1氮肥运筹对陇中旱农区玉米光合特性及产量的影响显示文摘全膜双垄沟播技术使玉米成为了陇中旱农区主要作物之一,但该技术下玉米的高产出导致土壤养分耗竭,影响玉米生产的可持续性。本研究依托2012年布设在陇中旱农区的田间定位试验,研究4个施氮水平(N0:不施肥;N1:100kg·hm^(-2)、N2:200kg·hm^(-2)、N3:300kg·hm^(-2))和2个施氮时期(T1:1/3基肥+1/3拔节期+1/3开花期、T2:1/3基肥+2/3拔节期)对玉米光合特性、叶绿素含量、叶面积指数、干物质积累和分配量及产量的影响。结果表明:1)随着施氮量的增加玉米光合性能也在增强,而N3和T2N2间差异不显著,T2时期提高玉米光合特性;2)全生育期内,N3处理下叶绿素含量较N2、N1、N0分别平均增加50.9%、17.0%、2.7%;叶面积指数也随施氮量的增加而增加,但N2和N3间无显著差异;T2下的叶绿素含量和叶面积指数在生育后期显著高于T1;3)干物质积累量和籽粒分配量表现为:N3>N2>N1>N0,T2下的干物质积累和籽粒分配量高于T1;4)籽粒产量和生物产量均随施氮量的增加而增加,N2和N3下的籽粒产量和生物产量显著高于N0,其中N3较N0增加79.2%和68.4%,N2较N0增加65.9%和54.1%,T2下的籽粒产量和生物产量分别较T1显著增加9.9%和13.5%,而T2N2与N3间无显著差异,因此,在陇中旱农区应用全膜双垄沟播技术种植玉米,施纯氮200kg·hm^(-2)左右,按照1/3基肥+2/3拔节期配施,可以增强光合作用,从而提高玉米籽粒产量和饲料产量,促进玉米生产可持续发展。王进斌 谢军红 李玲玲 Eunice Essel 彭正凯 邓超超 沈吉成 颉健辉 2019草业学报2019,28,1:42
2不同耕作措施对旱地作物生育期农田耗水结构和水分利用效率的影响显示文摘陇中黄土高原旱农区降水有限、水分利用效率低下是导致该区作物生产力水平低而不稳的主要原因。发展保护性耕作是保护水土资源、提高水分利用效率的重要途径。为揭示耕作措施影响水分利用效率的机制,2015—2016年在陇中黄土高原旱农区研究了不同耕作措施对土壤棵间蒸发、农田耗水量、作物蒸腾量、棵间蒸发与蒸散的比例、产量及水分利用效率的影响。试验设置传统耕作(T)、免耕秸秆覆盖(NTS)、免耕(NT)、传统耕作+秸秆翻入(TS)、传统耕作+覆膜(TP)、免耕覆膜(NTP)6个处理,春小麦和豌豆年间轮作。结果表明:(1)春小麦和豌豆全生育期棵间蒸发量NTS、TP、NTP比T显著减少6.52%~50.81%,NTS降低棵间蒸发量的作用主要在小麦开花后和豌豆结荚后,地膜覆盖在各个生育时期基本上都显著减少了棵间蒸发。(2)NTS对全生育期耗水量无显著影响,NTP的耗水量只在小麦地显著高于T。相比T,NTS显著提高了小麦开花-收获和豌豆结荚-收获期间的阶段耗水量及其占总耗水的比例。(3)NTS、TP、NTP均显著提高了春小麦和豌豆的蒸腾量,降低了田间的蒸发占蒸散的比例,降低了水分的无效损耗。(4)各年份春小麦和豌豆的产量NTS、TP、NTP比T提高了7.64%~62.79%,水分利用效率比T提高了0.43%~50.88%。因此,在陇中黄土高原旱农区,免耕秸秆覆盖、地膜覆盖等保护性耕作措施均能提高水分利用效率及小麦和豌豆的产量。免耕秸秆覆盖通过降低作物生长后期棵间蒸发量,提高作物生长后期耗水量,降低蒸发与蒸散的比例,从而提高春小麦和豌豆的水分利用效率及产量。而地膜覆盖处理主要是通过减少全生育期棵间蒸发量,增加作物全生育期蒸腾量,降低蒸发与蒸散的比例,从而实现作物水分高效利用,提高作物产量。彭正凯 李玲玲 谢军红 邓超超 EUNICE Essel 王进斌 颉健辉 沈吉成 康彩睿 2018水土保持学报2018,32,5:26
3Plasma exchange in patients with acute and acute-on-chronic liver failure: A systematic review显示文摘BACKGROUND Acute liver failure(ALF)and acute-on-chronic liver(ACLF)carry high short-term mortality rate,and may result from a wide variety of causes.Plasma exchange has been shown in a randomized control trial to improve survival in ALF especially in patients who did not receive a liver transplant.Other cohort studies demonstrated potential improvement in survival in patients with ACLF.AIM To assess utility of plasma exchange in liver failure and its effect on mortality in patients who do not undergo liver transplantation.METHODS Databases MEDLINE via PubMed,and EMBASE were searched and relevant publications up to 30 March,2019 were assessed.Studies were included if they involved human participants diagnosed with liver failure who underwent plasma exchange,with or without another alternative non-bioartificial liver assist device.RESULTS Three hundred twenty four records were reviewed,of which 62 studies were found to be duplicates.Of the 262 records screened,211 studies were excluded.Fifty-one articles were assessed for eligibility,for which 7 were excluded.Twenty-nine