1Overexpression of a dominant-negative mutant of SIRT1 in mouse heart causes cardiomyocyte apoptosis and early-onset heart failure显示文摘SIRT1,a mammalian ortholog of yeast silent information regulator 2(Sir2),is an NAD+-dependent protein deacetylase that plays a critical role in the regulation of vascular function.The current study aims to investigate the functional significance of deacetylase activity of SIRT1 in heart.Here we show that the early postnatal hearts expressed the highest level of SIRT1deacetylase activity compared to adult and aged hearts.We generated transgenic mice with cardiac-specific expression of a dominant-negative form of the human SIRT1(SIRT1H363Y),which represses endogenous SIRT1 activity.The transgenic mice displayed dilated atrial and ventricular chambers,and died early in the postnatal period.Pathological,echocardiographic and molecular phenotype confirmed the presence of dilated cardiomyopathy.Terminal deoxynucleotidyl transferase-mediated dUTP nick-end-labeling analysis revealed a greater abundance of apoptotic nuclei in the hearts of transgenic mice.Furthermore,we show that cardiomyocyte apoptosis caused by suppression of SIRT1 activity is,at least in part,due to increased p53acetylation and upregulated Bax expression.These results indicate that dominant negative form of SIRT1(SIRT1H363Y)overexpression in mouse hearts causes cardiomyocyte apoptosis and early-onset heart failure,suggesting a critical role of SIRT1 in preserving normal cardiac development during the early postnatal period.MU WenLi ZHANG QingJun TANG XiaoQiang FU WenYan ZHENG Wei LU YunBiao LI HongLiang WEI YuSheng LI Li SHE ZhiGang CHEN HouZao LIU DePei 2014Science China(Life Sciences)2014,57,9:12

3Phytoplankton composition and its ecological effect in subsurface cold pool of the northern Bering Sea in summer as revealed by HPLC derived pigment signatures显示文摘CHEMTAX analysis of high-performance liquid chromatography(HPLC) pigment was conducted to study phytoplankton community structure in the northern Bering Sea shelf, where a seasonal subsurface cold pool emerges. The results showed that fucoxanthin(Fuco) and chlorophyll a(Chl a) were the most abundant diagnostic pigments, with the integrated water column values ranging from 141 to 2 160 μg/m2 and 477 to 5 535 μg/m2, respectively. Moreover, a diatom bloom was identified at Sta. BB06 with the standing stock of Fuco up to 9 214 μg/m3. The results of CHEMTAX suggested that the phytoplankton community in the northern Bering Sea shelf was dominated by diatoms and chrysophytes with an average relative contribution to Chl a of 80% and 12%, respectively, followed by chlorophytes, dinoflagellates, and cryptophytes. Diatoms were the absolutely dominant algae in the subsurface cold pool with a relative contribution exceeding 90%, while the contribution of chrysophytes was generally higher in oligotrophic upper water. 5Zeolitic Imidazolate Framework 8-Derived Au@ZnO for Efficient and Robust Photocatalytic Degradation of Tetracycline显示文摘Summary of main observation and conclusion Tetracycline (TC) and other antibiotics accumulated in groundwater and soil pollute ecological environment and threaten human health. Gold nan oparticles doped on photocatalysts are able to enhance the photodegradation efficiency during removing these antibiotics, but preparation of Au nanoparticles of well-dispersion on photocatalysts remains challenging. In this work, zeolite imidazolate (ZIF-8) was employed as the precursor to prepare Au@ZnO photocatalyst via impregnation and in-situ reduction method to efficiently degrade the tetracycline in the aqueous solution. Au nanoparticles are of 10 nm in size and uniformly dispersed on the surfaces of ZnO microstructures. The as-prepared Au@ZnO is able to remove 85.5% of TC of 0.010 mg/mL within 2h, presenting higher photocatalytic activity than pure ZnO catalyst. Most importantly, the catalyst shows its superior stability after five cycles without structure and activity changing. 