2MicroRNA-663 induces immune dysregulation by inhibiting TGF-β1 production in bone marrow-derived mesenchymal stem cells in patients with systemic lupus erythematosus显示文摘Mesenchymal stem cells(MSCs)are critical for immune regulation.Although several microRNAs(miRNAs)have been shown to participate in autoimmune pathogenesis by affecting lymphocyte development and function,the roles of miRNAs in MSC dysfunction in autoimmune diseases remain unclear.Here,we show that patients with systemic lupus erythematosus(SLE)display a unique miRNA signature in bone marrow-derived MSCs(BMSCs)compared with normal controls,among which miR-663 is closely associated with SLE disease activity.MiR-663 inhibits the proliferation and migration of BMSCs and impairs BMSC-mediated downregulation of follicular T helper(Tfh)cells and upregulation of regulatory T(Treg)cells by targeting transforming growth factorβ1(TGF-β1).MiR-663 overexpression weakens the therapeutic effect of BMSCs,while miR-663 inhibition improves the remission of lupus disease in MRL/lpr mice.Thus,miR-663 is a key mediator of SLE BMSC regulation and may serve as a new therapeutic target for the treatment of lupus.Linyu Geng Xiaojun Tang Kangxing Zhou Dandan Wang Shiying Wang Genhong Yao Weiwei Chen Xiang Gao Wanjun Chen Songtao Shi Nan Shen Xuebing Feng Lingyun Sun 2019Cellular & Molecular Immunology2019,16,3:16

4Insight-HXMT observations of the first binary neutron star merger GW170817显示文摘Finding the electromagnetic(EM) counterpart of binary compact star merger, especially the binary neutron star(BNS) merger,is critically important for gravitational wave(GW) astronomy, cosmology and fundamental physics. On Aug. 17, 2017,Advanced LIGO and Fermi/GBM independently triggered the first BNS merger, GW170817, and its high energy EM counterpart,GRB 170817 A, respectively, resulting in a global observation campaign covering gamma-ray, X-ray, UV, optical, IR, radio as well as neutrinos. The High Energy X-ray telescope(HE) onboard Insight-HXMT(Hard X-ray Modulation Telescope) is the unique high-energy gamma-ray telescope that monitored the entire GW localization area and especially the optical counterpart(SSS17 a/AT2017 gfo) with very large collection area(~1000 cm^2) and microsecond time resolution in 0.2-5 MeV. In addition,Insight-HXMT quickly implemented a Target of Opportunity(ToO) observation to scan the GW localization area for potential X-ray emission from the GW source. Although Insight-HXMT did not detect any significant high energy(0.2-5 MeV) radiation from GW170817, its observation helped to confirm the unexpected weak and soft nature of GRB 170817 A. Meanwhile,Insight-HXMT/HE provides one of the most stringent constraints(~10^(-7) to 10^(-6) erg/cm^2/s) for both GRB170817 A and any other possible precursor or extended emissions in 0.2-5 MeV, which help us to better understand the properties of EM radiation from this BNS merger. 5The enhanced X-ray Timing and Polarimetry mission—eXTP显示文摘In this paper we present the enhanced X-ray Timing and Polarimetry mission—eXTP. eXTP is a space science mission designed to study fundamental physics under extreme conditions of density, gravity and magnetism. The mission aims at determining the equation of state of matter at supra-nuclear density, measuring effects of QED, and understanding the dynamics of matter in strong-field gravity. In addition to investigating fundamental physics, eXTP will be a very powerful observatory for astrophysics that will provide observations of unprecedented quality on a variety of galactic and extragalactic objects. 6IL-25 stimulates M2 macrophage polarization and thereby promotes mitochondrial respiratory capacity and lipolysis in adipose tissues against obesity显示文摘Obesity and associated metabolic diseases are characterized by a chronic low-grade inflammatory state with the infiltration of many inflammatory cells,especially macrophages.Immune molecules,including some cytokines,have a close relationship with metabolism.Interleukin(IL)-25 is a member of the IL-17 cytokine family that can regulate macrophages and alleviate some metabolic dysfunction;however,its role and mechanisms in lipid metabolism remain to be extensively clarified.Human serum and liver biopsy specimens,high-fat diet-induced obesity mice and DB/DB(Lepr−/−)animal models were used to examine IL-25 expression in obesity and nonalcoholic fatty liver diseases(NAFLD).To observe the role of IL-25 in lipid metabolism,model mice were administered with IL-25 or adoptively transferred with IL-25-educated macrophages in vivo,whereas bone marrow-derived macrophages,the macrophage cell line RAW264.7 and adipocytes differentiated from 3T3-L1 were used in vitro.IL-25 was decreased in NAFLD patients and obese mice.In addition,IL-25 reduced body weight gain and lipid accumulation,enhanced lipid uptake by macrophages and increased the expression of lipolysis andβ-oxidation enzymes via alternatively activating macrophages.IL-25 also promoted lipolysis and suppressed lipogenesis in adipocytes co-cultured with the IL-25-educated macrophages.Furthermore,IL-25 improved the mitochondrial respiratory capacity and oxygen consumption rate of macrophages and produced more NAD+/NADH and ATP.In conclusion,IL-25 can stimulate M2 macrophage polarization and thereby promote lipolysis and mitochondrial respiratory capacity,highlighting the potential for IL-25 to be used as a therapeutic agent against obesity and associated metabolic syndromes.Juan Feng Lingyi Li Zhiying Ou Qiao Li Baoyong Gong Zhenxian Zhao Weiwei Qi Ti Zhou Jun Zhong Weibin Cai Xia Yang Aiping Zhao Guoquan Gao Zhonghan Yang 2018Cellular & Molecular Immunology2018,15,5:10
