1Cell multiplication, apoptosis and p-Akt protein expression of bone mesenchymal stem cells of rat under hypoxia environment显示文摘Objective: To elucidate whether cell multiplication, apoptosis, glucose intake and p-Akt protein expression of bone Mesenchy-mal Stem Cells(MSCs) of rats is influenced by a hypoxic environment ex vivo. Methods: Passage 3 of bone marrow MSCs takenfrom Wistar rats, were cultured in a culturing chamber with 94%N2, 1%O2, 5%CO2 at 37℃. At different hypoxia time points, 0, 0.5,1, 4 and 8 h, glucose uptake was assayed by using radiation isotope 3H-G, Apoptotic Rate(AR) and dead rate(DR) were analyzedby flow cytometry(FCM) after Annexin V/PI staining, cell multiplication(by MTT methods) and p-Akt protein by immunocyto-chemistry and western blot. Results: Assay for CD29+, CD44+, CD71+, CD34- , Tn T+(after 5-azacytidine agent inducing) andALP+(after bone differentiation agent inducing) suggested these bone-derived cells were MSCs. The 3H-G intaking ratio(CPM/flask value: 157 ±11, 110 ±11, 107 ±13, 103 ±10, 100 ±9 and 98 ±10) of MSCs at different hypoxia time points, significantlydecreased compared to that of normoxia(P < 0.01) and tended to descend slowly with hypoxia time duration, for which there wasno statistical significance(P > 0.05). The AR(0.09 ±2.03%, 12.9 ±1.72%, 13.7 ±2.26%, 13.8 ±3.01%, 14.1 ±2.78% and 14.7 ±4.01% at 0, 0.5, 1, 4 and 8 h, respectively, P < 0.01) and DR (0.04 ±1.79%, 0.93 ±1.85%, 3.11 ±2.14%, 4.09 ±2.36%, 4.72 ±2.05% and 4.91 ±3.72% at 0, 0.5, 1, 4 and 8 h, respectively, P < 0.05) at different hypoxia time points significantly increasedcompared to those time in normoxia; The AR further went up with time (P < 0.05), however there was no statistical significance(P > 0.05) for the DR. Optical absorption value of MTT methods at different hypoxia time points significantly decreased com-pared to those with a corresponding normoxia time (P < 0.01) and degraded with time (in an hypoxic environment - P < 0.01).IOD of p-Akt protein of MSCs at different hypoxia time points significantly increased (0.367 ±0.031, 0.556 ±0.023, 0.579 ±0.013, 0.660 ±0.024, 0.685 ±0.039 and 0.685 ±0.011, respectively) compared to their equivalents in normoxia (P<0.05), how-ever, there was no statistical significance (P > 0.05) for different hypoxia time points. Hypoxia may result in ultramicrostructurechanges, such as defluvium of Microvilli, apoptotic body, 'margination'and so on and are further aggravated with hypoxia timestretching. 2The influence of pore structure on P-& S-wave velocities in complex carbonate reservoirs with secondary storage space显示文摘Secondary storage spaces with very complex geometries are well developed in Ordovician carbonate reservoirs in the Tarim Basin,which is taken as a study case in this paper.It is still not clear how the secondary storage space shape influences the P-& S-wave velocities (or elastic properties) in complex carbonate reservoirs.In this paper,three classical rock physics models (Wyllie timeaverage equation,Gassmann equation and the Kuster-Toks z model) are comparably analyzed for their construction principles and actual velocity prediction results,aiming at determining the most favourable rock physics model to consider the influence of secondary storage space shape.Then relationships between the P-& S-wave velocities in carbonate reservoirs and geometric shapes of secondary storage spaces are discussed from different aspects based on actual well data by employing the favourable rock physics model.To explain the influence of secondary storage space shape on V P-V S relationship,it is analyzed for the differences of S-wave velocities between derived from common empirical relationships (including Castagna's mud rock line and Greenberg-Castagna V P-V S relationship) and predicted by the rock physics model.We advocate that V P-V S relationship for complex carbonate reservoirs should be built for different storage space types.For the carbonate reservoirs in the Tarim Basin,the V P-V S relationships for fractured,fractured-cavernous,and fractured-hole-vuggy reservoirs are respectively built on the basis of velocity prediction and secondary storage space type determination.Through the discussion above,it is expected that the velocity prediction and the V P-V S relationships for complex carbonate reservoirs should fully consider the influence of secondary storage space shape,thus providing more reasonable constraints for prestack inversion,further building a foundation for realizing carbonate reservoir prediction and fluid prediction.Wang Haiyang Sam Zandong Sun Yang Haijun Gao Hongliang Xiao Youjun Hu Hongru 2011Petroleum Science2011,8,4:10
