1Effect of dry density on ^(125)I diffusion in GMZ bentonite显示文摘Gaomiaozi(GMZ) bentonite is regarded as the favorable candidate backfilling material for a potential repository in China.It is important to understand the diffusion behavior of 125 I in GMZ bentonite and compare the diffusion behavior in GMZ and other types of bentonite like MX-80,Avonlea,etc.Therefore,through-and out-diffusion experiments were conducted to obtain the effective diffusion coefficient(D e) and distribution coefficient(K d).A computer code named Fitting for diffusion coefficient(FDP) was used for the experimental data processing and theoretical modeling.At the dry density of GMZ bentonite from 1600-2000 kg/m 3,the D e values of 125 I were(2.4-20.4) × 10-12 m 2 /s and K d values were constants.At dry density above 1800 kg/m 3,the diffusion behaviors were almost the same,indicating that the anion exclusion was ineffective.Out-diffusion results showed that the species of 125 I may be changed during the diffusion processing.It was probably caused by some organic matters or reducing substances in GMZ bentonite.Since the main composition of bentonite is montmorillonite,similar diffusion parameters were obtained in GMZ and other types of bentonite.The relationship of D e and accessible porosity(acc) could be described by Archie's law with exponent n = 1.2-2.8 for 125 I diffusion in bentonite,whereas n = 2.0 in GMZ bentonite.Furthermore,bentonite with the dry density of 1800 kg/m 3 was proposed as the backfilling materials used in the construction of high level radioactivity waste repository.WU Tao LI JinYing DAI Wei XIAO GuoPing SHU FuJun YAO Jun SU YuLan SHI Lei 2012Science China Chemistry2012,55,9:11
2LncRNA Nron regulates osteoclastogenesis during orthodontic bone resorption显示文摘Activation of osteoclasts during orthodontic tooth treatment is a prerequisite for alveolar bone resorption and tooth movement.However,the key regulatory molecules involved in osteoclastogenesis during this process remain unclear.Long noncoding RNAs(lnc RNAs)are a newly identified class of functional RNAs that regulate cellular processes,such as gene expression and translation regulation.Recently,lnc RNAs have been reported to be involved in osteogenesis and bone formation.However,as the most abundant noncoding RNAs in vivo,the potential regulatory role of lnc RNAs in osteoclast formation and bone resorption urgently needs to be clarified.We recently found that the lnc RNA Nron(long noncoding RNA repressor of the nuclear factor of activated T cells)is highly expressed in osteoclast precursors.Nron is downregulated during osteoclastogenesis and bone ageing.To further determine whether Nron regulates osteoclast activity during orthodontic treatment,osteoclastic Nron transgenic(Nron c TG)and osteoclastic knockout(Nron CKO)mouse models were generated.When Nron was overexpressed,the orthodontic tooth movement rate was reduced.In addition,the number of osteoclasts decreased,and the activity of osteoclasts was inhibited.Mechanistically,Nron controlled the maturation of osteoclasts by regulating NFATc1 nuclear translocation.In contrast,by deleting Nron specifically in osteoclasts,tooth movement speed increased in Nron CKO mice.These results indicate that lnc RNAs could be potential targets to regulate osteoclastogenesis and orthodontic tooth movement speed in the clinic in the future.Ruilin Zhang Junhui Li Gongchen Li Fujun Jin Zuolin Wang Rui Yue Yibin Wang Xiaogang Wang Yao Sun 2020International Journal of Oral Science2020,12,2:9
3A general alternate loading technique and its applications in the inverse designs of centrifugal and mixed-flow pump impellers显示文摘For the inverse designs of centrifugal and mixed-flow pump impellers,clarifying the generation process of secondary flows and putting forward corresponding suppression measures is an important approach to improve the impeller performance.In this paper,to provide a better qualitative insight into the generation mechanism of secondary flows in the impeller,a simple kinematic equation is derived based on the ideal assumptions,which indicates that the potential rothalpy gradient(PRG)is the most important dynamic source that actively induces secondary vortical flows.Induced by the natural adverse PRG on the S1 and S2 stream surfaces,two typical secondary flows,H-S and P-S secondary flows,are clearly presented.To