applications in healthcare industry by artificial cultivation: A review显示文摘Andrias davidianus,i.e.Chinese giant salamander(CGS),is one of the largest and oldest amphibians existing in the world and is also one of the valuable biological resources of China.Wild CGS has been threatened with extinction in the past decades due to over capturing,deterioration of natural environment,the slow breeding and growth of the wild species in nature.However,in the past twenty years,with the breakthrough and progress of artificial breeding technology by artificial insemination,the number of artificially cultivated CGS has increased rapidly.Artificially cultivated CGS can either be released to the CGS living environment to increase the population in nature or legally applied in food and medicinal industry as a feedstock due to the unique nutritional and medicinal values of CGS as recorded historically.In this review,the nutritional components,bioactive components and medicinal activities of the artificially cultivated CGS will be summarized.The mucus,skin,meat and bone of CGS contain many different bioactive substances thereby having various medicinal activities including anti-aging,anti-fatigue,anti-tumor,therapy of burn and anti-infection and other physiological functions.This paper will further discuss the potential applications of the artificially cultivated CGS in healthcare industry and prospects of future technological development.Dong He Wenming Zhu Wen Zenga Jun Lin Yang Ji Yi Wang Chong Zhang Yuan Lu Daoquan Zhao Nan Su Xin-Hui Xing 2018Food Science and Human Wellness2018,7,1:28

6CRISPR/Cas9-mediated PINK1 deletion leads to neurodegeneration in rhesus monkeys显示文摘Dear Editor,PINK1 mutations cause autosomal recessive and early-onset Parkinson’s disease(PD)with selective neurodegeneration.Unfortunately,current PINK1 knockout(KO)mouse1,2 and pig models3,4 are unable to recapitulate the selective and overt neurodegeneration seen in PD patients.Furthermore,endogenous Pink1 in the mouse brain is expressed at very low levels and can only be detected via immunoprecipitation,5 meaning that PINK1’s function in the mammalian brain needs to be assessed using larger animals that are closer to humans.We previously used CRISPR/Cas9 to target the monkey gene in one-cell stage embryos.6 Using the same approach,we designed two gRNAs to target exon 2(T1)and exon 4(T2),which encode a kinase domain in the PINK1 gene of rhesus monkeys(Fig.1a).CRISPR/Cas9 and PINK1 gRNAs were injected into one-cell stage rhesus monkey embryos.A T7E1 assay and sequencing of PCR products from the injected embryos showed high efficiency(61.5%)in targeting PINK1(Fig.1b and Supplementary information,Fig.S1).Transfer of 87 embryos to 28 surrogate rhesus monkeys resulted in 11 pregnancies(39.2%)(Fig.1b).Eleven fetuses developed to term and were born naturally.Of these live monkeys,eight carried PINK1 mutations(M),and three were wild type(WT).However,three mutant monkeys(M1,M3 and M4)were newborn triplets that struggled to survive and died 3-4 days after birth.One WT newborn monkey also died after a difficult labor.Another mutant monkey(M2)died 7 days after birth without noticeable warning signs or symptoms.The other three mutant monkeys(M6,M7 and M8)have lived for three years;M5,however,reduced its food intake and showed weakness at the age of 1.5 years,and died 30 days after anesthesia for MRI examination.Weili Yang Yunbo Liu Zhuchi Tu Chong Xiao Sen Yan Xishan Ma Xiangyu Guo Xiusheng Chen Peng Yin Zhengyi Yang Su Yang Tianzi Jiang Shihua Li Chuan Qin Xiao-Jiang Li 2019Cell Research2019,29,4:12
7Genome-wide association study identifies favorable SNP alleles and candidate genes for waterlogging tolerance in chrysanthemums显示文摘Chrysanthemums are sensitive to waterlogging stress,and the development of screening methods for tolerant germplasms or genes and the breeding of tolerant new varieties are of great importance in chrysanthemum breeding.To understand the genetic basis of waterlogging tolerance(WT)in chrysanthemums,we performed a genome-wide association study(GWAS)using 92,811 single nucleotide polymorphisms(SNPs)in a panel of 88 chrysanthemum accessions,including 64 spray cut and 24 disbud chrysanthemums.The results showed that the average MFVW(membership function value of waterlogging)of the disbud type(0.65)was significantly higher than that of the spray type(0.55)at P<0.05,and the MFVW of the Asian accessions(0.65)was significantly higher than that of the European