6Magnetic performance and corrosion resistance of electroless plating CoWP film显示文摘The CoWP film with good magnetic performance and corrosion resistance was electrolessly plated from alkaline solution. The technical parameters of the electroless plating system were optimized. When the pH value of electroless plating solution was 11.0 and the reducing agent (NaH2PO2) content was 0.4 mol L 1, the target chemical reactions proceeded in the electroless plating solution smoothly with negligible interference and side effects. CoWP film prepared under optimal deposition condition contained more hexagonal close-packed (hcp) cobalt (ε-Co) of [110] and crystallographic orientation of Co [002]. VSM analysis reveals that the saturation magnetization of the CoWP film is 100~220 Am2 kg 1 and coercivity is (2.87~4.38)×104 A·cm 1. The corrosion behavior of CoWP film in 3.5% NaCl aqueous solution was studied by electrochemical experiments. The results prove that the corrosion resistance of the CoWP film deposited under the optimal depo-sition condition exhibites relatively lower corrosion current of (3.054~3.162)×10 6 A·cm 2. 7Tauroursodeoxycholic acid(TUDCA) inhibits influenza A viral infection by disrupting viral proton channel M2显示文摘Influenza is a persistent threat to human health and there is a continuing requirement for updating antiinfluenza strategies. Initiated by observations of different endoplasmic reticulum(ER) responses of host to seasonal H1N1 and highly pathogenic avian influenza(HPAI) A H5N1 infections, we identified an alternative antiviral role of tauroursodeoxycholic acid(TUDCA), a clinically available ER stress inhibitor, both in vitro and in vivo. Rather than modulating ER stress in host cells, TUDCA abolished the proton conductivity of viral M2 by disrupting its oligomeric states, which induces inefficient viral infection. We also showed that M2 penetrated cells, whose intracellular uptake depended on its proton channel activity,an effect observed in both TUDCA and M2 inhibitor amantadine. 9Inhibition of inflammatory mediator release from microglia can treat ischemic/hypoxic brain injury显示文摘Interleukin-1α and interleukin-1β aggravate neuronal injury by mediating the inflammatory reaction following ischemic/hypoxic brain injury. It remains unclear whether interleukin-1α and interleukin-1β are released by microglia or astrocytes. This study prepared hippocampal slices that were subsequently subjected to oxygen and glucose deprivation. Hematoxylin-eosin staining verified that neurons exhibited hypoxic changes. Results of enzyme-linked immunosorbent assay found that interleukin-1α and interleukin-1β participated in this hypoxic process. Moreover, when hypoxic injury occurred in the hippocampus, the release of interleukin-1α and interleukin-1β was mediated by the P2X4 receptor and P2X7 receptor. Immunofluorescence staining revealed that during ischemia/hypoxia, the P2X4 receptor, P2X7 receptor, interleukin-1α and interleukin-1β expression was detectable in rat hippocampal microglia, but only P2X4 receptor and P2X7 receptor expression was detected in astrocytes. Results suggested that the P2X4 receptor and P2X7 receptor, respectively, mediated interleukin-1α and interleukin-1β released by microglia, resulting in hippocampal ischemic/hypoxic injury. 10Microstructure and mechanical properties of C_(x)Hf_(0.25)NbTaW_(0.5) refractory high-entropy alloys at room and high temperatures显示文摘The microstructure and mechanical properties of as-cast and isothermally annealed C_(x)Hf_(0.25)NbTaW_(0.5)(x=0,0.05,0.15,0.25)refractory high-entropy alloys(RHEAs)were studied.Both the as-cast and annealed RHEAs consisted of disordered body-centered cubic solid solution phase and metal carbide(MC)phase with a face-centered cubic crystal structure(Fm-3 m space group).The primary carbides were enriched with Hf and C elements and tended to form lamellar eutectic-like microstructure in the interdendrites.The lamellar eutectic-like structure in the interdendrites would be formed from the decomposition of sub-carbide M_(2)C under the influence of Hf element.After isothermal annealing,slatted carbides were precipitated on the matrix,and the distribution became more uniform with high C content.The formation of carbides strongly influenced the mechanical properties both at room and high temperatures.The yield strength values of C_(x)Hf_(0.25)NbTaW_(0.5) RHEA at 1473 and 1673 K were 792 and 749 MPa,respectively.The result had exceeded the high temperature mechanical properties of currently known RHEAs.Moreover,this RHEA exhibited high-temperature performance stability and excellent plasticity,exceeding 30 and 50%at room and elevated temperatures(above 1273 K),respectively.During thermal deformation,carbon-containing RHEAs obtained more severe work hardening than that of ACHO RHEAs,and required greater dynamic recrystallization to achieve the dynamic equilibrium.Shiyu Wu Dongxu Qiao Haitao Zhang Junwei Miao Hongliang Zhao Jun Wang Yiping Lu Tongmin Wang Tingju Li 2022Journal of Materials Science & Technology2022,,2:4
