The Chromosome-Level Genome Sequence of the Autotetraploid Alfalfa and Resequencing of Core Germplasms Provide Genomic Resources for Alfalfa Research显示文摘Alfalfa(Medicago sativa)is one of the most important forage crops in the world;however, its molecular genetics and breeding research are hindered due to the lack of a high-quality reference genome. Here,we report a de novo assembled 816-Mb high-quality,chromosome-level haploid genome sequence for‘Zhongmu No.1’alfalfa,a heterozygous autotetraploid. The contig N50 is 3.92 Mb,and 49165 genes are annotated in the genome. The alfalfa genome is estimated to have diverged from M.truncatula approximately 8 million years ago.Genomic population analysis of 162 alfalfa accessions revealed high genetic diversity, weak population structure, and extensive gene flow from wild to cultivated alfalfa.Genome-wide association studies identified many candidate genes associated with important agronomic traits.Furthermore,we showed that MsFTa2,a Flowering Locus T homolog, whose expression is upregulated in salt-resistant germplasms, may be associated with fall dormancy and salt resistance.Taken together, these genomic resources will facilitate alfalfa genetic research and agronomic improvement. | Chen Shen Huilong Du Zhuo Chen Hongwei Lu Fugui Zhu Hong Chen Xiangzhao Meng Qianwen Liu Peng Liu Lihua Zheng Xiuxiu Li Jiangli Dong Chengzhi Liang Tao Wang | 2020 | Molecular Plant2020,13,9: | 21
Error Modeling and Sensitivity Analysis of a Parallel Robot with SCARA(Selective Compliance Assembly Robot Arm) Motions显示文摘Parallel robots with SCARA(selective compliance assembly robot arm) motions are utilized widely in the field of high speed pick-and-place manipulation. Error modeling for these robots generally simplifies the parallelogram structures included by the robots as a link. As the established error model fails to reflect the error feature of the parallelogram structures, the effect of accuracy design and kinematic calibration based on the error model come to be undermined. An error modeling methodology is proposed to establish an error model of parallel robots with parallelogram structures. The error model can embody the geometric errors of all joints, including the joints of parallelogram structures. Thus it can contain more exhaustively the factors that reduce the accuracy of the robot. Based on the error model and some sensitivity indices defined in the sense of statistics, sensitivity analysis is carried out. Accordingly, some atlases are depicted to express each geometric error's influence on the moving platform's pose errors. From these atlases, the geometric errors that have greater impact on the accuracy of the moving platform are identified, and some sensitive areas where the pose errors of the moving platform are extremely sensitive to the geometric errors are also figured out. By taking into account the error factors which are generally neglected in all existing modeling methods, the proposed modeling method can thoroughly disclose the process of error transmission and enhance the efficacy of accuracy design and calibration. | CHEN Yuzhen XIE Fugui LIU Xinjun ZHOU Yanhua | 2014 | Chinese Journal of Mechanical Engineering2014,27,4: | 17
Wnt and BMP signaling crosstalk in regulating dental stem cells:Implications in dental tissue engineering显示文摘Tooth is a complex hard tissue organ and consists of multiple cell types that are regulated by important signaling pathways such as Wnt and BMP signaling.Serious injuries and/or loss of tooth or periodontal tissues may significantly impact aesthetic appearance, essential oral functions and the quality of life.Regenerative dentistry holds great promise in treating oral/dental disorders. The past decade has witnessed a rapid expansion of our understanding of the biological features of dental stem cells, along with the signaling mechanisms governing stem cell self-renewal and differentiation. In this review,we first summarize the biological characteristics of seven types of dental stem cells, including dental pulp stem cells, stem cells from apical papilla, stem cells from human exfoliated deciduous teeth, dental follicle precursor cells, periodontal ligament stem cells, alveolar bone-derived mesenchymal stem cells(MSCs),and MSCs from