accessions(0.48)at P<0.01.The GWAS performed using the general linear model(GLM)and mixed linear model(MLM)identified 137 and 14 SNP loci related to WT,respectively,and 11 associations were commonly predicted.By calculating the phenotypic effect values for 11 common SNP loci,six highly favorable SNP alleles that explained 12.85—21.85%of the phenotypic variations were identified.Furthermore,the dosage-pyramiding effects of the favorable alleles and the significant linear correlations between the numbers of highly favorable alleles and phenotypic values were identified(r2=0.45;P<0.01).A major SNP locus(Marker6619-75)was converted into a derived cleaved amplified polymorphic sequence(dCAPS)marker that cosegregated with WT with an average efficiency of 78.9%.Finally,four putative candidate genes in the WT were identified via quantitative real-time PCR(qRT-PCR).The results presented in this study provide insights for further research on WT mechanisms and the application of molecular marker-assisted selection(MAS)in chrysanthemum WT breeding programs.Jiangshuo Su Fei Zhang Xinran Chong Aiping Song Zhiyong Guan Weimin Fang Fadi Chen 2019Horticulture Research2019,6,1:9

9Development of a Rapid Multi-residue Assay for Detecting β-lactams Using Penicillin Binding Protein 2x显示文摘Objective To develop a rapid multi-residue assay for detecting 16 demanded by the European Union (EU). Methods A recombinant penicillin-binding protein(PBP)2x* from Streptococcus pneumoniae R6 was expressed in vitro and six β-lactams were conjugated to HRP by four methods.A rapid multi-residue assay for β-lactams was established with PBP2x* and HRP-conjugate. Results PBP2x* was expressed and purified successfully and the ideal HRP-conjugate was identified. The multi-residue assay was developed.After optimization,penicillin G,ampicillin,amoxicillin, cloxacillin,dicloxacillin,oxacillin,nafcillin,cephalexin,ceftiofur,cefaIonium,cefquinome,cefazolin, cefoperazone,cephacetrile,and cephapirin can be detected at levels below MRL in milk with simple pretreatment. Conclusion This assay developed can detect all 16 β-lactams demanded by the European Union(EU). 10Prevalence of upper gastrointestinal bleeding risk factors among the general population and osteoarthritis patients显示文摘AIM To assess the prevalence of possible risk factors of upper gastrointestinal bleeding(UGIB) and their agegroup specific trend among the general population and osteoarthritis patients.METHODS We utilized data from the National Health InsuranceService that included claims data and results of the national health check-up program. Comorbid conditions(peptic ulcer, diabetes, liver disease, chronic renal failure, and gastroesophageal reflux disease), concomitant drugs(aspirin, clopidogrel, cilostazol, non-steroidal anti-inflammatory drugs, steroid, anticoagulants, and SSRI), personal habits(smoking, and alcohol consumption) were considered as possible UGIB risk factors. We randomly imputed the prevalence of infection in the data considering the age-specific prevalence of Helicobacter pylori(H. pylori) infection in Korea. The prevalence of various UGIB risk factors and the age-group specific trend of the prevalence were identified. Prevalence was compared between osteoarthritis patients and others.RESULTS A total of 801926 subjects(93855 osteoarthritis patients) aged 20 and above were included. The prevalence of individual and concurrent multiple risk factors became higher as the age increased. The prevalence of each comorbid condition and concomitant drug were higher in osteoarthritis patients. Thirty-five point zero two percent of the overall population and 68.50% of osteoarthritis patients had at least one or more risk factors of UGIB. The prevalence of individual and concurrent multiple risk factors in younger age groups were also substantial. Furthermore, when personal habits(smoking, and alcohol consumption) and H. pylori infection were included, the prevalence of concurrent multiple risk factors increased greatly even in younger age groups.CONCLUSION Prevalence of UGIB risk factors was high in elderly population, but was also considerable in younger population. Patient with osteoarthritis was at higher UGIB risk than those without osteoarthritis. 