1Effect of Sancaijiangtang on plasma nitric oxide and endothelin-1 levels in patients with type 2 diabetes mellitus and vascular dementia:a single-blind randomized controlled trial显示文摘OBJECTIVE: To observe the effect of Sancaijiangtang powders on plasma nitric oxide and endothelin-1 levels. We sought to identify the common pathological link and mechanism of action for Traditional Chinese medicine in type 2 diabetes mellitus and vascular dementia,and to explicate the material basis for treating the different diseases with the same method in Traditional Chinese Medicine.METHODS: In total,168 patients with type 2 diabetes mellitus and vascular dementia were enrolled in the study,and randomly divided into two groups by simple randomization. Patients in the treatment group received oral Sancaijiangtang powders with pioglitazone hydrochloride three times daily,while patients in the control group received pioglitazone hydrochloride alone. The treatment course was for12 weeks. Mini-mental state examinations(Chinese version) and Montreal Cognitive Assessments(Beijing version) were performed,and fasting plasma glucose,fasting insulin,hemoglobin A1 c,homeostasis model assessment of insulin resistance,plasma nitric oxide and endothelin-1 levels were measured before and after the treatment.RESULTS: The post-treatment levels for all measurements in both groups were better than pre-treatment levels(P < 0.05). The post-treatment levels for all measurements in the treatment group were better than the levels measured in the control group(P < 0.05).CONCLUSION: Type 2 diabetes mellitus and vascular dementia have common pathological mechanisms for insulin resistance and endothelium dysfunction. Sancaijiangtang powders could improve the release of nitric oxide and inhibit the secretion of endothelin-1. 2How does the motor relearning program improve neurological function of brain ischemia monkeys?显示文摘The motor relearning program can significantly improve various functional disturbance induced by ischemic cerebrovascular diseases. However, its mechanism of action remains poorly understood. In injured brain tissues, glial fibrillary acidic protein and neurofilament protein changes can reflect the condition of injured neurons and astrocytes, while vascular endothelial growth factor and basic fibroblast growth factor changes can indicate angiogenesis. In the present study, we induced ischemic brain injury in the rhesus macaque by electrocoagulation of the M1 segment of the right middle cerebral artery. The motor relearning program was conducted for 60 days from the third day after model establishment. Immunohistochemistry and single-photon emission CT showed that the numbers of glial fibrillary acidic protein-, neurofilament protein-, vascular endothelial growth factor- and basic fibroblast growth factor-positive cells were significantly increased in the infarcted side compared with the contralateral hemisphere following the motor relearning program. Moreover, cerebral blood flow in the infarcted side was significantly improved. The clinical rating scale for stroke was used to assess neurological function changes in the rhesus macaque following the motor relearning program. Results showed that motor function was improved, and problems with consciousness, self-care ability and balance function were significantly ameliorated. 3Gut microbiota drives macrophage-dependent self-renewal of intestinal stem cells via niche enteric serotonergic neurons显示文摘Lgr5+intestinal stem cells(ISCs)reside within specialized niches at the crypt base and harbor self-renewal and differentiation capacities.ISCs in the crypt base are sustained by their surrounding niche for precise modulation of self-renewal and differentiation.However,how intestinal cells in the crypt niche and microbiota in enteric cavity coordinately regulate ISC stemness remains unclear.Here,we show that ISCs are regulated by microbiota and niche enteric serotonergic neurons.The gut microbiota metabolite valeric acid promotes Tph2 expression in enteric serotonergic neurons via blocking the recruitment of the NuRD complex onto Tph2 promoter.5-hydroxytryptamine(5-HT)in turn activates PGE2 production in a PGE2+macrophage subset through its receptors HTR2A/3 A;and PGE2 via binding its receptors EP1/EP4,promotes Wnt/β-catenin signaling in ISCs to promote their self-renewal.Our findings illustrate a complex crosstalk among microbiota,intestinal nerve cells,intestinal immune cells and ISCs,revealing a new layer of ISC regulation by niche cells and microbiota.Pingping Zhu Tiankun Lu Jiayi Wu Dongdong Fan Benyu Liu Xiaoxiao Zhu Hui Guo Ying Du Feng Liu Yong Tian Zusen Fan 2022Cell Research2022,32,6:8
