| 1 | First identification of kdr allele F1534S in VGSC gene and its association with resistance to pyrethroid insecticides in Aedes albopictus populations from Haikou City,Hainan Island,China显示文摘Background:Aedes albopictus is distributed widely in China,as a primary vector of Dengue fever and Chikungunya fever in south of China.Chemical insecticide control is one of the integrated programmes to prevent mosquito-borne diseases.Long-term applications of pyrethroids have resulted in the development of resistance in Ae.albopictus populations in China.However,the susceptibility of Ae.albopictus to pyrethroids in Hainan Island was unclear.Knockdown resistance(kdr),caused by point mutations in the VGSC gene,is one of the mechanisms that confer resistance to DDT and pyrethroids.This study was to investigate the resistance level of Ae.albopictus populations in Haikou City to three pyrethroid insecticides,and elucidate the relationship between the resistant phenotype and kdr mutations.Methods:The Aedes albopictus samples were collected in Xinbu Island(XI),Longtang Town(LT),Shishan Town(ST),Baishamen Park(BP),and Flower Market(FM)from Haikou City,Hainan Island,China.The larval susceptibility to deltamethrin,permethrin and beta-cypermethrin was tested by larval bioassays,and adult susceptibility to deltamethrin and DDT was determined by adult bioassays.The degree of resistance was determined by resistance ratio value(RR50>3)for larvae and by mortality for adult.The kdr alleles at codon 1534 of the VGSC gene were genotyped.The relationship between kdr genotypes and resistant phenotypes was analyzed by Chi-square test.Results:Out of five populations,assessed by larval bioassays,XI was susceptible to deltamethrin and permethrin;LT was susceptible to permethrin and beta-cypermethrin;and ST was susceptible to permithrin.FM and BP both were resistant to all of the three pyrethroids,and FM showed the highest degree of resistance,with RR50 values from 65.17 to 436.36.A total of 493 individuals from the larval bioassays were genotyped for kdr alleles.Five alleles were detected,including two wildtype alleles,TTC(F)(67.04%)and TTT(F)(0.41%),and three mutant alleles,TGC(C)(0.30%),TCC(S)(31.54%)and TTG(L)(0.71%).There was a clear correlation between mutant alleles(or F1534S)and resistant phenotypes(P<0.01).Conclusion:Two novel kdr mutant alleles F1534S and F1534L were detected in the pyrethroid resistant populations of Ae.albopictus in Haikou Hainan,China.For the first time,the mutant F1534S was associated with pyrethroid resistance in Ae.albopictus. | Huiying Chen Kaili Li Xiaohua Wang Xinyan Yang Yi Lin Fang Cai Wenbin Zhong Chunyan Lin Zhongling Lin Yajun Ma | 2016 | Infectious Diseases of Poverty2016,5,1: | 19 |
| 2 | Integration, Quality Assurance, and Usage of Global Aircraft Observations in CRA显示文摘This paper presents a detailed description of integration, quality assurance procedure, and usage of global aircraft observations for China's first generation global atmospheric reanalysis(CRA) product(1979–2018). An integration method named 'classified integration' is developed. Aircraft observations from nine different sources are integrated into the Integrated Global Meteorological Observation Archive from Aircraft(IGMOAA), a new dataset from the National Meteorological Information Center(NMIC) of the China Meteorological Administration(CMA). IGMOAA consists of global aircraft temperature, wind, and humidity data from the surface to 100 h Pa, extending from 1973 to the present. Compared with observations assimilated in the Climate Forecast System Reanalysis(CFSR) of NCEP,the observation number of IGMOAA increased by 12.9% between 2010 and 2014, mainly as a result of adding more Chinese Aircraft Meteorological Data Relay(AMDAR) data. Complex quality control procedures for aircraft observations of NCEP are applied to detect data errors. Observations are compared with ERA-Interim reanalysis from 1979 to 2018 to investigate data quality of different types and aircraft, and subsequently to develop the blacklists for CRA. IGMOAA data have been assimilated in CRA in 2018 and are real-time updated at the CMA Data-as-a-Service(CMADaa S) platform. For CRA, the fits to observations improve over time. From 1994 to 2018, root-meansquare error(RMSE) of observations relative to CRA background decreases from 1.8 to 1.0℃ for temperature above 300 h Pa, and from 4.5 to 3 m s^(-1) for zonal wind. | 3 | Engineering Escherichia coli to improve tryptophan production via genetic manipulation of precursor and cofactor pathways显示文摘Optimizing the supply of biosynthetic precursors and cofactors is usually an effective metabolic strategy to improve the production of target compounds.Here,the combination of optimizing precursor synthesis and balancing cofactor metabolism was adopted to improve tryptophan production in Escherichia coli.First,glutamine synthesis was improved by expressing heterologous glutamine synthetase from Bacillus subtilis and Bacillus megaterium in the engineered Escherichia coli strain KW001,resulting in the best candidate strain TS-1.Then icd and gdhA were overexpressed in TS-1,which led to the accumulation of 1.060 g/L tryptophan.Subsequently,one more copy of prs was introduced on the chromosome to increase the flux of 5-phospho-α-D-ribose 1-diphosphate followed by the expression of mutated serA and thrA to increase the precursor supply of serine,resulting in the accumulation of 1.380 g/L tryptophan.Finally,to maintain cofactor balance,sthA and pntAB,encoding transhydrogenase,were overexpressed.With sufficient amounts of precursors and balanced cofactors,the engineered strain could produce 1.710 g/L tryptophan after 48 h of shake-flask fermentation,which was 2.76-times higher than the titer of the parent strain.Taken together,our results demonstrate that the combination of optimizing precursor supply and regulating cofactor metabolism is an effective approach for high-level production of tryptophan.Similar strategies could be applied to the production of other amino acids or related derivatives. | Zhu Li Dongqin Ding Huiying Wang Linxia Liu Huan Fang Tao Chen Dawei Zhang | 2020 | Synthetic and Systems Biotechnology2020,5,3: | 5 |
