2Construction and Identification of a Human Liver Specific MicroRNA Eukaryotic Expression Vector显示文摘MiR-122 is one of the non-coding RNAs which showed its effects on the lipo-metablism,virus infection and HCC forming through regulation of liver gene expression.Its eukaryotic expression vector was constructed by using pSuper which was widely applied in the siRNA expression.The precursor of human miR-122 gene was amplified by polymerase chain reaction(PCR)from the human genomic DNA.The positive clones were screened by PCR and restriction enzyme digestion.The new expression vector of miR-122 was named pHsa-m122.PHsa-m122 and its controls were transfected to HepG2 cells.The miR-122 expression activity was evaluated by GFP122i sensor reporter plasmid through fluorescence detection and Western blot.It was shown that the fluorescence intensity of GFP122si and pHsa-m122 co-transfection group was weaker than that of the controls,so the functional activity of expressed miR-122 was detected.When HepG2 cells were co-transfected with HBV1.3 and pHsa-m122 plasmids, the results showed miR-122 may down-regulate the gene expression of HBV.The human liver specific microRNA eukaryotic expression vector of miR-122 was constructed successfully,which may facilitate further study of its function in the development of liver virus infection diseases and HCC.Shanshan Chen Ming Ni Bing Yu Tingting Lv Mengji Lu Feili Gong 2007Cellular & Molecular Immunology2007,4,6:18

4MATRIEX imaging:multiarea two-photon real-time in vivo explorer显示文摘Two-photon laser scanning microscopy has been extensively applied to study in vivo neuronal activity at cellular and subcellular resolutions in mammalian brains.However,the extent of such studies is typically confined to a single functional region of the brain.Here,we demonstrate a novel technique,termed the multiarea two-photon real-time in vivo explorer(MATRIEX),that allows the user to target multiple functional brain regions distributed within a zone of up to 12mm in diameter,each with a field of view(FOV)of ~200μm in diameter,thus performing two-photon Ca2+imaging with single-cell resolution in all of the regions simultaneously.For example,we demonstrate real-time functional imaging of single-neuron activities in the primary visual cortex,primary motor cortex and hippocampal CA1 region of mice in both anesthetized and awake states.A unique advantage of the MATRIEX technique is the configuration of multiple microscopic FOVs that are distributed in three-dimensional space over macroscopic distances(>1 mm)both laterally and axially but that are imaged by a single conventional laser scanning device.In particular,the MATRIEX technique can be effectively implemented as an add-on optical module for an existing conventional single-beam-scanning two-photon microscope without requiring any modification to the microscope itself.Thus,the MATRIEX technique can be readily applied to substantially facilitate the exploration of multiarea neuronal activity in vivo for studies of brain-wide neural circuit function with single-cell resolution.Mengke Yang Zhenqiao Zhou Jianxiong Zhang Shanshan Jia Tong Li Jiangheng Guan Xiang Liao Bing Leng Jing Lyu Kuan Zhang Min Li Yan Gong Zhiming Zhu Junan Yan Yi Zhou Jian K Liu Zsuzsanna Varga Arthur Konnerth Yuguo Tang Jinsong Gao Xiaowei Chen Hongbo Jia 2019Light(Science & Applications)2019,8,1:4

9The genome of Magnolia biondii Pamp. provides insights into the evolution of Magnoliales and biosynthesis of terpenoids显示文摘Magnolia biondii Pamp.(Magnoliaceae,magnoliids)is a phylogenetically,economically,and medicinally important ornamental tree species widely grown and cultivated in the north-temperate regions of China.Determining the genome sequence of M.biondii would help resolve the phylogenetic uncertainty of magnoliids and improve the understanding of individual trait evolution within the Magnolia genus.We assembled a chromosome-level reference genome of M.biondii using~67,~175,and~154Gb of raw DNA sequences generated via Pacific Biosciences single-molecule real-time sequencing,10X Genomics Chromium,and Hi-C scaffolding strategies,respectively.The final genome assembly was~2.22Gb,with a contig N50 value of 269.11 kb and a BUSCO complete gene percentage of 91.90%.Approximately 89.17%of the genome was organized into 19 chromosomes,resulting in a scaffold N50 of 92.86Mb.The genome contained 47,547 protein-coding genes,accounting for 23.47%of the genome length,whereas 66.48%of the genome length consisted of repetitive elements.We confirmed