3The chemosensory protein of Chinese honeybee, Apis cerana cerana: Molecular cloning of cDNA, immunocytochemical localization and expression显示文摘Chemosensory proteins (CSPs) are ubiquitous soluble small proteins isolated from sensory organs thought to be involved in chemical communication. Here we report the first cDNA of CSPs, called Ac-ASP3, cloned and characterized from antennas of adult worker bees in Chinese honeybee, Apis cerana cerana. The Ac-ASP3 cDNA comprises 2 exons, with an ORF of 393-bp encoding 130 aa. Protein signature analyses show that the protein consists of four conserved cysteines and a signal peptide with 19 aa in the N-terminal sequence. The deduced protein sequence shares high homology with Am-ASP3 of Apis mellifera and low similarity with other species of insects. Immunocytochemical localization shows that Ac-ASP3 is only specifically expressed on the antenna contact chemosensilla such as sensilla trichodea B and sensilla basiconica, whereas Ac-ASP3 is scarcely expressed on olfactory chemosensilla such as sensilla placodea. Real-time PCR of Ac-ASP3 transcripts shows that Ac-ASP3 is highly expressed on wings and legs, but expression is lower on antenna. Temporal expression patterns suggest that Ac-ASP3 is expressed during the period of pupa and adults from 1-d to 6-d stages when bees act as house bees, cleaning the comb and taking care of the queen and larvae in comb. The above evidence suggests that Ac-ASP3 is unique in species and is generally not involved in olfaction during searching for honey and pollen. 4Pre-stack inversion for caved carbonate reservoir prediction:A case study from Tarim Basin,China显示文摘The major storage space types in the carbonate reservoir in the Ordovician in the TZ45 area are secondary dissolution caves.For the prediction of caved carbonate reservoir,post-stack methods are commonly used in the oilfield at present since pre-stack inversion is always limited by poor seismic data quality and insufficient logging data.In this paper,based on amplitude preserved seismic data processing and rock-physics analysis,pre-stack inversion is employed to predict the caved carbonate reservoir in TZ45 area by seriously controlling the quality of inversion procedures.These procedures mainly include angle-gather conversion,partial stack,wavelet estimation,low-frequency model building and inversion residual analysis.The amplitude-preserved data processing method can achieve high quality data based on the principle that they are very consistent with the synthetics.Besides,the foundation of pre-stack inversion and reservoir prediction criterion can be established by the connection between reservoir property and seismic reflection through rock-physics analysis.Finally,the inversion result is consistent with drilling wells in most cases.It is concluded that integrated with amplitude-preserved processing and rock-physics,pre-stack inversion can be effectively applied in the caved carbonate reservoir prediction.Zhang Yuanyin Sam Zandong Sun Yang Haijun Wang Haiyang HanJianfa Gao Hongliang Luo Chunshu Jing Bing 2011Petroleum Science2011,8,4:9
5A NAC transcription factor, NOR-like1, is a new positive regulator of tomato fruit ripening显示文摘Ripening of the model fruit tomato(Solanum lycopersicum)is controlled by a transcription factor network including NAC(NAM,ATAF1/2,and CUC2)domain proteins such as No-ripening(NOR),SlNAC1,and SlNAC4,but very little is known about the NAC targets or how they regulate ripening.Here,we conducted a systematic search of fruit-expressed NAC genes and showed that silencing NOR-like1(Solyc07g063420)using virus-induced gene silencing(VIGS)inhibited specific aspects of ripening.Ripening initiation was delayed by 14 days when NOR-like1 function was inactivated by CRISPR/Cas9 and fruits showed obviously reduced ethylene production,retarded softening and chlorophyll loss,and reduced lycopene accumulation.RNA-sequencing profiling and gene promoter analysis suggested that genes involved in ethylene biosynthesis(SlACS2,SlACS4),color formation(SlGgpps2,SlSGR1),and cell wall metabolism(SlPG2a,SlPL,SlCEL2,and SlEXP1)are direct targets of NOR-like1.Electrophoretic mobility shift assays(EMSA),chromatin immunoprecipitation-quantitative PCR(ChIP-qPCR),and dual-luciferase reporter assay(DLR)confirmed that NOR-like1 bound to the promoters of these genes both in vitro and in vivo,and activated their expression.Our findings demonstrate that NOR-like1 is a new positive regulator of tomato fruit ripening,with an important role in the transcriptional regulatory network.Ying Gao Wei Wei Xiaodan Zhao Xiaoli Tan Zhongqi Fan Yiping Zhang Yuan Jing Lanhuan Meng Benzhong Zhu Hongliang Zhu Jianye Chen Cai-Zhong Jiang Donald Grierson Yunbo Luo Da-Qi Fu 2018Horticulture Research2018,5,1:9
