2Indoor Allergen Levels and Household Distributions in Nine Cities Across China显示文摘Objective Chinese allergic subjects have high levels of sensitization to house dust mite(HDM) and other indoor allergens. This study quantifies common indoor allergen levels in Chinese households. Methods Dust samples were collected from nine cities. Major allergens Der p 1 and Der f 1 from Dermatophagoides pteronyssinus and D. farinae, and specific antigens of Blomia tropicalis, Tyrophagus putrescentiae, Acarus siro, and cockroach species Blattella germanica and Periplaneta americana were measured by ELISA. Results HDM allergens were found in dust samples from bedding in 95% of the Chinese households. The median levels varied from <0.006 to 9.2 μg/g of dust, depending on the city. The percentages of households having HDM allergen levels associated with the risk of developing allergy sensitization and asthma were 65% and 25%, respectively. Specific antigens of the storage mite and cockroach were only found in samples from the southern and tropical regions of China. Levels of mite allergens were generally higher in samples from bedding compared to samples from the living room, even for storage mites, whereas levels of cockroach antigens were higher in the living room samples. Conclusion HDM allergens are present in bedding dust samples from most Chinese households. Cities in southern and central China have relatively high levels of HDM major allergens compared to cities in northern and western China. 3Small molecule inhibitors reveal allosteric regulation of USP14 via steric blockade显示文摘The ubiquitin system is important for drug discovery,and the discovery of selective small-molecule inhibitors of deubiquitinating enzymes (DUBs) remains an active yet extremely challenging task. With a few exceptions,previously developed inhibitors have been found to bind the evolutionarily conserved catalytic centers of DUBs,resulting in poor selectivity. The small molecule IU1 was the first-ever specific inhibitor identified and exhibited surprisingly excellent selectivity for USP14 over other DUBs. However,the molecular mechanism for this selectivity was elusive. Herein,we report the high-resolution co-crystal structures of the catalytic domain of USP14 bound to IU1 and three IU1 derivatives. All the structures of these complexes indicate that IU1 and its analogs bind to a previously unknown steric binding site in USP14,thus blocking the access of the C-terminus of ubiquitin to the active site of USP14 and abrogating USP14 activity. Importantly,this steric site in USP14 is very unique,as suggested by structural alignments of USP14 with several known DUB X-ray structures. These results,in conjunction with biochemical characterization,Indicate a coherent steric blockade mechanism for U5P14 inhibition by compounds of the IU series. 4A new bone repair scaffold combined with chitosan/ hydroxyapatite and sustained releasing icariin显示文摘Icariin, a plant-derived flavonol glycoside, has been proved as an osteoinductive agent for bone tissue engineering. A new bone repair scaffold was generated by thorough mixing of icariin and chitosan/ hydroxyapatite (icariin-CS/HA) using freeze-drying technigue. Characteristics of morphology, mechanical properties, biocompatibility, drug release behavior and bone repair abilities in vivo were evaluated. The results show that drug loading process of icariin did not affect physical structure of CS/HA composite significantly but decreased mechanical properies of CS/HA composite, which happened with a high dosage; icariin-CS/HA had favorable cell compatibility and promoted osteogenic differentiation of hBMSCs; the controlled release of icariin was satisfactory and the release retained after 90 d in vitro. In addition, icariin-CS/HA scaffolds had favorable osteoconduction and osteoinduction in vivo, and could fill bone defect sites and stimulate newborn bone tissues formation at early stage. 