specially suppress these typical secondary flows,a general alternate loading technique(GALT)is proposed,aiming to adjust the real blade loadingδp to control the PRG features.At the blade fore part,theδp on the hub streamline should be slowly increased to avoid breakneck growth of the potential rothalpy to reduce adverse streamwise PRG on the S2 streamsurface.At the blade middle part,theδp should be moderately decreased to reduce adverse streamwise PRG on the S1 streamsurface.At the blade aft part,the difference in theδp between the shroud and hub streamlines should be decreased faster to control the exit uniformity.By applying the GALT to the impeller designs of three typical pump types in hydraulic engineering,the organizational effect of the PRG on fundamental flow structures is proven.The GALT can effectively control the PRG distributions and suppress the secondary flows,thereby widening the pump’s high-efficiency zone,improving flow uniformity and suppressing pressure fluctuations.Compared with the current Z-G method and the ALT,the GALT can meet the requirements of'de-experience'better,thereby enabling the designers to obtain good products explicitly and quickly.WANG ChaoYue WANG FuJun AN DongSen YAO ZhiFeng XIAO RuoFu LU Li HE ChengLian ZOU ZhiChao 2021Science China(Technological Sciences)2021,64,4:7
4Highly sensitive,sub-microsecond polymer photodetectors for blood oxygen saturation testing显示文摘Bottom surface of active layers and interface of indium tin oxide(ITO)electrodes and active layers play a crucial role in determining the performance of polymer photodetectors with photomultiplication(PM-PPDs).The interfacial trapped electron distribution closing to ITO electrodes will determine spectral response range and external quantum efficiency(EQE)of PMPPDs.The bottom interface is more sensitive than top interface when light is coming from the ITO side,and the larger density of generated charge on the bottom interfaces will induce interfacial band more bending for efficient charge tunneling injection.Highly sensitive and sub-microsecond PM-PPDs are achieved with PMBBDT:Y6(100:7,w/w)as active layers under forward bias,yielding EQE of 18,700%at 320 nm,21,700%at 600 nm and 16,400%at 810 nm under a bias of 7 V,respectively,as well as fast response time of 79μs.The high EQE of the PM-PPDs is attributed to efficient hole tunneling injection from ITO electrode under forward bias.The electron traps closing to ITO electrode will be quickly filled up when light is coming from ITO side,leading to interfacial band more bending for hole tunneling injection.Importantly,the PM-PPDs is performed to measure heart rate(HR)and blood oxygen saturation(SpO_(2)),and the measured data by the PM-PPDs are very similar with those obtained by commercial photodetectors.Zijin Zhao Baiqiao Liu Changlin Xie Yao Ma Jian Wang Ming Liu Kaixuan Yang Yunhua Xu Jian Zhang Weiwei Li Liang Shen Fujun Zhang 2021Science China Chemistry2021,64,8:2

11Heterogeneity of Surface Heat Exchange of Slopes and Potential Drivers of the Initiation of Thaw Slump,Qinghai‑Tibet Plateau显示文摘In the mountainous permafrost area,most thaw slumps are distributed in north or northeast-facing shady slope areas.It is commonly known that there is a heterogeneity in permafrost between diferent slope aspects,but there has been a lack of detailed measured data to quantitatively evaluate their relationships,and in-depth understandings on how the slope aspects are linked to the distribution of thaw slumps.This study examined the heterogenous thermal regime,soil moisture content,and surface radiation at two slope sites with opposing aspects in a warming permafrost region on the Qinghai-Tibet Plateau(QTP).The results indicate that similar air temperatures(T_(a))were monitored on the two slopes,but there were signifcant diferences in ground temperature and moisture content in the active layer from 2016 to 2021.The sunny slope exhibited a higher mean annual ground surface temperature(T_(s)),and over the fve years the mean annual temperature at the top of permafrost was 1.3–1.4℃warmer on the sunny slope than the shady slope.On the contrary,the near-surface soil moisture content was about 10–13%lower on the sunny slope(~22–27%)than the shady slope(~35–38%)during the thawing season(June–September).Radiation data indicate that signifcantly higher shortwave downward