4Bacillus cereus AR156 primes induced systemic resistance by suppressing miR825/825 and activating defense-related genes in Arabidopsis显示文摘Small RNAs play an important role in plant immune responses. However, their regulatory function in induced systemic resistance(ISR) is nascent. Bacillus cereus AR156 is a plant growth-promoting rhizobacterium that induces ISR in Arabidopsis against bacterial infection. Here,by comparing small RNA profiles of Pseudomonas syringae pv. tomato(Pst) DC3000-infected Arabidopsis with and without AR156 pretreatment, we identified a group of Arabidopsis micro RNAs(mi RNAs) that are differentially regulated by AR156 pretreatment. mi R825 and mi R825 are two mi RNA generated from a single mi RNA gene.Northern blot analysis indicated that they were significantly downregulated in Pst DC3000-infected plants pretreated with AR156, in contrast to the plants without AR156 pretreatment. mi R825 targets two ubiquitin-protein ligases,while mi R825 targets toll-interleukin-like receptor(TIR)-nucleotide binding site(NBS) and leucine-rich repeat(LRR)type resistance(R) genes. The expression of these target genes negatively correlated with the expression of mi R825 and mi R825. Moreover, transgenic plants showing reduced expression of mi R825 and mi R825 displayed enhanced resistance to Pst DC3000 infection, whereas transgenic plants overexpressing mi R825 and mi R825 were more susceptible. 5Effects of laser scanning strategies on selective laser melting of pure tungsten显示文摘Three types of scanning strategies,including the chessboard scanning strategy,the zigzag scanning strategy and the remelting scanning strategy,were conducted to study the effects of scanning strategies on surface morphology,microstructure,mechanical properties and the grain orientation of selective laser melted pure tungsten.The results showed that the pores and cracks were main defects in SLM-processed tungsten parts.The pores could be eliminated using the remelting scanning strategy.However,the cracks seemed to be inevitable regardless of the applied scanning strategies.The microstructures of SLM-processed tungsten were columnar grains and showed strong epitaxial growth along the building direction.A compressive strength of 923 MPa with an elongation of 7.7%was obtained when the zigzag scanning strategy was applied,which was the highest among the three scanning strategies.By changing the scanning strategies,the texture of SLM-processed tungsten in the direction of processing could be changed.Dongdong Gu Meng Guo Hongmei Zhang Yixuan Sun Rui Wang Lei Zhang 2020International Journal of Extreme Manufacturing2020,2,2:6
6Regulative effect of Taohong Siwu decoction(桃红四物汤) on extracellular matrix of endometrium in drug-induced abortion显示文摘OBJECTIVE: To explore the effects of Taohong Siwu decoction(桃红四物汤, THSWD) on the extracellular matrix of endometrium in rats following drug-induced abortion.METHODS: Thirty-six pregnant female rats were administered mifepristone and misoprostol to induce abortion, and amounts of uterine bleeding were recorded. Pathological damage and collagen accumulation were detected by hematoxylin-eosin staining and Masson’s trichrome staining in uterus, respectively. Myeloperoxidase was evaluated by immunohistochemistry.The expression levels of fibronectin, laminin, matrix metalloproteinase 9(MMP-9), and tissue inhibitor of metalloproteinase 1(TIMP-1) were quantified using western blotting.RESULTS: THSWD could promote endometrial protection in rats following drug-induced abortion.The contents of cellulose and myeloperoxidase were significantly decreased in uterine tissue of THSWD-treated groups. Moreover, THSWD significantly decreased the expression levels of fibronectin, laminin, and TIMP-1. THSWD also significantly increased MMP-9 expression and the MMP-9/TIMP-1 ratio.CONCLUSION: THSWD plays a critical role in endometrial protection by reducing extracellular matrix deposition and uterine fibrosis. 7Preparation of 5-fluorouracil-loaded chitosan nanoparticles and study of the sustained release in vitro and in vivo显示文摘The sustained-release properties of the biodegradable nano-drug delivery systems were used to improve the residence time of the chemotherapeutic agent in the body. These drug delivery systems were widely used to deliver chemotherapeutic drugs. The 5-fluorouracil loaded chitosan nanoparticles prepared in this paper have the above advantage. Here, we found that when the mass ratio of 5-fluorouracil and chitosan was 1:1, the maximum drug loading of nanoparticles was 20.13 ± 0.007%, the encapsulation efficiency was 44.28 ± 1.69%, the particle size was 283.9 ± 5.25 nm and the zeta potential was 45.3 ± 3.23 mV. The prepared nanoparticles had both burst-release and sustained-release phases in vitro release studies.In addition, the inhibitory effect of the prepared nanoparticles on gastric cancer SGC-7901 cells was similar to that of 5-fluorouracil injection, and the blank vector had no obvious inhibitory effect on SGC-7901 cells. 