1A two-step lineage reprogramming strategy to generate functionally competent human hepatocytes from fibroblasts显示文摘Terminally differentiated cells can be generated by lineage reprogramming,which is,however,hindered by incomplete conversion with residual initial cell identity and partial functionality. Here, we demonstrate a new reprogramming strategy by mimicking the natural regeneration route, which permits generating expandable hepatic progenitor cells and functionally competent human hepatocytes. Fibroblasts were first induced into human hepatic progenitor-like cells (hHPLCs), which could robustly expand in vitro and efficiently engraft in vivo. Moreover, hHPLCs could be efficiently induced into mature human hepatocytes (hiHeps) in vitro, whose molecular identity highly resembles primary human hepatocytes (PHHs). Most importantly, hiHeps could be generated in large quantity and were functionally competent to replace PHHs for drug-metabolism estimation, toxicity prediction and hepatitis B virus infection modeling. Our results highlight the advantages of the progenitor stage for successful lineage reprogramming. 3Effect of active control on optimal structures in wall turbulence显示文摘The effect of active control imposed at the wall on optimal structures in wall turbulence is studied by using a linear transient growth model.When the detection plane of the control is located in the buffer layer,the influence of the control on the transient growth of large scale motion becomes negligible as Reynolds number increases.However,if the control signal is detected at the plane located in the logarithm region,the transient growth at large scale can be greatly suppressed.New peak values of transient growth resulting from the strong blowing and suction on the wall exist.The study indicates that a proper selection of control imposed on the wall can suppress the large scale motion in the logarithmic region.DENG BingQing XU ChunXiao HUANG WeiXi CUI GuiXiang 2013Science China(Physics,Mechanics & Astronomy)2013,56,2:3
4Increasing the heterologous production of spinosad in Streptomyces albus J1074 by regulating biosynthesis of its polyketide skeleton显示文摘Spinosyns are natural broad-spectrum biological insecticides with a double glycosylated polyketide structure that are produced by aerobic fermentation of the actinomycete,Saccharopolyspora spinosa.However,their large-scale overproduction is hindered by poorly understood bottlenecks in optimizing the original strain,and poor adaptability of the heterologous strain to the production of spinosyn.In this study,we genetically engineered heterologous spinosyn-producer Streptomyces albus J1074 and optimized the fermentation to improve the production of spinosad(spinosyn A and spinosyn D)based on our previous work.We systematically investigated the result of overexpressing polyketide synthase genes(spnA,B,C,D,E)using a constitutive promoter on the spinosad titer in S.albus J1074.The supply of polyketide synthase precursors was then increased to further improve spinosad production.Finally,increasing or replacing the carbon source of the culture medium resulted in a final spinosad titer of~70 mg/L,which is the highest titer of spinosad achieved in heterologous Streptomyces species.This research provides useful strategies for efficient heterologous production of natural products.Ziheng An Hui Tao Yong Wang Bingqing Xia Yang Zou Shuai Fu Fang Fang Xiao Sun Renqiong Huang Yao Xia Zixin Deng Ran Liu Tiangang Liu 2021Synthetic and Systems Biotechnology2021,6,4:1
5Derivation of totipotent-like stem cells with blastocyst-like structure forming potential显示文摘It is challenging to derive totipotent stem cells in vitro that functionally and molecularly resemble cells from totipotent embryos.Here,we report that a chemical cocktail enables the derivation of totipotent-like stem cells,designated as totipotent potential stem(TPS)cells,from 2-cell mouse embryos and extended pluripotent stem cells,and that these TPS cells can be stably maintained long term in vitro.TPS cells shared features with 2-cell mouse embryos in terms of totipotency markers,transcriptome,chromatin accessibility and DNA methylation patterns.In vivo chimera formation assays show that these cells have embryonic and extraembryonic developmental potentials at the single-cell level.Moreover,TPS cells can be induced into blastocyst-like structures resembling preimplantation mouse blastocysts.Mechanistically,inhibition of HDAC1/2 and DOT1L activity and activation of RARγsignaling are important for inducing and maintaining totipotent features of TPS cells.Our study opens up a new path toward fully capturing totipotent stem cells in vitro.Yaxing Xu Jingru Zhao Yixuan Ren Xuyang Wang Yulin Lyu Bingqing Xie Yiming Sun Xiandun Yuan Haiyin Liu Weifeng Yang Yenan Fu Yu Yu Yinan Liu Rong Mu Cheng Li Jun Xu Hongkui Deng 2022Cell Research2022,32,6:1
6Numerical study on the effects of progressive gravity waves on turbulence显示文摘The wind-wave-ocean system, which contains complex interactive processes, is of great importance for the momentum, heat and mass transport in the atmosphere and ocean and at their interface. In this work, we perform wave-coupled phase-resolved numerical simulations to investigate the effect of progressive gravity waves on wind and ocean turbulence. Initially homogeneous turbulence under a finite-amplitude monochromatic surface wave is simulated to reveal how the wave influences the subsurface turbulence. For the interaction between wind-driven waves and shear turbulence in the ocean, new wave-phase-resolved simulation approaches are developed to capture Langmuir cells.