11Differences in Thermal Preference and Tolerance among Three Phrynocephalus Lizards (Agamidae) with Different Body Sizes and Habitat Use显示文摘We acclimated adults of two viviparous(Phrynocephalus guinanensis and P.vlangalii) and one oviparous(P.versicolor) species of toad-headed lizards(Agamidae) to 28 °C,33 °C and 38 °C to examine whether thermal preference(preferred body temperature,Tp) and thermal tolerance(critical thermal minimum,CTMin;critical thermal maximum,CTMax) were affected by acclimation temperature,and correlate with body size and habitat use.Both Tp and CTMax were highest in P.versicolor and lowest in P.vlangalii,with P.guinanensis in between.The two viviparous species did not differ in CTMin and thermal tolerance range,and they both were more resistant to low temperatures and had a wider range of thermal tolerance than the oviparous species.Both CTMin and CTMax shifted upward as acclimation temperature increased in all the three species.Tp was higher in the lizards acclimated to 33 °C than in those to 28 °C or 38 °C.The range of thermal tolerance was wider in the lizards acclimated to 28 °C than in those to 33 °C or 38 °C.The data showed that:1) thermal preference and tolerance were affected by acclimation temperature,and differed among the three species of Phrynocephalus lizards with different body sizes and habitat uses;2) both Tp and CTMax were higher in the species exchanging heat more rapidly with the environment,and CTMin was higher in the species using warmer habitats during the active season;and 3) thermal preference and tolerance might correlat with body size and habitat use in Phrynocephalus lizards.Zheng WANG Hongliang LU Li MA Xiang JI 2013Asian Herpetological Research2013,4,3:4
12Response of nutrients and the surface phytoplankton community to ice melting in the central Arctic Ocean显示文摘During the fourth Chinese National Arctic Research Expedition cruise in summer 2010, a time-series observation was carried out to examine the response of nutrients and phytoplankton community in the ice-water interface to the ice melting ice in the central Arctic Ocean. Phosphate and silicate in the ice-water interface were rich relative to dissolved inorganic nitrogen (DIN), based on the Redfield ratio (16N:1P:16Si), suggesting that DIN was the potential limiting nutrient. DIN concentrations in the sea ice were about 3-4 times that in the surface seawater, indicating that melting ice delivered DIN to the surface water. Pigment analysis showed that fucoxanthin and chlorophyll a contribute to carotenoids and chlorophylls in particles. The mean concentrations of chlorophyll c,diatoxanthin, diadinoxanthin and fucoxanthin from 15 August to 18 August were 6 μgm-3, 22 μgm-3, 73 μgm-3 and 922 μg·m-3, respectively, suggesting that diatoms dominated in the phytoplankton community composition.Furthermore, a notable enhancement in fucoxanthin and chlorophyll a during a large-scale ice melting was likely attributed to senescent diatoms released from the bottom sea-ice as well as phytoplankton diatoms growth in the water column due to the input of nutrients (i.e., DIN) and reducing light limitation from melting ice. 13Novel hierarchical structure of MoS_(2)/TiO_(2)/Ti_(3)C_(2)T_(x) composites for dramatically enhanced electromagnetic absorbing properties显示文摘In order to prevent the microwave leakage and mutual interference,more and more microwave absorbing devices are added into the design of electronic products to ensure its routine operation.In this work,we have successfully prepared MoS_(2)/TiO_(2)/Ti_(3)C_(2)T_(x) hierarchical composites by one-pot hydrothermal method and focused on the relationship between structures and electromagnetic absorbing properties.Supported by comprehensive characterizations,M0S_(2) nanosheets were proved to be anchored on the surface and interlayer of Ti_(3)C_(2)T_(x) through a hydrothermal process.Additionally,TiO_(2) nanoparticles were obtained in situ.Due to these hierarchical structures,the MoS_(2)/TiO_(2)/Ti_(3)C_(2)T_(x) composites showed greatly enhanced microwave absorbing performance.The MoS_(2)/TiO_(2)/Ti_(3)C_(2)T_(x) composites exhibit a maximum reflection loss value of -33.5 dB at 10.24 GHz and the effective absorption bandwidth covers 3.1GHz(13.9-17 GHz)at the thickness of 1.0 mm,implying the features of wide frequency and light weight.This work in the hierarchical structure of MoS_(2)/TiO_(2)/Ti_(3)C_(2)T_(x) composites opens a promising door to the exploration of constructing extraordinary electromagnetic wave absorbents.Heng DU Qipeng ZHANG Biao ZHAO Frank MARKEN Qiancheng GAO Hongxia LU Li GUAN Hailong WANG Gang SHAO Hongliang XU Rui ZHANG Bingbing FAN 2021Journal of Advanced Ceramics2021,10,5:4