studies were included for ALF only,and 9 studies for ACLF only.Six studies included both ALF and ACLF patients.A total of 44 publications were included.Of the included publications,2 were randomized controlled trials,14 cohort studies,12 case series,16 case reports.All of three ALF studies which looked at survival rate or survival days reported improvement in outcome with plasma exchange.In two out of four studies where plasma exchange-based liver support systems were compared to standard medical treatment(SMT)for ACLF,a biochemical improvement was seen.Survival in the non-transplanted patients was improved in all four studies in patients with ACLF comparing plasma exchange vs SMT.Using the aforementioned studies,plasma exchange based therapy in ACLF compared to SMT improved survival in non-transplanted patients at 30 and 90-d with a pooled OR of 0.60(95%CI 0.46-0.77,P<0.01).CONCLUSION The level of evidence for use of high volume plasma exchange in selected ALF cases is high.Plasma exchange in ACLF improves survival at 30-and 90-d in nontransplanted patients.Further well-designed randomized control trials will need to be carried out to ascertain the optimal duration and amount of plasma exchange required and assess if the use of high volume plasma exchange can be extrapolated to patients with ACLF.Eunice Xiang-Xuan Tan Min-Xian Wang Junxiong Pang Guan-Huei Lee 2020World Journal of Gastroenterology2020,26,2:26
4耕作措施对陇中旱农区土壤细菌群落的影响显示文摘为探究耕作措施对陇中旱农区麦豆轮作系统土壤细菌多样性的影响,借助2001年在陇中旱农区建立的不同耕作措施长期定位试验,应用高通量测序技术比较传统耕作(T)与3种保护性耕作(免耕(NT)、传统耕作+秸秆还田(TS)和免耕+秸秆覆盖(NTS))对麦豆轮作体系土壤有机碳、全氮、微生物生物量碳、微生物生物量氮及细菌群落结构影响。结果表明:1)相对于传统耕作,秸秆还田显著增加0~10 cm、10~30 cm土壤有机碳、全氮、微生物量碳和微生物量氮含量,其中NTS处理0~10 cm、10~30 cm土层有机碳、全氮、微生物量碳和微生物量氮含量较传统耕作分别提高28.27%、114.16%、13.51%、49.14%和39.86%、98.05%、10.78%、40.72%。2)酸杆菌门(26.42%)、变形菌门(19.86%)和放线菌门(19.44%)为陇中旱农区麦豆轮作系统土壤细菌的主要优势种群,NTS处理下酸杆菌门、放线菌门和变形菌门丰度较传统耕作显著提高35.11%、33.77%和30.17%。3)与传统耕作相比,保护性耕作提高了0~30cm土层土壤细菌的Observedspecies指数、Chao指数、香农指数、辛普森指数,以NTS处理提高最为显著。因此,在陇中旱农区推广以免耕秸秆覆盖为主的保护性耕作措施有利于增加土壤碳氮固存、提高土壤细菌群落丰度和多样性和土壤的生物活性,促进农业的可持续发展。邓超超 李玲玲 谢军红 彭正凯 王进斌 颉健辉 沈吉成 Eunice Essel 2019土壤学报2019,56,1:21
5保护性耕作对陇中旱作农田水分特征的影响显示文摘陇中旱农区生产力水平低而不稳,而保护性耕作措施是农业可持续发展的重要途径.本研究依托2001年建立在陇中旱农区的长期不同耕作措施的定位试验,研究了不同耕作措施对土壤水分入渗、蒸发、作物产量和水分利用效率的影响.该试验共设6个处理,分别为传统耕作(T)、免耕秸秆覆盖(NTS)、免耕(NT)、传统耕作+秸秆翻入(TS)、传统耕作+覆膜(TP)、免耕覆膜(NTP),春小麦和豌豆年间轮作.结果表明:与T处理相比,NTS处理的小麦地和豌豆地的土壤容重显著降低,总孔隙度显著增加.保护性耕作措施降低了豌豆地0~5 cm土壤渗吸率,NTS处理渗吸率比T处理降低56.2%.保护性耕作提高了土壤饱和导水率,无论小麦地和豌豆地,NTS均比T处理显著提高了饱和导水率,增幅为52.8%~107.1%.保护性耕作显著降低了作物生育期棵间蒸发量,NTP、TP、NTS比T处理降低了14.4%~50.8%,并减弱了雨后土壤蒸发.保护性耕作提高了作物产量和水分利用效率,NTS、TP、NTP的产量比传统耕作提高了9.5%~62.8%,水分利用效率比传统耕作提高了0.4%~50.9%.因此,在陇中旱农区,保护性耕作措施可以提高水分利用效率,增加作物产量.彭正凯 李玲玲 谢军红 康彩睿 ESSEL Eunice 王进斌 頡健辉 沈吉成 2018应用生态学报2018,29,12:14