radiation(DR)appeared at the sunny slope site.However,due to the greater surface albedo,the net radiation(Rn)was lower on the sunny slope.Slope aspect also afects the ground ice content due to its infuence on ground temperature,freeze-thaw cycles,and soil moisture.Shady slopes have a shallower burial of ice-rich permafrost compared to sunny slopes.The results highlight greatly diferent near-surface ground thermal conditions at the two slope sites with diferent aspects in a mountainous permafrost region.This helps identify the slope-related causes of increasing thaw slumps and provides a basis for predicting their future development.Xingwen Fan Wenjiao Li Xuyang Wu Miaomiao Yao Fujun Niu Zhanju Lin 2023International Journal of Disaster Risk Science2023,14,4:0
12Crystalline-magnetism action in biomimetic mineralization of calcium carbonate显示文摘The influence of minor environmental factors,such as the geomagnetic field,on the biomineralization of nacres,is often ignored but a great deal of research has confirmed its important role in the normal mineralization of calcium carbonate.Although the geomagnetic field is weak,its cumulative effects need to be considered given that the biomineralization process can take years.Accordingly,the authors of this paper have investigated the effects of weak magnetic fields(25 Gs or 50 Gs)on calcium carbonate mineralization and analyzed the mechanism involved.The results show that even a weak magnetic field conduces to the formation of vaterite or aragonite,in the induction order of precursor→vaterite→aragonite.The stronger the magnetic field and the longer the time,the more obvious the induction effect.The effect of a magnetic field is strongest in the aging stage and weakest in the solution stage.Inductions by egg-white protein and by a magnetic field inhibit each other,but they both restrict particle growth.These findings highlight the importance of minor environmental factors for biomineralization and can serve as a reference for biomimetic preparation of a CaCO_(3)nacre-like structure and for anti-scale technology for circulating cooling water.Chaoqun Wu Xun Liu Fujun Yao Xin Yang Yan Wang Wenyuan Hu 2023Chinese Journal of Chemical Engineering2023,59,7:0
13A SMOOTH C^1 INTERPOLATION FOR TWO-DIMENSIONAL CONTACT PROBLEMS USING PARAMETRIC CURVE TECHNIQUE显示文摘A smooth C1 interpolation for two-dimensional contact problems using paxametric curve technique was developed and implemented. The parametric curve can ensure C1 continuity of the contact surfaces and provide a unique surface normal vector. Some numerical examples were used to illustrate the advantages of the newly developed representation of contact surface. The results reveal a significant improvement in the prediction of contact stresses and contactarea. 14Loss of NEIL3 activates radiotherapy resistance in the progression of prostate cancer显示文摘Objective:To explore the genetic changes in the progression of castration-resistant prostate cancer(CRPC)and neuroendocrine prostate cancer(NEPC)and the reason why these cancers resist existing therapies.Methods:We employed our CRPC cell line microarray and other CRPC or NEPC datasets to screen the target gene NEIL3.Lentiviral transfection and RNA interference were used to construct overexpression and knockdown cell lines.Cell and animal models of radiotherapy were established by using a medical electron linear accelerator.Flow cytometry was used to detect apoptosis or cell cycle progression.Western blot and qPCR were used to detect changes in the protein and RNA levels.Results:TCGA and clinical patient datasets indicated that NEIL3 was downregulated in CRPC and NEPC cell lines,and NEIL3 was correlated with a high Gleason score but a good prognosis.Further functional studies demonstrated that NEIL3 had no effect on the proliferation and migration of PCa cells.However,cell and animal radiotherapy models revealed that NEIL3 could facilitate the radiotherapy sensitivity of PCa cells,while loss of NEIL3 activated radiotherapy resistance.Mechanistically,we found that NEIL3 negatively regulated the expression of ATR,and higher NEIL3 expression repressed the ATR/CHK1 pathway,thus regulating the cell cycle.Conclusions:We demonstrated that NEIL3 may serve as a diagnostic or therapeutic target for therapy-resistant patients.Qiong Wang Zean Li Jin Yang Shirong Peng Qianghua Zhou Kai Yao Wenli Cai Zhongqiu Xie Fujun Qin Hui Li Xu Chen Kaiwen Li Hai Huang 2022Cancer Biology & Medicine2022,19,8:0
