| 1 | The OsAMT1.1 gene functions in ammonium uptake and ammoniumepotassium homeostasis over low and high ammonium concentration ranges显示文摘Rice(Oryza sativa) grown in paddy fields is an ammonium(NH^+_4)-preferring crop;however,its AMT-type NH^+_4transporters that mediate root N acquisition have not been well characterized yet.In this study,we analyzed the expression pattern and physiological function of the OsAMT1.1 gene of the AMT1 subfamily in rice.Os AMT1.1 is located in the plasma membrane and is mainly expressed in the root epidermis,stele and mesophyll cells.Disruption of the Os AMT1.1 gene decreased the uptake of NH^+_4,and the growth of roots and shoots under both low NH^+_4and high NH^+_4conditions.Os AMT1.1 contributed to the short-term(5 min)^(15)NH^+_4influx rate by approximately one-quarter,irrespective of the NH^+_4concentration.Knockout of Os AMT1.1 significantly decreased the total N transport from roots to shoots under low NH^+_4conditions.Moreover,compared with the wild type,the osamt1.1 mutant showed an increase in the potassium(K)absorption rate under high NH^+_4conditions and a decrease under low NH^+_4conditions.The mutants contained a significantly high concentration of K in both the roots and shoots at a limited K(0.1 mmol/L)supply when NH^+_4was replete.Taken together,the results indicated that OsAMT1.1 significantly contributes to the NH^+_4uptake under both low and high NH^+_4conditions and plays an important role in Ne K homeostasis in rice. | Chang Li Zhong Tang Jia Wei Hongye Qu Yanjie Xie Guohua Xu | 2016 | Journal of Genetics and Genomics2016,43,11: | 12 |
| 2 | Effects of dietary leucine supplementation on the gene expression of mammalian target of rapamycin signaling pathway and intestinal development of broilers显示文摘This experiment was to investigate the effects of dietary leucine supplementation on the gene expression of mammalian target of rapamycin(mTOR) signaling pathway and intestinal development of broilers. A total of 384 one-day-old broilers were randomly assigned into 4 treatments with 6 replicates(16 broilers per replicate). Broilers in these treatment groups were offered the following diets with 1.37,1.77,2.17 and2.57% of leucine. These diet treatments were named 1.37 TM, 1.77 TM, 2.17 TM, and 2.57 TM. The experiment lasted 21 days and all birds had free access to feed and water. Results indicated that there was no significant difference in body weight, average daily gain and average feed intake among all treatments(P > 0.05). The broiler duodenal villus height in 2.57 TM was the lowest, but the highest occurred in1.37 TM on d 7 and 14(P < 0.05). The villus height in the jejunum and ileum increased along with leucine level from 1.37 to 2.17%. The villus height of jejunum was significantly higher in 2.17 TM than in 1.37 TM on d 7 and 14, and the ratio of villus height to crypt depth(V:C) in the duodenum, jejunum and ileum increased significantly(P < 0.05) on d 21. The gene expression level of mTOR in the duodenum decreased with increasing leucine level and was higher in 1.37 TM than in 2.57 TM on d 7 and 14(P < 0.05). On d 14 and 21 of the trial, the expression of S6 K1 in the duodenum was higher in 1.37 TM than in 2.57 TM(P < 0.05), and the expression of mTOR, S6 K1 in the jejunum and ileum increased with increasing leucine level form 1.37 to 2.17%, whereas a significant difference occurred between 1.37 TM and 2.17 TM(P< 0.05).In conclusion, the addition of leucine fails to enhance the growth performance of broilers. However,leucine can improve intestinal development by enhancing villus height and V:C ratio in the jejunum and ileum. | 3 | Molecular mechanisms of growth depression in broiler chickens(Gallus Gallus domesticus)mediated by immune stress:a hepatic proteome study显示文摘Background:Immunological stress decreases feed intake,suppresses growth and induces economic losses.However,the underlying molecular mechanism remains unclear.Label-free liquid chromatography and mass spectrometry(LC-MS)proteomics techniques were employed to investigate effects of immune stress on the hepatic proteome changes of Arbor Acres broilers(Gallus Gallus domesticus)challenged with Escherichia