6大豆蛋白体外酶解物中血管紧张素转化酶抑制剂活性肽研究显示文摘目的:体外模仿部分胃肠道消化酶解过程,考察大豆蛋白酶解消化能否产生血管紧张素转化酶抑制剂(ACEI)活性肽及其活性状况,以揭示大豆蛋白体内消化酶解与ACEI的活性关系。方法:模拟人体胃肠道消化过程,以胃蛋白酶结合胰蛋白酶,相继酶解大豆分离蛋白(SPI),经色谱分离,动态检测不同阶段ACEI肽片段及其活性大小。结果:胃蛋白酶酶解过程前20min内,酶解液ACEI活性达到最高点,随后在胰蛋白酶酶解阶段其抑制活性下降。180min后的酶解产物,其半抑制活性浓度IC50值为0.28±0.06 mg/ml。同时,未经酶解的SPI液在0.73mg/ml时无ACEI活性。SPI酶解液经各种色谱分离后的组分,其IC50值从0.13±0.03到0.93±0.08 mg/ml。低分子量和伴有疏水性基团的肽类最具ACE抑制活性。结论:体外模仿胃肠消化过程使用胃蛋白酶和胰蛋白酶酶解SPI可产生不同ACEI活性的肽片段,说明人体正常摄食消化大豆蛋白可产生血管紧张素转化酶抑制剂活性肽。刘志伟 刘秀梅 郑梦云 WENDY LO EUNICE LI-CHAN 2007营养学报2007,29,1:13
7氮肥运筹对旱作覆膜玉米产量及固碳减排效应研究显示文摘在黄土高原雨养农业区,依托布设于2012年的定位试验研究氮肥运筹对全膜双垄沟播玉米固碳效应和NH3挥发的影响。采用裂区设计,主处理施氮水平:不施氮对照(N0)、低施氮100 kg·hm^-2(N1)、中施氮200kg·hm^-2(N2)和高施氮300 kg·hm^-2(N3);副处理为施肥时期及比例:设基肥∶拔节肥=1∶2(T1)和基肥∶拔节肥∶大喇叭口肥=1∶1∶1(T2)。结果表明:将200 kg·hm^-2氮肥以基肥∶拔节肥=1∶2的比例施用能显著提高全膜双垄沟播玉米的籽粒产量和生物产量,分别较对照增加197%和131%,氮肥偏生产力和氮肥农学效率较高施氮水平分别降低48%和47%。氨挥发排放主要集中在追肥后的一周,穴施追肥后,NH3挥发速率迅速升高,1~4 d内达到峰值,随后逐渐降低至对照水平,且施氮水平越高排放峰值越大。高、中、低施氮水平的NH3挥发量较不施氮对照分别增加6.5、5.9、5.4倍;T1处理较T2处理显著降低了37%;N3T2的NH3挥发量最多,较其他处理显著增多了13%~47%。NH3挥发损失率变幅在13%~60%,随施氮水平增加而降低,高、中施氮水平较低施氮水平降低了59.6%和44.6%;与T2相比,T1处理NH3挥发损失率显著降低了40.4%,增加施氮水平和追肥次数潜在增加NH3挥发损失。综上所述,在黄土高原雨养农业区种植全膜双垄沟播玉米时,能提高玉米籽粒产量和土壤固碳减排能力的适宜施肥制度为:施纯氮200 kg·hm^-2,全部氮肥按基肥∶拔节肥=1∶2的比例施用。颉健辉 李玲玲 谢军红 彭正凯 邓超超 沈吉成 王进斌 Eunice Essel 2019中国土壤与肥料2019,0,6:9
8耕作措施和氮肥用量对陇中旱农区粮饲兼用玉米光合特性与水分利用效率的影响显示文摘全膜双垄沟播技术使玉米成为陇中旱农区主栽作物之一,但该技术下玉米的高产导致土壤养分和水分耗竭,影响玉米生产的可持续性,依托2012年布设在陇中旱农区的田间定位试验,研究4种耕作方式(CT:传统翻耕,RT:旋耕,SS:深松耕,NT:免耕)和4种施氮水平(N1:不施氮,N2:施纯氮100kg·hm^(-2),N3:施纯氮200kg·hm^(-2),N4:施纯氮300kg·hm^(-2))对粮饲兼用玉米光合特性、干物质积累量、产量及水分利用效率的影响。结果表明:深松耕和免耕显著增强玉米光合性能,且随着施氮量的增加玉米光合性能也增强;各生育时期干物质积累量均表为SS>NT>RT>CT、N4>N3>N2>N1,成熟期时SS较CT显著增加37.4%,N4较其他处理增加15.0%~85.0%;与传统耕作相比,深松耕和免耕可以提高玉米籽粒产量,2014年SS和NT较CT分别增加33.0%、18.8%,2015年分别增加19.7%、11.6%,且深松耕处理生物量显著高于传统耕作;施纯氮300kg·hm^(-2)和200kg·hm^(-2)均可以提高玉米籽粒产量、生物量和水分利用效率,但施纯氮200kg·hm^(-2)与300kg·hm^(-2)无显著差异,施纯氮200kg·hm^(-2)处理下籽粒产量、生物量和水分利用效率较不施肥处理分别增加68.4%、63.5%、52.2%,较施氮100kg·hm^(-2)分别增加40.2%、31.7%、30.5%。因此,在陇中旱农区应用全膜双垄沟播技术种植粮饲兼用玉米,覆膜前深松耕或免耕,施氮200kg·hm^(-2)左右比较适宜,既有利于粮饲兼用玉米籽粒产量、饲料产量的提高,也可以提高水氮利用效率,促进玉米生产的可持续发展。王进斌 李玲玲 谢军红 Eunice Essel 邓超超 颉健辉 彭正凯 沈吉成 2018西北农业学报2018,27,6:8

10The interplay of aging,genetics and environmental factors in the pathogenesis of Parkinson’s disease显示文摘Background:Parkinson’s disease(PD)is characterized by dopaminergic neuronal loss in the substantia nigra pars compacta and intracellular inclusions called Lewy bodies(LB).During the course of disease,misfoldedα-synuclein,the major constituent of LB,spreads to different regions of the brain in a prion-like fashion,giving rise to successive non-motor and motor symptoms.Etiology is likely multifactorial,and involves interplay among aging,genetic susceptibility and environmental factors.Main body:The prevalence of PD rises exponentially with age,and aging is associated with impairment of cellular pathways which