6Mining and fine-tuning sugar uptake system for titer improvement of milbemycins in Streptomyces bingchenggensis显示文摘Dramatic decrease of sugar uptake is a general phenomenon in Streptomyces at stationary phase,when antibiotics are extensively produced.Milbemycins produced by Streptomyces bingchenggensis are a group of valuable macrolide biopesticides,while the low yield and titer impede their broad applications in agricultural field.Considering that inadequate sugar uptake generally hinders titer improvement of desired products,we mined the underlying sugar uptake systems and fine-tuned their expression in this work.First,we screened the candidates at both genomic and transcriptomic level in S.bingchenggensis.Then,two ATP-binding cassette transporters named TP2 and TP5 were characterized to improve milbemycin titer and yield significantly.Next,the appropriate native temporal promoters were selected and used to tune the expression of TP2 and TP5,resulting in a maximal milbemycin A3/A4 titer increase by 36.9%to 3321 mg/L.Finally,TP2 and TP5 were broadly finetuned in another two macrolide biopesticide producers Streptomyces avermitilis and Streptomyces cyaneogriseus,leading to a maximal titer improvement of 34.1%and 52.6%for avermectin B1a and nemadectin,respectively.This work provides useful transporter tools and corresponding engineering strategy for Streptomyces.Pinjiao Jin Shanshan Li Yanyan Zhang Liyang Chu Hairong He Zhuoxu Dong Wensheng Xiang 2020Synthetic and Systems Biotechnology2020,5,3:1

8Analysis of COVID-19 Guideline Quality and Change of Recommendations:A Systematic Review显示文摘Background.Hundreds of coronavirus disease 2019(COVID-19)clinical practice guidelines(CPGs)and expert consensus statements have been developed and published since the outbreak of the epidemic.However,these CPGs are of widely variable quality.So,this review is aimed at systematically evaluating the methodological and reporting qualities of COVID-19 CPGs,exploring factors that may influence their quality,and analyzing the change of recommendations in CPGs with evidence published.Methods.We searched five electronic databases and five websites from 1 January to 31 December 2020 to retrieve all COVID-19 CPGs.The assessment of the methodological and reporting qualities of CPGs was performed using the AGREE II instrument and RIGHT checklist.Recommendations and evidence used to make recommendations in the CPGs regarding some treatments for COVID-19(remdesivir,glucocorticoids,hydroxychloroquine/chloroquine,interferon,and lopinavir-ritonavir)were also systematically assessed.And the statistical inference was performed to identify factors associated with the quality of CPGs.Results.We included a total of 92 COVID-19 CPGs developed by 19 countries.Overall,the RIGHT checklist reporting rate of COVID-19 CPGs was 33.0%,and the AGREE II domain score was 30.4%.The overall methodological and reporting qualities of COVID-19 CPGs gradually improved during the year 2020.Factors associated with high methodological and reporting qualities included the evidence-based development process,management of conflicts of interest,and use of established rating systems to assess the quality of evidence and strength of recommendations.The recommendations of only seven(7.6%)CPGs were informed by a systematic review of evidence,and these seven CPGs have relatively high methodological and reporting qualities,in which six of them fully meet the Institute of Medicine(IOM)criteria of guidelines.Besides,a rapid advice CPG developed by the World Health Organization(WHO)of the seven CPGs got the highest overall scores in methodological(72.8%)and reporting qualities(83.8%).Many CPGs covered the same clinical questions(it refers to the clinical questions on the effectiveness of treatments of remdesivir,glucocorticoids,hydroxychloroquine/chloroquine,interferon,and lopinavirritonavir in COVID-19 patients)and were published by different countries or organizations.Although randomized controlled trials and systematic reviews on the effectiveness of treatments of remdesivir,glucocorticoids,hydroxychloroquine/chloroquine,interferon,and lopinavir-ritonavir for patients with COVID-19 have been published,the recommendations on those treatments still varied greatly across COVID-19 CPGs published in different countries or regions,which may suggest that the CPGs do not make sufficient use of the latest evidence.Conclusions.Both the methodological and reporting qualities of COVID-19 CPGs increased over time,but there is still room for further improvement.The lack of effective use of available evidence and management of conflicts of interest were the main reasons for the low quality of the CPGs.The use of formal rating systems for the quality of evidence and strength of recommendations may help to improve the quality of CPGs in the context of the COVID-19 pandemic.During the pandemic,we suggest developing a living guideline of which recommendations are supported by a systematic review for it can facilitate the timely translation of the latest research findings to clinical practice.We also suggest that CPG developers should register the guidelines in a registration platform at the beginning for it can reduce duplication development of guidelines on the same clinical question,increase the transparency of the development process,and promote cooperation among guideline developers all over the world.Since the International Practice Guideline Registry Platform has been created,developers could register guidelines prospectively and internationally on this platform.Siya Zhao Shuya Lu Shouyuan Wu Zijun Wang Qiangqiang Guo Qianling Shi Hairong Zhang Juanjuan Zhang Hui Liu Yunlan Liu Xianzhuo Zhang Ling Wang Mengjuan Ren Ping Wang Hui Lan Qi Zhou Yajia Sun Jin Cao Qinyuan Li Janne Estill Joseph LMathew Hyeong Sik Ahn Myeong Soo Lee Xiaohui Wang Chenyan Zhou Yaolong Chen 2021Health Data Science2021,,1:1

