2Host metabolism dysregulation and cell tropism identification in human airway and alveolar organoids upon SARS-CoV-2 infection显示文摘The coronavirus disease 2019(COVID-19)pandemic is caused by infection with the severe acute respiratory syndrome coronavirus 2(SARS-CoV-2),which is spread primary via respiratory droplets and infects the lungs.Currently widely used cell lines and animals are unable to accurately mimic human physiological conditions because of the abnormal status of cell lines(transformed or cancer cells)and species differences between animals and humans.Organoids are stem cell-derived selforganized three-dimensional culture in vitro and model the physiological conditions of natural organs.Here we showed that SARS-CoV-2 infected and extensively replicated in human embryonic stem cells(hESCs)-derived lung organoids,including airway and alveolar organoids which covered the complete infection and spread route for SARS-CoV-2 within lungs.The infected ceils were ciliated,club,and alveolar type 2(AT2)cells,which were sequentially located from the proximal to the distal airway and terminal alveoli,respectively.Additionally,RNA-seq revealed early cell response to virus infection including an unexpected downregulation of the metabolic processes,especially lipid metabolism,in addition to the well-known upregulation of immune response.Further,Remdesivir and a human neutralizing antibody potently inhibited SARS-CoV-2 replication in lung organoids.Therefore,human lung organoids can serve as a pathophysiological model to investigate the underlying mechanism of SARS-CoV-2 infection and to discover and test therapeutic drugs for COVID-19.Rongjuan Pei Jianqi Feng Yecheng Zhang Hao Sun Lian Li Xuejie Yang Jiangping He Shuqi Xiao Jin Xiong Ying Lin Kun Wen Hongwei Zhou Jiekai Chen Zhili Rong Xinwen Chen 2021Protein & Cell2021,12,9:7
3Accelerated evolution of an Lhx2 enhancer shapes mammalian social hierarchies显示文摘Social hierarchies emerged during evolution,and social rank influences behavior and health of individuals.However,the evolutionary mechanisms of social hierarchy are still unknown in amniotes.Here we developed a new method and performed a genome-wide screening for identifying regions with accelerated evolution in the ancestral lineage of placental mammals,where mammalian social hierarchies might have initially evolved.Then functional analyses were conducted for the most accelerated region designated as placental-accelerated sequence 1(PAS1,P=3.15 × 10^-18).Multiple pieces of evidence show that PAS1 is an enhancer of the transcription factor gene Lhx2 involved in brain development.PAS1s isolated from various amniotes showed different c/s-regulatory activity in vitro,and affected the expression of Lhx2 differently in the nervous system of mouse embryos.PAS1 knock-out mice lack social stratification.PAS1 knock-in mouse models demonstrate that PAS1s determine the social dominance and subordinate of adult mice,and that social ranks could even be turned over by mutated PAS1.All homozygous mutant mice had normal huddled sleeping behavior,motor coordination and strength.Therefore,PAS1-Lhx2 modulates social hierarchies and is essential for establishing social stratification in amniotes,and positive Darwinian selection on PAS1 plays pivotal roles in the occurrence of mammalian social hierarchies.Yuting Wang Guangyi Dai Zhili Gu Guopeng Liu Ke Tang Yi-Hsuan Pan Yujie Chen Xin Lin Nan Wu Haoshan Chen Su Feng Shou Qiu Hongduo Sun Qian Li Chuan Xu Yanan Mao Yong Edward Zhang Philipp Khaitovich Yan-Ling Wang Qunxiu Liu Jing-Dong Jackie Han Zhen Shao Gang Wei Chun Xu Naihe Jing Haipeng Li 2020Cell Research2020,30,5:4
4High Thermoelectric Performance of Cu-Doped PbSe-PbS System Enabled by High-Throughput Experimental Screening显示文摘Recent advances in high-throughput(HTP)computational power and machine learning have led to great achievements in exploration of new thermoelectric materials.However,experimental discovery and optimization of thermoelectric materials have long relied on the traditional Edisonian trial and error approach.Herein,we demonstrate that ultrahigh thermoelectric performance in a Cu-doped PbSe-PbS system can be realized by HTP experimental screening and precise property modulation.Combining the HTP experimental technique with transport model analysis,an optimal Se/S ratio showing high thermoelectric performance has been efficiently screened out.Subsequently,based on the screened Se/S ratio,the doping content of Cu has been subtly adjusted to reach the optimum carrier concentration.As a result,an outstanding peak zT~1:6 is achieved at 873 K for a 1.8 at%Cu-doped PbSe_(0.6)S_(0.4) sample,which is the superior value among the n-type Te-free lead chalcogenides.We anticipate that current work will stimulate large-scale unitization of the HTP experimental technique in the thermoelectric field,which can greatly accelerate the research and development of new high-performance thermoelectric materials.Li You Zhili Li Quanying Ma Shiyang He Qidong Zhang Feng Wang Guoqiang Wu Qingyi Li Pengfei Luo Jiye Zhang Jun Luo 2020Research2020,,1:4

