1FGF1^(△HBS) prevents diabetic cardiomyopathy by maintaining mitochondrial homeostasis and reducing oxidative stress via AMPK/Nur77 suppression显示文摘As a classically known mitogen,fibroblast growth factor 1(FGF1)has been found to exert other pleiotropic functions such as metabolic regulation and myocardial protection.Here,we show that serum levels of FGF1 were decreased and positively correlated with fraction shortening in diabetic cardiomyopathy(DCM)patients,indicating that FGF1 is a potential therapeutic target for DCM.We found that treatment with a FGF1 variant(FGF1^(△HBS))with reduced proliferative potency prevented diabetes-induced cardiac injury and remodeling and restored cardiac function.RNA-Seq results obtained from the cardiac tissues of db/db mice showed significant increase in the expression levels of anti-oxidative genes and decrease of Nur77 by FGF1AHBS treatment.Both in vivo and in vitro studies indicate that FGF1^(△HBS) exerted these beneficial effects by markedly reducing mitochondrial fragmentation,reactive oxygen species(ROS)generation and cytochrome c leakage and enhancing mitochondrial respiration rate and β-oxidation in a 5;AMP-activated protein kinase(AMPK)/Nur77-dependent manner,all of which were not observed in the AMPK null mice.The favorable metabolic activity and reduced proliferative properties of FGF1^(△HBS) testify to its promising potential for use in the treatment of DCM and other metabolic disorders.Dezhong Wang Yuan Yin Shuyi Wang Tianyang Zhao Fanghua Gong Yushuo Zhao Beibei Wang Yuli Huang Zizhao Cheng Guanghui Zhu Zengshou Wang Yang Wang Jun Ren Guang Liang Xiaokun Li Zhifeng Huang 2021Signal Transduction and Targeted Therapy2021,6,4:6

3Suppressing thermal quenching via defect passivation for efficient quasi-2D perovskite light-emitting diodes显示文摘Emission thermal quenching is commonly observed in quasi-2D perovskite emitters,which causes the severe drop in luminescence eficiency for the quasi-2D perovskite light-emitting diodes(Pel EDs)during practical operations.However,this issue is often neglected and rarely studied,and the root cause of the thermal quenching has not been completely revealed now.Here,we develop a passivation strategy via the 2,7-dibromo-9,9-bis(3'-diethoxylphosphorylpropyl-fluorene to investigate and suppress the thermal quenching.The agent can effectively passivate coordination-unsaturated Pb2+defects of both surface and bulk of the flm without affecting the perovskite crysaliation,which helps to more truly demonstrate the important role of defects in thermal quenching.And our results reveal the root cause that the quenching will be strengthened by the defect promoted exciton-phonon coupling.Ultimately,the PeL EDs with defect passivation achieve an improved external quantum efficiengy(EQE)over 22%and doubled operation lifetime at room temperature,and can maintain about 85%of the initial EQE at 85℃,much higher than 17%of the control device.These findings provide an important basis for fabricating practical PeLEDs for lighting and displays.Dezhong Zhang Yunxing Fu Hongmei Zhan Chenyang Zhao Xiang Gao Chuanjiang Qin Lixiang Wang 2022Light(Science & Applications)2022,11,4:1

5A wall-thickness compensation strategy for wax pattern of hollow turbine blade显示文摘As an important index affecting the aerodynamic performance and the structural strength of hollow turbine blades, the wall-thickness precision of the blade is mainly inherited from the positional relationship between the corresponding wax pattern and the internal ceramic core.However, due to locating errors, the actual position of ceramic core is always deviated from the ideal position, which makes it difficult to guarantee the wall-thickness precision of the wax pattern.To solve this problem, a wall-thickness compensation strategy is proposed in this paper. Firstly,based on the industrial computed tomography(ICT) technique and curve matching algorithms, a model reconstruction method is developed, with which the 3D model of a trial wax pattern can be easily constructed. After that, focusing on eliminating the wall-thickness errors of the trial wax pattern, an optimization method for the pose of the ceramic core in the wax pattern is proposed. Then, by mapping the optimal pose of the ceramic core to length adjustments of the locating rods, the wall-thickness errors of the wax pattern can be greatly reduced. 