7Offline two-dimensional liquid chromatography coupled with ion mobility-quadrupole time-of-flight mass spectrometry enabling fourdimensional separation and characterization of the multicomponents from white ginseng and red ginseng显示文摘Inherent complexity of plant metabolites necessitates the use of multi-dimensional information to accomplish comprehensive profiling and confirmative identification.A dimension-enhanced strategy,by offline two-dimensional liquid chromatography/ion mobility-quadrupole time-of-flight mass spectrometry(2 D-LC/IM-QTOF-MS)enabling four-dimensional separations(2 D-LC,IM,and MS),is proposed.In combination with in-house database-driven automated peak annotation,this strategy was utilized to characterize ginsenosides simultaneously from white ginseng(WG)and red ginseng(RG).An offline 2 DLC system configuring an Xbridge Amide column and an HSS T3 column showed orthogonality 0.76 in the resolution of ginsenosides.Ginsenoside analysis was performed by data-independent high-definition MSE(HDMSE)in the negative ESI mode on a Vion?IMS-QTOF hybrid high-resolution mass spectrometer,which could better resolve ginsenosides than MSEand directly give the CCS information.An in-house ginsenoside database recording 504 known ginsenosides and 58 reference compounds,was established to assist the identification of ginsenosides.Streamlined workflows,by applying UNIFI?to automatedly annotate the HDMSEdata,were proposed.We could separate and characterize 323 ginsenosides(including 286 from WG and 306 from RG),and 125 thereof may have not been isolated from the Panax genus.The established 2 D-LC/IM-QTOF-HDMSEapproach could also act as a magnifier to probe differentiated components between WG and RG.Compared with conventional approaches,this dimensionenhanced strategy could better resolve coeluting herbal components and more efficiently,more reliably identify the multicomponents,which,we believe,offers more possibilities for the systematic exposure and confirmative identification of plant metabolites.Tiantian Zuo Chunxia Zhang Weiwei Li Hongda Wang Ying Hu Wenzhi Yang Li Jia Xiaoyan Wang Xiumei Gao Dean Guo 2020Journal of Pharmaceutical Analysis2020,10,6:9
8Colorectal cancer, radiotherapy and gut microbiota显示文摘Colorectal cancer is closely related to inflammation and immune response. Radiotherapy, as a major treatment for colorectal cancer, plays a central role in cancer control. Inflammation caused by ionizing radiation can exert either anti-or pro-tumorigenic effects. Additionally, radiotherapy can elicit an anti-tumor response not only in radiation of target lesions but also in radiation of remote lesions. However, the immune mechanism underlying this effect has not been thoroughly elucidated yet. The combination therapeutic regimen of radiotherapy with other therapeutic methods, including chemotherapy and immunotherapy, has been applied in clinical practice. Meanwhile, radiation toxicity and radiosensitivity have long been problems that affect a patient's quality of life and morbidity.Researchers have found that the abovementioned problems are closely associated with gut microbiota. 9Characteristics and Cause Analysis of Heavy Haze in Changchun City in Northeast China显示文摘Northeast China has been reported as having serious air pollution in China with increasing occurrences of severe haze episodes. Changchun City, as the center of Northeast China, has longstanding industry and is an important agricultural base. Additionally, Changchun City has a long winter requiring heating of buildings emitting pollution into the air. These factors contribute to the complexity of haze pollution in this area. In order to analyze the causes of heavy haze, surface air quality has been monitored from 2013 to 2015. By using satellite and meteorological data, atmospheric pollution status, spatio-temporal variations and formation have been analyzed. Results indicated that the air quality in 88.9% of days exceeding air quality index(AQI) level-1 standard(AQI >50) according to the National Ambient Air Quality Standard(NAAQS) of China. Conversely, 33.7% of the days showed a higher level with AQI > 100. Extreme haze events(AQI > 300) occurred frequently during agricultural harvesting period(from October 10 to November 10), intensive winter heating period(from Late-December to February) and period of spring windblown dust(April and May). Most daily concentrations of gaseous pollutants, i.e., NO_2(43.8 μg/m^3), CO(0.9 mg/m^3), SO_2(37.9 μg/m^3), and O_3(74.9 μg/m^3) were evaluated within level-1 concentration limits of NAAQS