5Effects of operational factors on soluble microbial products in a carrier anaerobic baffled reactor treating dilute wastewater显示文摘The effects of feed strength, hydraulic residence time (HRT), and operational temperatures on soluble microbial product (SMP) production were investigated, to gain insights into the production mechanism. A carrier anaerobic baffled reactor (CABR) treating dilute wastewater was operated under a wide range of operational conditions, namely, feed strengths of 300–600 mg/L, HRTs of 9– 18 h, and temperatures of 10–28°C. Generally, SMP production increased with increasing feed strength and decreasing temperature. At high temperature (28°C), SMP production increased with decreasing HRT. As the temperature was decreased to 18 and 10°C, the SMP production was at its peak for 12 h HRT. Therefore, temperature could be an important determinant of SMP production along with HRT. A higher SMP to soluble chemical oxygen demand (SCOD) ratio was found at high temperature and long HRT because of complete volatile fatty acid degradation. SMP accounted for 50%–75% of the SCOD in the last chamber of the CABR. 6中国5地无偿献血者HCV流行特征分析显示文摘目的了解我国献血者丙型肝炎病毒(HCV)的初筛阳性率和确证阳性率。方法采用2种不同ELA试剂对2013年6月-2016年12月我国5家血液中心/中心血站献血者血液标本进行ALT、HBsAg、抗-HCV、抗-HIV、抗-TP进行常规筛查,抗-HCV单试剂或双试剂筛查阳性标本进一步采用MP HCV Blot 3. 0进行确证检测。分别估算首次献血者的HCV流行率及重复献血者的HCV发病率,并采用χ~2检验比较不同人群体、不同献血类型之间HCV确证阳性率。结果在1 276 544人次献血样本中,有3 855 (0. 30%)为抗-HCV ELISA初筛反应性,其中只有30. 18%(904/2 995,)确证为抗-HCV阳性。本次研究中,首次献血者总计648607人次,其中1080份样本确证为抗-HCV阳性,即首次献血者HCV流行率为166. 56 (156. 04—177. 08)/100 000人次,年龄、受教育水平、职业均与HCV确证阳性率相关。重复献血者总计627 937人次,其中126份样本确证为抗-HCV阳性,即重复献血者HCV发病率为13. 04(9. 55—17. 81)/100 000人次,其中重复献血者抗-HCV确证阳性率明显高于单采血小板献血者(0. 02%vs0. 01%)。结论本研究表明我国献血者HCV仍处于较高水平流行状态。尽管现在NAT已成为我国血液检测的常规检测项,但仍需持续有效地对我国输血HCV传播风险进行监测和控制。同时,新的规范中规定可采用1种或2种EIA进行筛查,本意是为了提高检测敏感性,但本研究中EIA筛查阳性样本如此低的确证阳性率提示其特异性差,可能会造成许多合格的献血者丢失。因此,优化献血者HCV筛查策略将有助于防止血液浪费。付萍 Liao Dan 吕运来 张洪斌 何伟兰 黄梅 文秀琼 马红丽 何涛 柯玲 吴秉婷 刘鱼 何苗 Liu Jing Don Brambill 王憬惺 Paul Ness Shan Hua 2018中国输血杂志2018,31,9:10
7Effect of corrosive media on galvanic corrosion of complicated tri-metallic couples of 2024 Al alloy/Q235 mild steel/304 stainless steel显示文摘Galvanic corrosion of tri-metallic couples is more complicated than that of bi-metallic couples. In this study, the effect of the pH of corrosive media on the galvanic corrosion of 2024 A1 alloy/Q235 mild steel/304 stainless steel tri-metallic couples was investigated using potentiodynamic polarization, scanning electron microscopy, scanning vibrating electrode technique and a multi-channel galvanic corrosion meter. The results show that 2024 always acts as the only anode in 3.5 wt% NaCl at pH 5.56,9.72 and 12.0, while both Q235 and 2024 act as anodes at pH 2.39 in the initial stage and then the role of Q235 changes at longer coupling time, which can be attributed to the effect of pH on the surface film of 2024. It is also found that the galvanic current density of a tri-metallic couple is the superposition of two bi-metallic couples when cathodic reactions are controlled by the diffusion of oxygen, otherwise it is smaller than that of the sum of two bi-metallic couples. 9Separate lateral parametrial lymph node dissection improves detection rate of parametrial lymph node metastasis in early-stage cervical cancer: 10-year clinical evaluation in a single center in China显示文摘Objective: To investigate the clinical significance of separate lateral parametrial lymph node dissection(LPLND) in improving parametrial lymph node(PLN) and its metastasis detection rate during radical hysterectomy for early-stage cervical cancer.Methods: From July 2007 to August 2017, 2,695 patients with cervical cancer in stage IB1-IIA2 underwent radical hysterectomy were included. Of these patients, 368 underwent separate dissection of PLNs using the LPLND method, and 2,327 patients underwent conventional radical hysterectomy(CRH). We compared the surgical parameters, PLN detection rate and PLN metastasis rate between the two groups.Results: Compared with CRH group, the rate of laparoscopic surgery was higher(60.3% vs. 15.9%, P<0.001),and the blood transfusion rate was lower(19.0% vs. 29.0%, P<0.001) in the LPLND group. PLNs were detected in 356 cases(96.7%) in the LPLND group, and 270 cases(11.6%) in the CRH group(P<0.001), respectively. The number of PLNs detected in the LPLND group was higher than that in the CRH group(median 3 vs. 1, P<0.001).The PLN metastases were detected in 25 