7Proximity-enabled covalent binding of IL-2 to IL-2Rα selectively activates regulatory T cells and suppresses autoimmunity显示文摘Interleukin-2(IL-2)is a pleiotropic cytokine that orchestrates bidirectional immune responses via regulatory T cells(Tregs)and effector cells,leading to paradoxical consequences.Here,we report a strategy that exploited genetic code expansion-guided incorporation of the latent bioreactive artificial amino acid fluorosulfate-L-tyrosine(FSY)into IL-2 for proximity-enabled covalent binding to IL-2Rαto selectively promote Treg activation.We found that FSY-bearing IL-2 variants,such as L72-FSY,covalently bound to IL-2Rαvia sulfur-fluoride exchange when in proximity,resulting in persistent recycling of IL-2 and selectively promoting the expansion of Tregs but not effector cells.Further assessment of L72-FSY-expanded Tregs demonstrated that L72-FSY maintained Tregs in a central memory phenotype without driving terminal differentiation,as demonstrated by simultaneously attenuated expression of lymphocyte activation gene-3(LAG-3)and enhanced expression of programmed cell death protein-1(PD-1).Subcutaneous administration of L72-FSY in murine models of pristane-induced lupus and graft-versus-host disease(GvHD)resulted in enhanced and sustained therapeutic efficacy compared with wild-type IL-2 treatment.The efficacy of L72-FSY was further improved by N-terminal PEGylation,which increased its circulatory retention for preferential and sustained effects.This proximity-enabled covalent binding strategy may accelerate the development of pleiotropic cytokines as a new class of immunomodulatory therapies.Bo Zhang Jiaqi Sun Yeshuang Yuan Dezhong Ji Yeting Sun Yudong Liu Shengjie Li Xingxing Zhu Xunyao Wu Jin Hu Qiu Xie Ling Wu Lulu Liu Boyang Cheng Yuanjie Zhang Lingjuan Jiang Lidan Zhao Fei Yu Wei Song Min Wang Yue Xu Shiliang Ma Yunyun Fei Lihe Zhang Demin Zhou Xuan Zhang 2023Signal Transduction and Targeted Therapy2023,8,2:0
8Fabrication and properties of high quality InGaN-based LEDs with highly reflective nanoporous GaN mirrors显示文摘Distributed Bragg reflectors (DBRs) are essential components for the development of optoelectronic devices.In this paper, we first report the use of the nanoporous GaN (NP-GaN) DBR as a template for regrowth of InGaN-based light-emitting diodes (LEDs). The wafer-scale NP-GaN DBR, which is fabricated by electrochemical etching in a neutral solution, has a smooth surface, high reflectivity (>99.5%), and wide spectral stop band width (>70 nm). The chemical composition of the regrown LED thin film is similar to that of the reference LED, but the photoluminescence (PL) lifetime, PL intensity, and electroluminescence intensity of the LED with the DBR are enhanced several times compared to those of the reference LED. 10Improvement of SFC显示文摘The Heavy Ion Research Facility at Lanzhou (HIRFL) is a cyclotron complex in which a Sector Focusing Cyclotron (SFC) with energy constant K=69 as an injector was converted from the original existing conversional cyclotron with diameter 1.5 m. In the process of operation for about 20 years, some shortages were exposed. For example, the vacuum pressure of SFC is only 10^-4 Pa magnitude, in the case most of the heavier ions will be lost in it. Because the yoke of SFC was not changed in the fabricated process, while the diameter of accelerator is changed from 1.5 m to 1.7 m, so the magnetic saturation is very serious in the yoke. The stay magnetic field around the cyclotron is very strong. It affects not only the beam tuning in vertical process coming from ECR ion source, but also the extraction beam behind SFC. 11Sleep deprivation reorganizes the dynamic configurations of default mode network activity during recovery sleep显示文摘Sleep deprivation causes disturbances of the neural activity, leading to the impairment of brain functions. However, the exact mechanism of sleep deprivation and how it affects the dynamics of brain activity during the recovery sleep remains unclear. In the current study, we performed sleep deprivation experiments on ten adult rats, and recorded the local field potentials from default mode network(DMN) regions during sleep before and after sleep deprivation. The DMN dynamics was assessed with the configurations of coactive micropatterns(CAMPs) using our previously proposed CAMP method. Our analysis revealed that the effects of sleep deprivation on DMN dynamics in the slow-wave sleep(SWS) state and the rapid eye-movement sleep(REM)state were disparate. Dynamic configurations of DMN activity in the SWS state were significantly impaired after sleep deprivation, with increased occurrence of low-activity CAMP and reorganized transition structure across three CAMPs. Moreover,enhanced functional connectivity and improved efficiencies in all CAMP networks were observed during the SWS state in the recovery sleep. However, there were no significant alterations in either DMN dynamics or CAMP network structures in the REM sleep state after sleep deprivation. Our results described the alterations of DMN dynamics in different sleep states after sleep deprivation, and illustrated the differential effects of sleep deprivation on two sleep states.