a WGD event that occurred very close to the time of the split between the Magnoliales and Laurales.Functional enrichment of the Magnolia-specific and expanded gene families highlighted genes involved in the biosynthesis of secondary metabolites,plant–pathogen interactions,and responses to stimuli,which may improve the ecological fitness and biological adaptability of the lineage.Phylogenomic analyses revealed a sister relationship of magnoliids and Chloranthaceae,which are sister to a clade comprising monocots and eudicots.The genome sequence of M.biondii could lead to trait improvement,germplasm conservation,and evolutionary studies on the rapid radiation of early angiosperms.Shanshan Dong Min Liu Yang Liu Fei Chen Ting Yang Lu Chen Xingtan Zhang Xing Guo Dongming Fang Linzhou Li Tian Deng Zhangxiu Yao Xiaoan Lang Yiqing Gong Ernest Wu Yaling Wang Yamei Shen Xun Gong Huan Liu Shouzhou Zhang 2021Horticulture Research2021,8,1:4
10Exploring Location Pattern of Commercial Stores in Shichahai, Beijing from a Street Centrality Perspective显示文摘The location pattern of different commercial stores in Shichahai, a historic conservation area in Beijing, was investigated from a street centrality perspective. Many previous studies have investigated the relationship between street centrality and land use patterns or commercial activities at interurban or intraurban scales. We considered Shichahai in this study to determine if street centrality applied at the street scale and if the street network was the only factor influencing the selection of store location. First, the nearest neighbor index, nearest neighbor hierarchical spatial cluster(NNHSC), and kernel density estimation(KDE) methods were used to provide baseline spatial distributions of commercial stores. Second, urban network analysis(UNA) tools were used to measure the street centrality indices under two conditions, with and without the weighting of cultural relics calculated by a principal component analysis(PCA). Finally, both store locations and centrality values at nodes were transformed to one unit(raster pixel) for a correlation analysis.The results showed that three of the four store types were clustered and had their own hotspots that were mostly located in the eastern and central parts of city blocks. The most momentous findings were determined from the street centrality indices. Among the three store types with correlation coefficients above 0.5, all centrality indices with landmark weighting, except straightness, had higher correlations,with closeness with landmark weighting having the highest correlation, followed by betweenness with landmark weighting. Therefore,we statistically concluded that street centrality could apply at the street scale and that the street network was not the only factor that influenced store location pattern, with landmarks also playing a significant role. 11Clinical Prediction Models for Hepatitis B Virus-related Acute-on-chronic Liver Failure:A Technical Report显示文摘Background and Aims:It is critical but challenging to predict the prognosis of hepatitis B virus-related acute-on-chronic liver failure(HBV-ACLF).This study systematically summarized and evaluated the quality and performance of available clinical prediction models(CPMs).Methods:A keyword search of articles on HBV-ACLF CPMs published in PubMed from January 1995 to April 2020 was performed.Both the quality and performance of the CPMs were assessed.Results:Fifty-two CPMs were identified,of which 31 were HBV-ACLF specific.The modeling data were mostly derived from retrospective(83.87%)and single-center(96.77%)cohorts,with sample sizes ranging from 46 to 1,202.Three-month mortality was the most common endpoint.The Asian Pacific Association for the Study of the Liver consensus(51.92%)and Chinese Medical Association liver failure guidelines(40.38%)were commonly used for HBV-ACLF diagnosis.Serum bilirubin(67.74%),the international normalized ratio(54.84%),and hepatic encephalopathy(51.61%)were the most frequent variables used in models.Model discrimination was commonly evaluated(88.46%),but model calibration was seldom performed.The model for end-stage liver disease score was the most widely used(84.62%);however,varying performance was reported among the studies.Conclusions:Substantial limitations lie in the quality of HBV-ACLF-specific CPMs.Disease severity of study populations may impact model performance.The clinical utility of CPMs in predicting short-term prognosis of HBV-ACLF remains to be undefined.Xia Yu Yi Lu Shanshan Sun Huilan Tu Xianbin Xu Kai Gong Junjie Yao Yu Shi Jifang Sheng 2021Journal of Clinical and Translational Hepatology2021,9,6:3
