| 1 | CDH13 Genetic Polymorphisms, Adiponectin and Ischemic Stroke: a Chinese Family-based Sib-pair Study显示文摘Objective To understand the relationships between CDH13(T-cadherin) genetic polymorphisms, adiponectin levels and ischemic stroke, and possible interactions between CDH13 polymorphisms and other risk factors. Methods We recruited 342 Chinese ischemic stroke sib pairs. We genotyped rs4783244 and rs7193788 on CDH13 using time-of-flight mass spectrometry genotyping technology and measured total and high-molecular weight(HMW) adiponectin levels. We investigated associations between SNPs and ischemic stroke, and interactions between SNPs and other risk factors using multi-level mixed-effects regression model. Results In individuals without ischemic stroke, CDH13 rs4783244 was associated with total adiponectin levels(per T: Coef =-0.257, P = 0.001). CDH13 rs7193788 was associated with total adiponectin levels(per A: Coef =-0.221, P = 0.001) and HMW adiponectin levels(per A: Coef =-0.163, P = 0.003). rs7193788 was significantly associated with ischemic stroke(GA/AA vs. GG: OR = 1.55, 95% CI: 1.07 to 2.24, P = 0.020) after Bonferroni correction(α = 0.025). There was an interaction between rs7193788 and diabetes(P = 0.036). Compared to diabetes-free individuals with rs7193788 GG genotype, diabetes patients with rs7193788 GA/AA genotypes had higher risks for ischemic stroke(OR = 2.64, 95% CI: 1.58-4.40, P < 0.001). Conclusion CDH13 genetic polymorphisms are associated with adiponectin levels and ischemic stroke. | 3 | Identifying disease feature genes based on cellular localized gene functional modules and regulation networks显示文摘Identifying disease-relevant genes and functional modules, based on gene expression pro- files and gene functional knowledge, is of high im- portance for studying disease mechanisms and sub- typing disease phenotypes. Using gene categories of biological process and cellular component in Gene Ontology, we propose an approach to selecting func- tional modules enriched with differentially expressed genes, and identifying the feature functional modules of high disease discriminating abilities. Using the differentially expressed genes in each feature module as the feature genes, we reveal the relevance of the modules to the studied diseases. Using three data- sets for prostate cancer, gastric cancer, and leukemia, we have demonstrated that the proposed modular approach is of high power in identifying functionally integrated feature gene subsets that are highly rele- vant to the disease mechanisms. Our analysis has also shown that the critical disease-relevant genes might be better recognized from the gene regulation network, which is constructed using the characterized functional modules, giving important clues to the concerted mechanisms of the modules responding to complex disease states. combinations are proposed where necessary to improve our traditional species concepts and provide updates on their classification.In addition to extensive morphological descriptions and appropriate asexual and sexual connections,DNA sequence data are also analysed from concatenated datasets(rDNA,TEF-a,RBP2 and b-Tubulin)to infer phylogenetic relationships and substantiate systematic position of taxa within appropriate ranks.Wherever new species or combinations are being proposed,we apply an integrative approach(morphological and molecular data as well as ecological features wherever applicable).Notes on 125 fungal taxa are compiled in this paper,including eight new genera,101 new species,two new combinations,one neotype,four reference specimens,new host or distribution records | 15 | HACE1-mediated NRF2 activation causes enhanced malignant phenotypes and decreased radiosensitivity of glioma cells显示文摘HACE1,an E3 ubiquitin-protein ligase,is frequently inactivated and has been evidenced as a putative tumor suppressor in different types of cancer.However,its role in glioma remains elusive.Here,we observed increased expression of HACE1 in gliomas related to control subjects,and found a strong correlation of high HACE1 expression with poor prognosis in patients with WHO grade III and IV as well as low-grade glioma(LGG)patients receiving radiotherapy.HACE1 knockdown obviously suppressed malignant behaviors of glioma cells,while ectopic expression of HACE1 enhanced cell growth in vitro and in vivo.Further