coli lipopolysaccharide(LPS).Results:Proteomic analysis indicated that 111 proteins were differentially expressed in the liver of broiler chickens from the immune stress group.Of these,28 proteins were down-regulated,and 83 proteins were up-regulated in the immune stress group.Enrichment analysis showed that immune stress upregulated the expression of hepatic proteins involved in defense function,amino acid catabolism,ion transport,wound healing,and hormone secretion.Furthermore,immune stress increased valine,leucine and isoleucine degradation pathways.Conclusion:The data suggests that growth depression of broiler chickens induced by immune stress is triggered by hepatic proteome alterations,and provides a new insight into the mechanism by which immune challenge impairs poultry production. | Aijuan Zheng Anrong Zhang Zhimin Chen Shoaib Ahmed Pirzado Wenhuan Chang Huiyi Cai Wayne L.Bryden Guohua Liu | 2021 | Journal of Animal Science and Biotechnology2021,12,4: | 4 |

| 17 | Sea level change along the coast of China relative to the isostatic datum显示文摘On the basis of the analyses of significant periods for the sea level observation data taken from recent several decades at 12 tide stations, the monthly mean sea level observations are fitted by a model of linear trend of sea level change superimposed with several variations of different fixed periods. The trends of sea level relative changes and their errors are estimated by the LS method. The results are reduced to the isostatic datum proposed and established in the paper (Huang et al. , 1991, Seismology and Geology , 1, 1-15). The trends of sea level changes in the near future along the coast of China are studied. It is pointed out that the general trend of the sea level change along the coast of China is going up slowly and the rate of the change is not the same in different segments of the coasts. In a few segments, the sea level is even relatively going down. | 18 | Effects of dynamic segmentation of nutrient supply on growth performance and intestinal development of broilers显示文摘This experiment was to investigate the effects of dynamic segmentation of interval nutrient supply phase feeding on the growth performance, carcass characteristics, immune organs indexes and intestinal morphology of broilers. A total of 320 one-day-old broilers were randomly assigned into 4 feeding treatments, which included 4 interval nutrient supply phases as follows. Treatment A: a nutrient supply standard was used for every 14 d. Treatment B: a nutrient supply standard was used for every 7 d. Treatment C: a nutrient supply standard was used for every 3.5 d except that one nutrient supply standard was used for d 1 to 7. Treatment D: a nutrient supply standard was used for every 3.5 d including d 1 to 7. Each treatment was represented by 8 replicates with 10 broilers per replicate. The trial lasted for 42 days. Throughout the 42 d trial period, treatment A showed significantly higher average daily gain than treatments B and C(P < 0.05). The feed:gain ratio of treatment A was significantly lower than those of treatments C and D(P < 0.05). On d 28 and 42, body weight of broilers in treatment A was much higher than those of treatments B and C(P < 0.05). The slaughter rate of treatment A was significantly higher than that of treatment B(P < 0.05). Eviscerated percentage of treatment A was significantly higher than those of treatment B and D(P < 0.05). There were no significant differences among 4 treatments in immune organs indexes(P > 0.05). The crypt depth of duodenum was significantly greater in treatments A and B than in treatment C on day 42(P < 0.05).Meanwhile, the ratio of villus height to crypt depth(V:C ratio) of treatment D was significantly higher than that of treatment A(P < 0.05). No significant differences were found between treatments C and D in growth performance, carcass performance, immune organs indexes and intestinal structure(P> 0.05). In conclusion, this study revealed that the growth and carcass performance of broilers is the best for 14 days segmentation phase feeding, and 3.5 days segmentation interval phase feeding can promote small intestinal development of broilers.