12Climate warming shifts the time interval between flowering and leaf unfolding depending on the warming period显示文摘The timing of flowering(FL)and leaf unfolding(LU)determine plants’reproduction and vegetative growth.Global warming has substantially advanced FL and LU of temperate and boreal plants,but their responses to warming differ,which may influence the time interval between FL and LU(ΔLU-FL),thereby impacting plant fitness and intraspecific physiological processes.Based on twigs collected from two flowering-first tree species,Populus tomentosa and Amygdalus triloba,we conducted a manipulative experiment to investigate the effects of winter chilling,spring warming and photoperiod on theΔLU-FL.We found that photoperiod did not affect theΔLU-FL of Amygdalus triloba,but shortenedΔLU-FL by 5.1 d of Populus tomentosa.Interestingly,spring warming and winter chilling oppositely affected theΔLU-FL of both species.Specifically,low chilling accumulation extended theΔLU-FL by 3.8 and 9.4 d for Populus tomentosa and Amygdalus triloba,but spring warming shortened theΔLU-FL by 4.1 and 0.2 d℃^(-1).Our results indicate that climate warming will decrease or increase theΔLU-FL depending on the warming periods,i.e.,spring or winter.The shifted time interval between flowering and leaf unfolding may have ecological effects including affecting pollen transfer efficiency and alter the structure and functioning of terrestrial ecosystem.Shuxin Wang Zhaofei Wu Yufeng Gong Shubiao Wang Wei Zhang Shanshan Zhang Hans J.De Boeck Yongshuo H.Fu 2022Science China(Life Sciences)2022,65,11:2
13English Forum on Medical Humanities of SMMU:An interdisciplinary innovative teaching practice显示文摘To conform to the college English teaching reform from English for General Purposes (EGP) to English for Academic Purposes (EAP) in the vital transfer era, the Foreign Language Department of Second Military Medical University (SMMU) launched an English for Medical Academic Purpose (EMAP) curriculum with English Forum on Medical Humanities (EFMH), a project-based teaching practice. Based on Rogers' learner-centered humanistic teaching approach which emphasizes students' involvement and experiences in learning, EFMH puts students and their practice in a central place. After four-year exploration and operation, the Forum has developed from a small-scaled embryo into an internationalized, standardized and professionalized event, having produced fruitful outcomes. 15Association of an Increased Risk of Pre-eclampsia and Fetal Growth Restriction in Singleton and Twin Pregnancies with Female Fetuses显示文摘Objective: To investigate whether the fetal gender affects the incidence of pre-eclampsia (PE) and fetal growth restriction (FGR) in singleton and twin pregnancies.Methods: This was a 10-year single-center, retrospective, cohort study from January 2009 to January 2019. A total of 57,129 singleton and 3699 twin pregnancies aged between 18-55 years old were recruited at the Third Affiliated Hospital of Guangzhou Medical University, China. We used multivariable logistic regression to analyze the effect of fetal gender on the incidence of PE and FGR.Results: In singleton pregnancies, the incidence rates of PE and FGR with a female fetus were higher than those with a male fetus (6.4% (1713/26,793)vs. 5.9% (1803/30,336),P < 0.05 and 3.5% (932/26,793)vs. 2.4% (745/30,336),P < 0.05, respectively). A female fetus was an independent risk factor for either PE or FGR (adjusted odds ratio: 1.169 or 1.563;95% confidence interval: 1.036-1.319 or 1.349-1.810, respectively). In twin pregnancies, the incidence of early-onset PE was greater in pregnancies with two females compared with two males or one male plus one female (4.6% (46/1003)vs. 4.1% (54/1305)vs. 2.4% (33/1391),P < 0.05). 