11Capecitabine with radiation is an effective adjuvant therapy in gastric cancers显示文摘AIM:To analyze the outcome of patients who received concurrent capecitabine(Xeloda) and radiation(XRT) compared to the established concurrent 5-fluorouracil(5-FU) with radiation(5FU-RT) and fluoropyrimidine-based chemotherapy alone as adjuvant treatment in gastric cancers.METHODS:All patients with gastric cancers who received adjuvant treatment at the National Cancer Centre Singapore between 1996 and 2006 were reviewed.Treatment outcomes of patients who received XRT were compared with those who had 5FU-RT or chemotherapy alone as adjuvant therapy for gastric cancers.RESULTS:A total of 108 patients were reviewed.Median age at diagnosis was 60.The majority of the patients(64.8%) had advanced stage Ⅲ and Ⅳ disease(with no distant metastasis).All except 4 patients had D2 gastrectomy.Twenty one patients(19.4%) had positive surgical resection margins.Thirty three patients received XRT compared with 52 who had 5FU-RT and 23 who received chemotherapy alone.For the patients in the chemotherapy-only group,all had fluoropyrimidine-based therapy,with added cisplatin in 7 patients and epirubicin in 2 patients.Median recurrence-free survival was longer for the XRT group(52 mo) compared to the 5FU-RT(35 mo) and chemotherapy-only groups(25 mo)(P=0.48).The patients in the XRT group achieved similar median overall survival(53 mo) as the 5FU-RT(54 mo) and the chemotherapy-only groups(44 mo)(P=0.5).CONCLUSION:Capecitabine with concurrent radiation was as effective as concurrent 5FU with radiation or fluoropyrimidine-based chemotherapy alone when used as adjuvant treatment in patients with gastric cancers.Chee Kian Tham Su Pin Choo Donald Yew Hee Poon Han Chong Toh Simon Yew Kuang Ong Sze Huey Tan Michael Lian Chek Wang Kian Fong Foo 2010World Journal of Gastroenterology2010,16,29:6
12Analytical methods for investigating in vivo fate of nanoliposomes:A review显示文摘Nanoliposomes are considered to be the most successful nanoparticle drug delivery system, but their fate in vivo has not been fully understood due to lack of reliable bioanalytical methods, which seriously limits the development of liposomal drugs. Hence, an overview of currently used bioanalytical methods is imperative to lay the groundwork for the need of developing a bioanalytical method for liposome measurements in vivo. Currently, major analytical methods for nanoliposomes measurement in vivo include fluorescence labeling, radiolabeling, magnetic resonance imaging(MRI), mass spectrometry and computed tomography. In this review, these bioanalytical methods are summarized, and the advantages and disadvantages of each are discussed. We provide insights into the applicability and limitations of these analytical methods in the application of nanoliposomes measurement in vivo, and highlight the recent development of instrumental analysis techniques. 13Burn image segmentation based on Mask Regions with Convolutional Neural Network deep learning framework:more accurate and more convenient显示文摘Background:Burns are life-threatening with high morbidity and mortality.Reliable diagnosis supported by accurate burn area and depth assessment is critical to the success of the treatment decision and,in some cases,can save the patient’s life.Current techniques such as straight-ruler method,aseptic film trimming method,and digital camera photography method are not repeatable and comparable,which lead to a great difference in the judgment of burn wounds and impede the establishment of the same evaluation criteria.Hence,in order to semi-automate the burn diagnosis process,reduce the impact of human error,and improve the accuracy of burn diagnosis,we include the deep learning technology into the diagnosis of burns.Method:This article proposes a novel method employing a state-of-the-art deep learning technique to segment the burn wounds in the images.We designed this deep learning segmentation framework based on the Mask Regions with Convolutional Neural Network(Mask R-CNN).For training our framework,we labeled 1150 pictures with the format of the Common Objects in Context(COCO)data set and trained our model on 1000 pictures.In the evaluation,we compared the different backbone networks in our framework.These backbone networks are Residual Network-101 with Atrous Convolution in Feature Pyramid Network(R101FA),Residual Network-101 with Atrous Convolution(R101A),and InceptionV2-Residual Network with Atrous Convolution(IV2RA).Finally,we used the Dice coefficient(DC)value to assess the model accuracy.Result:The R101FA backbone network gains the highest accuracy 84.51%in 150 pictures.Moreover,we chose different burn depth pictures to evaluate these three backbone