cases(6.8%) in the LPLND group, and 18 cases(0.8%) in the CRH group(P<0.001), respectively. In multivariable analysis, LPLND is an independent factor not only for PLN detection [odds ratio(OR)=228.999, 95% confidence interval(95% CI): 124.661-420.664;P<0.001], but also for PLN metastasis identification(OR=10.867, 95% CI: 5.381-21.946;P<0.001).Conclusions: LPLND is feasible and safe. 10Effects of Temperature and Hydraulic Residence Time (HRT) on Treatment of Dilute Wastewater in a Carrier Anaerobic Baffled Reactor显示文摘Objective To examine the effect of hydraulic residence time (HRT) on the performance and stability, to treat dilute wastewater at different operational temperatures in a carrier anaerobic baffled reactor (CABR), and hence to gain a deeper insight into microbial responses to hydraulic shocks on the base of the relationships among macroscopic performance, catabolic intermediate, and microcosmic alternation. Methods COD, VFAs, and microbial activity were detected with constant feed strength (300 mg/L) at different HRTs (9-18 h) and temperatures (10℃-28℃) in a CABR. Results The removal efficiencies declined with the decreases of HRTs and temperatures. However, the COD removal load was still higher at short HRT than at long HRT. Devastating reactor performance happened at temperature of 10℃ and at HRT of 9 h. HRTs had effect on the VFAs in the reactor slightly both at high and low temperatures, but the reasons differed from each other. Microbial activity was sensitive to indicate changes of environmental and operational parameters in the reactor. 11Hyperthyroid heart disease in pregnancy: Retrospective analysis of a case series and review of the literature显示文摘BACKGROUND Hyperthyroidism in pregnancy may pose a great threat to maternal and fetal health.The risk of hyperthyroid heart disease(HHD),even heart failure,is significantly elevated in pregnant women.AIM To investigate the clinical characteristics,prognosis,and therapy of HHD in pregnant women.METHODS We searched the patient registry data at West China Second University Hospital of Sichuan University in Chengdu,China,following the approval by the Ethics Committee.We retrospectively analyzed the clinical characteristics of pregnant women diagnosed with HHD.The medical records of women with HHD during pregnancy from January 2012 to December 2017 were obtained from the electronic medical records system.All the included patients were followed in outpatient clinics and by telephone interviews until October 2018.RESULTS A total of 155 patients were diagnosed with thyrotoxicosis,of whom six were diagnosed with HHD.Three of them had regular antenatal care.Two patients were complicated with acute heart failure attacks,and one of them had a stillbirth.Both of these patients had a long history of Graves’disease with poor treatment compliance.Treatments of precipitating factors such as the control of infection could relieve the symptoms and prolong gestation for a better prognosis.Hyperthyroid heart failure could be controlled with aggressive diuretics and management of the coexisting complications.Intense monitoring and timely anti-heart failure treatment were crucial in patients with severe cardiac damage.Our findings indicated the importance of regular antenatal care and treatment adherence in patients with hyperthyroidism.CONCLUSION The timely and accurate diagnosis of HHD and the implementation of effective management are important for a better prognosis in pregnant women with HHD.Improvement in patients’awareness of thyrotoxicosis is needed.Dan Shan Yi Bai Qiu-He Chen Yu-Xia Wu Qian Chen Ya-Yi Hu 2019World Journal of Clinical Cases2019,7,19:5