15Building digital life systems for future biology and medicine显示文摘The rapid development of biological technology (BT) and information technology (IT) especially of genomics and artificial intelligence (AI) is bringing great potential for revolutionizing future medicine. We propose the concept and framework of Digital Life Systems or dLife as a new paradigm to unleash this potential. It includes the multi-scale and multi-granule measure and representation of life in the digital space, the mathematical and/or computational modeling of the biology behind physiological and pathological processes, and ultimately cyber twins of healthy or diseased human body in the virtual space that can be used to simulate complex biological processes and deduce effects of medical treatments. 19Radon Measure Solutions to Riemann Problems for Isentropic Compressible Euler Equations of Polytropic Gases显示文摘We solve the Riemann problems for isentropic compressible Euler equations of polytropic gases in the class of Radon measures,and the solutions admit the concentration of mass.It is found that under the requirement of satisfying the over-compressing entropy condition:(i)there is a unique delta shock solution,corresponding to the case that has two strong classical Lax shocks;(ii)for the initial data that the classical Riemann solution contains a shock wave and a rarefaction wave,or two shocks with one being weak,there are infinitely many solutions,each consists of a delta shock and a rarefaction wave;(iii)there are no delta shocks for the case that the classical entropy weak solutions consist only of rarefaction waves.These solutions are self-similar.Furthermore,for the generalized Riemann problem with mass concentrated initially at the discontinuous point of initial data,there always exists a unique delta shock for at least a short time.It could be prolonged to a global solution.Not all the solutions are self-similar due to the initial velocity of the concentrated point-mass(particle).Whether the delta shock solutions constructed satisfy the over-compressing entropy condition is clarified.This is the first result on the construction of singular measure solutions to the compressible Euler system of polytropic gases,that is strictly hyperbolic,and whose characteristics are both genuinely nonlinear.We also discuss possible physical interpretations and applications of these new solutions.Yunjuan Jin Aifang Qu Hairong Yuan 2023Communications on Applied Mathematics and Computation2023,5,3:0
20Design and experiment of a Chinese chive harvester显示文摘Recognizing the labor intensity of current domestic Chinese chive harvesting and the need for its mechanized harvesting,a self-propelled electric Chinese chive harvester is designed.The machine is composed of a control system,supporting mechanism,cutting parts,conveying and clamping mechanisms,frame,and collection system.It can complete Chinese chive feeding,cutting,transmission,and binning operations at one time.It has a cutting width of 0.25 m and wheelbases of 260 mm and 460 mm.Two 12 V,42 Ah lead-acid batteries were used in series as the power supply.The mechanical properties of Chinese chive were analyzed,and shear and compression tests were carried out to determine that the average value of the maximum shear force on the chives is 129 N.Up to a compressive force of 225 N,Chinese chives are not damaged.According to the physical characteristics of Chinese chives and the requirements of the harvesting operations,design and calculation of the key components of the harvester’s transmission mechanism and the strength of the whole machine frame were completed.An embedded control system and Cortex-M3 microprocessor were used to control the harvester and realize its functions of rolling and harvesting while regulating its speed and protecting its motor.Using the Box-Behnken test design method,the machine’s operating speed,cutter speed,and cutter height were selected as influencing factors,and the lodging rate and loss rate were evaluated as a test.A mathematical regression model of influencing factors and evaluation indexes was established to analyze the influence of these factors and indexes and optimize test parameters.The optimal parameter combination is determined to be:operating speed,1.1 km/h;cutter speed,220 r/min;cutter height 2.0 cm.The parameters were verified from field tests and the results showed that under the optimized parameter combination,the average lodging rate was 3.96% and the average loss rate was 3.23%,indicating that the Chinese chive harvester meets the agronomic requirements of Chinese chive harvesting.This harvester could be modified to harvest other crops such as chrysanthemums and Mongolian Chinese chive.Yulong Feng Xuchao Yin Hairong Jin Wenyu Tong Xiaofeng Ning 2023International Journal of Agricultural and Biological Engineering2023,16,2:0