15Protective effects of (-)-epigallocatechin-3-gallate on D-galactose-induced neuronal apoptosis in mice显示文摘BACKGROUND: The neuroprotective effects of (-)-epigallocatechin-3-gallate (EGCG), the mainpolyphenolic constituent of green tea, have been widely reported. However, the action mechanisms,in particular in D-galactose-induced aging mice, remain poorly understood.OBJECTIVE: The present study investigated the protective effects of EGCG onD-galactose-induced hippocampus neuronal apoptosis in aging mice, as well as the relationship withexpression of p75ICD, JNK2, and p53 proteins.DESIGN, TIME AND SETTING: A randomized, controlled, molecular biological, animal experimentwas performed at the Laboratory of Pharmacology, Pharmaceutical College of China MedicalUniversity, China, from September 2006 to July 2008.MATERIALS: D-galactose and EGCG (Sigma, USA), as well as terminal deoxynucleotidyltransferase-mediated dUTP-biotin nick-end labeling (TUNEL) In Situ Cell Apoptosis Detection Kit(Promega, USA), were used in this study.METHODS: A total of 64 mice were equally and randomly divided into D-galactose model, low-doseEGCG, high-dose EGCG, and control groups. Mice in the D-galactose model, low-dose EGCG,and high-dose EGCG groups were subcutaneously injected with 3% D-galactose (150 mg/kg),daily for 6 weeks, to establish a mouse model of aging. Mice in the control group were treated withsaline (5 mL/kg). At 3 weeks following injection, mice in the low-dose EGCG and high-dose EGCGgroups were orally administered EGCG at a dose of 2 mg/kg and 6 mg/kg, respectively, once a day,for 4 consecutive days. Mice in the control and D-galactose model groups received distilled water(5 mL/kg).MAIN OUTCOME MEASURES: Memory function was evaluated using a step-through passiveavoidance test. Neuronal apoptosis in the mouse hippocampus was detected using TUNEL staining.Expression levels of the intracellular domain of the p75 neurotrophin receptor (p75NTR)-p75ICD,JNK2, and p53 proteins in the hippocampus were determined using Western blot analysis.RESULTS: The aging mouse model was induced by subcutaneous injection of D-galactose, whichresulted in obvious memory impairment, increased apoptotic index, and increased proteinexpression levels of p75ICD, JNK2, and p53 in the hippocampus, compared with control mice (P <0.01). Oral EGCG administration (2 or 6 mg/kg) for 4 weeks significantly improved levels of memorydeficit in the aging mice and reduced apoptotic indices and protein expression levels of p75ICD,JNK2, and p53 in the mouse hippocampus (P < 0.01).CONCLUSION: Results from this study demonstrated increased protein expression levels ofp75ICD, JNK2, and p53, as well as increased hippocampal neuronal apoptosis in aD-galactose-induced mouse model of aging. 16Generation of functional oligopeptides that promote osteogenesis based on unsupervised deep learning of protein IDRs显示文摘Deep learning(DL)is currently revolutionizing peptide drug development due to both computational advances and the substantial recent expansion of digitized biological data.However,progress in oligopeptide drug development has been limited,likely due to the lack of suitable datasets and difficulty in identifying informative features to use as inputs for DL models.Here,we utilized an unsupervised deep learning model to learn a semantic pattern based on the intrinsically disordered regions of~171 known osteogenic proteins.Subsequently,oligopeptides were generated from this semantic pattern based on Monte Carlo simulation,followed by in vivo functional characterization.A five amino acid oligopeptide(AIB5P)had strong bone-formation-promoting effects,as determined in multiple mouse models(e.g.,osteoporosis,fracture,and osseointegration of implants).Mechanistically,we showed that AIB5P promotes osteogenesis by binding to the integrinα5 subunit and thereby activating FAK signaling.In summary,we successfully established an oligopeptide discovery strategy based on a DL model and demonstrated its utility from cytological screening to animal experimental verification.Mingxiang Cai Baichuan Xiao Fujun Jin Xiaopeng Xu Yuwei Hua Junhui Li Pingping Niu Meijing Liu Jiaqi Wu Rui Yue Yong Zhang Zuolin Wang Yongbiao Zhang Xiaogang Wang Yao Sun 2022Bone Research2022,10,2:0