6Targeted delivery of chlorogenic acid by mannosylated liposomes to effectively promote the polarization of TAMs for the treatment of glioblastoma显示文摘Tumor-associated macrophages(TAMs)generally display an immunosuppressive M2 phenotype and promote tumor progression and metastasis,suggesting their potential value as a target in cancer immunotherapy.Chlorogenic acid(CHA)has been identified as a potent immunomodulator that promotes the polarization of TAMs from an M2 to an M1 phenotype.However,rapid clearance in vivo and low tumor accumulation have compromised the immunotherapeutic efficacy of CHA in clinical trials.In this study,mannosylated liposomes are developed for targeted delivery of CHA to TAMs.The immunoregulatory effects of CHA,along with the overall antitumor efficacy of CHA-encapsulated mannosylated liposomes,are investigated through in vitro and in vivo experiments.The prepared CHA-encapsulated mannosylated liposomes exhibit an ideal particle size,favorable stability,and preferential accumulation in tumors via the mannose receptor-mediated TAMs-targeting effects.Further,CHA-encapsulated mannosylated liposomes inhibit G422 glioma tumor growth by efficiently promoting the polarization of the pro-tumorigenic M2 phenotype to the anti-tumorigenic M1 phenotype.Overall,these findings indicate that CHA-encapsulated mannosylated liposomes have great potential to enhance the immunotherapeutic efficacy of CHA by inducing a shift from the M2 to the M1 phenotype.Jun Ye Yanfang Yang Jing Jin Ming Ji Yue Gao Yu Feng Hongliang Wang Xiaoguang Chen Yuling Liu 2020Bioactive Materials2020,5,3:9
7Zeolitic Imidazolate Framework 8-Derived Au@ZnO for Efficient and Robust Photocatalytic Degradation of Tetracycline显示文摘Summary of main observation and conclusion Tetracycline (TC) and other antibiotics accumulated in groundwater and soil pollute ecological environment and threaten human health. Gold nan oparticles doped on photocatalysts are able to enhance the photodegradation efficiency during removing these antibiotics, but preparation of Au nanoparticles of well-dispersion on photocatalysts remains challenging. In this work, zeolite imidazolate (ZIF-8) was employed as the precursor to prepare Au@ZnO photocatalyst via impregnation and in-situ reduction method to efficiently degrade the tetracycline in the aqueous solution. Au nanoparticles are of 10 nm in size and uniformly dispersed on the surfaces of ZnO microstructures. The as-prepared Au@ZnO is able to remove 85.5% of TC of 0.010 mg/mL within 2h, presenting higher photocatalytic activity than pure ZnO catalyst. Most importantly, the catalyst shows its superior stability after five cycles without structure and activity changing. 8Seismic attributes and integrated prediction of fractured and caved carbonate reservoirs in the Tarim Basin,China显示文摘The carbonate reservoirs in the Tarim Basin are characterized by anisotropy and strong heterogeneity.Combined with an integrated analysis of data from seismic,geology,and drilling results,a series of attributes which are suitable for fractured and caved carbonate reservoir prediction is discussed,including amplitude,coherence analysis,spectra decomposition,seismic absorption attenuation analysis and impedance inversion.Moreover,3-D optimization of these attributes is achieved by integration of multivariate discriminant analysis and principle component analysis,where the logging data are taken as training samples.Using the optimized results,the spatial distribution and configuration features of the caved reservoirs can be characterized in detail.This technique not only improves the understanding of the spatial distribution of current reservoirs but also provides a significant basis for the discovery and production of carbonate reservoirs in the Tarim Basin.Liu Lifeng Sam Zandong Sun Yang Haijun Han Jianfa Gao Hongliang Jing Bing Zhu Xinghui 2011Petroleum Science2011,8,4:6