17Revisiting dynamics and models of microsegregation during polycrystalline solidification of binary alloy显示文摘Microsegregation formed during solidification is of great importance to material properties.The conventional Lever rule and Scheil equation are widely used to predict solute segregation.However,these models always fail to predict the exact solute concentration at a high solid fraction because of theoretical assumptions.Here,the dynamics of microsegregation during polycrystalline solidification of refined Al-Cu alloy is studied via two-and three-dimensional quantitative phase-field simulations.Simulations with different grain refinement level,cooling rate,and solid diffusion coefficient demonstrate that solute segregation at the end of solidification(i.e.when the solid fraction is close to unit)is not strongly correlated to the grain morphology and back diffusion.These independences are in accordance with the Scheil equation which only relates to the solid fraction,but the model predicts a much higher liquid concentration than simulations.Accordingly,based on the quantitative phase-field simulations,a new analytical microsegregation model is derived.Unlike the Scheil equation or the Lever rule that respectively overestimates or underestimates the liquid concentration,the present model predicts the liquid concentration in a pretty good agreement with phase-field simulations,particularly at the late solidification stage.Tongzhao Gong Yun Chen Shanshan Li Yanfei Cao Dianzhong Li Xing-Qiu Chen Guillaume Reinhart Henri Nguyen-Thi 2021Journal of Materials Science & Technology2021,,15:1
18Long non-coding RNA AFAP1-AS1 accelerates lung cancer cells migration and invasion by interacting with SNIP1 to upregulate c-Myc显示文摘Actin filament associated protein 1 antisense RNA 1(named AFAP1-AS1)is a long non-coding RNA and overexpressed in many cancers.This study aimed to identify the role and mechanism of AFAP1-AS1 in lung cancer.The AFAP1-AS1 expression was firstly assessed in 187 paraffin-embedded lung cancer and 36 normal lung epithelial tissues by in situ hybridization.The migration and invasion abilities of AFAP1-AS1 were investigated in lung cancer cells.To uncover the molecular mechanism about AFAP1-AS1 function in lung cancer,we screened proteins that interact with AFAP1-AS1 by RNA pull down and the mass spectrometry analyses.AFAP1-AS1 was highly expressed in lung cancer clinical tissues and its expression was positively correlated with lung cancer patients'poor prognosis.In vivo experiments confirmed that AFAP1-AS1 could promote lung cancer metastasis.AFAP1-AS1 promoted lung cancer cells migration and invasion through interacting with Smad nuclear interacting protein 1(named SNIP1),which inhibited ubiquitination and degradation of c-Myc protein.Upregulation of c-Myc molecule in turn promoted the expression of ZEB1,ZEB2,and SNAIL gene,which ultimately enhanced epithelial to mesenchymal transition(EMT)and lung cancer metastasis.Understanding the molecular mechanism by which AFAP1-AS1 promotes lung cancer's migration and invasion may provide novel therapeutic targets for lung cancer patients'early diagnosis and therapy.Yu Zhong Liting Yang Fang Xiong Yi He Yanyan Tang Lei Shi Songqing Fan Zheng Li Shanshan Zhang Zhaojian Gong Can Guo Qianjin Liao Yujuan Zhou Ming Zhou Bo Xiang Xiaoling Li Yong Li Zhaoyang Zeng Guiyuan Li Wei Xiong 2021Signal Transduction and Targeted Therapy2021,6,7:1
19Design spherical particles of potassium peroxymonosulfate compound salt with high stability and good powder properties via an agglomeration-dissolution mechanism显示文摘With the outbreak of COVID-19,disinfection protection has become a necessary measure to prevent infection.As a new type of disinfectant,potassium peroxymonosulfate compound salt(PMS)has the advantages of good bactericidal effect,non-toxicity,high safety and stability.However,the current PMS products with irregular particle shapes lead to poor flowability,high hygroscopicity,poor stability of reactive oxygen species(ROS)and serious caking problems.In this work,an agglomeration-dissolution mechanism was designed to prepare spherical PMS particles with large size(>300μm)and high sphericity(up to 90%),effectively addressing the above problems.Shaping(dissolution and abrasion)is the key to improving sphericity,which is mainly controlled by the design of the heating mode,residence time and stirring rate.Compared with the irregular PMS particles,the large spherical particles present better flowability(angle of repose decreased by 35.80%,Carr's index decreased by 64.29%,Hausner's ratio decreased by 19.14%),lower hygroscopicity(decreased by 38.0%),lower caking ratio(decreased by 84.50%),and higher stability(the monthly loss of ROS was reduced by 61.68%).The agglomeration-dissolution mechanism demonstrates the crystallization,agglomeration,dissolution and abrasion process of inorganic salt crystals,providing an opportunity to prepare high-end inorganic crystal materials with high-quality morphologies.Shanshan Feng Changyou Yu Shilin Guo Jinbo Ouyang Fei Yin Xiaodong Zhang Yongjian Zhang Mingyang Chen Dandan Han Junbo Gong Jingkang Wang 2023Particuology2023,,9:0
