| 1 | A landscape shape index-based sampling approach for land cover accuracy assessment显示文摘Spatial sampling design is one of the key steps in land cover accuracy assessment,and many traditional sampling approaches may not achieve credible spatial sampling due to the high spatial heterogeneity of land cover.This paper characterizes the spatial heterogeneity with three-level LSIs and determines the subsequent sample sizes and their spatial distributions.The three-level LSIs are rLSI in a region,cLSI for each land cover class and uLSI in each geographic sampling unit in the region.The rLSIs are used to derive appropriate sample sizes in the target regions.The cLSIs are used to assure that larger sample numbers are allocated to land cover classes with higher spatial heterogeneity.The uLSIs provide useful measures for selecting optimal geographic units in which sample sites will be located.This LSI-based sampling approach can derive the sample sizes and determine their distributions in an adaptive way according to the spatial heterogeneity.An experimental case study further demonstrates that the LSI-based sampling approach obtains more appropriate sample sizes for each region,sufficient sample numbers for rare classes,and optimal sample distributions in the geographical space. | CHEN Fei CHEN Jun WU Hao HOU DongYang ZHANG WeiWei ZHANG Jun ZHOU XiaoGuang CHEN LiJun | 2016 | Science China(Information Sciences)2016,59,12: | 17 |
| 2 | miR-137 inhibits melanoma cell proliferation through downregulation of GLO1显示文摘Late-stage melanoma is refractory to current therapies. MicroRNAs (miRNAs) can modulate many physiological and pathological processes of melanoma. Studies have demonstrated that miR-137 acts as a tumor suppressor by inhibiting the proliferation of melanoma cells through targeting multiple mRNAs. The glyoxalase system member glyoxalase 1 (GLO1) is the principal scavenging enzyme of methylglyoxal (MG), a toxic byproduct of glycolysis. Using 35S in vivo/vitro labelling analysis for dynamic proteomics (SiLAD), we found that miR-137 downregulated the expression of GLO1 in melanoma cells.Bioinformatics analysis predicted that GLO1 is a direct target of miR-137. This was validated by dual luciferase reporter assay.Quantitative RT-PCR (qRT-PCR) and western blot analysis indicated that miR-137 could decrease endogenous GLO1 expression.Furthermore, siRNA targeting of GLO1 mimicked inhibition of melanoma cell proliferation caused by miR-137 overexpression.Re-expression of GLO1 was able to restore miR-137-mediated suppression of melanoma cell proliferation. | 9 | Mitochondrial genome sequences of Artemia tibetiana and Artemia urmiana:assessing molecular changes for high plateau adaptation显示文摘Brine shrimps,Artemia(Crustacea,Anostraca),inhabit hypersaline environments and have a broad geographical distribution from sea level to high plateaus.Artemia therefore possess significant genetic diversity,which gives them their outstanding adaptability.To understand this remarkable plasticity,we sequenced the mitochondrial genomes of two Artemia tibetiana isolates from the Tibetan Plateau in China and one Artemia urmiana isolate from Lake Urmia in Iran and compared them with the genome of a low-altitude Artemia,A.franciscana.We compared the ratio of the rate of nonsynonymous(Ka) and synonymous(Ks) substitutions(Ka/Ks ratio) in the mitochondrial protein-coding gene sequences and found that atp8 had the highest Ka/Ks ratios in comparisons of A.franciscana with either A.tibetiana or A.urmiana and that atp6 had the highest Ka/Ks ratio between A.tibetiana and A.urmiana.Atp6 may have experienced strong selective pressure for high-altitude adaptation because although A.tibetiana and A.urmiana are closely related they live at different altitudes.We identified two extended termination-associated sequences and three conserved sequence blocks in the D-loop region of the mitochondrial genomes.We propose that sequence variations in the D-loop region and in the subunits of the respiratory chain complexes independently or collectively contribute to the adaptation of Artemia to different altitudes. | ZHANG HangXiao LUO QiBin SUN Jing LIU Fei WU Gang YU Jun WANG WeiWei | 2013 | Science China(Life Sciences)2013,56,5: | 4 |