10Spatial heterogeneity of lake eutrophication caused by physiogeographic conditions:An analysis of 143 lakes in China显示文摘In order to identify the effect of geographic characteristics on the variations of nutrient concentrations and the utilization efficiency of nutrients by phytoplankton, data from 143 lakes, from 2008 to 2010, including three very different types of topography, i.e., the first topography ladder(FTL), second topography ladder(STL), and third topography ladder(TTL), were statistically analyzed. Lakes in the FTL and STL, located at high elevation(above1000 m) and low longitudes(lower than 105° E), were sporadically oligotrophic, whereas lakes in the TTL were almost all mesotrophic and eutrophic. The trophic level index(TLI)became higher with the rise of longitude. Two turning points(5 and 15°C) on the curve of TLI as function of the average annual temperature(AAT) corresponded with the AAT in different lake regions in the STL and TTL. Because the responses of TLI to AAT differ significantly, there were variations of nutrient and algal biomass concentrations in different lake regions in the same type of topography ladder. According to the differences in utilization efficiency of total nitrogen or total phosphorus by phytoplankton, China could be partitioned into six lake regions. 11Novel hierarchical structure of MoS_(2)/TiO_(2)/Ti_(3)C_(2)T_(x) composites for dramatically enhanced electromagnetic absorbing properties显示文摘In order to prevent the microwave leakage and mutual interference,more and more microwave absorbing devices are added into the design of electronic products to ensure its routine operation.In this work,we have successfully prepared MoS_(2)/TiO_(2)/Ti_(3)C_(2)T_(x) hierarchical composites by one-pot hydrothermal method and focused on the relationship between structures and electromagnetic absorbing properties.Supported by comprehensive characterizations,M0S_(2) nanosheets were proved to be anchored on the surface and interlayer of Ti_(3)C_(2)T_(x) through a hydrothermal process.Additionally,TiO_(2) nanoparticles were obtained in situ.Due to these hierarchical structures,the MoS_(2)/TiO_(2)/Ti_(3)C_(2)T_(x) composites showed greatly enhanced microwave absorbing performance.The MoS_(2)/TiO_(2)/Ti_(3)C_(2)T_(x) composites exhibit a maximum reflection loss value of -33.5 dB at 10.24 GHz and the effective absorption bandwidth covers 3.1GHz(13.9-17 GHz)at the thickness of 1.0 mm,implying the features of wide frequency and light weight.This work in the hierarchical structure of MoS_(2)/TiO_(2)/Ti_(3)C_(2)T_(x) composites opens a promising door to the exploration of constructing extraordinary electromagnetic wave absorbents.Heng DU Qipeng ZHANG Biao ZHAO Frank MARKEN Qiancheng GAO Hongxia LU Li GUAN Hailong WANG Gang SHAO Hongliang XU Rui ZHANG Bingbing FAN 2021Journal of Advanced Ceramics2021,10,5:4

13Comparative transcriptional profiling under drought stress between upland and lowland rice (Oryza sativa L.) using cDNA-AFLP显示文摘The continuous growth of lowland rice (LR) in paddy fields supplied with enough water over the years, and of upland rice (UR) in naturally rain-fed soils, has resulted in greater resistance to drought stress in UR compared to LR. To elucidate their differential regulation mechanisms of drought-resistance, genome-wide transcript regulation under drought stress in UR and LR was investigated using cDNA-AFLP. The results indicated that over 90% of gene expression was not affected by drought stress in the two rice genotypes, more than 8% was regulated by drought stress in both, and less than 1% was specifically expressed in UR or LR. Fifty-seven genes were specifically expressed in UR and thirty-eight specifically in LR. Genes specifically expressed in UR included cell rescue and defence genes functioning in drought-resistance, signal transduction molecules, nucleotides and amino acid biosynthesis genes required for plant growth, and the regulatory genes for growth and development. In LR, genes specifically expressed were related to protein and nucleotide degradation. Some genes were upregulated earlier in UR, and downregulated genes were inclined to be downregulated earlier in UR compared to LR, implying that more rapid regulation mechanisms caused earlier responses of UR to drought stress. Expression levels of upregulated genes in UR were higher than those in LR. 15Lycopene Accumulation Affects the Biosynthesis of Some Carotenoid-related Volatiles Independent of Ethylene in Tomato显示文摘For elucidating the regulatory mechanism of ethylene on carotenoid-related volatiles (open chain) compounds and the relationship between lycopene and carotenoid-related volatiles, transgenic tomato fruits in which ACC synthase was suppressed were used. The transgenic tomato fruit showed a significant reduction of lycopene and aroma volatiles with low ethylene production. 