studies revealed that HACE1 enhanced protein stability of nuclear factor erythroid 2-related factor 2(NRF2)by competitively binding to NRF2 with another E3 ligase KEAP1.Besides,HACE1 also promoted internal ribosome entry site(IRES)-mediated mRNA translation of NRF2.These effects did not depend on its E3 ligase activity.Finally,we demonstrated that HACE1 dramatically reduced cellular ROS levels by activating NRF2,thereby decreasing the response of glioma cells to radiation.Altogether,our data demonstrate that HACE1 causes enhanced malignant phenotypes and decreased radiosensitivity of glioma cells by activating NRF2,and indicate that it may act as the role of prognostic factor and potential therapeutic target in glioma. | Chenxing Da Jun Pu Zhe Liu Jing Wei Yiping Qu Yongxing Wu Bingyin Shi Jian Yang Nongyue He Peng Hou | 2021 | Signal Transduction and Targeted Therapy2021,6,12: | 1 |

| 18 | Joint Acaricidal Action of Diafenthiuron and Fenbutatin-oxide Against Tetranychus cinnabarinus(Boisduval)(Acari: Tetranychidae)显示文摘[Objective]This study was conducted to obtain a new high-efficiency acaricide mixture.[Method]The synergism acaricidal of diafenthiuron and fenbutatin-oxide against carmine spider mite was investigated,and the optimal ratio of both acaricides were determined by means of co-toxicity factors and co-toxicity coefficients(CTC).[Result]At 24 h post-treatment,the LC50values of diafenthiuron and fenbutatin-oxide against T.cinnabarinus female adults were 154.67 and 93.26 mg/L,respectively,and the synergistic mass ratios of diafenthiuron and fenbutatin-oxide ranged from 1:0.06 to 1:0.87,and the optimal diafenthiuron-to-fenbutatin-oxide ratio was 1:0.39 with the LC50and CTC of 67.87 mg/L and 188.93(CTC significantly above 100 strongly indicated synergism),respectively,and the toxicity relative to diafenthiuron and fenbutatin-oxide was 2.28 and 1.37 times,respectively.[Conclusion]The joint acaricidal action of diafenthiuron and fenbutatin-oxide strongly indicates synergism.These findings proved scientific theoretical basis for mixing diafenthiuron and fenbutatin-oxide to control spider mites and developing a new synergisic acaricide. | Zhenguo YANG Daoyan XIE Aisi DA Xiujun JIANG Jing NI Ping HUANG Jianping CHAI Yanjie LUO | 2014 | Agricultural Science & Technology2014,15,12: | 0 |

The surface composition of implanted electrode and concentration-depth profiles of various elements were measured by AES. The chemical state of Pt in glassy carbon electrode implanted with platinum (Pt/GC) was detected by X-ray Photoelectron Spectroscopy (XPS). The electro-oxidation of HCOOH and HCHO have been investigated on Pt/GC and smooth Pt electrodes. | 20 | GTPase-activating protein TBC1D5 coordinates with retromer to constrain synaptic growth by inhibiting BMP signaling显示文摘Formation and plasticity of neural circuits rely on precise regulation of synaptic growth.At Drosophila neuromuscular junction(NMJ),Bone Morphogenetic Protein(BMP)signaling is critical for many aspects of synapse formation and function.The evolutionarily conserved retromer complex and its associated GTPase-activating protein TBC1D5 are critical regulators of membrane trafficking and cellular signaling.However,their functions in regulating the formation of NMJ are less understood.Here,we report that TBC1D5 is required for inhibition of synaptic growth,and loss of TBC1D5 leads to abnormal presynaptic terminal development,including excessive satellite boutons and branch formation.Ultrastructure analysis reveals that the size of synaptic vesicles and the density of subsynaptic reticulum are increased in TBC1D5mutant boutons.Disruption of interactions of TBC1D5 with Rab7 and retromer phenocopies the loss of TBC1D5.Unexpectedly,we find that TBC1D5 is functionally linked to Rab6,in addition to Rab7,to regulate synaptic growth.Mechanistically,we show that loss of TBC1D5 leads to upregulated BMP signaling by increasing the protein level of BMP type Ⅱ receptor Wishful Thinking(Wit)at NMJ.Overall,our data establish that TBC1D5 in coordination with retromer constrains synaptic growth by regulating Rab7 activity,which negatively regulates BMP signaling through inhibiting Wit level. | Xiu Zhou Guangming Gan Yichen Sun Mengzhu Ou Junhua Geng Jing Wang Xi Yang Shu Huang Da Jia Wei Xie Haihuai He | 2023 | Journal of Genetics and Genomics2023,50,3: | 0 |