increases susceptibility of dopaminergic neurons to cell death.However,the majority of those over the age of 80 do not have PD,thus other factors in addition to aging are needed to cause disease.Discovery of neurotoxins which can result in parkinsonism led to efforts in identifying environmental factors which may influence PD risk.Nevertheless,the causality of most environmental factors is not conclusively established,and alternative explanations such as reverse causality and recall bias cannot be excluded.The lack of geographic clusters and conjugal cases also go against environmental toxins as a major cause of PD.Rare mutations as well as common variants in genes such as SNCA,LRRK2 and GBA are associated with risk of PD,but Mendelian causes collectively only account for 5%of PD and common polymorphisms are associated with small increase in PD risk.Heritability of PD has been estimated to be around 30%.Thus,aging,genetics and environmental factors each alone is rarely sufficient to cause PD for most patients.Conclusion:PD is a multifactorial disorder involving interplay of aging,genetics and environmental factors.This has implications on the development of appropriate animal models of PD which take all these factors into account.Common converging pathways likely include mitochondrial dysfunction,impaired autophagy,oxidative stress and neuroinflammation,which are associated with the accumulation and spread of misfoldedα-synuclein and neurodegeneration.Understanding the mechanisms involved in the initiation and progression of PD may lead to potential therapeutic targets to prevent PD or modify its course.Shirley Yin-Yu Pang Philip Wing-Lok Ho Hui-Fang Liu Chi-Ting Leung Lingfei Li Eunice Eun Seo Chang David Boyer Ramsden Shu-Leong Ho 2019Translational Neurodegeneration2019,8,1:5
11培肥模式对陇中旱农区粮饲兼用玉米产量和品质的影响显示文摘依托2012年布设在陇中旱农区的田间定位试验,在2014-2015年研究4种土壤培肥模式[无培肥(NA)、猪粪(SM)、秸秆(MS)、猪粪+化肥(SC)]对粮饲兼用玉米纤维素质量分数和积累率,粗蛋白质量分数及产量的影响。结果显示,玉米各生育时期干物质累积量、籽粒和饲草产量均SC>SM>MS>NA。2014年和2015年,与NA比较,SC处理籽粒和饲草产量分别提高67.24%、49.40%和78.69%、55.09%。随着玉米生育进程的推进,粗蛋白质量分数降低,酸性洗涤纤维(ADF)和中性洗涤纤维(NDF)质量分数增加,玉米各生育时期纤维素质量分数及积累率各处理表现为SCSM>MS>NA。可见,猪粪与化肥相结合提高粮饲兼用玉米产量,降低纤维素质量分数,是有利于陇中旱农区粮饲兼用玉米高产优质可持续生产的土壤培肥模式。沈吉成 李玲玲 谢军红 彭正凯 Eunice Essel 邓超超 王进斌 颉健辉 2018西北农业学报2018,27,9:3
12长期不同耕作措施对黄绵土体积质量和作物水分利用效率的影响显示文摘借助2001年在陇中黄土高原建立的不同耕作措施长期定位试验,研究5种保护性耕作[免耕(NT)、传统耕作+秸秆还田(TS)、免耕+秸秆覆盖(NTS)、传统耕作+地膜覆盖(TP)、免耕+地膜覆盖(NTP)]和传统耕作(T)对黄绵土体积质量、水分含量、作物耗水量、产量及水分利用效率的影响。结果表明:保护性耕作均可降低0~30cm土层土壤体积质量,提升土壤孔隙度;其中NTS显著降低0~10cm土层土壤体积质量,并显著提升该土层土壤孔隙度,且随着应用年限的延长,效果更为突出。保护性耕作显著提高播种期0~10cm土层土壤含水量,尤以NTS和NTP效果最佳。与传统耕作相比,NTS和NTP在增加作物耗水量的同时也提高了作物产量和水分利用效率,但NTP不利于土壤结构的持续改善。因此,长期施用免耕秸秆覆盖耕作措施能显著改善土壤结构,提高土壤水分利用效率,进而提高作物产量,促进农业可持续发展。邓超超 李玲玲 谢军红 彭正凯 王进斌 颉健辉 沈吉成 Eunice Essel 2018西北农业学报2018,27,7:3