8Microbial population dynamics and changes in main nutrients during the acidification process of pig manures显示文摘This study evaluated the impact of pig manure acidification on anaerobic treatment and composition of the fecal microbial community.According to the different chemical oxygen demand (COD) in the anaerobic treatment processes,pig manure was diluted 4 times (×4),16 times (×16),or 64 times (×64) and subjected to acidification.During the acidification process,pH,soluble chemical oxygen demand (SCOD),volatile fatty acids (VFAs),nitrogen (N),phosphorus (P) and potassium (K) were determined along with microbial population dynamics.The pH of the three dilutions first declined,and then slowly increased.The total VFAs of ×4 and ×16 dilutions peaked on day 15 and 20,respectively.The content of acetic acid,propanoic acid,butanoic acid and valeric acid of the × 4 dilution were 23.6,11.4,8.8 and 0.6 g/L respectively,and that of the ×16 dilution was 5.6,2.3,0.9 and 0.2 g/L respectively.Only acetic acid was detected in the ×64 dilution,and its level peaked on day 10.The results showed that the liquid pig manure was more suitable to enter the anaerobic methanogenic bioreactors after two weeks of acidification.During the acidification process,total P concentration increased during the first ten days,then dropped sharply,and rose again to a relatively high final concentration,while total N concentration rose initially and then declined.Based on the analysis of denaturing gradient gel electrophoresis (DGGE) and 16S rRNA gene clone library,we concluded that the acidification process reduced the number of pathogenic bacteria species in pig manure.Dongdong Zhang Xufeng Yuan Peng Guo Yali Suo Xiaofen Wang Weidong Wang Zongjun Cui 2011Journal of Environmental Sciences2011,23,3:4
9Hd1,Ghd7,and DTH8 synergistically determine the rice heading date and yield-related agronomic traits显示文摘Heading date determines the seasonal and regional adaptation of rice(Oryza sativa L.)varieties and is mainly controlled by photoperiod sensitivity(PS).The core heading date genes Hd1,Ghd7,DTH8,and PRR37 act synergistically in regulating the PS.In this study,we systematically analyze the heading date,PS,and agronomic traits of eight homozygous lines with various combinations of Hd1,Ghd7,and DTH8 alleles in the prr37 background under long-day(LD)and short-day(SD)conditions,respectively.We find that Hd1 alone promotes heading,regardless of the day length.However,under LDs,Hd1 suppresses flowering,in coordination with functional Ghd7 or with Ghd7 and DTH8.These loci cooperate to negatively regulate the Ehd1-Hd3 a/RFT1 pathway and delay heading.Under SDs,Hd1 competes with various heading suppressors to promote heading.Therefore,the dual function of Hd1 is vital for PS.The lines carrying Hd1 alone show reduced plant height with fewer primary and secondary branches in panicles.Lines carrying Ghd7 and DTH8(with hd1)show delayed heading and improve agronomic traits.Overall,our results reveal the regulation of rice PS flowering by the core heading date genes and their effects on agronomic traits,providing valuable information for the selection of rice varieties for adaptation to different light and temperature conditions.Kangli Sun Minghui Huang Wubei Zong Dongdong Xiao Chen Lei Yanqiu Luo Yingang Song Shengting Li Yu Hao Wanni Luo Bingqun Xu Xiaotong Guo Guangliang Wei Letian Chen Yao-Guang Liu Jingxin Guo 2022Journal of Genetics and Genomics2022,49,5:4
10A parameterization scheme for upward lightning in the cloud model and a discussion of the initial favorable environmental characteristics in the cloud显示文摘The upward lightning(UL) initiated from the top of tall buildings(at least above 100 m) is a type of atmospheric discharge. Currently, we understand the nature of the UL from ground observations, but the corresponding theoretical research is lacking. Based on an existing bidirectional leader stochastic model, a stochastic parameterization scheme for the UL has been built and embedded in an existing two-dimensional thundercloud charge/discharge model. The ULs simulated from the experiments with two-dimensional high resolution agree generally with the observation results. By analyzing the charge structure of thunderstorm clouds, we determined the in-cloud environmental characteristics that favor the initiation of conventional cloud-to-ground(CG) flashes and analyzed the differences and similarities of some characteristics of the positive and the negative UL. Simulation results indicate that the positive