18(Ca,Sr,Ba)ZrO_(3):A promising entropy-stabilized ceramic for titanium alloys smelting显示文摘A novel entropy-stabilized(ES)(Ca,Sr,Ba)ZrO_(3) ceramic has been designed and synthesized by pressureless sintering of CaZr03,SrZr03 and BaZr03 powders mixtures at 1450℃,1500℃and 1550℃for 3 h.X-ray diffraction,scanning electron microscopy and transmission electron microscopy analyses collectively indicate that a single solid solution is formed with a homogeneous distribution of metal elements after sintering at 1550℃.The relative density and hardness of the ES(Ca,Sr,Ba)ZrO_(3) ceramic sintered at 1550℃are 97.79%and 10.840.33 GPa,re s pectively.This ES(Ca,Sr,Ba)ZrO_(3) exhibits lower thermal conductivity from 373 K to 1073 K than their constituting zirconates,CaZrO_(3),SrZrO_(3) and BaZrO_(3).Most importantly,the ES(Ca,Sr,Ba)ZrO_(3) ceramic possesses good corrosion resistance to TiNi alloy melt and no distinct reaction layer exists between TiNi alloy and ES(Ca,Sr,Ba)ZrO_(3) ceramic in the contact region.The combination of these properties indicates that ES(Ca,Sr,Ba)ZrO_(3) ceramic is promising for use as a novel crucible material for the melting of titanium alloys.Shuaihang Qiu Mingliang Li Gang Shao Hailong Wang Jinpeng Zhu Wen Liu Bingbing Fan Hongliang Xu Hongxia Lu Yanchun Zhou Rui Zhang 2021Journal of Materials Science & Technology2021,,6:3
19Lycopene Accumulation Affects the Biosynthesis of Some Carotenoid-related Volatiles Independent of Ethylene in Tomato显示文摘For elucidating the regulatory mechanism of ethylene on carotenoid-related volatiles (open chain) compounds and the relationship between lycopene and carotenoid-related volatiles, transgenic tomato fruits in which ACC synthase was suppressed were used. The transgenic tomato fruit showed a significant reduction of lycopene and aroma volatiles with low ethylene production. 6-methyl-5-hepten-2-one, 6-methyl-5-hepten-2-ol and geranylacetone, which were suspected to be lycopene degradation products, were lower than those in wild type tomato fruits. In order to identify whether lycopene accumulation effects the biosynthesis of some carotenoid-related volatiles independent of ethylene in tomato or not, the capability of both wild type and transgenic tomato fruits discs to convert lycopene into carotenoid-related volatiles was evaluated. The data showed that external lycopene could convert into 6-methyl-5-hepten-2-one and 6-methyl-5-hepten-2-ol in vivo, indicating that the strong inhibition of ethylene production had no effect on enzymes in the biosynthesis pathway of some carotenoid-related volatiles. Therefore, in ACS-suppression transgenic tomato fruits, the low levels of 6-methyl-5-hepten-2-one, 6-methyl-5-hepten-2-ol was due to decreased lycopene accumulation, not ethylene production. 20Satb2 expression in Foxci-promoted osteogenic differentiation of MC3T3-E1 cells is negatively regulated by microRNA-103-3p显示文摘The forkhead tran scripti on factor C1 (Foxci) is a cell-fate-determining factor that controls cranial bone development and osteogenic differentiation. Previously, it was demonstrated that various microRNAs (miRNAs) play important roles in osteogenesis and regulate the complex process of osteogenic differentiation. However, it remains unclear how miRNA expression changes during Foxci-promoted osteogenic differentiation. In this study, we successfully overexpressed the Foxci gene in MC3T3-E1 cells and investigated the alterations in the miRNA expression profile on day 3 after osteogenic induction by using a miRNA microarray. Nine downregulated miRNAs and eight upregulated miRNAs were found to be differentially expressed. Among these miRNAs, miR-103-3p was consistently downregulated in the Foxc7-overexpressing MC3T3-E1 cells and was identified as a negative regulator of osteogenic differentiation by using a gain-and Iose-of-funotion assay. The special AT-rich sequence-binding protein 2 (Satb2), a pivotal osteogenic transcription factor, was identified as the miR-103-3p targeting gene and was verified by real-time polymerase chain reaction, western blot analysis, and luciferase assay. Overexpression of miR-103-3p markedly inhibited the expression of Satb2 and attenuated Foxci-promoted osteogenic differentiation.