6-methyl-5-hepten-2-one, 6-methyl-5-hepten-2-ol and geranylacetone, which were suspected to be lycopene degradation products, were lower than those in wild type tomato fruits. In order to identify whether lycopene accumulation effects the biosynthesis of some carotenoid-related volatiles independent of ethylene in tomato or not, the capability of both wild type and transgenic tomato fruits discs to convert lycopene into carotenoid-related volatiles was evaluated. The data showed that external lycopene could convert into 6-methyl-5-hepten-2-one and 6-methyl-5-hepten-2-ol in vivo, indicating that the strong inhibition of ethylene production had no effect on enzymes in the biosynthesis pathway of some carotenoid-related volatiles. Therefore, in ACS-suppression transgenic tomato fruits, the low levels of 6-methyl-5-hepten-2-one, 6-methyl-5-hepten-2-ol was due to decreased lycopene accumulation, not ethylene production. 16Response of phytoplankton community to different water types in the western Arctic Ocean surface water based on pigment analysis in summer 2008显示文摘Nutrients and photosynthesis pigments were investigated in the western Arctic Ocean during the 3rd Chinese Arctic Research Expedition Cruise in summer 2008.The study area was divided into five provinces using the Kmeans clustering method based on the physical and chemical characteristics of the sea water,and to discuss the distribution of the phytoplankton community structure in these provinces.CHEMTAX software was performed using HPLC pigments to estimate the contributions of eight algal classes to the total chlorophyll a(TChl a).The results showed that on the Chukchi Shelf,the Pacific Ocean inflow mainly controlled the Chl a biomass and phytoplankton communities by nutrient concentrations.The high nutrient Anadyr Water and Bering Shelf Water(An W and BSW) controlled region have high Chl a levels and the diatom dominated community structure.In contrast,in the region occupied by low-nutrient like Alaska Coastal Water(ACW),the Chl a biomass was low,with pico-and nano-phytoplankton as dominated species,such as prasinophytes,chrysophytes and cryptophytes.However,over the off-shelf,the ice cover condition which would affect the physical and nutrient concentrations of the water masses,in consequence had a greater impact on the phytoplankton community structure.Diatom dominated in ice cover region and its contribution to Chl a biomass was up to 75%.In the region close to the Mendeleev Abyssal Plain(MAP),controlled by sea-ice melt water with relatively high salinity(MW-HS),higher nutrient and Chl a concentrations were found and the phytoplankton was dominated by pico-and nano-algae,while the diatom abundance reduced to 33%.In the southern Canada Basin,an ice-free basin(IfB) with the lowest nutrient concentrations and most freshened surface water,low Chl a biomass was a consequence of low nutrients.The ice retreating and a prolonged period of open ocean may not be beneficial to the carbon export efficiency due to reducing the Chl a biomass or intriguing smaller size algae growth.JIN Haiyan ZHUANG Yanpei LI Hongliang CHEN lianfang GAO Shengquan JI Zhongqiang ZHANG Yang 2017Acta Oceanologica Sinica2017,36,8:2

18Maternal Thermal Effects on Female Reproduction and Hatchling Phenotype in the Chinese Skink(Plestiodon chinensis)显示文摘We maintained gravid Chinese skinks(Plestiodon chinensis) at three constant temperatures(25, 28 and 31 °C) during gestation, and randomly assigned eggs from each female to one of the same three temperatures for incubation to determine maternal thermal effects on female reproduction and hatchling phenotype. Maternal temperature affected egg-laying date, hatching success and hatchling linear size(snout-vent length, SVL) but not clutch size, egg size, egg component, and embryonic stage at laying. More specifically, females at higher temperatures laid eggs earlier than did those at low temperatures, eggs laid at 31 °C were less likely to hatch than those laid at 25 °C or 28 °C, and hatchlings from eggs laid at 31 °C were smaller in SVL. Our finding that maternal temperature(pre-ovipositional thermal condition) rather than incubation temperature(post-ovipositional thermal condition) affected hatching success indicated that embryos at early stages were more vulnerable to temperature than those at late stages.