ULs are typically other-lightning-triggered ULs(OLTUL) and are usually a discharge phenomenon between the ground and the lower positive charge region appearing below the main middle negative charge region. The effect of the previous in-cloud lightning(IC) process of space electrical field provides favorable conditions for the initiation of a positive UL. Its entire discharge process is limited, and the branches of the leader are fewer in number as its discharge is not sufficient. A negative UL is generally a discharge phenomenon of the dipole charge structure between the ground and the main negative charge region. The lower temperature stratification and the sinking of the hydrometeors typically initiate a negative UL. Negative ULs develop strongly and have more branches. 11Discovery of Late Carboniferous strata and their tectonic significance in the southern margin of the North China Block(Huoqiu and Gushi areas)显示文摘Late Carboniferous fossils(such as Boultonia? sp., Tabulata, and spiriferoid specimens with smooth shells), bioclastic material(such as crinoid stems and sponge fragments), and Late Ordovician microfossils of the conodont Belodina have been discovered in the lower part of the strata typically referred to as the Neoproterozoic on the boundary of the provinces of Anhui and Henan in the southern margin of the North China Block. These findings prove that the strata contain macrofossils belonging to the Late Carboniferous, which belonged to a carbonate debris flow deposit that was formed under a carbonate slope environment. The conodont fossils might belong to a detrital deposit. 12Fast mapping of a chlorophyll b synthesis-deficiency gene in barley(Hordeum vulgare L.) via bulked-segregant analysis with reduced-representation sequencing显示文摘Bulked-segregant analysis coupled with next-generation sequencing(BSA-seq) has emerged as an efficient tool for genetic mapping of single genes or major quantitative trait loci controlling(agronomic) traits of interest. However, such a mapping-by-sequencing approach usually relies on deep sequencing and advanced statistical methods. Application of BSA-Seq based on construction of reduced-representation libraries and allele frequency analysis permitted anchoring the barley pale-green(pg) gene on chromosome 3 HL. With further marker-assisted validation, pg was mapped to a 3.9 Mb physical-map interval. In the pg mutant a complete deletion of chlorophyllide a oxygenase(HvCAO) gene was identified.Because the product of this gene converts Chl a to Chl b, the pg mutant is deficient in Chl b.An independent Chl b-less mutant line M4437_2 carried a nonsynonymous substitution(F263 L) in the C domain of HvCAO. 15The Development Characteristics and Distribution Predictions of the Paleogene Coal-measure Source Rock in the Qiongdongnan Basin,Northern South China Sea显示文摘There are known to be enormous Cenozoic coal-type oil and gas resources located in the basins of the South China Sea,among which the Paleogene coal-measure source rock are one of the main source rock.In order to more effectively analyze the distribution laws of coal-measure source rock in marginal sea basins and guide coal-type oil and gas explorations,the Oligocene coal-measure source rock in the Qiongdongnan Basin were selected as examples in order to systematically analyze the types,development characteristics,control factors,and distribution prediction methods of coalmeasure source rock in marginal sea basins.The Qiongdongnan Basin is located in the northern region of the South China Sea.Previous explorations of the area have determined that the Oligocene coal-measure source rock in the Qiongdongnan Basin have typical'binary structures',which include coal seams and terrigenous marine mudstone.Among those,the terrigenous marine mudstone has been found to greatly expand the scope of the coal-measure source rock.In addition,the coal seams which have been exposed by drilling have been observed to have the characteristics of thin single layer thickness,many layers,and poor stability.Meanwhile,the terrigenous marine mudstone has the characteristics of large thickness and wide distribution.The development of coal-measure source rock is known to be controlled by many factors,such as paleoclimate and paleobotany condition,paleo-structure and topography,paleo-geography,rise and fall of base level,and so on.In accordance with the comprehensive analyses of various control factors of coal-measure source rock,and the changes in water body energy in sedimentary environments,a genetic model of the'energy belt'for the development and distribution of coal-measure source rock was proposed.Also,the development and distribution characteristics of coalmeasure source rock in different types