| 1 | Perioperative'remote'acute lung injury:recent update显示文摘Perioperative acute lung injury(ALI) is a syndrome characterised by hypoxia and chest radiograph changes.It is a serious post-operative complication,associated with considerable mortality and morbidity.In addition to mechanical ventilation,remote organ insult could also trigger systemic responses which induce ALI.Currently,there are limited treatment options available beyond conservative respiratory support.However,increasing understanding of the pathophysiology of ALI and the biochemical pathways involved will aid the development of novel treatments and help to improve patient outcome as well as to reduce cost to the health service.In this review we will discuss the epidemiology of peri-operative ALI;the cellular and molecular mechanisms involved on the pathological process;the clinical considerations in preventing and managing perioperative ALI and the potential future treatment options. | Zhaosheng Jin Ka Chun Suen Daqing Ma | 2017 | The Journal of Biomedical Research2017,31,3: | 7 |
| 2 | Vegfa signaling regulates diverse artery/vein formation in vertebrate vasculatures显示文摘Vascular endothelial growth factor A(Vegfa) signaling regulates vascular development during embryogenesis and organ formation.However,the signaling mechanisms that govern the formation of various arteries/veins in various tissues are incompletely understood.In this study,we utilized transcription activator-like effector nuclease(TALEN) to generate zebrafish vegfaa mutants.vegfaa^(-/-) embryos are embryonic lethal,and display a complete loss of the dorsal aorta(DA) and expansion of the cardinal vein.Activation of Vegfa signaling expands the arterial cell population at the expense of venous cells during vasculogenesis of the axial vessels in the trunk.Vegfa signaling regulates endothelial cell(EC) proliferation after arterial-venous specification.Vegfa deficiency and overexpression inhibit the formation of tip cell filopodia and interfere with the pathfinding of intersegmental vessels(ISVs).In the head vasculature,vegfaa^(-/-) causes loss of a pair of mesencephalic veins(MsVs) and central arteries(CtAs),both of which usually dvelop via sprouting angiogenesis.Our results indicate that Vegfa signaling induces the formation of the DA at the expense of the cardinal vein during the trunk vasculogenesis,and that Vegfa is required for the angiogenic formation of MsVs and CtAs in the brain.These findings suggest that Vegfa signaling governs the formation of diverse arteries/veins by distinct cellular mechanisms in vertebrate vasculatures. | Daqing Jin Diqi Zhu Yabo Fang Yiwei Chen Gaihong Yu Weijun Pan Dong Liu Fen Li Tao P.Zhong | 2017 | Journal of Genetics and Genomics2017,44,10: | 6 |

| 5 | Induction of Wnt signaling antagonists and p21-activated kinase enhances cardiomyocyte proliferation during zebrafish heart regeneration显示文摘Heart regeneration occurs by dedifferentiation and proliferation of pre-existing cardiomyocytes(CMs).However,the signaling mechanisms by which injury induces CM renewal remain incompletely understood.Here,we find that cardiac injury in zebrafish induces expression of the secreted Wnt inhibitors,including Dickkopf 1(Dkkl),Dkk3,secreted Frizzled-related protein 1(sFrpl),and sFrp2,in cardiac tissue adjacent to injury sites.Experimental blocking of Wnt activity via Dkkl overexpression enhances CM proliferation and heart regeneration,whereas ectopic activation of Wnt8 signaling blunts injury-induced CM dedifferentiation and proliferation.Although Wnt signaling is dampened upon injury,the cytoplasmic β-catenin is unexpectedly increased at disarrayed CM sarcomeres in myocardial wound edges.Our analyses indicated that p21-activated kinase 2(Pak2)is induced at regenerating CMs,where it phosphorylates cytoplasmic β-catenin at Ser 675 and increases its stability at disassembled sarcomeres.Myocardial-specific induction of the phospho-mimeticβ-catenin(S675E)enhances CM dedifferentiation and sarcomere disassembly in response to injury.Conversely,inactivation of Pak2 kinase activity reduces the Ser 675-phosphorylatedβ-catenin(pS675-β-catenin)and attenuates CM sarcomere disorganization and dedifferentiation・Taken together,these findings demonstrate that coordination of Wnt signaling inhibition and Pak2/pS675-βYatenin signaling enhances zebrafish heart regeneration by supporting CM dedifferentiation and proliferation. | Xiangwen Peng Kaa Seng Lai Peilu She Junsu Kang Tingting Wang Guobao Li Yating Zhou jianjian Sun Daqing Jin Xiaolei Xu Lujian Liao Jiandong Liu Ethan Lee Kenneth D.Poss Tao P.Zhong | 2021 | Journal of Molecular Cell Biology2021,13,1: | 3 |