12CHIP regulates bone mass by targeting multiple TRAF family members in bone marrow stromal cells显示文摘Carboxyl terminus of Hsp70-interacting protein(CHIP or STUB1) is an E3 ligase and regulates the stability of several proteins which are involved in different cellular functions. Our previous studies demonstrated that Chip deficient mice display bone loss phenotype due to increased osteoclast formation through enhancing TRAF6 activity in osteoclasts. In this study we provide novel evidence about the function of CHIP. We found that osteoblast differentiation and bone formation were also decreased in Chip KO mice. In bone marrow stromal(BMS) cells derived from Chip^(-/-) mice, expression of a panel of osteoblast marker genes was significantly decreased. ALP activity and mineralized bone matrix formation were also reduced in Chip-deficient BMS cells. We also found that in addition to the regulation of TRAF6, CHIP also inhibits TNFα-induced NF-κB signaling through promoting TRAF2 and TRAF5 degradation. Specific deletion of Chip in BMS cells downregulated expression of osteoblast marker genes which could be reversed by the addition of NF-κB inhibitor. These results demonstrate that the osteopenic phenotype observed in Chip^(-/-) mice was due to the combination of increased osteoclast formation and decreased osteoblast differentiation. 1317α-Ethynylestradiol biodegradation in different river-based groundwater recharge modes with reclaimed water and degradation-associated community structure of bacteria and archaea显示文摘This study investigated 17α-ethynylestradiol(EE2) biodegradation process and primary metabolic pathways associated with community structures of microorganism during groundwater recharge using reclaimed water. The attenuation rate is 1.58 times higher in wetting and drying alternative recharge(WDAR) than in continual recharge(CR). The primary biotransformation pathways of EE2 in WDAR system began with the oxidation of C-17 on ring D to form a ketone group, and D-ring was subsequently hydroxylated and cleaved. In the CR system, the metabolic pathway changed from the oxidation of C-17 on ring D to hydroxylation of C-4 on ring A, and ring A or B subsequently cleaved; this transition was related to DO, and the microbial community structure. Four hundred fifty four pyrosequencing of 16 s r RNA genes indicated that the bacterial communities in the upper layer of the WDAR system were more diverse than those found in the bottom layer of the CR system; this result was reversed for archaea. Unweighted Uni Frac and taxonomic analyses were conducted to relate the change in bacterial community structure to the metabolic pathway. Microorganism community diversity and structure were related to the concentrations of dissolved oxygen, EE2 and its intermediates in the system. Five known bacterial classes and one known archaeal class, five major bacterial genera and one major archaeal genus might be involved in EE2 degradation. 14IL-17-triggered downregulation of miR-497 results in high HIF-1α expression and consequent IL-1β and IL-6 production by astrocytes in EAE mice显示文摘Interleukin 17(IL-17)is increasingly recognized as a key factor that contributes to the pathogenesis of multiple sclerosis(MS)and its experimental mouse autoimmune encephalomyelitis(EAE)model.However,the roles and regulatory mechanisms of IL-17-induced pro-inflammatory cytokine production in EAE mice remain largely unclear.In this study,the expression of IL-17,hypoxia inducible factor-1α(HIF-1α),IL-1β,IL-6 and microRNA-497(miR-497),as well as their intrinsic associations,was investigated using EAE model mice and cultured astrocytes exposed to IL-17 in vitro.We observed markedly increased production of IL-17,HIF-1α,IL-1βand IL-6 in the brain tissues of EAE mice,while the expression and secretion of HIF-1α,IL-1βand IL-6 were also significantly increased when cultured primary astrocytes from mice were stimulated with IL-17.Meanwhile,the expression of miR-497 was downregulated both in vivo and in vitro.Subsequent in vitro experiments revealed that IL-17 induced the production of IL-1βand IL-6 in astrocytes through the upregulation of HIF-1αas a transcriptional factor,indicating that IL-17-mediated downregulation of miR-497 enhanced HIF-1αexpression.Furthermore,astrocytespecific knockdown of IL-17RA and HIF-1αor astrocyte-specific overexpression of miR-497 by infection with different lentiviral vectors containing an astrocyte-specific promotor markedly decreased IL-1βand IL-6 production in brain tissues and alleviated the pathological changes and score of EAE mice.Collectively,these findings indicate that decreased miR-497 expression is responsible for IL-17-triggered high HIF-1αexpression and consequent IL-1βand IL-6 production by astrocytes in EAE mice.Kai Shan Rongrong Pang Chenhui Zhao Xiaomei Liu Wenxing Gao Jing Zhang Dan Zhao Yingwei Wang Wen Qiu 2017Cellular & Molecular Immunology2017,14,11:5