11Facet-induced coordination competition for highly ordered CsPbBr_(3) nanoplatelets with strong polarized emission显示文摘Controllable anisotropic growth of perovskite nanocrystals(NCs)is challenging since it is difficult to separate the nucleation and growth processes.Here,a two-step nucleation strategy is proposed to control the binding interaction between surface ligands and NCs,resulting in facet-induced coordination competition.Oleic acid as surface activated ligand leads to the formation of defective lead bromine octahedron,and the binding interaction between 4-dodecylbenzenesulfonic acid and lead atoms promotes the formation of two kinds of binding interactions.Based on this strategy,the anisotropic growth of CsPbBr_(3) nanoplatelet(NPLs)with adjusted length from 11.4 to 24 nm,and the evolution of NPLs from stacked to tongue-shaped have been realized.Elemental line scan reveals the sulfur atoms mainly distribute at the edge of NPLs.Furthermore,binding energy calculation and experimental results illustrate the coordination competition of different binding interaction on specific facets induces the anisotropic growth of NPLs.Importantly,strong emission anisotropy of highly ordered NPLs with polarization ratio up to 0.58 is illustrated.This work not only deepens our understanding of the controllable synthesis of perovskite NCs,but also provides a reference for the regulation of light emitting diode and soler cells.Dandan Yang Xiaoming Li Yuelei Li Bo Cai Lin Su Shengli Zhang Zhili Juan Cuifang Meng Dongling Geng Jiaxin Chen Feng Xu Haibo Zeng 2022Nano Research2022,15,1:0

13Mechanical joint performances of friction self-piercing riveted carbon fiber reinforced polymer and AZ31B Mg alloy显示文摘Carbon fiber reinforced polymer(CFRP) and AZ31B Mg alloy were joined by the friction self-piercing riveting(F-SPR) with different steel rivet shank sizes. With the increase of rivet shank size, lap shear fracture load and mechanical interlock distance increased. Ultrafine grains were formed at the joint in AZ31B as a result of dynamic recrystallization, which contributed to the higher hardness. Fatigue life of the CFRP-AZ31B joint was studied at various peak loads of 0.5, 1, 2, and 3 kN and compared with the resistance spot welded AZ31B-AZ31B from the open literature. The fatigue performance was better at higher peak load(>2 kN) and comparable to that of resistance spot welding of AZ31B to AZ31B at lower peak loads(<1 kN). From fractography, the crack initiation for lower peak load(<1 kN) case was observed at the fretting positions on the top and bottom surfaces of AZ31B sheet. 18Galvanic corrosion of AZ31B joined to dual-phase steel with and without Zn layer by ultrasonic and friction stir welding显示文摘Galvanic corrosion of AZ31B joined with bare or Zn-coated DP590 steel by ultrasonic spot welding or linear friction stir welding was quantitatively studied by pre-defining anode and cathode in the lap joint samples. Corrosion volume and depth from Mg anode surfaces exposed to 0.1 M sodium chloride solution was analyzed as functions of cathode surface type and welding method. Characterization of as-welded joints was performed to identify any microstructural feature of the bonding zone that could impact galvanic corrosion behavior.COMSOL modeling with modified user subroutine was conducted to simulate the progression of Mg corrosion in the same joint and electrode configurations used for the corrosion experiments. 19Effects of Seasonal and Perennial Grazing on Soil Fauna Community and Microbial Biomass Carbon in the Subalpine Meadows of Yunnan, Southwest China显示文摘Grazing and over-grazing may drive changes in the diversity and functioning of below-ground meadow ecosystems. A field soil survey was conducted to compare microbial biomass carbon(Cmin) and soil fauna communities in the two main grassland management systems in subalpine regions of Yunnan Province, China: perennial grazing currently practiced due to increasing herd sizes and traditional seasonal grazing. A three-year exclosure experiment was then conducted to further compare the effects of different grazing practices,including treatments of no mowing, perennial grazing(NM + G), mowing followed by seasonal grazing(M + G), mowing and no grazing(M + NG), and no mowing or grazing(NM + NG). The comparative survey result revealed that Cminand total density of soil fauna were significantly lower at a perennially grazed site than at a seasonally grazed site. The experiment results showed that in comparison to non-grazing treatments(M + NG and NM + NG), grazing(NM + G and M + G) reduced total fauna density(by 150 individuals m^(-2)) and the number of taxonomic groups present(by 0.32 taxa m^(-2)). Mowing decreased Cmin(by 0.31 mg g^(-1)). Furthermore, the NM + G treatment(perennial grazing) had the lowest density of Collembola(16.24 individuals m^(-2)), one of the two most common taxonomic groups, although other taxonomic groups responded differently to the treatments. Treatment effects on soil fauna were consistent with those on above-ground grasses, in which C:N ratios were greatly reduced by grazing, with this effect being the greatest for the NM + G treatment. In contrast, different grazing treatments had little effect on C:N ratio of soil. Furthermore, the traditional grazing method(mowing followed by seasonal grazing) may have less severe effects on some taxonomic groups than perennial grazing.