gingiva. We then focus on how these stem cells are regulated by bone morphogenetic protein(BMP)and/or Wnt signaling by examining the interplays between these pathways.Lastly,we analyze the current status of dental tissue engineering strategies that utilize oral/dental stem cells by harnessing the interplays between BMP and Wnt pathways. We also highlight the challenges that must be addressed before the dental stem cells may reach any clinical applications. Thus,we can expect to witness significant progresses to be made in regenerative dentistry in the coming decade. | Fugui Zhang Jinlin Song Hongmei Zhang Enyi Huang Dongzhe Song Viktor Tollemar Jing Wang Jinhua Wang Maryam Mohammed Qiang Wei Jiaming Fan Junyi Liao Yulong Zou Feng Liu Xue Hu Xiangyang Qu Liqun Chen Xinyi Yu Hue H.Luu Michael J.Lee Tong-Chuan He Ping Ji | 2016 | Genes & Diseases2016,3,4: | 16

Differentiation trajectories and biofunctions of symbiotic and un-symbiotic fate cells in root nodules of Medicago truncatula显示文摘The root nodule is a complex symbiotic nitrogen fixation factory,in which cells are highly heterogeneous. However, the differentiation trajectories and interconnection of nodule cells remain largely unknown. In this study,we set up a modified protocol for nodule protoplast preparation and performed a single-cell RNA sequencing profiling of the indeterminate Medicago truncatula nodule. We designated 13 cell clusters with specific expression patterns in 14-day post inoculation nodules and constructed a spatial and functional cellular map based on experimental data and bioinformatic analyses.Pseudotime analysis further revealed that two groups of apical meristematic cells develop into symbiotic and un-symbiotic fate cells along their particular trajectories.Biofunction analysis of each cell cluster revealed their particularity and interrelation, especially that the un-infected cells in nitrogen fixation zone are also involved in nitrogen assimilation by undertaking the asparagine synthesis.Collectively, our data offer an important resource for investigating the mechanism of nodule organogenesis and symbiotic nitrogen fixation. | Qinyi Ye Fugui Zhu Fanghao Sun Tai-Cheng Wang Jiale Wu Peng Liu Chen Shen Jiangli Dong Tao Wang | 2022 | Molecular Plant2022,15,12: | 1
SATB2:A versatile transcriptional regulator of craniofacial and skeleton development, neurogenesis and tumorigenesis, and its applications in regenerative medicine显示文摘SATB2(special AT-rich sequence-binding protein 2)is a member of the special AT-rich binding protein family. As a transcription regulator,SATB2 mainly integrates higher-order chromatin organization.SATB2 expression appears to be tissue-and stage-specific,and is governed by several cellular signaling molecules and mediators.Expressed in branchial arches and osteoblast-lineage cells,SATB2 plays a significant role in craniofacial pattern and skeleton development. In addition to regulating osteogenic differentiation,SATB2 also displays versatile functions in neural development and cancer progression. As an osteoinductive factor,SATB2 holds great promise in improving bone regeneration toward bone defect repair. In this review,we have summarized our current understanding of the physiological and pathological functions of SATB2 in craniofacial and skeleton development, neurogenesis, tumorigenesis and regenerative medicine. | Xia Huang Qiuman Chen Wenping Luo Mikhail Pakvasa Yuxin Zhang Liwen Zheng Shuang Li Zhuohui Yang Huan Zeng Fang Liang Fugui Zhang Daniel A.Hu Kevin H.Qin Eric J.Wang David S.Qin Russell R.Reid Tong-Chuan He Aravind Athiviraham Mostafa El Dafrawy Hongmei Zhang | 2022 | Genes & Diseases2022,9,1: | 0