14Liver resection versus liver transplantation for hepatocellular carcinoma within Milan criteria:a meta-analysis of 18,421 patients显示文摘Background:Outcomes after liver resection(LR)and liver transplantation(LT)for hepatocellular carcinoma(HCC)are heterogenous and may vary by region,over time periods and disease burden.We aimed to compare overall survival(OS)and disease-free survival(DFS)between LT versus LR for HCC within the Milan criteria.Methods:Two authors independently searched Medline and Embase databases for studies comparing survival after LT and LR for patients with HCC meeting the Milan criteria.Meta-analyses and meta-regression were conducted using random-effects models.Results:We screened 2,278 studies and included 35 studies with 18,421 patients.LR was associated with poorer OS[hazard ratio(HR)=1.44;95%confidence interval(CI):1.14-1.81;P<0.01]and DFS(HR=2.71;95%CI:2.23-3.28;P<0.01)compared to LT,with similar findings among intention-to-treat(ITT)studies.In uninodular disease,OS in LR was comparable to LT(P=0.13)but DFS remained poorer(HR=2.95;95%CI:2.30-3.79;P<0.01).By region,LR had poorer OS versus LT in North America and Europe(P≤0.01),but not Asia(P=0.25).LR had inferior survival versus LT in studies completed before 2010(P=0.01),but not after 2010(P=0.12).Cohorts that underwent enhanced surveillance had comparable OS after LT and LR(P=0.33),but cohorts undergoing usual surveillance had worse OS after LR(HR=1.95;95%CI:1.24-3.07;P<0.01).Conclusions:Mortality after LR for HCC is nearly 50%higher compared to LT.Survival between LR and LT were similar in uninodular disease.The risk of recurrence after LR is threefold that of LT.Jin Hean Koh Darren Jun Hao Tan Yuki Ong Wen Hui Lim Cheng Han Ng Phoebe Wen Lin Tay Jie Ning Yong Mark D.Muthiah Eunice X.Tan Ning Qi Pang Beom Kyung Kim Nicholas Syn Alfred Kow Brian K.P.Goh Daniel Q.Huang 2022Hepatobiliary Surgery and Nutrition2022,11,1:3

16Quantitative scintigraphic analysis of the apical seal in Thermafil/Topseal and Real Seal 1/Realseal filled root canals显示文摘AIM: To investigate the microleakage of two different root canal obturation systems, using the nuclear medicine approach.METHODS: Twenty-six single-rooted extracted teeth were selected. The crowns were sectioned to obtain 15-mm long root segments and each tooth was prepared using rotary Pro File instruments. The roots were divided into 2 experimental groups using Real Seal 1 and Real Seal sealer or Thermafil and Top Seal sealer as well as two control groups. On the 7th and the 28 th day the apices were submersed in a solution of 99 m TcPertechnetate during 3 h. The radioactivity was counted using a γ camera. RESULTS: The present study showed that none of the root canal-filled teeth was leakage free. The statistical analyses were made using Kruskal-Wallis and