| 4 | 4-Formyl-2-(2H-benzotriazol-2-yl)-phenol: an ESIPT chromophore显示文摘In the present work, excited state intramolecular proton transfer(ESIPT) emission properties of a new benzotriazole derivative 4-formyl-2-(2H-benzotriazol-2-yl)-phenol(C1) were studied. 4-Formyl-2-(2H-benzotriazol-2-yl)-methoxy-benzene(C2), 4-formyl-2-(2H-benzotriazol-2-yl)-phenyl acetate(C3) and 4-methyl-2-(2H-benzotriazol-2-yl)-phenol(C4) were used as the reference molecules.1H NMR chemical shift of hydroxy group in C1 was located at more down field than that of C4 or p-hydroxy-benzaldehyde(C5), respectively. C1 showed two absorption bands in the range of 260–400 nm zones in various solvents, while C2 and C3 exhibited single absorption band. The equal molar mixtures of C2/C5 or C3/C5 showed single absorption band.C2 and C3 displayed single fluorescence emission band in various solvents, while C1 exhibited dual emission bands in some strong polar solvents. Furthermore, the second emission band in these strong polar solvents showed the large Stokes shift. The results show that the second emission band of C1 was produced by ESIPT. C2 and C3 could not undergo ESIPT due to no hydroxy group. | 5 | Depression accelerates gastric cancer invasion and metastasis by inducing a neuroendocrine phenotype via the catecholamine/β_(2)-AR/MACC1 axis显示文摘Background:Depression is a common,easily ignored,accompanied disease of gastric cancer(GC)patients and is often observed with elevated plasma catecholamine levels.Depression frequently promotes GC progression and leads to poor clinical outcomes;however,the molecular mechanisms underlying depression-induced GC progression remain poorly understood.We aimed to study the effects of depression on GC progression and explore possible mechanisms mediating the action of depression-associated catecholamines on GC.Methods:Depression states of GC patients were graded using the Patient Health Questionnaire-9,and plasma catecholamine levels were examined by high performance liquid chromatography coupled with tandem mass spectrometry.Migrative and invasive GC cells were examined using transwell assays,and metastatic GC niches were imaged using bioluminescence technology in a depression mouse model established with chronic unpredictable mild stress.Mouse depression-like behaviors were assessed through sucrose preference,forced swimming,and tail suspension tests.Characteristics of the neuroendocrine phenotype were observed via RT-PCR,Western blotting,flow cytometry,and transmission electron microscopy.Results:Fifty-one GC patients(age:53.61±1.79 years;cancer duration:3.71±0.33 months;depression duration:2.37±0.38 months;male-to-female ratio:1.55:1)were enrolled in the study.Depression grade was significantly higher in GC patients showing higher plasma levels of catecholamines(epinephrine:P=0.018;noradrenaline:P=0.009),higher oncogene metastasis-associated in colon cancer-1(MACC1)level(P=0.018),and metastasis(P<0.001).Further,depression-associated catecholamine specifically bound to the beta-2 adrenergic receptor(β_(2)-AR)and upregulated MACC1 expression,and thus promoting neuroendocrine phenotypic transformation through direct binding betweenMACC1 and synaptophysin.Eventually,the neuroendocrine phenotypic transformation accelerated GC invasion in vitro and metastasis in vivo.However,β_(2)-AR antagonist ICI-118,551 or MACC1 silencing effectively blocked the catecholamineinduced neuroendocrine phenotypic transformation and eliminated depressionenhanced GC migration and invasion.Moreover,β_(2)-AR blocking or MACC1 silencing prevented GC metastasis attributed to a neuroendocrine phenotype in a depression mouse model.Conclusions:Catecholamine-induced neuroendocrine phenotypes of GC cells led to depression-accelerated GC invasion and metastasis via the β_(2)-AR/MACC1 axis,while β_(2)-AR antagonist or MACC1 silencing could reverse it,showing promising potential therapeutic strategies for improving the outcome of GC patients with comorbid depression. | Changqie Pan Jianhua Wu Siting Zheng Huiying Sun Yisheng Fang Zhenhua Huang Min Shi Li Liang Jianping Bin Yulin Liao Jinzhang Chen Wangjun Liao | 2021 | Cancer Communications2021,41,10: | 4 |
| 6 | Photocatalytic activity of TiO2 containing anatase nanoparticles and rutile nanoflower structure consisting of nanorods显示文摘A series of TiO2 with diferent crystal phases and morphologies was synthesized via a facile hydrothermal process using titanium n- butoxide and concentrated hydrochloric acid as raw materials. The photocatalytic activity of the samples was evaluated by degradation of Methyl Orange in aqueous solution under UV-Visible light irradiation. On the basis of detailed analysis of the characterizing results of high-resolution transmission electron microscopy, X-ray powder difraction measurements, X-ray photoelectron spectroscopy and Brunauer-Emmett-Teller measurement, it was concluded that the photo-activity of the catalyst is related directly to the 3D morphology and the crystal phase composition. An excellent catalyst should have both a rutile 3D flower-like structure and anatase granulous particles. The 3D flower-like structure could enhance light harvesting, as well as the transfer of reactant molecules from bulk solution to the reactive sites on TiO2.