15Small-scale spatial analysis of intermediate and definitive hosts of Angiostrongylus cantonensis显示文摘Background:Angiostrongyliasis is a food-borne parasitic zoonosis.Human infection is caused by infection with the third-stage larvae of Angiostrongylus cantonensis.The life cycle of A.cantonensis involves rodents as definitive hosts and molluscs as intermediate hosts.This study aims to investigate on the infection status and characteristics of spatial distribution of these hosts,which are key components in the strategy for the prevention and control of angiostrongyliasis.Methods:Three villages from Nanao Island,Guangdong Province,China,were chosen as study area by stratified random sampling.The density and natural infection of Pomacea canaliculata and various rat species were surveyed every three months from December 2015 to September 2016,with spatial correlations of the positive P.canaliculata and the infection rates analysed by ArcGIS,scan statistics,ordinary least squares(OLS)and geographically weighted regression(GWR)models.Results:A total of 2192 P.canaliculata specimens were collected from the field,of which 1190 were randomly chosen to be examined for third-stage larvae of A.cantonensis.Seventy-two Angiostrongylus-infected snails were found,which represents a larval infection rate of 6.1%(72/1190).In total,110 rats including 85 Rattus norvegicus,10 R.flavipectus,one R.losea and 14 Suncus murinus were captured,and 32 individuals were positive(for adult worms),representing an infection rate of 29.1%of the definitive hosts(32/110).Worms were only found in R.norvegicus and R.flavipectus,representing a prevalence of 36.5%(31/85)and 10%(1/10),respectively in these species,but none in R.losea and S.murinus,despite testing as many as 32 of the latter species.Statistically,spatial correlation and spatial clusters in the spatial distribution of positive P.canaliculata and positive rats existed.Most of the spatial variability of the host infection rates came from spatial autocorrelation.Nine spatial clusters with respect to positive P.canaliculata were identified,but only two correlated to infection rates.The results show that corrected Akaike information criterion,R2,R2 adjusted andσ2 in the GWR model were superior to those in the OLS model.Conclusions:P.canaliculata and rats were widely distributed in Nanao Island and positive infection has also been found in the hosts,demonstrating that there was a risk of angiostrongyliasis in this region of China.The distribution of positive P.canaliculata and rats exhibited spatial correlation,and the GWR model had advantage over the OLS model in the spatial analysis of hosts of A.cantonensis.Qiu-An Hu Yi Zhang Yun-Hai Guo Shan Lv Shang Xia He-Xiang Liu Yuan Fang Qin Liu Dan Zhu Qi-Ming Zhang Chun-Li Yang Guang-Yi Lin 2018Infectious Diseases of Poverty2018,7,1:4
16Testosterone attenuates pulmonary epithelial inflammation in male rats of COPD model through preventing NRF1-derived NF-κB signaling显示文摘Testosterone deficiency is common in male patients with chronic obstructive pulmonary disease (COPD) and may correlate with the deterioration of COPD. Clinical research suggests that testosterone replacement therapy may slow the COPD progression, but the specific biological pathway remains unclear. In this study, we explored the effect of testosterone on pulmonary inflammation in male COPD rats. The animals were co-treated with lipopolysaccharide (LPS) and cigarette to induce COPD. In COPD rats, nuclear respiratory factor 1 (NRF1) and NF-κB p65 were upregulated. In cigarette smoke extract (CSE)-, LPS-, or the combination of CSE and LPS-treated L132 cells, NRF1 and p65 were also upregulated. Silencing NRF1 resulted in the downregulation of p65. ChIP‒seq, ChIP‒qPCR, and luciferase results showed that NRF1 transcriptionally regulated p65. Both male and female COPD rats showed an upregulated NRF1 level and similar pulmonary morphology. But NRF1 was further upregulated in male castrated rats. Further supplementing testosterone in castrated male rats significantly reduced NRF1, pulmonary lesions, and inflammation. Supplementation of testosterone also reduced the phosphorylation of p65 and IKKβ induced by LPS or CSE in L132 cells. 