| 2 | A dynamic risk-based early warning monitoring system for population-based management of cardiovascular disease显示文摘Risk prediction tools are crucial for population-based management of cardiovascular disease(CVD).However,most prediction models are currently used to assess long-term risk instead of the risk of short-term CVD onset.We developed a Dynamic Risk-based Early wAming Monitoring(DREAM)system using large-scale,real-time electronic health record data from 2010 to 2020 from the CHinese Electronic health Records Research in Yinzhou study.The dynamic risk scores were derived from a 1:5 matched nested case-control set comprising 70,470 individuals(11,745 CVD events)and then validated in a cohort of 81,205 individuals(5950 CVD events).The individuals were Chinese adults aged 40-79 years without a history of CVD at baseline.Eleven predictors related to vital signs,laboratory tests,and health service utilization were selected to establish the dynamic scores.The proposed scores were significantly associated with the subsequent CVD onset(adjusted odds ratio,1.21;95%confidence interval,1.20-1.23).The area under the receiver operating characteristic curves(AUCs)was 0.6010(0.5929-0.6092)and 0.6021(0.5937-0.6105)for the long-term 10-year CVD risk<10%and≥10%groups in the derivation set,respectively.In the long-term 10-year CVD risk>10%group in the validation set,the change in AUC in addition to the long-term risk was 0.0235(0.0155-0.0315).By increasing the risk threshold from 7 to 16 points,the proportion of true subsequent CVD cases among those given alerts increased from 40.61%to 85.31%.In terms of management efficiency,the number needed to manage per CVD case ranged from 2.46 to 1.17 using the risk scores.With the increasing popularity and integration of EHR systems with wearable technology,the DREAM scores can be incorporated into an early-warning system and applied in dynamic,real-time,EHR-based,automated management to support healthcare decision making for individuals,general practitioners,and policymakers. | Xiaofei Liu Qianqian Li Weiye Chen Peng Shen Yexiang Sun Qi Chen Jinguo Wu Jingyi Zhang Ping Lu Hongbo Lin Xun Tang Pei Gao | 2021 | Fundamental Research2021,1,5: | 1 |

| 7 | The Effect of the Cyp19a1 Gene Methylation Modification on Temperature-dependent Sex Determination of Reeves' Turtle(Mauremys reevesii)显示文摘Temperature-dependent sex determination(TSD) is a type of environmental sex determination in which the sex of the embryos depends on the ambient temperature; however,the molecular mechanisms governing this process remain unknown.Aromatase,encoded by the cyp19a1 gene,which converts androgens into estrogens in animals,was considered to be the key gene for TSD.In this study,the 5'-flanking region of the cyp19a1 gene in Reeves' turtle(Mauremys reevesii) was cloned,and the promoter region was identified using the luciferase reporter assay.Then the eggs of Reeves' turtle were incubated at different temperatures(26°C: male-biased temperature; 29°C: non-sex-biased temperature and 32°C: female-biased temperature).During the thermosensitive period,the adrenal kidney gonad complexes(AKG) were sampled.DNA methylation analysis of the AKG samples showed that the promoter region of the cyp19a1 gene was significantly de-methylated in the female-biased temperature regime(P<0.01).Quantitative analysis of the cyp19a1 gene and estrogen by q PCR and Elisa assay showed that the expression level of the cyp19a1 gene and estrogen content were both upregulated significantly at the female-biased temperature(P<0.01).These results indicated that the de-methylation response of the cyp19a1 gene to incubation temperature,especially at the female-biased temperature,could lead to temperature-specific expression of aromatase and increased estrogen levels,which may further determine gonadal development in Reeves' turtle.These findings provide insights into the genetic mechanisms underlying TSD. | Wenxiu RU Liushuai HUA Yufeng WEI Weiye LI Dainan CAO Yan GE Hong CHEN Xianyong LAN Shiping GONG | 2017 | Asian Herpetological Research2017,18,3: | 1 |