| 6 | Inhibitory effects of lidocaine on cerebral vasospasm in a rabbit model of subarachnoid hemorrhage显示文摘Following subarachnoid hemorrhage, vasoconstrictor substances, cellular apoptosis, blood coagulation, and vascular cell proliferation affect the onset of cerebral vasospasm. Previous studies from our laboratory have revealed that injection of lidocaine (2 mg) into the cisterna magna reduces cerebral vasospasm and nerve functional impairment in an animal model of subarachnoid hemorrhage. The present study determined the optimal lidocaine dose for vasospasm and brain injury by injecting different doses of lidocaine into the cisterna magna in a rabbit model of subarachnoid hemorrhage. Results showed that endothelin, tumor necrosis factor-α, and interleukin-6 levels significantly increased in plasma, and calcitonin gene-related peptide levels significantly decreased in plasma (P < 0.05). The number of neurons was decreased, the number of cells expressing c-Fos increased in the hippocampus, and cross-sections and diameters of basilar arteries were reduced (P < 0.05). These changes significantly improved following injection of lidocaine (1, 2, 4, and 6 mg) into the cisterna magna. | 7 | Toxicity mechanisms and bioavailability of copper to fish based on an adverse outcome pathway analysis显示文摘Copper(Cu)exists in a variety of forms in different aquatic environments,and affects their bioavailability.In this study we provide a systematic review on toxicity of Cu which focuses on identifying evidence in the mechanisms of Cu toxicity,and apply an adverse outcome pathway(AOP)analysis to identify multiple potential mechanisms and their interactions of Cu toxicity to fish.This analysis process included the mechanisms of behavior toxicant,oxidative toxicant,ion regulation disruption toxicity,as well as endocrine disruption toxicity.It was found that at low levels of Cu exposure,swimming,avoid predators,locating prey and other sensory functions will be impaired,and the organism will suffer from metabolic alkalosis and respiratory acidosis following the inhibition of the carbonic anhydrase active.The main pathway of acute toxicity of Cu to fish is the inhibition of the Na^(+)/K^(+)-ATPase enzyme,and lead to reduced intracellular sodium absorption,as well as Cu-induced increased cell permeability,in turn resulting in increased sodium ion loss,leading to cardiovascular collapse and respiratory insufficiency.The endocrine disruption toxicity of Cu to fish caused growth inhibition and reproductive reduction.In addition,there are several key pathways of Cu toxicity that are affected by hardness(e.g.,Ca^(2+))and intracellular DOC concentrations,including inhibiting Cu-induction,improving branchial gas exchange,altering membrane transport functions,decreasing Na+loss,and increasing Na+uptake.The results of the AOP analysis will provide a robust framework for future directed research on the mechanisms of Cu toxicity. | Wei Liao Ziwei Zhu Chenglian Feng Zhenfei Yan Yajun Hong Daqing Liu Xiaowei Jin | 2023 | Journal of Environmental Sciences2023,,5: | 1 |

| 13 | Peptidomic Analysis Reveals that Novel Peptide LDP2 Protects Against Hepatic Ischemia/Reperfusion Injury显示文摘Background and Aims:Hepatic ischemia/reperfusion(I/R)injury has become an inevitable issue during liver transplantation,with no effective treatments available.However,peptide drugs provide promising regimens for the treatment of this injury and peptidomics has gradually attracted increasing attention.This study was designed to analyze the spectrum of peptides in injured livers and explore the potential beneficial peptides involved in I/R injury.Methods:C57BL/6 mice were used to establish a liver I/R injury animal model.Changes in peptide profiles in I/R-injured livers were analyzed by mass spectrometry,and the functions of the identified peptides were predicted by bioinformatics.AML12 cells were used to simulate hepatic I/R injury in vitro.After treatment with candidate liver-derived peptides(LDPs)1–10,the cells were collected at various reperfusion times for further study.Results:Our preliminary study demonstrated that 6 h of reperfusion caused the most liver I/R injury.Peptidomic results suggested that 10 down-regulated peptides were most likely to alleviate I/R injury by supporting mitochondrial function.Most importantly,a novel peptide,LDP2,was identified that alleviated I/R injury of AML12 cells.It increased cell viability and reduced the expression of inflammation-and apoptosis-related proteins.In addition,LDP2 inhibited the expression of proteins related to autophagy.Conclusions:Investigation of changes in the profiles of peptides in I/R-injured livers led to identification of a novel peptide,LDP2 with potential function in liver protection by inhibiting inflammation,apoptosis,and autophagy. | Qun Bao Zhengxin Wang Sheng Cheng Jin Zhang Qiuli Liu Yunpeng Zhang Daqing Cheng Xirong Guo Xingyun Wang Bo Han Peng Sun | 2023 | Journal of Clinical and Translational Hepatology2023,11,2: | 0 |