standards. However, particulate matter(PM_(2.5) and PM10) concentrations(67.3 μg/m^3 and 115.2 μg/m^3, respectively) were significantly higher than their level-1 limits. Severe haze in spring was caused by offsite transported dust and windblown surface soil. Heavy haze periods during fall and winter were mainly formed by intensive emissions of atmospheric pollutants and steady weather conditions(i.e., low wind speed and inversion layer). The overlay emissions of widespread straw burning and coal combustion for heating were the dominant factors contributing to haze in autumn, while intensive coal burning during the coldest time was the primary component of total emissions. In addition, general emissions including automobile exhaust, road and construction dust, residential and industrial activities, have significantly increased in recent years, making heavy haze a more frequent occurrence. 10Distributed Adaptive Fault-Tolerant Output Regulation of Heterogeneous Multi-Agent Systems With Coupling Uncertainties and Actuator Faults显示文摘In this paper, we consider the distributed adaptive fault-tolerant output regulation problem for heterogeneous multiagent systems with matched system uncertainties and mismatched coupling uncertainties among subsystems under the influence of actuator faults. First, distributed finite-time observers are proposed for all subsystems to observe the state of the exosystem. Then, a novel fault-tolerant controller is designed to compensate for the influence of matched system uncertainties and actuator faults. By using the linear matrix inequality technique, a sufficient condition is provided to guarantee the solvability of the considered problem in the presence of mismatched coupling uncertainties. 11Effects of capture and captivity on plasma corticosterone and metabolite levels in breeding Eurasian Tree Sparrows显示文摘Background: Bringing free-living animals into captivity subjects them to the stress of both capture and captivity, leading to the alteration of normal physiological processes and behaviors through activation of the hypothalamic– pituitary–adrenal axis. In free-living birds, although elevated plasma corticosterone (CORT) is an important adaptation regulating physiological and behavioral responses during the process of capture and captivity stress, little information is currently available on the effects of such stress on plasma metabolite levels. Methods: We examined the effects of immediate capture and 24-h captivity on body mass, body condition, plasma CORT, and metabolite levels including glucose (Glu), triglyceride (TG), total cholesterol (TC), uric acid (UA), in breeding Eurasian Tree Sparrows (Passer montanus). Results: CORT and Glu levels were increased significantly by the stress of capture, whereas TC and UA levels decreased. Body mass, body condition declined notably after 24 h in captivity, but CORT, Glu, and UA levels increased. Furthermore, male sparrows had lower TG levels after both capture and captivity than those of females. The relationships between plasma CORT and metabolite levels varied between sexes. Conclusions: Our results revealed that the metabolic status of Eurasian Tree Sparrows could be dramatically altered by capture and captivity. 13Protection of Li metal anode by surface-coating of PVDF thin film to enhance the cycling performance of Li batteries显示文摘Lithium metal, the ideal anode material for next-generation high-energy batteries, suffers from the severe safety problem of Li dendrites. Herein, we report a simple approach to effectively maintain the morphology of Li-metal anode and enhance the cycling performance of Li batteries by surface coating of a porous polyvinylidene fluoride (PVDF) thin film. In symmetrical cells testing, the cells with the Li@PVDF electrode display stable cycling performance more than 1300 h (650 cycles) at the current density of 0.5 mA/cm^2 with a stripping/plating capacity of 0.5 mAh/cm^2. 15Numerical study on the correlation of transonic single-degree-of-freedom flutter and buffet显示文摘Transonic single-degree-of-freedom(SDOF) flutter and transonic buffet are the typical and complex aeroelastic phenomena in the transonic flow. In this study, transonic aeroelastic issues of an elastic airfoil are investigated using Unsteady Reynolds-Averaged Navier-Stokes(URANS) equations. The airfoil is free to vibrate in SDOF of pitching. It is found that, the coupling system may be unstable and SDOF self-excited pitching oscillations occur in pre-buffet flow condition, where the free-stream angle of attack(AOA) is lower than the buffet onset of a stationary airfoil. In the theory of classical aeroelasticity, this unstable phenomenon is defined as flutter. However, this transonic SDOF flutter is closely related to transonic buffet(unstable aerodynamic models) due to the following reasons. Firstly, the SDOF flutter occurs only when the free-stream AOA of the spring suspended airfoil is slightly lower than that of buffet onset, and the ratio of the structural characteristic frequency to the buffet frequency is within a limited range. Secondly, the response characteristics show a high correlation between the SDOF flutter and buffet. A similar 'lock-in' phenomenon exists, when the coupling frequency follows the structural characteristic frequency. Finally, there is no sudden change of the response characteristics in the vicinity of buffet onset, that is, the curve of response amplitude with the free-stream AOA is nearly smooth. 