17Comparison of the corrosion behavior of AM60 Mg alloy with and without self-healing coating in atmospheric environment显示文摘Coatings on the surface of Mg alloys are inevitably damaged during their practical applications,and corrosion can easily initiate from the damaged areas to accelerate the failure of Mg parts.A dual self-healing coating has already been developed to solve this problem in our previous work.Considering the practical application of this coating,it is necessary to investigate its service behavior in atmospheric environment.Thus,the corrosion behavior of AM60 Mg substrate with and without the self-healing coating was compared in Shenyang industrial atmospheric environment.The results show that the enrichment of sediments and rainwater on the scratch areas can accelerate the corrosion of the exposed Mg substrate.The inhibitors can be released from the damaged coating to inhibit corrosion.The dual self-healing coating shows better inhibition ability to narrow scratches due to the higher inhibitor concentration and less resumption.Also,the coating with wide scratches displays enough inhibition ability as well.The dual self-healing coating is a good alternative for Mg alloy parts in the practical applications.Yingwei Song Dan Liu Weineng Tang Kaihui Dong Dayong Shan En-Hou Han 2021Journal of Magnesium and Alloys2021,9,4:4

19Discovery of Novel Androgen Receptor Ligands by Structure-based Virtual Screening and Bioassays显示文摘Androgen receptor(AR) is a ligand-activated transcription factor that plays a pivotal role in the development and progression of many severe diseases such as prostate cancer, muscle atrophy, and osteoporosis. Binding of ligands to AR triggers the conformational changes in AR that may affect the recruitment of coactivators and downstream response of AR signaling pathway.Therefore, AR ligands have great potential to treat these diseases. In this study, we searched for novel AR ligands by performing a docking-based virtual screening(VS) on the basis of the crystal structure of the AR ligand binding domain(LBD) in complex with its agonist. A total of 58 structurally diverse compounds were selected and subjected to LBD affinity assay, with five of them(HBP1-3, HBP1-17, HBP1-38, HBP1-51, and HBP1-58) exhibiting strong binding to AR-LBD. The IC50 values of HBP1-51 and HBP1-58 are 3.96 m M and 4.92 m M, respectively, which are even lower than that of enzalutamide(Enz, IC50= 13.87 m M), a marketed second-generation AR antagonist. Further bioactivity assays suggest that HBP1-51 is an AR agonist, whereas HBP1-58 is an AR antagonist. In addition, molecular dynamics(MD) simulations and principal components analysis(PCA) were carried out to reveal the binding principle of the newlyidentified AR ligands toward AR. Our modeling results indicate that the conformational changes of helix 12 induced by the bindings of antagonist and agonist are visibly different. 20Human factors quantification via boundary identification of flight performance margin显示文摘A systematic methodology including a computational pilot model and a pattern recognition method is presented to identify the boundary of the flight performance margin for quantifying the human factors. The pilot model is proposed to correlate a set of quantitative human factors which represent the attributes and characteristics of a group of pilots. Three information processing components which are influenced by human factors are modeled: information perception, decision making, and action execution. By treating the human factors as stochastic variables that follow appropriate probability density functions, the effects of human factors on flight performance can be investigated through Monte Carlo(MC) simulation. Kernel density estimation algorithm is selected to find and rank the influential human factors. Subsequently, human factors are quantified through identifying the boundary of the flight performance margin by the k-nearest neighbor(k-NN) classifier.