networks.The R101FA backbone network gains the best segmentation effect in superficial,superficial thickness,and deep partial thickness.The R101A backbone network gains the best segmentation effect in full-thickness burn.Conclusion:This deep learning framework shows excellent segmentation in burn wound and extremely robust in different burn wound depths.Moreover,this framework just needs a suitable burn wound image when analyzing the burn wound.It is more convenient and more suitable when using in clinics compared with the traditional methods.And it also contributes more to the calculation of total body surface area(TBSA)burned.Chong Jiao Kehua Su Weiguo Xie Ziqing Ye 2019Burns & Trauma2019,7,1:4

15Celecoxib attenuates hepatocyte apoptosis by inhibiting endoplasmic reticulum stress in thioacetamide-induced cirrhotic rats显示文摘BACKGROUND Endoplasmic reticulum(ER)stress is an important mechanism in the progression of chronic and acute liver diseases,especially in the progression and recovery of liver fibrosis.Excessive and long-term ER stress induces apoptosis.ER stressinduced apoptosis is considered to be an important pathway in the development of liver fibrosis.Cyclooxygenase-2(COX-2)induction is also closely related to ER stress.In our previous studies,we showed that celecoxib,a COX-2 inhibitor,improves liver fibrosis and portal hypertension.However,the role and mechanism of celecoxib in alleviating liver fibrosis remain unclear.AIM To investigate whether celecoxib alleviates liver fibrosis by inhibiting hepatocyte apoptosis via the ER stress response.METHODS Cirrhosis was induced by intraperitoneal injections of thioacetamide(TAA)for 16 wk(injection dose is 200 mg/kg per 3 d for the first 8 wk and 100 mg/kg per 3 d after 8 wk).Thirty-six male Sprague-Dawley rats were randomly divided into three groups,namely,control group,TAA group,and TAA+celecoxib group.In the last 8 wk,TAA-induced cirrhotic rats received celecoxib(20 mg/kg/day)or the vehicle by gastric gavage.After 16 wk,the rats were sacrificed,and serum alanine aminotransferase(ALT),aspartate aminotransferase(AST),and albumin(ALB)were detected.The hepatic fibrosis areas were evaluated by Sirius red staining and the degree of fibrosis was assessed by measuring the level of hydroxyproline.ER stress levels were evaluated by detecting the marker proteins glucose-regulated protein 78(GRP78),CCAAT/enhancer binding protein homologous protein(CHOP),PKR-like ER protein kinase(PERK),activating transcription factor 6(ATF6),and inositol-requiring enzyme 1 alpha(IRE1α).Apoptosis levels were evaluated by detecting caspase-12 and caspase-3.RESULTS The serum ALT and AST levels in the liver were significantly reduced by celecoxib;however,the serum ALB had no significant changes.Celecoxib significantly reduced the degree of liver fibrosis and the levels of hydroxyproline(-38%and-25.7%,respectively,P<0.01).Celecoxib ameliorated ER stress by reducing the level of GRP78 compared to the TAA group(P<0.05).Consistently,after celecoxib administration,the upregulation of TAA-induced hepatic apoptosis markers(caspase-12 and caspase-3)and CHOP were significantly inhibited.In addition,after celecoxib treatment,the expression of key molecules associated with ER stress(PERK,ATF6,and IRE1)was decreased(P<0.05).CONCLUSION Therapeutic administration of celecoxib effectively reduces hepatic apoptosis in TAA-induced cirrhotic rats.The mechanism of action may be attributed to the suppression of CHOP expression,which subsequently inhibits ER stress.Wei Su Yang Tai Shi-Hang Tang Yan-Ting Ye Chong Zhao Jin-Hang Gao Bi-Guang Tuo Cheng-Wei Tang 2020World Journal of Gastroenterology2020,26,28:4

17Impact of damping on high speed 850 nm VCSEL performance显示文摘High speed VCSELs are important optical devices in short-reach optical communication links and interconnects because of their low cost and high modulation speeds. In this paper, the impact of damping on the their static and dynamic characteristics is analyzed and demonstrated. Through the shallow corrosion of the top layer DBR, the VCSELs with different damping is designed and fabricated. With the increase of the surface etch depth from 0 to ~55 nm for 9 μm oxide-aperture VCSEL, the K factor related with the damping is reduced from0.31 to 0.23 ns^(-1). When the etch depth of the VCSEL with 9 μm oxide-aperture is decreased to ~25 nm,output power is increased from 4.03 to 4.70 mW and small signal modulation bandwidth is also increased from15.46 to 16.37 GHz.