Integrating molybdenum sulfide selenide-based cathode with C-O-Mo heterointerface design and atomic engineering for superior aqueous Zn-ion batteries显示文摘Transition metal dichalcogenides(TMDs)have been regarded as promising cathodes for aqueous zinc-ion batteries(AZIBs)but suffer from sluggish reaction kinetics due to their poor conductivity and the strong electrostatic interaction between Zn-ion and cathode materials.Herein,a well-defined structure with MoSSe nanosheets vertically anchored on graphene is used as the cathode for AZIBs.The dissolution of Se into MoS2 lattice together with heterointerface design via developing C-O-Mo bonds improves the inherent conductivity, enlarges interlayer spacing, and generates abundant anionic vacancies. As a result, the Zn2+intercalation/deintercalation process is greatly improved, which is confirmed by theoretical modeling and ex-situ experimental results.Remarkably, the assembled AZIBs exhibit high-rate capability(124.2 mAh·g^(−1)at 5 A·g^(−1))and long cycling life(83%capacity retention after 1,200 cycles at 2 A·g^(−1)).Moreover,the assembled quasi-solid-state Zn-ion batteries demonstrate a stable cycling performance over 100 cycles and high capacity retention over 94%after 2,500 bending cycles. This study provides a new strategy to unlock the electrochemical activity of TMDs via interface design and atomic engineering, which can also be applied to other TMDs for multivalent batteries. | Hong Li Biao Chen Runhua Gao Fugui Xu Xinzhu Wen Xiongwei Zhong Chuang Li Zhihong Piao Nantao Hu Xiao Xiao Feng Shao Guangmin Zhou Jinlong Yang | 2023 | Nano Research2023,16,4: | 0
Yes-associated protein contributes to magnesium alloy-derivedinflammation in endothelial cells显示文摘Magnesium alloy(Mg alloy)has attracted massive attention in the potential applications of cardiovascular stents because of its good biocompatibility and degradability. However, whether and how the Mg alloy induces inflammation in endothelial cells remains unclear. In the present work,we investigated the activation of Yes-associated protein(YAP)upon Mg alloy stimuli and unveiled the transcriptional function in Mg alloy-induced inflammation.Quantitative RT–PCR,western blotting and immunofluorescence staining showed that Mg alloy inhibited the Hippo pathway to facilitate nuclear shuttling and activation of YAP in human coronary artery endothelial cells(HCAECs).Chromatin immunoprecipitation followed sequencing was carried out to explore the transcriptional function of YAP in Mg alloy-derived inflammation. This led to the observation that nuclear YAP further bonded to the promoter region of inflammation transcription factors and co-transcription factors. This binding event activated their transcription and modified mRNA methylation of inflammation-related genes through regulating the expression of N6-methyladenosine modulators(METTL3,METTL14,FTO and WTAP).This then promoted inflammation-related gene expression and aggravated inflammation in HCAECs.In YAP deficiency cells,Mg alloy-induced inflammation was reduced.Collectively, our data suggest that YAP contributes to the Mg alloy-derived inflammation in HCAECs and may provide a potential therapeutic target that alleviates inflammation after Mg alloy stent implantation. | Hongchi Yu Zhe Hou Nuoya Chen Rifang Luo Li Yang Michael Miao Xiaoyi Ma Lifeng Zhou Fugui He Yang Shen Xiaoheng Liu Yunbing Wang | 2022 | Regenerative Biomaterials2022,9,1: | 0

The biological responses and mechanisms of endothelial cells to magnesium alloy显示文摘Due to its good biocompatibility and degradability, magnesium alloy(Mg alloy)has shown great promise in cardiovascular stent applications.Rapid stent re-endothelialization is derived from migrated and adhered endothelial cells(ECs),which is an effective way to reduce late thrombosis and inhibit hyperplasia. However, fundamental questions regarding Mg alloy affecting migration and adhesion of ECs are not fully understood. Here,we evaluated the effects of Mg alloy on the ECs proliferation, adhesion and migration.A global gene expression profiling of ECs co-culturing with Mg alloy was conducted, and the adhesion-and migration-related genes were examined. We found that Mg alloy had no adverse effects on ECs viability but significantly affected ECs migration and adhesion.Co-cultured with Mg alloy extract,ECs showed contractive adhesion morphology and decreased motility, which was supported by the down-regulation of adhesion-related genes(Paxillin and Vinculin)and migration-related genes(RAC 1,Rho A and CDC 42).Accordingly,the re-endothelialization of Mg alloy stent was inhibited in vivo. Our results may provide new inspiration for improving the broad application of Mg alloy stents. | Zhe Hou Maolong Xiang Nuoya Chen Xiao Cai Bo Zhang Rifang Luo Li Yang Xiaoyi Ma Lifeng Zhou Fugui He Hongchi Yu Yunbing Wang | 2021 | Regenerative Biomaterials2021,8,3: | 0
