| 1 | Deep seismic reflection profile across the juncture zone between the Tarim Basin and the West Kunlun Mountains显示文摘Fine structures of the crust and upper mantle of the basin-and-range juncture on the northwestern margin of the Qinghai-Tibetan Plateau are first delineated by the deep seismic reflection profile across the juncture zone between the Tarim Basin and the West Kunlun Mountains. Evidence is found for the northward subduction of the northwest marginal lithosphere of the Qinghai-Tibetan Plateau and its collision with Tarim lithosphere beneath the West Kunlun Mountains. | 2 | Transcriptional landscape of rice roots at the single-cell resolution显示文摘There are two main types of root systems in flowering plants,namely taproot systems of dicots and fibrous root systems found in monocots.Despite this fundamental split,our current knowledge of cellular and molecular mechanism driving root development is mainly based on studies of the dicot model Arabidopsis.However,the world major crops are monocots and little is known about the transcriptional programs underlying cell-type specification in this clade.Here,we report the transcriptomes of more than 20000 single cells derived from root tips of two agronomically important rice cultivars.Using combined computational and experimental analyses we were able to robustly identify most of the major cell types and define novel cell-type-specific marker genes for both cultivars.Importantly,we found divergent cell types associated with specific regulatory programs,including phytohormone biosynthesis,signaling,and response,which were well conserved between the two rice cultivars.In addition,we detected substantial differences between the cell-type transcript profiles of Arabidopsis and rice.These species-specific features emphasize the importance of analyzing tissues across diverse model species,including rice.Taken together,our study provides insight into the transcriptomic landscape of major cell types of rice root tip at single-cell resolution and opens new avenues to study cell-type specification,function,and evolution in plants. | Qing Liu Zhe Liang Dan Feng Sanjie Jiang Yifan Wang Zhuoying Du Ruoxi Li Guihua Hu Pingxian Zhang Yanfei Ma Jan ULohmann Xiaofeng Gu | 2021 | Molecular Plant2021,14,3: | 9 |

| 6 | Real-time simultaneous recording of electrophysiological activities and dopamine overflow in the deep brain nuclei of a non-human primate with Parkinson’s disease using nano-based microelectrode arrays显示文摘Parkinson’s disease(PD)is characterized by a progressive degeneration of nigrostriatal dopaminergic neurons.The precise mechanisms are still unknown.Since the neuronal communications are inherently electrical and chemical in nature,dual-mode detection of PD-related neuroelectrical and neurochemical information is essential for PD research.Subthalamic nucleus(STN)highfrequency stimulation(HFS)can improve most symptoms of PD patients and decrease the dosage of antiparkinsonian drugs.The mechanism of STN-HFS for PD still remains elusive.In this study,a silicon-based dual-mode microelectrode array(MEA)probe was designed and fabricated,and systematic dual-mode detection methods were established.The recording sites were modified using Pt nanoparticles and Nafion to improve the signal-to-noise(SNR)ratio.To evaluate its applicability to PD research,in vivo electrophysiological and electrochemical detection was performed in normal and hemiparkinsonian models,respectively.Through comparison of the dual-mode signals,we demonstrated the following in a PD monkey:(1)the maximum dopamine concentration in the striatum decreased by 90%;(2)the spike firing frequency increased significantly,especially in the region of the cortex;(3)the spectrogram analysis showed that much power existed in the 0–10 Hz frequency band;and(4)following repeated subthalamic nucleus high-frequency stimulation trials,the level of DA in the striatum increased by 16.5μM,which led to a better elucidation of the mechanism of HFS.The dual-mode MEA probe was demonstrated to be an effective tool for the study of neurological disorders. | Song Zhang Yilin Song Mixia Wang Guihua Xiao Fei Gao Ziyue Li Guoxian Tao Ping Zhuang Feng Yue Piu Chan Xinxia Cai | 2018 | Microsystems & Nanoengineering2018,4,1: | 2 |

| 8 | Core-satellite metal-organic framework@upconversion nanoparticle superstructures via electrostatic self-assembly for efficient photodynamic theranostics显示文摘The nanoplatforms based on upconversion nanoparticles(UCNPs)have shown great promise in amplified photodynamic therapy(PDT)triggered by near-infrared(NIR)light.However,their practical in vivo applications are hindered by the overheating effect of 980 nm excitation and low utilization of upconversion luminescence(UCL)by photosensitizers.To solve these defects,core-satellite metal-organic framework@UCNP superstructures,composed of a single metal-organic framework(MOF)NP as the core and Nd3+-sensitized UCNPs as the satellites,are designed and synthesized via a facile electrostatic self-assembly strategy.The superstructures realize a high co-loading capacity of chlorin e6(Ce6)and rose bengal(RB)benefitted from the highly porous nature of MOF NPs,showing a strong spectral overlap between maximum absorption of photosensitizers and emission of UCNPs.The in vitro and in vivo experiments demonstrate that the dual-photosensitizer superstructures have trimodal(magnetic resonance(MR)/UCL/fluorescence(FL))imaging functions and excellent antitumor effectiveness of PDT at 808 nm NIR light excitation,avoiding the laser irradiation-induced overheating issue.This study provides new insights for the development of highly efficient PDT nanodrugs toward precision theranostics. | Zhike Li Xi Qiao Guihua He Xin Sun Danhua Feng Liefeng Hu Hua Xu Hai-Bing Xu Shengqian Ma Jian Tian | 2020 | Nano Research2020,13,12: | 2 |
| 9 | Crystal structure of histidine-containing phosphocarrier protein from Thermoanaerobacter tengcongensis MB4 and the implications for thermostability显示文摘Protein thermostability is an inherent characteristic of proteins from thermophilic microorganisms,and therefore enables these organisms to survive at extreme temperatures.Although it is well-known that thermostable proteins are critical for the growth of thermophilic organisms,the structural basis of protein thermostability is not yet fully understood.The histidine-containing phosphocarrier (HPr) protein,a phosphate shuttle protein in the phosphoenolpyruvate-dependent sugar transport system (PTS) of bacterial species,is an ideal model for investigating protein thermostability with respect to its small size and deficiency in disulphide bonds or cofactors.In this study,the HPr protein from Thermoanaerobacter tengcongensis (TtHPr) is cloned and purified.Crystal structure with good quality has been determined at 2.3 resolution,which provides a firm foundation for exploring the thermostable mechanism.However,it shows that the crystal structure is conserved and no clue can be obtained from this single structure.Furthermore,detailed comparison of sequence and structure with the homologs from mesoor thermophilic bacteria shows no obvious rule for thermostability,but the extra salt-bridge existing only in thermophilic bacteria might be a better explanation for thermostability of HPr.Thus,mutations are performed to interrupt the salt-bridge in HPrs in thermophilic bacteria.Using site-directed mutations and the circular dichroism method,thermostability is evaluated,and the mutational variations are shown to have a faster denaturing rate than for wild-type viruses,indicating that mutations cause instability in the HPrs.Understanding the higher-temperature resistance of thermophilic and hyperthermophilic proteins is essential to studies on protein folding and stability,and is critical in engineering efficient enzymes that can work at a high temperature. | FENG ChunYan GAO Feng LIU YiWei WANG GuiHua PENG Hao MA YanHe YAN JingHua GAO George Fu | 2011 | Science China(Life Sciences)2011,54,6: | 2 |