| 10 | ZIF-8 coated gold nanospheres: A multi-responsive drug delivery system promoting the killing effect of photothermal therapy against osteosarcoma cells显示文摘Photothermal therapy(PTT)has been widely used in the treatment of tumors,but its efficacy is greatly limited by the inability of precise drug delivery and the increase of heat shock proteins(HSPs)caused by high temperature.This article describes a therapeutic strategy to enhance PTT with starvation therapy in conjunction with ferroptosis mechanism.A nanoparticle platform ZIF-8@GA was constructed by wrapping together glucose oxidase(GOX)and gold nanospheres(AuNPs)loaded with dihydroartemisinin(DHA)with zeolitic imidazolate framework-8(ZIF-8).This platform can take advantage of the micro-environment of osteosarcoma(OS)cells,featuring low pH and high reactive oxygen species(ROS),for precision drug delivery.GOX can effectively catalyze glucose to produce gluconic acid and H_(2)O_(2),and DHA can also induce ROS production in OS cells.ROS produced by GOX and DHA can further generate lipid peroxidation(LPO)and lead to ferroptosis of OS cells.At the same time,ROS and LPO produced can inhibit the expression of HSPs,thereby increasing the therapeutic effect of PTT.In vitro experiments show that the nanoparticles are pH and ROS responsive.1μg/mL GOX combined with 0.2μg/mL DHA promotes ferroptosis of OS cells,and increases the killing effect of near-infrared(NIR)on OS cells.Further in vivo experiments showed that the nano drug-delivery platform combined with PTT can effectively inhibit the growth of OS cells.Meanwhile,this study provides a new idea for the treatment of OS with biomaterials combined with various treatment methods. | Miao Lei Weiye Cai Luetao Zou Bin Yu Lin Chen Yang Cao Shanlin Xiang Chao Song Jiandu Lei Wei Jiang Zhenming Hu | 2024 | Nano Research2024,17,3: | 0 |
| 11 | Adenosquamous Carcinoma of the Larynx: A Case Report and Literature Review显示文摘Objective:To retrospectively analyze the clinical characteristics of laryngeal adenosquamous carcinoma(ASC) and review the related literature. Methods: We retrospectively analyzed five male patients with confirmed ASC of the larynx between 2005 and 2010 in our hospital, accounting for 0.2% of the laryngeal malignant tumors in the same period. Results: All patients underwent surgeries including horizontal partial laryngectomy(2 cases), vertical partial laryngectomy(1 case) and total laryngectomy(2 cases). One of the two who underwent total laryngectomy also received post-operative chemoradiotherapy. Four also underwent neck dissections. The mean follow-up time was 77.4±14.98 months. At the end of follow-up, two suffered regional metastasis and another local relapse. They all underwent related operations again and were still alive. The therapeutic approach might follow that of squamous cell carcinoma of the larynx. | 13 | Chondroitin Polymerizing Factor (CHPF) promotes cell proliferation and tumor growth in human osteosarcoma by inhibiting SKP2's ubiquitination while activating the AKT pathway显示文摘Osteosarcoma is a common malignant tumor occurring in children and young adults. Chondroitin sulfate (CS) participates in cell adhesion, cell division, and the formation of neural networks in the body, the biosynthesis of which requires the participation of glycosyltransferases. CHPF, a glycosyltransferase, plays a role in the extension of CS. Recently, CHPF's biological roles and functional importance in human diseases including malignant tumors have been widely discussed. However, whether CHPF is involved in osteosarcoma development and growth has not been revealed. The present work aimed to investigate the expression levels, functional significance and molecular mechanism of CHPF in osteosarcoma progression. Our results revealed that CHPF is strongly expressed in osteosarcoma tissues and cells. Furthermore, CHPF serves as a tumor promoter in the development and progression of osteosarcoma through enhancing cell proliferation and migration while suppressing apoptosis. Exploration of the mechanism by which CHPF promotes osteosarcoma indicated that CHPF promotes osteosarcoma through counteracting SKP2's ubiquitination and activating the Akt signaling pathway.