16Collection and preservation of urinary proteins, using a fluff pulp diaper显示文摘Change is the most fundamental property of a biomarker. In contrast to the blood, which is under homeostatic controls, urine reflects changes in the body earlier and is more sensitive, thus making it a better biomarker source. Moreover, drawing blood from infants and toddlers is difficult and not tolerated well. For patients limited by language, communicating their chief complaint is difficult. Thus, monitoring biomarkers in urine can provide valuable clues for the diagnosis of diseases, especially pediatric diseases. Collecting urine from young children and some adult patients is more challenging than collecting it from healthy adults.Here, we propose a method that uses a fluff pulp diaper to collect urine. Urinary proteins are then eluted and adsorbed onto a piece of nitrocellulose membrane, which can be dried and stored in a vacuum bag. SDS-PAGE and LC-MS/MS analysis indicated that this method is reproducible, and similar proteins were identified as those obtained by an acetone precipitation method. 18Evolution and Domestication Footprints Uncovered from the Genomes of Coix显示文摘Coix lacryma-jobi,a plant species closely related to Zea and Sorghum,is an important food and medicinal crop in Asia.However,no reference genome of this species has been reported,and its exact phylogeny within the Andropogoneae remains unresolved.Here,we generated a high-quality genome assembly of coix comprising~1.73 Gb with 44485 predicted protein-coding genes.We found coix to be a typical diploid plant with an overall 1-to-1 syntenic relationship with the Sorghum genome,despite its drastic genome expansion(~2.3-fold)due mainly to the activity of transposable elements.Phylogenetic analysis revealed that coix diverged with sorghum~10.41 million years ago,which was~1.49 million years later than the divergence between sorghum and maize.Resequencing of 27 additional coix accessions revealed that they could be unambiguously separated into wild relatives and cultivars,and suggested that coix experienced a strong genetic bottleneck,resulting in the loss of about half of the genetic diversity during domestication,even though many traits have remained undomesticated.Our data not only provide novel comparative genomic and evolutionary insights into the Andropogoneae lineage,but also an important resource that will greatly benefit molecular breeding of this important crop.Hongbing Liu Junpeng Shi Zexi Cai Yumin Huang Menglu Lv Huilong Du Qiang Gao Yi Zuo Zhaobin Dong Wei Huang Rui Qin Chengzhi Liang Jinsheng Lai Weiwei Jin 2020Molecular Plant2020,13,2:4
19Do migrant and resident species differ in the timing of increases in reproductive and thyroid hormone secretion and body mass? A case study in the comparison of pre-breeding physiological rhythms in the Eurasian Skylark and Asian Short-toed Lark显示文摘Background:Physiological preparation for reproduction in small passerines involves the increased secretion of reproductive hormones, elevation of the metabolic rate and energy storage, all of which are essential for reproduction. However, it is unclear whether the timing of the physiological processes involved is the same in resident and migrant species that breed in the same area. To answer this question, we compared temporal variation in the plasma concentration of luteinizing hormone(LH), testosterone(T), estradiol(E_2), triiothyronine(T_3) and body mass, between a migrant species, the Eurasian Skylark(Alauda arvensis) and a resident species, the Asian Short-toed Lark(Calandrella cheleensis), both of which breed in northeastern Inner Mongolia, China, during the 2014 and 2015 breeding seasons.Methods:Twenty adult Eurasian Skylarks and twenty Asian Short-toed Larks were captured on March 15, 2014 and 2015 and housed in out-door aviaries. Plasma LH, T(males), E_2(females), T_3 and the body mass of each bird were measured every six days from March 25 to May 6.Results:With the exception of T, which peaked earlier in the Asian Short-toed Lark in 2014, plasma concentrations of LH, T, E_2 and T_3 of both species peaked at almost the same time. However, Asian Short-toed Larks attained peak body mass earlier than Eurasian Skylarks. Plasma T_3 concentrations peaked 12 days earlier than plasma LH in both species. Generally, plasma LH, T, E_2, T_3 and body mass, peaked earlier in both species in 2014 than 2015.Conclusions:The timing of pre-reproductive changes in the endocrine system and energy metabolism can be the same in migrant and resident species; however, residents may accumulate energy reserves faster than migrants. Although migration does not affect the timing of pre-breeding reproductive and metabolic changes, migrant species may need more time to increase their body mass.