| 10 | OsPKpa_1 encodes a plastidic pyruvate kinase that affects starch biosynthesis in the rice endosperm显示文摘Pyruvate kinase(PK) is a key enzyme in glycolysis and carbon metabolism. Here, we isolated a rice(Oryza sativa) mutant, w59, with a white-core floury endosperm. Map-based cloning of w59 identified a mutation in OsPKpa_1, which encodes a plastidic isoform of PK(PKp). OsPKpa_1 localizes to the amyloplast stroma in the developing endosperm, and the mutation of OsPKpa_1 in w59 decreases the plastidic PK activity, resulting in dramatic changes to the lipid biosynthesis in seeds. The w59 grains were also characterized by a marked decrease in starch content. Consistent with a decrease in number and size of the w59 amyloplasts, large empty spaces were observed in the central region of the w59 endosperm, atthe early grain-filling stage. Moreover, a phylogenetic analysis revealed four potential rice isoforms of OsPKp. We validated the in vitro PK activity of these OsPKps through reconstituting active PKp complexes derived from inactive individual OsPKps, revealing the heteromeric structure of rice PKps, which was further confirmed using a protein–protein interaction analysis. | 12 | Three-dimensional reconstruction of Picea wilsonii Mast. pollen grains using automated electron microscopy显示文摘Three-dimensional electron microscopy(3 D-EM) has attracted considerable attention because of its ability to provide detailed information with respect to developmental analysis. However, large-scale high-resolution 3 D reconstruction of biological samples remains challenging. Herein, we present a 3 D view of a Picea wilsonii Mast. pollen grain with 100 nm axial and38.57 nm lateral resolution using AutoCUTS-SEM(automatic collector of ultrathin sections-scanning electron microscopy). We established a library of 3,127 100 nm thick serial sections of pollen grains for preservation and observation, demonstrating that the protocol can be used to analyze large-volume samples. After obtaining the SEM images, we reconstructed an entire pollen grain comprising 734 serial sections. The images produced by 3D reconstruction clearly revealed the main components of the P.wilsonii pollen grain, i.e., two sacci and pollen corpus, tube cell, generative cell, and two degenerated prothallial cells, and their internal organization. In addition, we performed a quantitative analysis of the different pollen grain cells, including sacci, and found that there were 202 connections within a saccus SEM image. | 15 | Loss-of-function mutation of rice SLAC7 decreases chloroplast stability and induces a photoprotection mechanism in rice显示文摘Plants absorb sunlight to power the photochemical reactions of photosynthesis, which can potentially damage the photosynthetic machinery. However, the mechanism that protects chloroplasts from the damage remains unclear. In this work, we demonstrated that rice(Oryza sativa L.) SLAC7 is a generally expressed membrane protein. Lossof-function of SLAC7 caused continuous damage to the chloroplasts of mutant leaves under normal light conditions.Ion leakage indicators related to leaf damage such as H_2O_2 and abscisic acid levels were significantly higher in slac7-1 than in the wild type. Consistently, the photosynthesis ef ficiency and Fv/Fm ratio of slac7-1 were significantly decreased(similar to photoinhibition). In response to chloroplast damage, slac7-1 altered its leaf morphology(curled or fused leaf) by the synergy between plant hormones and transcriptional factors to decrease the absorption of light, suggesting that a photoprotection mechanism for chloroplast damage was activated in slac7-1. When grown in dark conditions, slac7-1 displayed a normal phenotype. SLAC7 under the control of the At SLAC1 promoter could partially complement the phenotypes of Arabidopsis slac1 mutants, indicating a partia conservation of SLAC protein functions. | 18 | Saccades of video head impulse test in Meniere's disease and Vestibular Migraine: What can we learn from?显示文摘Background:Saccades are often observed on video head impulse tests(vHIT)in patients with Meniere's Disease(MD)and Vestibular Migraine(VM).However,their saccadic features are not fully described.Objective:This study aims to identify the saccades characteristics of MD and VM.Methods:75 VM patients and 103 definite unilateral MD patients were enrolled in this study.First raw saccades were exported and analyzed.The VM patients were divided into left and right based on their ears,while the MD patients were separated into affected and unaffected subgroups based on their audiograms and symptoms.Results:The MD patients have more saccades on the affected side(85%vs.69%),and saccade velocity is more consistent than the contralateral side(shown by the coefficient of variation).The saccades occurrence rates on both sides are similar in VM(77%vs.76%),as are other saccadic parameters.The MD patients have more significant inter-aural differences than the VM patients,manifested in higher velocity(p-value 0.000),earlier arriving(p-value 0.010),and more time-domain gathered(p-value 0.003)on the affected side.Conclusions:Bilateral saccades are commonly observed in MD and VM.In contrast to MD,saccades on VM are subtle,scattered,and late-arrived.Furthermore,the MD patients showed inconsistent saccadic distribution with more velocity-uniform saccades on the affected side. | Yi Du Xingjian Liu Lili Ren Yu Wang Fei Ji Weiwei Guo Ziming Wu | 2023 | Journal of Otology2023,18,2: | 1 |
