1Conservation of Jiaohe ancient earthen site in China显示文摘Earthen sites,which are mainly made of earth,are cultural heritages with historical,artistic and scientific values.Many extremely valuable earthen sites have been preserved in the arid areas in China.The earthen site of Jiaohe Ancient City is one of the earliest National Protected Important Cultural Heritage Sites.The Jiaohe Ancient City site exhibits all kinds of deteriorations,which can be found in the earthen sites in arid environments in China.Through a case study of the Jiaohe ancient earthen site,we present in this paper the comprehensive conservation technologies,including the mud bricklaying and reparation,wooden rod anchorage,crack grouting,surface potassium silicate(PS) penetration consolidation,and suspended steel beam ceiling,etc.Results of this case study showed that better conservation effects could be achieved by selecting proper PS penetration and crack grouting processes based on the deterioration characteristics of the earthen sites.The technology of mud bricklaying and reparation was also an effective method for preventing the earthen body from collapsing.Compared with traditional conservation technologies,the suspended steel beam ceiling technology could effectively reduce the negative impacts to the original state of the earthen site.As for unstable cliffs,a new method using composite anchor rod of bamboo and steel with massive loose earth was applied.Deformation monitoring and temporary supports were critical and indispensable measures for the safe of site conservation projects.Through years of monitoring and practical operation at the Jiaohe ancient earthen site,deterioration at the site has been effectively controlled.Zuixiong Li Xudong Wang Manli Sun Wenwu Chen Qinglin Guo Huyuan Zhang 2011Journal of Rock Mechanics and Geotechnical Engineering2011,3,3:6
2Inhibition of cyclooxygenase-2 activity in subchondral bone modifies a subtype of osteoarthritis显示文摘Osteoarthritis(OA) causes the destruction of joints. Its pathogenesis is still under investigation, and there is no effective diseasemodifying therapy. Here, we report that elevated cyclooxygenase-2(COX-2) expression in the osteocytes of subchondral bone causes both spontaneous OA and rheumatoid arthritis(RA). The knockout of COX-2 in osteocytes or treatment with a COX-2 inhibitor effectively rescues the structure of subchondral bone and attenuates cartilage degeneration in spontaneous OA(STR/Ort)mice and tumor necrosis factor-α transgenic RA mice. Thus, elevated COX-2 expression in subchondral bone induces both OAassociated and RA-associated joint cartilage degeneration. 3Three dimensional distribution of surfactant in microspheres revealed by synchrotron radiation X-ray microcomputed tomography显示文摘This study investigated the formulation mechanism of microspheres via internal surfactant distribution. Eudragit L100 based microspheres loaded with bovine serum albumin were prepared by solid in oil in oil emulsion solvent evaporation method using acetone and liquid paraffin system containing sucrose stearate as a surfactant. The fabricated microspheres were evaluated for encapsulation efficiency, particle size, production yield, and in vitro release characteristics. The internal structures of microspheres were characterized using synchrotron radiation X-ray microcomputed tomography(SR-μCT). The enhanced contrast made the sucrose stearate distinguished from Eudragit to have its three dimensional(3D) distribution. Results indicated that the content and concentration determined the state of sucrose stearate and had significant influences on the release kinetics of protein. The dispersity of sucrose stearate was the primary factor that controlled the structure of the microspheres and further affected the encapsulation efficiency, effective drug loading, as well as in vitro release behavior. In conclusion, the 3D internal distribution of surfactant in microspheres and its effects on protein release behaviors have been revealed for the first time. 5Facile and efficient fabrication of g-C3N4 quantum dots for fluorescent analysis of trace copper(Ⅱ) in environmental samples显示文摘The facile preparation of g-C3N4 QDs with high fluorescent performance has become an important direction in the last decade. Herein, we develop a facile, rapid approach to synthesize highly fluorescent QDs based on recrystallization and ultrasonic exfoliation. Size-controllable graphitic carbon nitride(gC3N4) QDs can be obtained from the precursor of recrystallized dicyandiamide, only 90 min is needed and the size of QDs is adjusted from 5 nm to 200 nm by controlling the ultrasonic time. Moreover, better fluorescent efficiency is also obtained comparing to traditional g-C3N4 QDs. 6Assembly of layer-by-layer films of superoxide dismutase and gold nanorods:A third generation biosensor for superoxide anion显示文摘Based on the layer-by-layer self-assembly of positively charged cetyltrimethylammonium bromide (CTAB) wrapped gold na- norods (AuNRs) and negatively charged superoxide dismutase (SOD) from their aqueous solutions on cysteine modified gold electrode (Cys/Au), a third generation electrochemical biosensor ((SOD/AuNRs)2/Cys/Au) for superoxide anion (O2·-) was developed. The two layers assembly of SOD/AuNRs can significantly enhance the direct electron transfer between SOD and the electrode. The functional enzymatic activities of the SOD offer an electrochemical approach to the determination of O2·-. 7Numerical simulation of the behaviors of test square for prehistoric earthen sites during archaeological excavation显示文摘During the process of archaeological excavation in the regions of Southeast China, collapse of test square usually occurs due to poor site-specific conditions. In this paper, the fast Lagrangian analysis of continua in three dimensions(FLAC3D) is employed to reveal the behaviors of test square. Taking the archaeological works in Liangzhu prehistoric earthen sites as the research background, the paper first introduces the geological setting, excavation procedure and monitoring scheme of the studied test square. Then, the deformation of four sides of the test square is modeled using FLAC3D. By comparison, it shows that the numerical results are consistent with the results from field monitoring. 19Hybridoma-derived neutralizing monoclonal antibodies against Beta and Delta variants of SARS-CoV-2 in vivo显示文摘Neutralizing monoclonal antibodies(mAb)are a major therapeutic strategy for the treatment of severe acute respiratory syndrome coronavirus-2(SARS-CoV-2)infection.The continuous emergence of new SARS-CoV-2 variants worldwide has increased the urgency for the development of new mAbs.In this study,we immunized mice with the receptor-binding domain(RBD)of the SARS-CoV-2 prototypic strain(WIV04)and screened 35 RBDspecific mAbs using hybridoma technology.Results of the plaque reduction neutralization test showed that 25 of the mAbs neutralized authentic WIV04 strain infection.The 25 mAbs were divided into three categories based on the competitive enzyme-linked immunosorbent assay results.A representative mAb was selected from each category(RD4,RD10,and RD14)to determine the binding kinetics and median inhibitory concentration(IC_(50))of WIV04 and two variants of concern(VOC):B.1.351(Beta)and B.1.617.2(Delta).RD4 neutralized the B.1.617.2 variant with an IC50 of 2.67 ng/mL;however,it completely lost neutralizing activity against the B.1.351 variant.RD10 neutralized both variants with an IC50 exceeding 100 ng/mL;whereas RD14 neutralized two variants with a higher IC50(>1 mg/mL).Animal experiments were performed to evaluate the protective effects of RD4 and RD10 against various VOC infections.RD4 could protect Adv-hACE2 transduced mice from B.1.617.2 infection at an antibody concentration of 25 mg/kg,while RD10 could protect mice from B.1.351 infection at an antibody concentration of 75 mg/kg.These results highlight the potential for future modifications of the mAbs for practical use.Qianran Wang Lu Peng Yanqiu Nie Yanni Shu Huajun Zhang Zidan Song Yufeng Li Hengrui Hu Liushuai Li Xi Wang Jia Liu Jiang Li Zhengli Shi Fei Deng Yu Guo Yiwu Zhou Bing Yan Zhihong Hu Manli Wang 2023Virologica Sinica2023,38,2:0