of energy belts were clarified.Then,based on the development and distribution characteristics of coal-measure source rock and their controlling factors,prediction methods of the distribution ranges of coal-measure source rock were proposed from both qualitative and(semi-)quantitative aspects.That is to say,a method for(semi-)quantitative predictions of the distribution ranges of coal-bearing intervals based on model wave impedance inversion and neural network wave impedance inversion,along with a method for(semi-)quantitative predictions of the distribution ranges of terrigenous marine mudstone based on topographical slopes,were introduced in this study.WANG Dongdong ZHANG Gongcheng LI Zengxue DONG Guoqi SUN Rui GUO Shuai WU Yue 2021Acta Geologica Sinica(English Edition)2021,95,1:3
16Genetic causes of oil-rich and oil-poor reservoirs: Implications from two Cenozoic basins in the eastern North China Craton显示文摘The Bohai Bay and Hehuai(southern North China) rift basins in the eastern part of the North China Craton are southnorth-adjacent. They have shown synchronous evolutionary processes, and possess generally identical superficial and shallow structural characteristics as well as similar basin areas. However, there is a large difference in the richness of oil resources between the two basins. The Bohai Bay Basin has extremely abundant oil reserves, while commercial oil reserves have not been found in the Hehuai Basin. The deep tectonic structures, magmatic activities, and modern and paleo-geothermal fields of the two basins are significantly different. Compared with the Hehuai Basin, the Bohai Bay Basin has a thinner crust and more complex structure with multiple low-velocity layers. It is also characterized by intense magmatic activity, high modern and paleogeothermal fields, frequent seismic activity, and active deep interactions, small effective elastic thickness of the isotropic lithosphere, and shorter balanced transformation wavelength of the lithosphere with a high likelihood of local compensation. The Hehuai Basin has a simple deep structure and homogeneous crustal composition, with a high likelihood of regional compensation. The characteristics of the deep structures mentioned above are generally similar to those of the southern part of the stable Ordos Basin, except for the smaller crust thickness. This indicates the presence of differences in Mesozoic destruction between the southern and northern zones in the eastern part of the North China Craton. The northern zone was subjected to significant destruction, while the southern zone was subjected to modifications, primarily in the form of local changes in the structures and/or properties of the crust or lithospheric mantle, with the overall structure and stability of the craton kept intact. The formation of high-quality source rock is primarily influenced by the abnormal flourishment of organisms in water bodies during the syndepositional period, and is also strongly associated with the high geothermal setting of basins and nutrients from hydrothermal solutions and volcanoes. In other words, it is mainly controlled by deep processes and deep-major fault activity. The differences in the deep structures and modern and paleo geothermal fields of the two basins correspond to the difference in richness of oil resources, suggesting that there is an important internal or causal relationship between the two aspects. This viewpoint coincides with the conditions and environments required for the development of high-quality source rock in hydrocarbon-rich basins(sags)in China and other countries, and is evidenced by the modern lake basin of the East African Rift. 18Sparse channel recovery with inter-carrier interference self-cancellation in OFDM显示文摘A new sparse channel estimation method of orthogonal frequency division multiplexing(OFDM) system based on intercarrier interference(ICI) self-cancellation is investigated. Firstly,based on the characteristic that the ICI generated by a subcarrier to the two adjacent subcarriers is approximately equal, a data pair with opposite sign and equal magnitude is modulated onto two adjacent subcarriers as pilot pair to eliminate the effect of ICI on pilots. Secondly, a new OFDM channel estimation model based on linear time-varying(LTV) model and compressed sensing(CS) is constructed, which obtains the mean of the gains of the multipath.Finally, a pilot pair optimization algorithm based on two layers loop is used to realize the minimization of the mutual coherence of the measurement matrix. For time-varying channel scenes with different numbers or delay of multipath and maximum Doppler frequency shift, the performances of several channel estimation methods are verified by simulation.