statistical significance was assessed using α = 0.05. Although apical leakage measured in counts per minute(cpm) in the Thermafil/Top Seal group was lower than in the Real Seal/Real Seal group(363 916 ± 180 707.7 cpm vs533 427 ± 414 020.6 cpm) on 7th day and(1 678 200 ± 567 217.4 cpm vs 2 240 518 ± 383 356.7 cpm) on 28 th day, there was no statistical difference(P > 0.05). In the Thermafil/Top Seal group and Real Seal 1/Real Seal group it was found that over time, the number of counts increased between 7 d and 28 d(363 916 ± 180 707.7 cpm vs 1 678 200 ± 567 217.4 cpm) and(533 427 ± 414 020.6 cpm vs 2 240 518 ± 383 356.7 cpm), respectively, with statistically significant differences(Thermafil/Top Seal group, P = 0.015 and Real Seal 1/Real Seal group, P = 0.036).CONCLUSION:Both carrier-based Realseal 1 and Thermafil techniques showed a similar sealing effect,but none of the materials was leakage free.Manuel Marques Ferreira Margarida Abrantes Eunice Virgínia Carrilho Maria Filomena Botelho 2013World Journal of Stomatology2013,2,2:2
17LRRK2, GBA and their interaction in the regulation of autophagy: implications on therapeutics in Parkinson’s disease显示文摘Mutations in leucine-rich repeat kinase 2 (LRRK2) and glucocerebrosidase (GBA) represent two most common genetic causes of Parkinson’s disease (PD). Both genes are important in the autophagic-lysosomal pathway (ALP), defects of which are associated with α-synuclein (α-syn) accumulation. LRRK2 regulates macroautophagy via activation of the mitogen activated protein kinase/extracellular signal regulated protein kinase (MAPK/ERK) kinase (MEK) and the calcium-dependent adenosine monophosphate (AMP)-activated protein kinase (AMPK) pathways. Phosphorylation of Rab GTPases by LRRK2 regulates lysosomal homeostasis and endosomal trafficking. Mutant LRRK2 impairs chaperone-mediated autophagy, resulting in α-syn binding and oligomerization on lysosomal membranes. Mutations in GBA reduce glucocerebrosidase (GCase) activity, leading to glucosylceramide accumulation, α-syn aggregation and broad autophagic abnormalities. LRRK2 and GBA influence each other: GCase activity is reduced in LRRK2 mutant cells, and LRRK2 kinase inhibition can alter GCase activity in GBA mutant cells. Clinically, LRRK2 G2019S mutation seems to modify the effects of GBA mutation, resulting in milder symptoms than those resulting from GBA mutation alone. However, dual mutation carriers have an increased risk of PD and earlier age of onset compared with single mutation carriers, suggesting an additive deleterious effect on the initiation of PD pathogenic processes. Crosstalk between LRRK2 and GBA in PD exists, but its exact mechanism is unclear. Drugs that inhibit LRRK2 kinase or activate GCase are showing efficacy in pre-clinical models.
